가미마전오자환 추출물이 웅성 백서의 성기능에 미치는 효과 (Effects of Gamimajeonojahwan Extract on the Sexual Function in Male Rats)

  • To evaluate effect of Gamimajeonojahwan(GMOH) water extract on sexual functions, we orally administered GMOH exract(1,100㎎/㎏) for four weeks in male rats impaired by 2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD) and ketoconazole(KZ). And we investigated the sexual behaviors such as mount latency, mount frequency, intromission latency, intromission frequency, ejaculation period and ejaculation latency. Serum testosterone concentrations also are evaluated in GMOH-and/or vehicle-treated male rats. The results were obtained as follows: GMOH inhibited the increase of intromission latency by TCDD in male rats. GMOH inhibited the decrease of intromission frequency by TCDD in male rats. GMOH inhibited the increase of ejaculation latency by TCDD in male rats. GMOH inhibited the decrease of serum testosterone concentration by TCDD in male rats. GMOH inhibited the decrease of serum testosterone concentration by KZ in male rats. These results suggest that GMOH water extract is active in improving sexual functions and protecting a decrease of testosterone concentrations induced by TCDD and KZ in male rats.

서울지역 청소년 및 성인의 타우린 섭취량, 혈중 농도 및 소변내 배설량에 관한 연구 (Dietary Intakes, Plasma Levels and Urinary Excretions of Taurine in Adolescents and Adults Residing in Seoul Area)

  • As diverse physiological functions of taurine have been reported, taurine-containing health drinks and products are marketed worldwide for the treatment of various conditions such as improvements of liver, heart and circulatory functions or as an aid to athletic performance. Although animal studies have shown that taurine is fairly safe when supplemented in the diet for an extended period, the effective dose range of taurine for dietary supplements is in controversy. Reports on dietary taurine intakes have been sparse, and would serve as a guideline for determining an appropriate taurine dosage. The present study was aimed to estimate dietary intake level of taurine using the taurine content database of commonly used food stuffs established recently in our laboratory, and also to evaluate plasma concentration and urinary excretion of taurine in adolescents and adults residing in Seoul area. Dietary taurine intakes of the subjects were 219$\pm$16.9mg/day for 16-19 years old(n=123), 177$\pm$18.1mg/day for adults older than 20 years old(n=123). Male subjects(n=115) consumed 216$\pm$21.1mg of taurine/day, while female subjects(n=131) consumed 181$\pm$14.3mg of taurine/day(p<0.05). The level of dietary taurine intake was positively correlated with the levels of dietary intakes of energy, carbohydrate, total lipids, cholesterol, vitamin A, vitamin B$_1$, niacin, vitamin C, calcium, phosphorous, sodium and potassium at p<0.01, and with dietary intakes of iron and animal lipids at p<0.05, respectively. Plasma taurine concentration of subjects were 135$\pm$5.9$\mu$mol/L, which is considered to be within a normal range for healthy subjects. The subjects excreted 1158$\pm$72.7nmol/ of tarine mg creatinine in their urine, which is approximated as 150-170mg of taurine/24hr urine based on the assumption that 18mg creatinine/kg/day is excreted in the urine of healthy adults, and this would be about 80% of the daily taurine intake observed in the same subjects. Dietary taurine intake level was positively correlated with plasma taurine concentration, as well as with urinary taurine excretion corrected by creatinine excretion at p<0.05. The present study was the first report of taurine intake, and plasma concentration and urinary excretion of taurine in a Korean population so far, and these results would serve as an index for the future study evaluating taurine status in a diverse population within and outside Korea. 운동과 면역반응에 대한 고찰 (Immunological Aspects of Contemporary Exercise)

  • 면역학이 발점함에 따라서 인간의 면역체계에 영향을 미치는 많은 요인들에 대한 수많은 연구들이 진행되고 있다. 연령, 영야상태, 스트레스, 질병, 비만, 고지혈증, 장애 등 무수히 많은 변인들이 면역방응에 직접적인 영향을 주고 있으나 운동 또한 상당한 영향력이 있어, 운동시 나타나는 면역 반응에 대하여 고찰하게 되었다. 규칙적인 운동은 초기면역과 그 이후의 적응면역을 증진시키며 특히 적응면역 중 세포 매개 면역반응과 항체매개 면역반응에 영향을 미치는 것으로 알려져 있다. 하지만 이러한 운동도 몸의 상태가 건강하거나 몸에 맞는 운동을 규칙적으로 실시해야 효과적이며, 운동의 경험이 없거나 스트레스나 질병이 있는 상태에서의 운 동은 오히려 신체에 부담을 주어 면역력을 악화하는 결과를 초래하게 된다. 또한 일회성의 운동도 체력수준에 맞게 수행되어야 하는데, 장시간 너무 무리하게 수행하면 에너지 고갈과 함께 면역력의 감소를 초래하여 상기도 감염과 같은 질환을 야기하게 된다. 따라서 무리한 운동 후 'open window' 시기에 적절한 이온음료의 섭취, 영양분 공급 및 휴식은 건강 관리에 필수적이라 할 수 있다. 또한 수행한 운동이 규칙적이라 할지라도 운동은 운동 환경이나 선천적인 요소에 따라 운동 유발성 천식을 포함하는 운동 유발성 알레르기 질환을 야기하는 환경을 조성할 수 있으므로 본론에서 제시한 지식을 바탕으로 마라톤과 같은 장기간 운동 시 특히 주의해야 할 것으로 여겨진다.

YGF251 투여에 의한 인체내 혈중의 IGF-1 분비량 변화 효과 (Effect of YGF251 on Secretion Of IGF-1 in Human Blood)

  • 인체의 다양한 기능을 증진시키는 작용을 하는 것으로 알려진 IGF-I의 생체내 분비를 촉진시키기 위해 천연 생약재성분으로 구성된 YGF251을 개발하였고 YGF251의 효능을 측정하였다. 이중 맹검 시험 방식으로 40세에서 70세 사이의 성인 남녀 31명을 대상으로 IGF-I을 비롯하여 체중, 혈압, 간기능 검사 및 신장기능 검사를 실시하였다. YGF251 및 위약 투석 후의 IGF-I 함량변화에 대한 대응표본 검정결과는 YGF251 투여군에서 투여 전에는 245.6 ng/mL 이었는데 1개월 투여 후에는 269.3 ng/mL, 2개월 투여 후에는 275.6 ng/mL으로 유의하게 증가하였다 (p<0.05). 한편 위약 투여군에서는 투여 전에는 280.0 ng/mL이었으나 1개월 투여 후에는 239.2 ng/mL, 2개월 투여 후에는 230.2 ng/mL으로 유의하게 감소하는 결과를 보였다(p<0.05). 혈중 insulin함량은 실험군의 경우 YGF251 을 1개월 투여후 평균치로써 2배 정도의 증가를 보였으며 위약 투여군에서는 36% 정도 감소된 것으로 나타났다. 또한 YGF251투여에 의한 체중 및 혈압의 변화는 거의 나타나지 않았다. YGF251 투여에 의해 나타날수 있는 간기능 및 신장기능 변화를 관찰 하기 위해 여러 항목들의 수치는 측정 오차 범위내에서의 변화만을 나타내었다.

섬유소의 이용과 butyrate의 최근 연구 (Use of Cellulose and Recent Research into Butyrate)

  • 지구상에는 약 5,400여 종의 포유동물이 있고 그 중 약 1,000여 종은 풀을 뜯어 먹고 사는 초식동물이다. 초식동물 중에서 약 250여 종이 반추동물로 알려져 있다. 반추동물인 소와 양은 반추위에서 주로 발효가 일어나지만 비반추동물인 돼지와 사람은 맹장과 결장, 직장에서 주로 발효가 일어난다. 반추위 미생물의 종류와 우점도 Bacteroidetes 51%, Firmicutes 43% 존재하며, 사람의 대장미생물의 우점도Firmicutes 65%, Bacteroidetes 25%로 존재한다. 풀의 세포벽 구성성분은 미생물에 의해 분해, 발효에 의해 SCFA (short chain fatty acid)를 생성하게 되고 acetate, propionate, butyrate 생성비율은 60:25:15이다. 장내 primary butyrate transporter인 MCT1(monocarboxylatetransports-1)에 의해서 흡수된 butyrate는 SCFA receptor GPR43과 GPR41을 활성화시킨다. Butyrate는 강력한 anti-tumorigenic 기능을 가지고 있다. Butyrate는 다양한 cancer cell에 효과를 나타내며 세포내의 기능 조절에 기여하고, 암세포사멸을 유도하는 특성이 있다. Butyrate는 caspase의 활성화, HDAC (histone deacetylase) 활성을 억제하여apoptosis를 유도하고, p53 발현증가로 cell cycle arrest와 apoptosis를 유도한다. SCFA의 항 염증작용으로는 장 상피세포에서 IL-8 발현 감소, NO합성과 NF-${\kappa}B$ (nuclear factor ${\kappa}B$)의 활성을 억제하여 염증으로 인한 암 발생을 억제한다. Butyrate는 장 점막의 생리적 기능을 유지하는데 중요한 역할을 하며 IBD (inflammatory bowel disease) 치료법으로 이용되고 있다.

Contradictory Effects of Superoxide and Hydrogen Peroxide on $K_{Ca}3.1$ in Human Endothelial Cells

  • Reactive oxygen species (ROS) are generated in various cells, including vascular smooth muscle and endothelial cells, and regulate ion channel functions. $K_{Ca}3.1$ plays an important role in endothelial functions. However, the effects of superoxide and hydrogen peroxide radicals on the expression of this ion channel in the endothelium remain unclear. In this study, we examined the effects of ROS donors on $K_{Ca}3.1$ expression and the $K^+$ current in primary cultured human umbilical vein endothelial cells (HUVECs). The hydrogen peroxide donor, tert-butyl hydroperoxide (TBHP), upregulated $K_{Ca}3.1$ expression, while the superoxide donors, xanthine/xanthine oxidase mixture (X/XO) and lysophosphatidylcholine (LPC), downregulated its expression, in a concentration-dependent manner. These ROS donor effects were prevented by antioxidants or superoxide dismustase. Phosphorylated extracellular signal-regulated kinase (pERK) was upregulated by TBHP and downregulated by X/XO. In addition, repressor element-1-silencing transcription factor (REST) was downregulated by TBHP, and upregulated by X/XO. Furthermore, $K_{Ca}3.1$ current, which was activated by clamping cells with 1 ${\mu}M$ $Ca^{2+}$ and applying the $K_{Ca}3.1$ activator 1-ethyl-2-benzimidazolinone, was further augmented by TBHP, and inhibited by X/XO. These effects were prevented by antioxidants. The results suggest that hydrogen peroxide increases $K_{Ca}3.1$ expression by upregulating pERK and downregulating REST, and augments the $K^+$ current. On the other hand, superoxide reduces $K_{Ca}3.1$ expression by downregulating pERK and upregulating REST, and inhibits the $K^+$ current. ROS thereby play a key role in both physiological and pathological processes in endothelial cells by regulating $K_{Ca}3.1$ and endothelial function.

Neoagarohexaose-mediated activation of dendritic cells via Toll-like receptor 4 leads to stimulation of natural killer cells and enhancement of antitumor immunity

  • ${\beta}$-Agarase cleaves the ${\beta}$-1,4 linkages of agar to produce neoagarooligosaccharides (NAO), which are associated with various physiological functions. However, the immunological functions of NAO are still unclear. In this study, we demonstrated that ${\beta}$-agarase DagA-produced neoagarohexaose (DP6), an NAO product, promoted the maturation of dendritic cells (DCs) by Toll-like receptor 4 (TLR4). DP6 directly and indirectly enhanced the activation of natural killer (NK) cells in a TLR4-dependent manner in vitro and in vivo. Finally, the antitumor activity of DP6 against B16F1 melanoma cells was inhibited in NK cell-depletion systems by using NK-cell depleting antibodies in vivo. Collectively, the results indicated that DP6 augments antitumor immunity against B16F1 melanoma cells via the activation of DC-mediated NK cells in a TLR4-dependent manner. Thus, DP6 is a potential candidate adjuvant that acts as an immune cell modulator for the treatment of melanoma.

모듈형 환자감시기와 중앙 환자감시기로 구성되는 환자감시시스템 시제품의 개발: 전체구조 및 사양 (Development of a Prototype Patient Monitoring System with Module-Based Bedside Units and Central Stations: Overall Architecture and Specifications)

  • We have developed a prototype patient monitoring system including module-based bedside units, interbed network, and central stations. A bedside unit consists of a color monitor and a main CPU unit with peripherals including a module controller. It can also include up to 3 module cases and 21 different modules. In addition to the 3-channel recorder module, six different physiological parameters of ECG, respiration, invasive blood pressure, noninvasive blood pressure, body temperature, and arterial pulse oximetry with plethysmogaph are provided as parameter modules. Modules and a module controller communicate with up to 1Mbps data rate through an intrabed network based on RS-485 and HDLC protocol. Bedside units can display up to 12 channels of waveforms with any related numeric informations simultaneously. At the same time, it communicates with other bedside units and central stations through interbed network based on 10Mbps Ethernet and TCP/IP protocol. Software far bedside units and central stations fully utilizes gaphical user interface techniques and all functions are controlled by a rotate/push button on bedside unit and a mouse on central station. The entire system satisfies the requirements of AAMI and ANSI standards in terms of electrical safety and performances. In order to accommodate more advanced data management capabilities such as 24-hour full disclosure, we are developing a relational database server dedicated to the patient monitoring system. We are also developing a clinical workstation with which physicians can review and examine the data from patients through various kinds of computer networks far diagnosis and report generation. Portable bedside units with LCD display and wired or wireless data communication capability will be developed in the near future. New parameter modules including cardiac output, capnograph, and other gas analysis functions will be added.

COVID-19 감염병 대응 의료진용 개인보호복의 동작성 및 생리적 부담 평가를 위해 개발된 모의 작업 프로토콜의 타당도 (Validity of a Simulated Practical Performance Test to Evaluate the Mobility and Physiological Burden of COVID-19 Healthcare Workers Wearing Personal Protective Equipment)

  • This study evaluated the validity of a newly developed mobility protocol examining the comfort functions and requirements of personal protective equipment (PPE) for COVID-19 healthcare workers. Eight males (age: 24.7 ± 3.0 y, height: 173.4 ± 2.3 cm, and body weight 69.9 ± 3.7 kg) participated in the following three PPE conditions: (1) Plastic gown ensemble, (2) Level D ensemble, and (3) Powered air purifying respirator (PAPR) ensemble. The mobility protocol consisted of 10 different tasks in addition to donning and doffing. The 10 tasks were repeated twice at an air temperature of 25oC with 74% RH. The results showed significant differences among the three PPE conditions in mean skin temperature, local skin temperatures (the forehead, thigh, calf, and foot), clothing microclimate (the chest and back), thermal sensation, thermal comfort, and humidity sensation, while there were no significant differences in heart rate or total sweat rate. At rest, the subjects felt less warm and more comfortable in the PAPR than in the Level D condition (P<0.05). However, subjective perceptions in the PAPR and Level D conditions became similar as the tasks progressed and mean skin and leg temperature became greater for the PAPR than the Level D condition (P<0.05). An interview was conducted just after completing the mobility test protocol, and suggestions for improving each PPE item were obtained. To sum up, the mobility test protocol was valid for evaluating the comfort functions of PPE for healthcare workers and obtaining requirements for improving the mobility of each PPE item.

Heat Shock Causes Oxidative Stress and Induces a Variety of Cell Rescue Proteins in Saccharomyces cerevisiae KNU5377

  • In this study, we attempted to characterize the physiological response to oxidative stress by heat shock in Saccharomyces cerevisiae KNU5377 (KNU5377) that ferments at a temperature of $40^{\circ}C$. The KNU5377 strain evidenced a very similar growth rate at $40^{\circ}C$ as was recorded under normal conditions. Unlike the laboratory strains of S. cerevisiae, the cell viability of KNU5377 was affected slightly under 2 hours of heat stress conditions at $43^{\circ}C$. KNU5377 evidenced a time-dependent increase in hydroperoxide levels, carbonyl contents, and malondialdehyde (MDA), which increased in the expression of a variety of cell rescue proteins containing Hsp104p, Ssap, Hsp30p, Sod1p, catalase, glutathione reductase, G6PDH, thioredoxin, thioredoxin peroxidase (Tsa1p), Adhp, Aldp, trehalose and glycogen at high temperature. Pma1/2p, Hsp90p and $H^+$-ATPase expression levels were reduced as the result of exposure to heat shock. With regard to cellular fatty acid composition, levels of unsaturated fatty acids (USFAs) were increased significantly at high temperatures ($43^{\circ}C$), and this was particularly true of oleic acid (C18:1). The results of this study indicated that oxidative stress as the result of heat shock may induce a more profound stimulation of trehalose, antioxidant enzymes, and heat shock proteins, as well as an increase in the USFAs ratios. This might contribute to cellular protective functions for the maintenance of cellular homeostasis, and may also contribute to membrane fluidity.
