TTRAP is a multi-functional protein that is involved in multiple aspects of cellular functions including cell proliferation, apoptosis and the repair of DNA damage. Here, we demonstrated that the lentivirus-mediated overexpression of TTRAP significantly inhibited cell growth and induced apoptosis in osteosarcoma cells. The ectopic TTRAP suppressed the growth and colony formation capacity of two osteosarcoma cell lines, U2OS and Saos-2. Cell apoptosis was induced in U2OS cells and the cell cycle was arrested at G2/M phase in Saos-2 cells. Exogenous expression of TTRAP in serum-starved U2OS and Saos-2 cells induced an increase in caspase-3/-7 activity and a decrease in cyclin B1 expression. In comparison with wild-type TTRAP, mutations in the 5'-tyrosyl-DNA phosphodiesterase activity of TTRAP, in particular $TTRAP^{E152A}$, showed decreased inhibitory activity on cell growth. These results may aid in clarifying the physiological functions of TTRAP, especially its roles in the regulation of cell growth and tumorigenesis.
Vitamin E has been a generic term for all tocopherol and tocotrienol derivatives. The most active form of vitamin E isoforms in vivo is regarded to be $\alpha$-tocopherol which is the only form defined as vitamin E in the US Food and Nutrition Board, causing controversy over setting dietary reference intake (DRI) of vitamin E. However, most of the countries other than the US maintain the original concept that all isoforms are accepted as vitamin E but with different biopotency. The roles of the isoforms of vitamin E other than $\alpha$-tocopherol have received continuous attention. Among them those of $\gamma$-tocopherol and $\alpha$-tocotrienol have been most studied in comparison with $\alpha$-tocopherol, since $\gamma$-tocopherol comprises major form of vitamin E in many plant seeds and those of the both vitamin E have been implicated in unique physiological functions. This review summarizes findings that have led a better understanding of vitamin E absorption, transport, tissue storage and various functions common and specific to vitamin E isoforms focusing $\alpha$- and $\gamma$-tocopherol as well as tocotrienols. It is expected to help redefining vitamin E and setting its DRI for Koreans.

Clothes acting as a psychological protection might be of (1) influence of magic and of spirits: (2) moral danger; (3) the general unfriendliness of the world as a whole; a reassurance against the lack of love. Clothes should provide the maximum of satisfaction in accordance with the full recognition of reality. The reality should aim at the formulation of general laws concerning the most pleasing forms, colors, and styles to suit. The complex physiological anatomical and psychological aspects of individual personalities.
Caveolin-1 is a principal protein in the plasma membrane microdomains called caveolae. Caveolae play an important role in the transcytosis and pinocytosis. Therefore, caveolin-1 is most likely to work for the membrane dynamic events. In addition, caveolin-1 interacts with various signaling molecules. Although caveolin-1 possesses a variety of physiological functions, its structural properties were little construed. Here we analyzed the structural dynamics of the N-terminal caveolin-1 (residues 1-101), in order to better understand the structural properties in terms of its versatile functionality. We first analyzed its oligomeric form using GST-fused N-terminal domain, revealing that it equilibrates between a dimer and monomers in av concentration-dependent manner. The N-terminal domain of caveolin-1 was previously found to form a heptamer, so that our data suggest the dimeric form as an intermediate structure for the heptamer formation. Then, we obtained the folding profile, which indicated that $\DeltaG_{H2O}\;is\;about\;0.5\;\pm0.03$ kcal/mol. The stability of N-terminal domain is relatively low, indicating that N-terminal domain may not be crystalline. Conclusively, the dynamic and flexible structure of N-terminal domain appears more favorable to maintain the versatile functions of caveolin-1.

Objectives : This study explores perspectives, reasons, functions of dreams in order to develop a treatment model. Methods : We searched CNKI, KISS, NDSL. Papers published since 2002, which are classified in categories. Results : 1. The Recognitions about the dream of Oriental Medicine is based on "Hwangjenaegyeong Eumsabalmong(黃帝內經 淫邪發夢)". 2. By each era, There are a variety of etiological causes, stages. 3. Dreams are generated by the internal stimuli Physiological activities of the Jangbu(臟腑),Deficiencies of yin and yang(陰陽), Pathological changes in the Jangbu(臟腑), Pathology products, external stimuli Yukeum(六淫), Eumsiksang(飮食傷), and unusual feelings. 4. Spirit(神) comes from brain(腦), not heart(心). So what we call brain spirit(腦神) is right. Dreams are the mental activity related to the brain. 5. Dreams are the reflection of the pathological changes of the human body. And it shows the development, changes and prognosis of disease. Dreams sometimes illustrate the incidence of the disease Conclusions : We organized perspectives, Causes and Functions of dreams mentioned in Oriental Medicine Literatures. Health is closely related to the dream. Therefore, we propose that the dream is worth for the diagnosis and treatment in oriental medicine.
Potassium ($K^+$) is one of the most abundant cations in higher plant. It comprises about 10% of plant dry weight and it plays roles in numerous functions such as osmo- and turgor regulation, charge balance of plasma membrane and control of stomata and organ movement. Several potassium transporters and potassium channels regulate $K^+$ homeostasis in response to $K^+$ uptake systems. In this review, we describe the biological, biochemical and physiological characteristics of shaker like potassium channels in higher plant. Especially, we searched the rice genome databases and analysized expressed genes, genome structures and protein domain characteristics of shaker like potassium channels.

It has been suggested that the structure and function of nuclear receptors are evolutionally conserved. Here, we compare the molecular functions of the nile tilapia (Oreochromis niloticus) small heterodimer partner (nSHP/NR0B2) and the Dosage-sensitive sex reversal AHC critical region on X chromosome gene 1 (nDAX-1/NR0B1) with those of human SHP and DAX-1 (hSHP and hDAX-1, respectively). We found that, upon transient cotransfection of human cells, nDAX-1 repressed the activity of tilapia SF-1 (nSF-1) but not that of human SF-1, although the physical interaction with human SF-1 was retained. Similarly, nSHP repressed the activity of nSF-1, whereas hSHP did not, pointing to divergent evolution of SHP/SF-1 in fish and human. We thus propose that the repressive functions of SHP and DAX-1 have been conserved in fish and mammals although with different transcriptional targets and mechanisms. These differences provide new insights into the physiological diversification of atypical orphan nuclear receptors during vertebrate evolution.

Janus kinase 2 (JAK2), a non-receptor tyrosine kinase, is a critical component of cytokine and growth factor signaling pathways regulating hematopoietic cell proliferation. JAK2 mutations are associated with multiple myeloproliferative neoplasms. Although physiological and pathological functions of JAK2 in hematopoietic tissues are well-known, such functions of JAK2 in the nervous system are not well studied yet. The present study demonstrated that JAK2 could negatively regulate neuronal differentiation of mouse embryonic stem cells (ESCs). Depletion of JAK2 stimulated neuronal differentiation of mouse ESCs and activated glycogen synthase kinase 3β, Fyn, and cyclin-dependent kinase 5. Knockdown of JAK2 resulted in accumulation of GTP-bound Rac1, a Rho GTPase implicated in the regulation of cytoskeletal dynamics. These findings suggest that JAK2 might negatively regulate neuronal differentiation by suppressing the GSK-3β/Fyn/CDK5 signaling pathway responsible for morphological maturation.
Spermatogenesis and testis development are highly structured physiological processes responsible for post-pubertal fertility in stallions. Spermatogenesis comprises spermatocytogenesis, meiosis, and spermiogenesis. Although germ cell degeneration is a continuous process, its effects are more pronounced during spermatocytogenesis and meiosis. The productivity and efficiency of spermatogenesis are directly linked to pubertal development, degenerated germ cell populations, aging, nutrition, and season of the year in stallions. The multiplex interplay of germ cells with somatic cells, endocrine and paracrine factors, growth factors, and signaling molecules contributes to the regulation of spermatogenesis. A cell-tocell communication within the testes of these factors is a fundamental requirement of normal spermatogenesis. A noteworthy development has been made recently on discovering the effects of different somatic cells including Leydig, Sertoli, and peritubular myoid cells on manipulation the fate of spermatogonial stem cells. In this review, we discuss the self-renewal, differentiation, and apoptotic roles of somatic cells and the relationship between somatic and germ cells during normal spermatogenesis. We also summarize the roles of different growth factors, their paracrine/endocrine/autocrine pathways, and the different cytokines associated with spermatogenesis. Furthermore, we highlight important matters for further studies on the regulation of spermatogenesis. This review presents an insight into the mechanism of spermatogenesis, and helpful in developing better understanding of the functions of somatic cells, particularly in stallions and would offer new research goals for developing curative techniques to address infertility/subfertility in stallions.
