Effect of Pine Needle Extract (PNE) on Physiological Activity of SD Rats II. Feeding Effect of PNE on Oxygen Radicals and Their Scavenger Enztmes in Brain Membranes of SD Rats (흰쥐의 생리활성에 미치는 송엽 추출물(PNE)의 영향 II. 뇌세포막의 산소라디칼 및 제거효소의 활성에 미치는 PNE의 투여효과)

  • Pine(Pinus densiflora Sieb et Zucc.) is one of rhe popular plant drugs which has been used as a medicine in Asia. To investigate the effect of pine needle extract (PNE) on oxygen radicals and their scavenger enzymes in brain membranes of Sprague- Dawley (SD), make SD rats were fed basic diets(control group), and experimental diets (PNE group) with 0.5 and 1.0% of PNE 6 weeks. Mitochondrial hydroxyl radical levels in brain of 0.5%-PNE and 1.0%-PNE groups were significantly inhibited to 30% and 25%, respectively, and microsomal hydrogen peroxide levels in brain of 0.5%-PNE and 1.0%-PNE groups were significantly inhibited to 15% compared with control group. Cytosolic superoxide rdical levels in 1.0%-PNE group were significantly inhibited to 20% compared with control group. Lipid peroxide(LPO) levels in brain mitochondria of 0.5%-PNE and 1.0%-PNE groups were significantly lower(25% and 35%) than that in control group. Mn-superoxide disumtase (SOD) activities in brain of 0.5%-PNE and 1.0%-PNE groups were significantly higher(18% and 12%) than those in control groups, but Cu,Zn-SOD activities in brain of 0.5%-PNE were significantly activated to 15% compared with control group. Glutathione peroxidase(GSHPx) activities in brain of 1.5%-PNE and 1.0% PNE groups were significantly higher(14% and 12%) than those in control group. These results suggest that more beneficial effects such as inhibition of oxygen radicals and lipid peroxide(LPO). and oncreases of scavenger enzymes in brain membranes of SD rats may be effectively modulated by administration of pine needle extract (PNE)

  • This study was edsigne to investgate the effect of pine needle extract (PNE) on lipid adn oxygen radical metabolisms in serum of Sprague-Dawly (SD). Pine(Pinus densiflora Sieb et Zucc.) is one of the popular plant drugs which gas been used as a medicine in Asia. Male SD rats were fed basic (control group) and experimental diets(RGE group) with 0.5 and 1.0% of PNE for 6 weeks. Body weights gain in 0.5-PNE and 1.0-PNE groups were slightly lower than that in control group, but ther is no significant differences brtween these groups. Total energy intake, feed and gross efficiencies showed almost no change in these groups. Total cholesterol and triglyceride (TG) levels in serumof SD rats in .50-PNE and 0.1-PNE froups sho9wed almost no change colpared with control group. Serum LDL-cholesterol levels significantly decreased (12%) in 0.5-PNE group, while serum HDL-cholesterol levels significantly increased(14%) in 1.0-PNE group compared with control group. Malondialdehyde (MDA) and lipid peroxide (LPO) levels in 50-PNE and 0.1-PNE groups were significantly lower (17 and 13%, 24% and 12%, respectively) than those in control group. Superoxide disumutase (SOD) activity in 50-PNE and 0.1-PNE groups significantly increased about 30 to 15% compared with control group. There results suggest that lower LDL-cholesterol and LPO levels, higher HDL-cholsterol level, and higher SOD activity in serum of rats may be effectively modulated by administration of pine needle extract(PNE).

  • This study was designed to investigate the effects of pine (Pinus densiflora Sieb et Zucc) needle extract (PNE) on membrane fluidity and oxidative stress in liver membranes of Sprague-Dawley (SD) rats as a study on investigation of anti-aging effect and determination of chemical structures of PNE through the animal experiments. Male SD rats were fed basic diets (control group) and experimental diets (0.5% and 1.0%-PNE group) for 6 weeks. Administrations of 0.5% and 1.0%-PNE resulted in a marked decreases (15∼25% and 23∼26%, respectively) in cholesterol accumulations of liver mitochondria and microsomes compared with control group. Membrane fluidities were significantly increased (15∼25%) in liver microsomes of 0.5% and 1.0%-PNE groups compared with control group. Formations of basal and induced oxygen radicals (BOR and IOR) in liver mitochondria were significantly inhibited (11∼12% and 10∼15%, respectively) by administrations of 0.5% and 1.0%-PNE compared with control group. Lipid peroxide (LPO) levels were remarkbly decreased about 20% in liver mitochondria and microsomes of 0.5% and 1.0%-PNE groups compared with control group. Oxidized protein levels calculated with carbonyl group were significantly decreased about 15% in liver mitochondria of 1.0%-PNE group compared with control group. These results suggest that PNE may play a effective role in a attenuating a oxidative stress and increasing a membrane fluidity.

  • These studies were designed to investigate the effects of pine (Pinus densiflora Sieb et Zucc.) needle extract (PNE) on oxygen radicals and their scavenger enzymes in liver membranes of Sprague-Dawley (SD) rats as a study on investigation of anti-aging effect and determination of chemical structures of PNE through the animal experiments. Male SD rats were fed basic diets (control group) and experimental diets (0.5% and 1.0%-PNE group) for 6 weeks. There were no significant differences in hydroxyl radical (·OH) formations of liver mitochondria and microsomes in 0.5%-PNE group, while ·OH formations were significantly decreased (10% and 18%, respectively) in liver mitochondria and microsomes of 1.0%-PNE group compared with control group. Microsomal hydrogen peroxides and cytosolic superoxide radicals were remarkably decreased (20% and 20∼25%, respectively) in 0.5% and 1.0%-PNE groups compared with control group. Mn-SOD activities in mitochondria were significantly increased about 10% in 1.0%-PNE group, while Mn-SOD activities in mocrosomes were remarkably increased (16∼20%) in 0.5% and 1.0%-PNE groups compared with control group. There were no significant differences in Cu, Zn-SOD activities of liver cytosol in 0.5% and 1.0%-PNE groups, while glutathione peroxidase (GSHPx) and catalase (CAT) activities were significantly decreased (28∼30% and 15∼30%, respectively) in liver cytosols of 0.5% and 1.0%-PNE groups compared with control group. These results suggest that these PNE may play a effective role in a attenuating a oxygen radical formations and increasing a scavenger enzyme activities.

Effects of Pine Needle Ethyl Acetate Fraction on Membrane Fluidity and Oxidative Stress in Liver Membranes of Rats (간장 세포막의 유동성과 산화적 스트레스에 미치는 솔잎(Pine Needle) 에틸아세테이트획분의 영향)

  • This study was designed to investigate the effects of ethyl acetate (EtOAc) fraction of pine (Pinus densiflora Sieb et Zucc) needle extract on membrane fluidity (MF), basal and induced oxygen radical (BOR and IOR), lipid peroxide(LPO) and oxidized protein (OP) as an oxidative stress, and lipofuscin(LF) in liver membranes of male Sprague-Dawley rats. Rats were fed basic diets (control) and experimental diets (EtOAc-25, EtOAc-50 and EtOAc-100) for 45days. MFs were significantly increased (about 10%) in mitochondria of EtOAc-100 group compared with control group. BOR and IOR formations in mitochondria were significantly inhibited (about 12∼18% and 9 ∼l2%, respectively) in EtOAc-50 and EtOAc-100 groups, while BOR and IOR formations in microsomes were significantly inhibited (about 9∼l3% and 18∼19%, respectively) compared with control group. LPO levels were significantly inhibited (about 10% and 12∼13%, respectively) in mitochondria of EtOAc-100 and microsomes of EtOAc-50 and EtOAc-100 groups, whereas OP levels were significantly inhibited (about 13∼14%) in mitochondria of EtOAc-50 and EtOAc.-100 groups compared with control group. LF formations were significantly inhibited (about 10∼14%) in these three EtOAc groups. These results suggest that ethyl acetate fraction of pine needle may play an effective role in attenuating an oxidative stress and increasing a membrane fluidity.

These results suggest that administrations of ethyl acetate fraction of pine needle may play an effective role in an attenuating an oxidative stress and in increasing membrane fluidity.

Nail Withdrawal Behavior for Domestic Small Diameter Logs

  • Nail withdrawal tests were conducted on clear wood of domestic small diameter logs. Nails were driven into the cross and longitudinal sections of each specimen, then nail withdrawal tests were performed. Nail withdrawal loads are strongly dependent on the direction of nail positions. The average load values for the nail withdrawal both in cross section and longitudinal section are higher in high specific gravity (SG) wood of sawtooth oak (Quercus acutissima Carr.) than those in low SG wood of Korean red pine (Pinus densiflora Sieb. et Zucc.) and pitch pine (Pinus rigida Mill.). The average ratio of the nail withdrawal loads for side-grain and end-grain are higher in the low SG wood than that in the high SG of wood. Both linear and non-linear regression analyses were conducted on nail withdrawal load with SG, good correlations were obtained between nail withdrawal load and SG.

  • To understand the effect of supplement of water extract of pine needle(WEPN) on shelf-life enhancement of the kimchi, activities of four enzymes and number of lactic acid bacteria, during fermentation of the kimchi, were assayed. Enzyme activities of kimchi fermented for 7 days with supplement by 2% water extract of pine needle showed amylase of 86.4%, protease of 85.8%, polygalacturonase of 61.5% and $\beta$-galactosidase of 58.8% against the control kimchi. WEPN showed weak inhibitory effect when it was applied to the isolated enzymes in vitro then those menifested by the kimchi in vivo. Number of total bacterial cell of WEPN supplemented kimchi increased by 10 folds than control between 7 to 14 days of fermentation. On contrast, number of lactic acid bacteria decreased maximaly to 21% of control by fermentation. The clear zone formed on paper disk by WEPN against L. plantarum was larger than that of Leu. mesenteroides.

  • In order to elucidate the metabolism of the systemic insecticide, phosphamidon(2-chloro-2-diethylcarbamoyl-1-methylvinyl dimethyl phosphate), treated to pine trees against pine leaf gall midges (Thecodiplosis japonensis Uchida et Inouye), $[vinyl,\;carbonyl-^{14}C]$phosphamidon was implanted into the trunks of 10-year-old Korean red pine (Pinus densiflora Sieb. et Zucc.) and Japanese black pine (Pinus thunbergii Parl.), respectively. This chemical was degraded very quickly in pine trees after implanting, as confirmed by TLC/autoradiography of the extracts of pine needles. Phosphamidon metabolites in phosphate buffer extracts of pine needles include the major metabolite, ${\alpha}-chloroacetoacetic$ acid diethyl-amide, ${\alpha}-chloroacetoacetic$ acid ethylamide, 3-hydroxy-N,N-d iethylbutanamide, acetoacetamide, and trimethyl phosphate. The metabolism within pine trees is expected to be similar to this. Based on these findings, it is believed that the major pathway leading to the metabolites would be related to the P-O-vinyl hydrolysis of the chemical structure.

  • This study was carried out to investigate the effects of soil fertilizers on wax content, contact angle, mineral nutrient content of Japanese red pine (Pinus densiflora Sieb, et Zucc.) leaves and soil acidity of Japanese red pine communities in Namsan and Kwangnung to see whether they can recover forest decline. Japenese red pine communities were treated with $Ca(OH)_2$, $Mg(OH)_2$, $Ca(OH)_2+Mg(OH)_2$+C.F.(compound fertilizer) in a randomized complete block design with 3 replication from November, 1990 through October, 1993. Wax content, contact angle value and mineral nutrient content of Japanese red pine leaves and soil pH of communities were measured and the results obtained are as follows: 1. Contact angle value and wax content of Japenese red pine leaves increased when the fertilizers were applied in soil. The order leaves grew, the smaller their contact angle values. 2. K and Ca contents of Japanese red pine leaves were higher in Namsan than in Kwangnung, whereas Mg content was higher in Kwangnung. K and Mg contents of the leaves increased with fertilization both in Namsan and Kwangnung. 3. Soil acidity of pH 4.2 ~ 4.3 was shown in Namsan and pH 4.6 ~ 4.9 in Kwangnung. No acidity changes were shown when the fertilizers were applied in soil. However with the lapse of the soil fertilizer application time, there was the indication that soil pH became higher in the fertilizer treatments than in the control.
