안정만 패류양식해역의 표층퇴적물의 지화학적특성과 중금속 함량 (Physiochemical Characteristics and Heavy Metal in the Surface Sediments of Marine Shellfish Farming Waters in Anjung Bay, Korea)

  • 굴양식이 성행하는 안정만 해역은 인근 연안에 LNG비축기지, 조선소 등의 공단이 가동되고 있어 일부 양식장은 품종을 전환하고 있다. 이 연구는 안정만 해역의 연안생태계에 영향을 미치는 저질환경 특성을 조사하기 위하여 2009년부터 2010년까지 2회에 걸쳐 20개 정점의 표층퇴적물을 채집, 분석하였다. 안정만 해역의 표층퇴적물의 IL은 6.41-11.68% (평균 $9.52{\pm}1.28%$)의 범위였고, COD는 17.99-43.84 mg/g (평균 $27.53{\pm}6.39mg/g$), 그리고 AVS는 0.05-0.61 mg/g (평균 $0.17{\pm}0.14mg/g$)의 범위였다. 유기물함량은 저질의 수분함량과 밀접한 상관관계가 있었고, 이는 양식품종의 분포 패턴과도 관계가 있어 흰멍게로 양식품종이 변화된 북부해역의 COD가 비교적 높게 나타났다. 일부 조사정점을 제외하고 대부분의 중금속함량은 NOAA의 ERL 기준치 이하였고, 중금속 부화지수는 0.96-1.12의 범위로 중금속 오염에서는 비교적 청정한 것으로 판단되지만, 증가하는 선박 통행량을 고려할 때, 지속적인 저질환경 모니터링이 요구된다.

  • 동래온천지역 지열수 및 그 주변지역 지하수의 수리지구화학적 특성을 밝히기 위해서, 20개의 시료(11개 지하수 시료와 9개 온천수 시료)를 채수하여 화학성분을 분석하였다. 지하수와 온천수의 화학적 성질을 조사하고 이들의 화학 조성을 비교.분석하였다. 요인분석과 상관분석을 통하여 화학분석 자료를 몇 개의 요인으로 묶어서 요인들간의 상호연관성을 고찰하였다. 파이퍼 다이아그램에 도시한 결과, 지하수는 $Ca-HCO_3$형에 그리고 온천수는 Na-Cl 형에 속하는 것으로 나타났다. 동래온천수의 염소이온 함량은 우리나라의 다른 화강암지역의 온천수보다 높게 나타난다. 온천수의 Na/Cl 비는 0.7~l.3이다. 상평형도에 의하면, 지하수는 카오리나이트 안정 영역에 놓인다. 한편, 온천수는 미사장석이나 카오리나이트 안정영역에 놓인다. Na-K, Na-K-Ca 및 Na-K-Ca-Mg 지온계에 의하면, 지하 심부의 지열대수층의 온도는 $115~145^{\circ}C$로 추정된다.

  • 물거미는 한국에서 유일하게 연천군 은대리의 좁은 습지에서만 제한적으로 분포하고 있다. 이 연구에서는 물거미 개체군에 영향을 주는 서식 환경요인을 규명하기 위하여 습지내 물거미의 개체수와 수생태계의 다양한 물리화학 및 생물학적 환경요인과의 관계를 조사하였다. 그 결과 물거미의 개체수는 서식지의 물리화학적 환경요인 중에서 습지의 면적이 좁고 암모니아태질소와 전기전도도가 높을수록 증가하였다. 물거미는 포식자나 먹이인 피식자와는 관련성이 낮고 왕우렁이의 개체수가 많을수록 감소하였다. 또한 정수식물의 피도와는 관련성이 없었으나 부엽식물의 피도가 높을수록 물거미수는 감소하였고, 그 반대로 침수식물의 피도가 높을수록 이에 비례하여 물거미 수는 증가하였다. 이러한 결과는 물거미의 서식환경에서 수생식물이 중요하게 관련됨을 의미하고, 현재 이 서식지에서 외래종인 왕우렁이는 수생식물의 최대 섭식자로 작용하고 있으므로 이에 대한 제어대책이 시급히 필요하다.

진해만에서 물리화학적 환경요인이 식물플랑크톤 색소 및 군집조성의 시공간적 변화에 미치는 영향 (Effects of Physicochemical and Environmental Factors on Spatial and Temporal Variations in Phytoplankton Pigment and its Community Composition in Jinhae Bay)

  • 본 연구에서는 ultra performance liquid chromatography(UPLC)를 이용하여 진해만의 식물플랑크톤 생체량 및 군집구조의 시공간적 분포에 미치는 환경요인의 영향을 조사하였다. 이를 위해 2019년 4월에서 12월까지 총 5회에 걸쳐 7개 정점에 대한 식물플랑크톤 색소분석과 수온, 염분, 용존산소(DO), 영양염(DIN, DIP, Si(OH)4) 등의 환경요인 분석을 행하였다. 조사기간 중 식물플랑크톤의 생체량(Chl-a)은 7월(평균 15.4±4.3 ㎍/L)에 가장 높았고, 12월(평균 3.5±0.6 ㎍/L)에 가장 낮았다. 보조색소의 경우 fucoxanthin이 가장 많이 검출되었고 그 다음으로 peridinin, Chl-b 순으로 나타났으며, 이들의 월 변동은 Chl-a와 유사한 경향을 보였다. 식물플랑크톤 군집분석결과, 규조류가 평균 70 %로 가장 우점하였으나, 일부 녹조류, 은편모조류, 와편모조류가 출현하기도 하였다. 우점종인 규조류는 특히 수온 및 N:P ratio와 밀접하게 연관되어 있어서 여름철 고온환경 및 육상으로부터의 영양염 유입에 민감하게 반응하는 것으로 추론되었다. 또한 식물플랑크톤 색소 및 종조성은 전반적으로 계절에 따른 물리화학적 환경요인의 변화 및 지형적 특성과 연관되어 있으며 강우로 인한 담수 및 영양염 공급에 큰 영향을 받는 것으로 추정되었다.

An Approach for a Substitution Matrix Based on Protein Blocks and Physicochemical Properties of Amino Acids through PCA

  • Amino acid substitution matrices are essential tools for protein sequence analysis, homology sequence search in protein databases and multiple sequence alignment. The PAM matrix was the first widely used amino acid substitution matrix. The BLOSUM series then succeeded the PAM matrix. Most substitution matrixes were developed by using the statistical frequency of substitution between each amino acid at blocks representing groups of protein families or related proteins. However, substitution of amino acids is based on the similarity of physiochemical properties of each amino acid. In this study, a new approach was used to obtain major physiochemical properties in multiple sequence alignment. Frequency of amino acid substitution in multiple sequence alignment database and selected attributes of amino acids in physiochemical properties database were merged. This merged data showed the major physiochemical properties through principle components analysis. Using factor analysis, these four principle components were interpreted as flexibility of electronic movement, polarity, negative charge and structural flexibility. Applying these four components, BAPS was constructed and validated for accuracy. When comparing receiver operated characteristic ($ROC_{50}$) values, BAPS scored slightly lower than BLOSUM and PAM. However, when evaluating for accuracy by comparing results from multiple sequence alignment with the structural alignment results of two test data sets with known three-dimensional structure in the homologous structure alignment database, the result of the test for BAPS was comparatively equivalent or better than results for prior matrices including PAM, Gonnet, Identity and Genetic code matrix.

Effects of Onion Powder and Baking Temperature on the Physicochemical Properties of Cookies

  • Response surface methodology (RSM) was used for the optimization of the baking process of cookies made with onion powders (onion cookie). Experiments were carried out according to a central composite design, selecting onion powder content $(1.15{\sim}8.86%)$ and baking temperature $(155.9{\sim}184.1^{\circ}C)$ as independent variables and pH, titratable acidity, moisture content, density, spread factor, CIE color parameters $(L^{\ast}-$, $a^{\ast}-$, and $b^{\ast}-values)$, and hardness as response variables. pH decreased with increasing amount of onion powder and baking temperature. Moisture content also decreased as the baking temperature increased. $L^{\ast}-$ and $b^{\ast}-values$ decreased but $a^{\ast}-values$ increased with increasing onion content and baking temperature. Most polynomial models developed by RSM were highly effective to describe the relationships between the studied factors and the responses. Overall optimization, conducted by overlaying the contour plots under investigation, was able to point out the optimal range of the independent variables within which the six responses were simultaneously optimized. The point chosen as representative of this optimal region corresponded to 4.01% onion powder content and $161.84^{\circ}C$ baking temperature. The predicted dependent or response values in the optimal region were: pH=6.87, moisture content=2.77%, $L^{\ast}-value=68.45$, $a^{\ast}-value=1.98$, $b^{\ast}-value=34.64$, and spread factor=9.41.

Anti-Inflammatory Effects of Streamed Platycodon grandiflorum against UVB Radiation-Induced Oxidative Stress in Human Primary Dermal Fibroblast

  • Ultraviolet B (UVB) exposure is a risk factor for skin damage resulting in oxidative stress, inflammation, and cell death. The purpose of this study was to investigate the physicochemical properties of Platycodon grandiflorum (PG) to improve its biological activities using a three-step steaming process. We investigated the protective effects of PG and steamed PG extracts on human dermal fibroblasts (HDFs) against UVB radiation-induced oxidative stress and inflammation as well as the underlying mechanisms. The antioxidant potential of the PG extracts was evaluated by measuring the 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) scavenging activity. ABTS and DPPH were shown by the 0, 30, and 70% ethanol extracts of 2S-PG and 3S-PG ($IC_{50}$, 28~45 and $27{\sim}30{\mu}g/mL$, respectively). Treatment of UVB-irradiated cells with steamed PG ($25{\sim}400{\mu}g/mL$) did not affect their viability. The streamed PG extract suppressed UVB-induced generation of reactive oxygen species (ROS). In addition, streamed PG extract reduced cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) protein expression in UVB-irradiated HDF, regulating nuclear factor $(NF)-{\kappa}B$ expression. These findings suggest that steamed PG extract may be potentially effective against inflammation associated with UVB-induced oxidation stress.

반응표면분석법을 이용한 쌀과자의 제조 최적화 (Optimization of the Preparation of Rice Snack by Response Surface Methodology)

  • This study aimed at the optimization and development of rice snack by response surface methodology (RSM). The effect of rice soaking time and additional ratio of soybean milk on physicochemical properties and sensory evaluation of dried rice flour for non-fried snack was studied. After soaking for 0 to 12 days, Baekjinju rice was milled and then air-dried. A quadratic model was selected for weight, volume, hardness, taste and appearance. Two-factor interaction model was selected for expansibility, color and overall preference. A linear model was selected for yellowness and flavor. Weight, hardness and appearance were increased and decreased thereafter as rice soaking time increased, but the volume was reversed. Expansibility, color, taste and flavor were decreased as rice soaking time increased, but yellowness was reversed. Weight and overall preference decreased as additional ratio of soybean milk increased, but volume, expansibility and yellowness were reversed. Hardness, taste and appearance were increased after initial decrease as additional ratio of soybean milk increased, color and flavor were not changed by additional ratio of soybean milk. The optimum conditions of each factor were set to where the rice soaking time and additional ratio of soybean milk were at their minimum. Weight, volume, expansibility, yellowness, hardness, color, taste, flavor, appearance and overall preference were established at maximum level where the objective of the optimum was in level. Our data indicated the statistically predicted values of the highest desirability was 1.24 day of rice soaking time and 15.80% of additional ratio of soybean milk.

Carassius auratus(goldfish)를 이용한 BPMC와 Carbaryl의 생물농축계수의 측정 (Determination of Bioconcentration Factor on BPMC and Carbaryl in Carassius auratus(goldfish))

  • Bioconcentration Factor (BCF) is known as important criteria for ecotoxicology on hazardous chemicals. But there is no standard method for determining BCF and reported BCFs were slightly different in accordance with authors. This study was performed with aims to determine BCFs on BPMC and Carbaryl. Carassius auratus(goldfish) be chosen as test organism and test period were 3-day, 5-day and 10-day. Extract solvents were n-hexane and acetonitrile. GC-ECD was used to detecting carbamates. The obtained results were as follows: 1. It was possible to determine short term BCF$_s$ of Carbaryl or BPMC through relatively simple procedure. 2. BCF$_3$ of Carbaryl in concentration of 1, 2, 5, 10 ppm were 0.34 $\pm$ 0.06, 0.18 $\pm$ 0.02, 0.10 $\pm$ 0.01, 0.06 $\pm$ 0.01 respectively. BCF$_5$ of Carbaryl were 0.34 $\pm$ 0.05, 0.18 $\pm$ 0.02, 0.13 $\pm$ 0.01 and 0.07 $\pm$ 0.01, BCF$_{10}3$ of Carbaryl were 0.45 $\pm$ 0.05, 0.27 $\pm$ 0.02, 0.16 $\pm$ 0.02 and 0.09 $\pm$ 0.01. BCF$_3$ of BPMC in concentration of 1, 2, 5 ppm were 4.66 $\pm$ 0.17, 2.64 $\pm$ 0.49, 1.88 $\pm$ 0.24 respectively. BCF$_5$ of BPMC were 4.09 $\pm$ 0.50, 2.42 $\pm$ 0.37 and 1.83 $\pm$ 0.15. 3. BCF$_s$ of BPMC were decreased as increasing concentration. However, BPMC concentration in fish were increased in contrast to BCF. But more concentrated BPMC was found in fish 3-day test than found concentration in fish 5-day test. 4. Same trend appeared in Carbaryl. BCF$_s$ of Carbaryl were decreased as increasing concentration and prolonging test period. But found Carbaryl concentration in fish were increased. 5. BCF$_s$ of BPMC were higher than that of Carbaryl by 10 times, in spite of the physicochemical properties of the two carbamates were similar to each other. Further study is recommended to find out the reason of the difference.

부산 해운대지역 지하수와 지열수의 수리화학적 특성 (Hydrochemical characteristics of ground and geothermal waters in the Haeundae hot-spring area, Pusan, Korea)

  • Twenty-two water samples(fifteen groundwater and seven geothermal water samples) were collected to elucidate chemical characteristics of the ground and geothermal waters in the Haeundae hot spring area and its vicinity. Major and honor elements were analyzed for ground and geothermal water samples. The concentrations of $K^+$, Na+$, $Ca^{2+}$, $SO_4^{2-}$, $Cl^-$, ^F^-$ and $SiO_2$ were higher in the geothermal water samples than the groundwater samples except $HCO_3^- and Mg^{2+}$ ions. Based on the contents of Fe, Zn, Cu, Al, Mn and Pb, some of the ground and geothermal water samples are contaminated by anthropogenic sources. The ground waters shown on the Piper diagram belong to $Ca-HCO_3$ type, while the geothermal waters Na-Cl type. The graphs of $Cl^-$ versus $Na^+$, $Ca^{2+}, Mg^{2+}, K^+, SO_4^{2-} and HCO_3^-$ indicate that the groundwater is related partly with mineral-water reaction and partly with anthropogenic contamination, while the geothermal water is related with saline water. On the phase stability diagram, groundwater and thermal water mostly fall in the field of stability of kaolinite. This indicates that the ground and geothermal waters proceed with forming kaolinite. Factor and correlation analyses were carried out to simplify the physicochemical data into grouping some factors and to find interaction between them. Based on the Na-K, Na-K-Ca and Na-K-Ca-Mg geothermometers and silica geothermometers, the geothermal reservoir is estimated to have equilibrium temperature between 125${$\mid$circ}C$ and 160${$\mid$circ}C$.
