Little Leaf and Yellowing Symptoms on Castanea crenata are Associated with Phytoplasma in Korea

  • For unknown reasons, a few trees in a private chestnut orchard in Icheon si, Gyunggi-do suffered leaf chlorosis and growth decline. Based on symptoms, phytoplasma was a probable cause. Leaf samples were collected from two symptomatic and non-symptomatic trees in the orchard for phytoplasma detection. An amplicon of about 1.2 bp size was obtained from both symptomatic trees by PCR with the universal 16S rDNA primers. Sequences of these amplicons were found to have 99% nucleotide sequence identity to the corresponding genomic region of 16SrIII (X-disease group). More than 100 phytoplasma isolates, such as Candidatus phytoplasma pruni, Milkweed yellows phytoplasma, Goldenrod yellows phytoplasma, Tsuwabuki witches'-broom phytoplasma, Candidatus Phytoplasma trifolii, etc. were involved in the list. Phylogenetic analysis revealed that the sequence obtained in this study closely clustered with Candidatus phytoplasma groups. While one of the amplicons shared 91% identity with the Candidatus phytoplasma castaneae, the other shared only 47%. It needs further analysis and investigation to determine the exact taxonomy. Meanwhile, based on the analysis of the sequences, chlorosis, and small leaves were associated with phytoplasma.

Specific Primer for Detection of Jujube Witches' Broom Phytoplasma Group (16SrV) in Korea

  • In order to diagnose and differentiate jujube witches' broom (JWB) phytoplasma rapidly, oligonucleotide primer pair, 16Sr(V) F/R, for polymerase chain reactions (PCRs) was designed on the basis of 16S rRNA sequences of JWB phytoplasma. The PCR employing phytoplasma universal primer pair P1/P7 consistently amplified DNA in all tested phytoplasma isolates. But no phytoplasma DNA was detected from healthy jujube seedlings. The nested PCR, the primer pair 16S(V) F/R, about 460 bp fragment, amplified DNA in all tested JWB and related phytoplasmas including ligustrum witches' broom phytoplasma of the 16S rRNA group V, but no DNA amplification was detected from other phytoplasma strains such as groups 16SrI (Aster yellows) and 16SrXII (Stolbur group) in which mulberry dwarf phytoplasma and chrysanthemum witches' broom phytoplasma belong to, respectively. The same results were obtained from both Korean and Chinese isolates of JWB phytoplasma. Nested-PCR using phytoplasma universal primer pair P1/P7 and 16SrV group-specific primer pair 16S(V) F/R could detect group V phytoplasmas rapidly and easily, in particular JWB phytoplasma.

Nested-PCR using phytoplasma universal primer pair P1/P7 and 16S rRNA group V specific primer pair 16S(V) F/R could detect group V phytoplasma rapidly and easily, in particular JWB phytoplasma.

Association of Aster Yellow Phytoplasma with Witches′ Broom Disease of Ash(Fraxinus rhynchophylla Hence) in Korea

  • Typical whiches broom symptoms caused by phytoplasma were observed in Ash (Fraxinus rhynchophylla Hence) in Korea. The symptoms were showing abnormally small leaves, short internodes, and proliferation of shoots. Fluorescence and electron microscopy of leaf midribs revealed phytoplasma positive DAPI fluorescence and numerous phytoplasma bodies localized in the phloem sieve tubes. Phytoplasma DNA of 1.8 Kb was detected consistently from all symptomatic samples by the amplification of phytoplasma DNA with the phytoplasma specific primer pair Pl/P7. But no phytoplasma DNA was detected in healthy ash seedlings. Based on sequence analyses of an amplified region, this phytoplasma is closely related to Eqilodium phyllody, Mulberry dwarf, and Aster yellows phytoplasmas with the homology of 99.95 %, 99.79 % and 99.78 %, respectively, This phylogenetic analyses indicate that ash witches broom phytoplasma but is evidently distinct from the ash yellows group 16SrⅦ and should be classified into the Aster yellows group 16SrⅥ.

Candidatus Phytoplasma trifolii Associated with Witches' broom of Lespedeza cyrtobotrya M.

  • The Symptoms of witches' broom disease caused by phytoplasma including general stunting and yellowing, were observed in leafy lespedeza (Lespedeza cyrtobotrya M.) on Doam-myeon, Pyeongchang-gun, in 2006. Based on the sequence analysis of PCR-amplified 16S ribosomal DNA and 16S-23S spacer region DNA products using universal phytoplasma primers, the phytoplasma associated with leafy lespedeza witches' broom (LLWB) disease was identified as a member of Candidatus Pytoplasma trifolii. It was most closely related to alsike clover proliferation phytoplasma (99.8% similarity, accession no. AY390261), Candidatus Pytoplasma trifolii strain. RFLP patterns generated with AluI, HpaII clearly differentiated LLWB phytoplasma from the referenced phytoplasma strains, water dropwort witches' broom, mulberry dwarf, glehni aster yellow dwarf and jujube witches' broom. This paper is the first report on Candidatus Phytoplasma trifolii in leafy lespedeza identified at a molecular level.

Occurrence of Petunia Flattened Stem Caused by Phytoplasma

  • This study describes a phytoplasmal disease occurring in Petunia leaves grown in the glasshouse of the National Horticultural Research Institute, Suwon, Korea. Abnormal growth like flattened stem with flower malformation or phyllody was observed from the plant. The DNA extracted from the diseased leaves was amplified using a universal primer pair of P1/P6 derived from the conserved 16S rRNA gene of Mollicutes giving the expected polymerase chain reaction(PCR) product of 1.5 kb. In the nested PCR assays, the expected DNA fragment of 1.1 kb was amplified with the specific primer pair R16F1/R16R1 that was designed on the basis of aster yellows(AY) phytoplasma 16S rDNA sequences. The 1.1 kb PCR products were cloned and nucleotide sequences were determined, and the sequences of the cloned 168 rRNA gene were deposited in the GenBank database under the accession no. of EU267779. Analysis of the homology percent of the 168 rDNA of PFS-K showed the closest relationship with Hydrangea phyllody phytoplasma(AY265215), Brassica napus phytoplasma(EU123466) and AY phytoplasma CHRY(AY180956). Phytoplasma isolated from the diseased Petunia was designated as Petunia flat stem phytoplasma Korean isolate(PFS-K) in this study. Flattened stem occurring in Petunia was confirmed as infection of AY group of phytoplasma by determination of 16S rRNA gene sequences of phytoplasma and microscopic observation of phytoplasma bodies. This is the first report on the phytoplasmal disease in Petunia in Korea.

Detection and Molecular Characterization of a Stolbur Phytoplasma in Lilium Oriental Hybrids

  • Stolbur Phytoplasma was detected from Lilium Oriental hybrids showing flattened stem and flower clustering. The presence of phytoplasma was demonstrated using polymerase chain reaction(PCR) assays with phyto-plasma-universal(P1/P6)and stolbur phytoplasma-specific 16F1/R1-S primer pairs amplifying phytoplasma 16S rDNA regions. Nucleotide suquences of the phytoplasma 16S rDNA were determined. Nucleic acid extracted from lily amplified 1.5 kb DNA with a phytoplasma universal primer pair. In nested PCR, 1.1 kb PCR product was obtained using specific primer pair, indicating an isolate of stolbur phytoplasma. Nucleotide sequence of phytoplasma 16S rDNA reported in this study showed 99.5% and 99.1% identities with two known stolbur phytoplamas (16Sr XII-A). Also, it exhibited a sequence homology of 98.0% with phormium yellow leaf (16Sr XII-B), and 97.9% with Australian grapevine yellows (16Sr XII-B). Meanwhile, it showed 98.1% identity with strawberry green petal phytoplama, (16Sr1-C), and 94.7 % with American aster yellows (16Sr1-B). Homology percentage of the 16S rDNA nucleotide sequence suggests that this phytoplama could be classified into the stolbur phytoplasma, subgroup A (16Sr XII-A), as a type strain stolbur.

"Candidatus phytoplasma asteris" Group에 속하는 아까시나무 빗자루병 검출 (Detection of "Candidatus Phytoplasma Asteris" Associated with Black Locust Witches' Broom in Korea)

  • 전형적인 파이토플라스마성 빗자루증상을 보이는 아까시나무가 전북지방에서 발견되었으며, 일반적으로 작은 잎들이 총생하였고, 마디와 마디 사이가 좁고, 가지가 위축되어 있는 증상을 보였다. 파이토플라스마 검출 primer인 P1/P7(약 1.8 kb)과 R16F2n/R2(약 1.2 kb)를 이용하여 증폭한 결과 빗지루증상을 보인 아까시나무에서 파이토플라스마 검출되었으며, 건전한 아카시에서는 PCR산물이 증폭되지 않았다. 염기서열 분석에 의한 유의성 검정결과 아까시나무 빗자루병 파이토플라스마는 aster yellow, 뽕나무오갈병, maize bushy stunt, 물푸레나무 빗자루병, 붉나무 빗자루병 파이토플라스마와 99.2% 이상의 유의성을 보였으며, 아까시나무 빗자루병 (GeneBank Accession No. AF 244363) 및 대추나무 빗자루병 파이토플라스마와는 각 각 88.6% 및 87.7%의 유의성을 보여 본 연구에서 분석한 아까시나무 빗자루병 파이토플라스마는 Candidatus phytoplasma asteris(16Sr I) group에 속하였으며, 16Sr III(peach X-disease phytoplasma group) group에 속하는 아카시나무 빗자루병 파이토플라스마와는 다른 계통으로 분류되었다.

Two Groups of Phytoplasma from Chrysanthemum (Dendranthema grandiflorum) Distinguished by Symptoms and 16S rRNA Gene Sequence in Korea

  • Two groups of phytoplasma were identified in chrysanthemum(Dendranthema grandiflorum) cv. Chunkwang showing distinct symptoms. Isolate Ph-ch1 showed symptoms of dwarf, witches'-broom, rosette and root death. The other isolate, Ph-ch2, revealed symptoms of dwarf, yellowing, leaf cupping, vein clearing and root death. The presence of phytoplasma structures in chrysanthemum leaf tissue was confirmed by transmission electron microscopy. The 16S rRNA gene was amplified from isolates Ph-ch1 and Ph-ch2 by PCR and cloned, and the nucleotide sequences were determined. In RFLP analysis, isolate Ph-ch2 showed profiles identical to Ph-ch1, except with restriction enzymes HhaI and MseI. The sequence data showed that isolate Ph-ch1 was most closely related to the aster yellows (AY) phytoplasma, and isolate Ph-ch2 was more closely related to stolbur phytoplasma than to AY phytoplasma. This is the first reported observation of stolbur phytoplasma in chrysanthemum species.

Polymerase Chain Reaction(PCR)을 이용한 오동나무, 라일락, 미역취의 Phytoplasma 검출 및 유연 관계 (Detection of Phytoplasmas from Paulownia tomentosa, Syringa vulgaris and solidago vir-aurea var. gigantea Using Polymerase Chain Reaction (PCR) and Their Relationships)

  • 위축, 황화, 총생 증상 등 전형적인 병징을 나타내는 phytoplasma에 감염된 식물에서 phytoplasma만을 특이적으로 검출하기 위하여 polymerase chain reaction(PCR) 방법을 이용하였다. Phytoplasma의 16S rRNA gence의 DNA 단편을 증폭하기 위하여 1.4 kb primers (forward, 5` -GTTGATCCTGGCTCAGGATT-3` 와 reverse, 5` -AACCCCGAGAACGTATTCACC -3`)를 사용하여 증폭한 결과, phytoplasma에 이병된 오동나무, 라일락 및 미역취에서는 약 1.4 kbp의 위치에서 특이 band가 검출되었으나 control로 사용한 건전주에서는 어떠한 band 검출되지 않았다. 위의 결과를 재확인 하기 위하여 약 0.5 kb의 primers(forward, 5` -ACGAAAGCGTGGGGAGCAAA-3` 와 reverse, 5` -GAAGTCGAGTTGCAGACTTC-3`)를 사용하여 증폭한 결과, 0.5 kb의 위치에서 특이 band가 검출되었으나 control로 사용한 건전주에서는 어떠한 band도 검출되지 않았다. Phytoplasma에 이병된 식물의 PCR 반응산물을 제한효소인 AluI으로 처리하 sruf과, 오동나무와 라일락에서는 동일한 band pattern을 나타내어 서로 유연관계가 가까운 phytoplasma인 것으로 생각되며, 미역취에서는 이들과는 다른 band pattern을 나타내어 오동나무와 라일락의 phytoplasma와는 유연관계가 먼 것으로 추측된다.
