Phenanthrene-induced changes in blood organic components, transaminase and ALP activities of Paralichthys olivaceus

  • Phenanthrene-induced changes in blood organic components. transaminase and ALP activities of Paralichthys oIivaceus were investigaled. The aim of this study was to obtain a holistic view of the toxic responses, and compensations of fish exposed to waterborne phenanthrene. Fish were exposed to 0, 0.5, 1, 2$\mu$M of phenanthrene for 4 weeks. The most noticeable changes were concentration-dependent increase in levels of blood GOT and GPT activities. At concentrations 1.0 and 2.0 $\mu$M of phenanthrene GOT, GPT and ALP enzyme activities in plasma were increased. Plasma albumin concentration was stable in phenanthrene-exposed fish, however, plasma total protein level was reduced significantly at the 4-week sampling points (2.0$\mu$M). Plasma glucose levels were found to be increased significantly over the cootrol throughout duration of the experiment with phenanthrene. Overall the results suggest that phenanthrene has the potential to alter some physiological functions in olive flounder.

Application of hybrid LRR technique to protein crystallization

  • LRR family proteins play important roles in a variety of physiological processes. To facilitate their production and crystallization, we have invented a novel method termed "Hybrid LRR Technique". Using this technique, the first crystal structures of three TLR family proteins could be determined. In this review, design principles and application of the technique to protein crystallization will be summarized. For crystallization of TLRs, hagfish VLR receptors were chosen as the fusion partners and the TLR and the VLR fragments were fused at the conserved LxxLxLxxN motif to minimize local structural incompatibility. TLR-VLR hybridization did not disturb structures and functions of the target TLR proteins. The Hybrid LRR Technique is a general technique that can be applied to structural studies of other LRR proteins. It may also have broader application in biochemical and medical application of LRR proteins by modifying them without compromising their structural integrity.

  • Protein phosphatase 4 (PP4), one of serine/threonine phosphatases, is involved in many critical cellular pathways, including DNA damage response (DNA repair, cell cycle regulation, and apoptosis), tumorigenesis, cell migration, immune response, stem cell development, glucose metabolism, and diabetes. PP4 has been steadily studied over the past decade about wide spectrum of physiological activities in cells. Given the many vital functions in cells, PP4 has great potential to develop into the finding of key working mechanisms and effective treatments for related diseases such as cancer and diabetes. In this review, we provide an overview of the cellular and molecular mechanisms by which PP4 impacts and also discuss the functional significance of it in cell health.

  • Regulation of pyrimidine nucleotide synthesis has been studied extensively in enteric bacteria and Bacillus species. Varieties of control modes have been proposed for regulation of pyrimidine nucleotide biosynthetic (pyr) genes. In Bacillus caldolyticus and B. subtilis, it has been proved that pyrimidine de novo biosynthetic operon is controlled by a regulatory protein PyrR-mediated attenuation. Another Gram-positive bacteria including Enterococcus faecalis, Lactobacillus plantarum, and wctococcus lactis have been found to constitute a pyr gene cluster containing the pyrR gene. In addition, it has been proposed that the structure of the 5' leader region of the Gram-negative extreme thermophile Thermus strain Z05 pyr operon provides a novel mechanism of PyrR-dependent coupled transcription-translation attenuation. Bacterial genome sequencing projects have identified the PyrR homologues in Haemophilus influenzae, Synechocystis sp., Mycobacterium tuberculosis, Streptococcus pneumoniae, S. pyogenes, and Clostridium acetobutylicum, which are currently investigating for their physiological functions.

  • We have researched quantitative structure activity relationships between molecular structure of medicines and physiological activity. Since they are non-linear, neural networks are useful tool to research them. There are many ranks for the non-linearity; therefore, the neuron function must be selected carefully. As the results of some trial calculations, Ire find the sigmoid-linear functions pair. We call the neural network constructed of the pair as ANN. The inter- or extrapolation abilities of the ANN are excellent; therefere, ANN is a superior predictor for the relationships. We evaluated the anticarcinogenic medicines, Carboquinone derivatives, by the developed ANN and leave-one-out method.

PreSMo Target-Binding Signatures in Intrinsically Disordered Proteins

  • Intrinsically disordered proteins (IDPs) are highly unorthodox proteins that do not form three-dimensional structures under physiological conditions. The discovery of IDPs has destroyed the classical structure-function paradigm in protein science, 3-D structure = function, because IDPs even without well-folded 3-D structures are still capable of performing important biological functions and furthermore are associated with fatal diseases such as cancers, neurodegenerative diseases and viral pandemics. Pre-structured motifs (PreSMos) refer to transient local secondary structural elements present in the target-unbound state of IDPs. During the last two decades PreSMos have been steadily acknowledged as the critical determinants for target binding in dozens of IDPs. To date, the PreSMo concept provides the most convincing structural rationale explaining the IDP-target binding behavior at an atomic resolution. Here we present a brief developmental history of PreSMos and describe their common characteristics. We also provide a list of newly discovered PreSMos along with their functional relevance.

  • The relationship between selenium and signal molecules is not well elucidated yet. It was found that physiological concentration of selenite, less than 3 $\mu$M, reduced ASKl activity and induced of PI3-Kinase/Akt pathways in HT1080 cells. Duration of these signal molecules by selenite was much longer than that by growth factors and other stresses. The longer duration time of these signal molecules may be important to maintain normal functions against stresses.(omitted)

  • There are several methods to implement the logical machine reasoning such as a frame theory and a production system of artificial intelligence. And these algorithms can explain the obtained result through the inference processes. However, emotional (KANSEI) patterns are not so easily implement. One of reason is that some emotional expression is the result of process from unconscious level to conscious level, and not easily identified the original unconscious causes. Therefore, a function of KANSEI database needs to structuralize unconscious level. Our approach is to develop the computerized counseling support system which can structuralize the unconscious brain functions from the view point of the psychology with focusing physiological and emotional responses. Especially, development of the algorithm that can form the network from unconscious to conscious using the image recollection is the application of the structured association technique (SAT). The developed system was implemented on the Web using CGI and emotional network database.

Hypolipidemic and Antioxidative Properties of Tocotrienol-rich Fraction (TRF) Supplementation in High Fat-fed Rats

  • In this study, we investigated a dose-dependent hypolipidemic and antioxidant effects of tocotrienol-rich fraction (TRF) from grape seeds. After induction of hyperlipidemia for 4 weeks, rats were supplemented with different dose (5, 25, and 50 mg/kg BW/day) of TRF for 1 week. Oral administration of TRF (50 mg/kg BW/day) decreased the plasma triglyceride (TG, 162.6 mg/dL), total cholesterol (TC, 83.7 mg/dL), low density lipoprotein-cholesterol (LDL-C, 20.3 mg/dL), malondialdehyde contents (MDA, 3.3 nmol/dL), and atherogenic index (AI, 2.0) compare to high-fat diet group. These data suggest that TRF supplementation has significant health benefits through the modulation of physiological functions that include various atherogenic lipid profiles and antioxidative status in hyperlipidemia.

  • Collagenases are generally defined as enzymes capable of degrading the polypeptide backbone of native collagen under conditions which do not denature the protein. Two types of proteases with collagenolytic activity have been reported and thought to play different physiological functions. Metallo-collagenases, firstly discovered in tadpole tissue explants are zinc-containing enzymes requiring calcium for optimum activity and stability, and These enzymes have been widely studied from various mammalian tissues as well as from bacteria, such as Bacillus cereus, Clostridium histolyticum, Achromobacter, Vibrio alginolyticus and Clostridium perfringens and snake venoms.
