Elevated level of serum cholesterol in humans is a risk factor correlated with the development of coronary heart disease. We have screened lactic acid bacteria from pig and human feces for the development of probiotics which have an anti-cholesterol effect. We have used special media to isolate only lactic acid bacteria and they were subjected to the experiments such as oxgal test, carbohydrate fermentation test. Results from the acid tolerance test and growth test in the presence of oxgal demonstrated that some strains would likely survive in thuman stomach, where acidity is high, and in small intestine, where bile fluid is present. In conclusion, we were able to screen lactic acid bacteria which were tolerant against bile acid and supposed to be prominent to lower the cholesterol level in human serum.

Maximum residue limits (MRL) for pesticides in feed have been set to protect public health and produce safe livestock products. In vivo experiments to establish MRL are essential, as livestock are commonly used to obtain reliable in vivo quantitative information. Here, we aimed to evaluate whether small laboratory animals can replace or reduce monogastric livestock in experiments to quantify pesticide residues in vivo after oral consumption through feed. First, 24 pigs and rats were randomly assigned to four groups and fed 0, 3, 9, or 30 mg/kg of sulfoxaflor. After four weeks, serum, muscle, fat, liver, kidney, and small intestine samples were collected, and sulfoxaflor residues were analyzed using liquid chromatography - tandem mass spectrometry. Sulfoxaflor residues in pig tissues were significantly correlated with those in rat tissues. Model equations were formulated based on the residual sulfoxaflor amount in pig and rat tissues. The calculated and measured sulfoxaflor residues in pigs and rats showed more than 90% similarity. Sulfoxaflor did not affect body weight gain, feed intake, or the feed conversion ratio. Therefore, we concluded that pesticide residue quantification in vivo to establish MRL could be performed using small laboratory animals instead of livestock animals. This would contribute to obtaining in vivo pesticide residue information and reducing large-scale livestock animal experiments.
This study was based on 3 necropsies of patients affected with Balantidium coli dysentery from two pig farms. Grossly, the lesions involved the large intestine from cecum to rectum. Many variable-sized ulcers were diffusely scattered on the mucosa of the cecum and the colon which were covered with a necrotic, grayish white, slate black, or black membrane. The mucosa affected reddened and swollen. Microscopically, numerous balantidia penetrated In the mucosa of the colon and there they induced necrosis and desquamation of the epithelial cells, where inflammatory cells such as macrophages, neutrophils, eosinophils, lymphocytes and plasma cells were infiltrated.
Ethylacetate fraction from Orobanche crenata, contained two phenylpropanoid glycosides, exhibited some pharmacological properties. It was found to be non-toxic to rats in oral doses up to 500mg/100gm body weight. In large doses, it lowered the arterial blood pressure of anaethetised rats, and produced significant analgesic effect in mice and diuretic effect in rats. It further showed smooth muscle relaxant and antispasmodic effects in the isolated rabbit intestine and guinea-pig ileum respectively.
Pleiomorphic adenoma gene-like1 (PLAGL1) encodes a zinc-finger (ZF) protein with seven ZFs of the C2H2-type which is a regulator of apoptosis and cell cycle arrest, and also regulates the secretion of insulin. In both human and mouse, PLAGL1 is a candidate gene for tumor suppressor and transient neonatal diabetes mellitus (TNDM). In this study, a 2,238 bp fragment covering the complete coding region was obtained and deposited to GenBank (accession number: DQ288899). The reverse transcriptase-polymerase chain reaction (RT-PCR) indicated that PLAGL1 was expressed almost equally in heart, liver, spleen, lung, kidney, stomach, small intestine, skeletal muscle, fat, uterus and ovary. Comparing the sequences of Large White and Meishan pigs, a C-T transition in exon 6 was found. The polymorphism could be detected by TaqI and was genotyped in five purebreds (Large White, Landrace, Meishan, Tongcheng and Bamei). Association analysis was performed between the polymorphism and carcass traits in 276 pigs of a "Large White${\times}$Meishan" F2 resource population. As a consequence, significant associations of the genotypes with shoulder backfat thickness (SFT) and internal fat rate (IFR) were observed. Pigs with TT genotype had low SFT and high IFR compared with TC or CC genotypes.
본 실험은 음식물류 폐기물을 돼지발효사료로 만들기 위해 공기주입과 보조용양원으로 돼지혈분의 이용에 대해 알아보았다. 발효용 종모균주로는 돼지의 내장에서 분리한 유산균인 Lactobacillus brevis와 딸기에서 분리한 효모인 Saccharomyces cerevisiae가 이용되었다. 배지원료로는 당밀과 유청이 효모엑스와 함께 사용되었다. 실험결과 공기주입은 생균수를 높여주었고 사멸기를 저지하는 효과를 보였다. 보조 영양원으로 돈혈분이 효모엑스를 대치할 경우에도 생균수의 변화는 거의 없었다. 종모발효액은 상온에서 저장할 경우 1주일간 Lactobacillus brevis와 Saccharomyces cerevisiae생균수가 모두 8 log cfu/mL대를 유지했다. 종모발효액이 음식물류 폐기물과 혼합될 경우 이틀 뒤부터는 잡균이 증가되기 시작했으나 5일 이내에는 E.coli가 검출되지 않았다.
The growth rate of the young pig is generally much less than it's potential and may be constrained by endocrine status as well as nutrient intake. Growth factors are present in relatively high quantities in colostrum and play an important part in gut development. It is possible that supplementation of colostrum protein isolate may stimulate gut and whole body growth in the pig. Eight male and 8 female (Large Whitex${\times}$Landrace) piglets were weaned at 1 d of age after each pig had obtained colostrum from their dam, and were trained to consume one of two liquid diets. The two diets were based on either a colostrum protein isolate (n = 4 males and 4 females) or whey protein concentrate (n = 4 males and 4 females) and were formulated to contain equal levels of crude protein and amino acids. Pigs were fed their diets ad libitum for 28 days after which time 12 pigs were euthanised and various tissues and organs weighed. Pigs were bled for IGF-I analyses at 21 and 28 days of age. Daily gain was higher in pigs consuming the colostrum isolate (171 vs. 216 g/d, p = 0.010), particularly between 2 and 4 weeks of age (212 vs. 298 g/d, p = 0.010). Pigs tended to consume more of the liquid feed containing colostrum isolate (25.5 vs. 29.1 kg, p = 0.074) and gained more live weight per unit of liquid feed (0.203 vs. 0.223 g/g, p = 0.056). There were no effects of sex on growth performance. Pigs consuming the diet supplemented with colostrum isolate had higher (p<0.05) full gut weight (445 vs. 554 g, p = 0.026), empty gut weight (356 vs. 463 g, p = 0.008), stomach weight (42.2 vs. 54.4 g, p = 0.001), small intestine weight (222 vs. 275 g, p = 0.025) and large intestine weight (63.7 vs. 98.0 g, p = 0.005). Plasma IGF-I (99 vs. 150 ng/ml, p<0.001) and IGF-II (265 vs. 406 ng/ml, p<0.001) were higher in pigs fed colostrum isolate. Pigs consuming colostrum protein isolate ate more, grew faster and had higher plasma IGF-I concentrations than pigs consuming a diet with similar macronutrient content but devoid of growth factors.
Glucose is the main energy source for mammalian cells and its absorption is co-mediated by two different families of glucose transporters, sodium/glucose co-transporters (SGLTs) and facilitative glucose transporters (GLUTs). Here, we report the cloning and tissue distribution of porcine GLUT2. The GLUT2 was cloned by RACE and its cDNA was 2,051 bp long (GenBank accession no. EF140874). An AAATAA consensus sequence at nucleotide positions 1936-1941 was located upstream of the poly $(A)^+$ tail. Open reading frame analysis suggested that porcine GLUT2 contained 524 amino acids, with molecular weight of 57 kDa. The amino acid sequence of porcine GLUT2 was 87% and 79.4% identical with human and mouse GLUT2, respectively. GLUT2 mRNA was detected at highest level in porcine liver, at moderate levels in the small intestine and kidney, and at low levels in the brain, lung, muscle and heart. In the small intestine, the highest level was in the jejunum. In conclusion, the mRNA expression of GLUT2 was not only differentially regulated by age, but also differentially distributed along the small intestine of piglets, which may be related to availability of different intestinal luminal substrate concentrations resulting from different food sources and digestibility.
The aim of this study is to investigate how the gut microbiome shifts when pigs were exposed with low concentrations of mycotoxins, deoxynivalenol (DON) and zearalenone (ZEN) in feed. Fifteen of pigs, 15 kg in weight which were negative for PRRSV and PCV2 were purchased, acclimatized until 20 kg in weight, and randomly divided into 3 groups; the DON group (DON treated), the ZEN group (ZEN treated) and the CTL (untreated negative control). DON and ZEN administered to each group for 30 days at 0.8 mg/kg (800 ppb) and 0.20 mg/kg (200 ppb) in feed, respectively. After extraction of microbial DNA from intestine and fecal samples, sequencing procedures were performed in the Ion PGM using an Ion 316 V2 chip and Ion PGM sequencing 400 kit. The results suggested that the bacterial communities in duodenum, jejunum and ileum of the DON and ZEN groups presented low-abundant OTUs compared with the CTL group. OTUs in cecum, colon and feces were determined more than in small intestine of all three groups. However, the CTL group yielded more OTUs than other two groups in inter-group comparison. It is not fully clarified how the richness and abundance in microbiome functions in the health condition of animals, however, the exposure to DON and ZEN has caused microbial population shifts representing microbial succession and changes following the diversity and abundance of porcine gut microbiome. The metabolomic analysis correlate with microbiome analysis is needed for further study.
돼지의 장 및 분변에서 분리한 14종의 유산균으로부터 항세균 활성물질을 분석하였다. 각 균주의 세포배양액을 대상으로 여러 종류의 단백질 분해효소와 열, 알칼리 처리 등을 수행한 후 활성도를 조사한 결과, 효소 및 열처리 시료에서는 활성도가 유지되었으나, 1M의 NaOH로 중화 처리한 시료에서는 활성도가 사라짐에 따라, 항세균 활성물질이 단백질성이라기보다 유기산일 가능성이 큰 것으로 나타났다. Sep-pak $C_{18}$ Cartridges을 이용한 음이온 추출실험과 Bio-LC system을 이용한 high performance anion exchange chromatography를 실시하여 물질을 정제한 결과, 주요 활성물질로 옥살산, 구연산, 젖산 및 초산 등, 4종류의 유기산이 검출되었다. 이중 젖산은 각 균주에 따라 전체 유기산의 54~77%를 차지하는 것으로 나타나 유기산 중에서도 일차적인 항세균 활성물질일 것으로 추정된다.
