Antibacterial Activity of Selected Fruit Juices against Multidrug-Resistant Bacterial Pathogens Involved in Urinary Tract and Sexually Transmitted Infections among Tribal Women in Madhya Pradesh, India

  • Objectives: The aim of this study was to evaluate the effect of fruit juices on Multi-Drug Resistant (MDR) bacterial pathogens involved in Urinary Tract Infections (UTIs) and Sexually Transmitted Infections (STIs) among tribal women in the district Anuppur, Madhya Pradesh, India. Methods: Fresh juices of lemon (Citrus limon), amla/Indian gooseberry (Phyllanthus emblica), pineapple (Ananas comosus), mosambi/sweet lime (Citrus limetta), orange (Citrus sinensis), kiwi (Actinidia deliciosa), and pomegranate (Punica granatum) fruits were evaluated for in vitro antibacterial activity against bacterial pathogens involved in UITs and STIs among tribal women. Physico-chemical analysis of fresh fruits was also carried out by measuring the pH, moisture, protein, fat, crude fibre, carbohydrate, and ascorbic acid content. Results: Lemon and amla juice showed better antibacterial activity against the pathogens as compared to other juices. MIC results fruit juices against UTIs and STIs pathogens vary depending on the specific pathogen and juice chemical constituents. The physico-chemical analysis showed that the moisture content was highest in mosambi (90%), followed by orange (87%). Ascorbic acid content was found highest in amla (540 mg/100 g), followed by kiwi (90.3 mg/100 g). Pomegranate showed highest concentration of carbohydrate (15.28 g/100 g), fat (1.28 g/100 g), and protein (1.65 g/100 g). Lemon juice had lowest pH of 2.20, followed by amla 2.67. Conclusion: The lemon juice showed highest antibacterial activity against MDR bacterial pathogens involved in UTIs and STIs among tribal women in district Anuppur, Madhya Pradesh, India. The low pH of lemon may be responsible for its high antibacterial activity as compared to other juices.

Efficacy of Flaxseed Flour as Bind Enhancing Agent on the Quality of Extended Restructured Mutton Chops

  • Consumers have become very conscious about their nutrition and well being due to changes in their socio-economic lifestyle and rapid urbanization. Therefore, development of technology for production of low cost and functional meat products is urgently required. One such approach is innovative restructuring technology in which binding of meat pieces still remains the main challenge and extension of product is generally associated with poor binding and texture. Thus, the present study was envisaged as an attempt to solve this problem by the incorporation of flaxseed flour (FF) as bind enhancing agent. The FF was used at three different levels viz., 0.5%, 1%, and 1.5% to replace lean meat in pre-standardized restructured mutton chops formulation. The products were subjected to analysis for physico-chemical, sensory and textural properties. Cooking yield, moisture percentage and fat percentage increased with increase in the level of incorporation of FF, however, protein percent and pH decreased with increase in the level of incorporation. Shear force value of product incorporated with 1.5% FF was significantly higher (p<0.01) than control and product containing 0.5% FF level. Among the sensory attributes, product with 1% flaxseed flour showed significantly higher values (p<0.05) for general appearance, binding, texture and overall acceptability. Hardness showed significant increasing (p<0.01) values with increasing levels of incorporation of flaxseed flour, however all other parameters of texture profile analysis showed a decreasing trend. On the basis of sensory scores and physico-chemical properties, the optimum incorporation level of FF was adjudged as 1%. Products incorporated with optimum level of flaxseed flour (1%) were also assessed for water activity and microbiological quality during the storage period of 15 days. It was found that the extended restructured product could be safely stored under refrigeration ($4^{\circ}C{\pm}1^{\circ}C$) in low density polyethylene (LDPE) pouches for 15 days without marked deterioration in sensory and microbiological quality. Thus, it was concluded that flaxseed flour can be used as a good bind enhancing agent in extended restructured meat products at an economic cost.

  • Composting is among the most effective integrated waste management strategies used to recycle sewage sludge (SS) waste and generate a useful product. An encapsulated lifting system is a relatively new industrial-scale composting technology. The objective of this study was to evaluate the effectiveness of composting dewatered stabilized SS mixed with green waste using this new technology. The composting process was monitored by changes in the physico-chemical properties, UV-visible spectra, and fourier transform infrared (FTIR) spectra. The composting temperature was steady in the thermophilic range for 24 and 12 d in the intensive and maturation phases, respectively, which fulfilled the disinfection requirement. Moreover, the temperature increased rapidly to 76.8℃ within three days, and the thermophilic temperatures peaked twice and lasted longer than in traditional composting, which accelerated SS degradation and decreased the composting period necessary to obtain mature compost. FTIR spectroscopic analysis showed a diminished in methyl group derived from methylene C-H aliphatic groups because of organic matter degradation by microorganisms and an increased number of aromatic chains. The new technology may be a viable and sustainable alternative for SS management that converts waste into compost that is useful as a soil amendment.

  • 최근 물질의 특성과 구조를 해석하기 위해서 수치해석학적 모델과 컴퓨터 시뮬레이션이 많이 사용된다. 이러한 관점에서 물질의 미세구조를 해석하는 데 있어 분자동역학 해석법은 매우 유용한 방법이다. 이번 연구에서는 점토광물에 대한 확산계수 및 점착력과 같은 물리화학적 특성을 계산하기 위한 수치해석학적 방법을 소개한다. 이번연구에서 지질학적으로 심부에 위치하는 포화된 점토광물과 물을 포함한 점토광물에 대한 특성을 분자동역학을 이용해서 계산하고 균질화해석법을 활용하여 점착력과 같은 외부조건에 따라 결정되는 점토광물의 확산거동을 해석하였다. 그 결과 수치해석에 의한 해석결과 값과 기존의 실내 투수실험에 의한 결과 값이 매우 흡사한 결과를 보인다는 것을 알 수 있다. 이는 여러 가지 복합적인 조건하에서의 점토광물의 물리화학적 거동을 해석하는 데 수치해석학적 방법이 매우 유용하게 사용될 수 있다는 것을 의미한다.

FE-SEM/EDX 분석법을 이용한 석탄화력발전소에서 배출되는 입자상물질의 확인자 개발 (Identification Factor Development of Particulate Matters Emitted from Coal-fired Power Plant by FE-SEM/EDX Analysis)

  • Coal-fired power plants emit various Particulate Matter(PM) at coal storage pile and ash landfill as well as the stack, and affect the surrounding environment. Field Emission Scanning Electron Microscopy and Energy Dispersive X-ray analyzer(FE-SEM/EDX) were used to develop identification factor and the physico-chemical analysis of PM emitted from a power plant. In this study, three samples of pulverized coal, bottom ash, and fly ash were analyzed. The pulverized coal was spherical particles in shape and the chemical composition of C-O-Si-Al and C/Si and C/Al ratios were 200~300 on average. The bottom ash was spherical or non-spherical particles in shape, chemical composition was O-C-Si-Al-Fe-Ca and C/Si and C/Al ratios were $4.3{\pm}4.6$ and $8.8{\pm}10.0$. The fly ash was spherical particles in shape, chemical composition was O-Si-Ai-C-Fe-Ca and C/Si and C/Al ratios were $0.5{\pm}0.2$ and $0.8{\pm}0.5$.

Physico-Chemical Properties of Various Oxide Nanoparticles Synthesized with Electron Beam Irradiation System

  • Various oxide particles are synthesized in an ambient condition using an electron beam irradiation system. The formation of oxide particles is confirmed by FT-IR spectra results and the X-ray photoelectron spectrum. It is also shown that the morphology and size of oxide particles are significantly influenced by the type and flowrate of dilution gas. The energy dispersive X-ray (EDX) analysis also confirms that various oxide particles are synthesized.

  • Cigarettes were prepared using cut tobacco blends containing 5 levels of expanded tobacco. The effect of expanded tobacco content and tobacco weight on the draw resistance, tar, nicotine, static burning rate, net weight, and carbon monoxide deliveries were examined. At the specified hardness, tar, and tricot me significantly decreased with increasing expanded tobacco rates as a consequence of the increasing cigarette draw resistance. In the view of the experts, the smoke analysis of the cigarettes containing the expanded tobacco was investigated.

  • Ginsing polysaccharide, known to have an immune regulating effect, was administered to 23 randomly selected healthy male subjects with a mean age of 23 years in accordance with an IRB approval. Then, these subjects underwent physico-chemical tests and serum proteome was analyzed from the blood sample taken from these subjects. Analyses of proteome involved image analysis, protein sections and protein identification in sequence after two-dimensional electrophoresis was carried out. During the physico-chemical test, 4 subjects were excluded from the study. In the proteome analysis, identified were 5 spots such as SP40, 40, Cytokeratin 9, hypothetical protein LOC544932, Apolipoprotein E ,similar to Human albumin, which showed differences in the amount of protein expression. In conclusion, changes of 5 proteins were remarkable before and after administration of ginsing polysaccharides. In certain cases, hepatic and renal slight injury occurred. Thus, further clinical study on dosage regimen would be necessary for securing the basis for concentration-dependent effectiveness and safety.

Structural Bioinformatics Analysis of Disease-related Mutations

  • In order to understand the protein functions that are related to disease, it is important to detect the correlation between amino acid mutations and disease. Many mutation studies about disease-related proteins have been carried out through molecular biology techniques, such as vector design, protein engineering, and protein crystallization. However, experimental protein mutation studies are time-consuming, be it in vivo or in vitro. We therefore performed a bioinformatic analysis of known disease-related mutations and their protein structure changes in order to analyze the correlation between mutation and disease. For this study, we selected 111 diseases that were related to 175 proteins from the PDB database and 710 mutations that were found in the protein structures. The mutations were acquired from the Human Gene Mutation Database (HGMD). We selected point mutations, excluding only insertions or deletions, for detecting structural changes. To detect a structural change by mutation, we analyzed not only the structural properties (distance of pocket and mutation, pocket size, surface size, and stability), but also the physico-chemical properties (weight, instability, isoelectric point (IEP), and GRAVY score) for the 710 mutations. We detected that the distance between the pocket and disease-related mutation lay within $20\;{\AA}$ (98.5%, 700 proteins). We found that there was no significant correlation between structural stability and disease-causing mutations or between hydrophobicity changes and critical mutations. For large-scale mutational analysis of disease-causing mutations, our bioinformatics approach, using 710 structural mutations, called "Structural Mutatomics," can help researchers to detect disease-specific mutations and to understand the biological functions of disease-related proteins.
