한국산 및 중국산 천궁과 당귀의 Phthalide류 함량 비교 (Comparison on the Phthalides content of Cinidil Rhizoma and Angelicae Radix cultivated in Korea and China)

  • 천궁에서는 butylidene phthalide, butyl phthalide, senkyunolide, cnidilide + neocnidilide 및 ligustilide가 동정되었으며, 이들 성분의 70% 이상을 ligustilide와 senkyunolide가 차지하였다. Phthalide류 총량으로 볼 때 연길산 및 안국산의 중국재배 천궁은 한국재배종보다 약간 높은 경향이었으나, 길림산은 한국 재배종보다 뒤떨어지는 것으로 나타났으며 각 성분의 함량도 낮아 품질이 나쁜 것으로 확인되었다. 또한, 중국의 연길산 천궁은 ligustilide와 senkyunolide의 광분해생성물인 butylidene phthalide 및 butyl phthalide함량이 높아 유통기간과 보관상의 문제가 있었다고 판단되어져 약재 수입시 유의하여야 할 것으로 사료된다. 당귀에서는 butylidene phthalide와 ligustilide가 동정 되었으며, 전체의 약 90%를 ligustilide가 차지하였다. 우리나라 재배종 당귀 중의 ligustilide 함량은 중국 연길산의 상품 및 하품보다 높았으나, 안국산 및 길림산보다는 낮았다. 그러나 길림산을 제외한 중국산 당귀는 ligustilide의 광분해산물로 생성되는 butylidene phthalide함량이 우리 나라 재배종에 비하여 훨씬 높은 것으로 나타나, 천궁에서와 같이 중국산 약재의 유통과 보관상의 문제점이 제기되었다.

천궁의 세포배양에 의한 정유성분의 생산 (The Production of Essential Oils by Tissue Culture of Cnidium officinale)

  • Callus was derived from the shoots of Cnidium officinale. The growth rate of callus and the production of essential oils were studied under different culture conditions. The essential oils in the rhizome of Cnidium officinale and the cultivated callus were analyzed and compared by gas chromatography and mass spectrometry. It appeared that NAA induced higher growth rate and production of essential oils than 2,4-D. The compositions of essential oils were influenced by the illumination. Butyl phthalide, cnidilide, senkyunolide, butylidene phthalide, ligustilide, grandisol, tricosane, 3-methylphenol and 2-pentylthiophene were identified in the cultivated callus.

고본뿌리의 정유성분 (The Essential Oils of Ligusticum tenuissimum Roots)

  • Ligusticum tenuissimum belongs to Umbelliferae, and its root is generally used in headache, common cold and arthralgia L. We have extracted and isolated butylidene phthal ide, 3-butylidene-4,5-dihydrophthalide of phthalide group and l-limonene, etc. of terpenoid group from the root by ordinary method. In Ligusticum sinense, L. jeholense, L. tenuissimum, L. jeholense var. tenuisectum, and L. officinale etc., the components of cnidilide, ligustilide of phthalide group are common features and specific constituents in genus Ligusticum. Therefore comparing that there is no phthalide group in genus Angelica, we think it is able to classify the difference of genus between Ligusticum and Angelica of Umbelliferae in chemotaxonomy.

토천궁과 일천궁의 Phthalide류 성분비교 (Phthalide Content of Cnidium Rhizome)

  • Cnidium Rhizome(Chuan-Xing) used as sedatives, the treatment of anemia, woman's disease and ozena etc. has been one of the important oriental medicines. This paper deals with a comparison of butylidene phthalide and ligustilide as main components between rhizome of Conioselinum sp. and rhizome of Cnidium officinale by HPLC. The contents of butylidene phthalide in rhizome of Conioselinum sp. and rhizome of Cnidium officinale were found to be 0.083%, 0.067% and that of ligustilide to be 0.304%, 0.272% respectively.

Effects of the Essential Oil Components from Ligusticum chuanxiong on Proinflammatory Mediators of RAW264.7 Macrophage Cells

  • The essential oil fraction was obtained from the underground parts o of Ligusticum chuanxiong (Umbelliferae) by steam distillation, and its main components, Z-ligustilide and butylidene phthalide, were isolated by column chromatography. Its essential oil fraction and the isolated main components were examined for effects on their anti-inflammatory properties in RAW 264.7 macrophage cells to develop a new natural anti-inflammatory drug. The results showed that the L. chuanxiong essential oil fraction and its main components, Z-ligustilide and butylidene phthalide, inhibited the production of nitric oxide significantly in lipopolysaccharide (LPS)-treated RAW 264.7 cells. LPS-induced interleukin-$1{\beta}$ (IL-$1{\beta}$), interleukin-6 (IL-6), and tumor necrosis factor-alpha (TNF-$\alpha$) production was also decreased in a dose-dependent manner. In addition, western blot analysis revealed that the L. chuanxiong essential oil fraction and also its main components, Z-ligustilide, and butylidene phthalide reduced the expression levels of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS).

  • The effects of various kind and amount of nitrogen fertilizers on the plant grorth. physiological , yield and extracts. ligustilide, butylidene phthalide contents of curdsdrug "Tou-ki" (AnTelica acutiloba Kitagawa) were investigated in 1989. Five plots variouskind of nitrogen fertilizers. Namely, no nitrogen, urea. ammonium nitrate, ammoniumchloride and control plot of ammonium sulfate. The other, by providing five plotsdifferent composed ratio of nitrogenous fertilizers. containing no nitrogen (No. o) .0.5-fold nitrogen (No s), control plot of 1.0-fold nitrogen(Nl.o). 1.5-fold nitrogen(Nl s)and 2.0-fold nitrogen Na. o), but nitogen was used the ammonium sulfate. The results showedthat the crude drug "Tou-ki" can ammonium chloride be produced in good yield, displayingboth good plant growth and remarkable physiologically-active conditions, and it can beproduced such that the extracts is maximized. Additionally, using methods of gaschromatography (GC) , it was established that ligustilide and butylidene phthalide, majorcomponents in the crude drug was recovered in a good yield from the fully grown plants.The other, the plant growth, the physiologically-active, the weight of whole plant, theyield of extracts and ligustilide, butylidene phthalide were seen to be best at the plotof 2.0-fold nitrogen and according to the increase or decrease of nitrogen decreasedgradually. Therefore. about 2 fold of standard quantity seems to be the most suitablequantity of nitrogen for "Tou-ki" cultivation .uot;Tou-ki" cultivation .ion .

Anti-Aspergillus Activities of the Ligusticum chuanxiong Essential Oil Alone and in Combination with Antibiotics

  • The present study aimed to assess the antifungal properties of the essential oil fraction from Ligusticum chuanxiong (Umbelliferae) and its components against five clinically important Aspergillus species. The essential oil fraction was extracted from the underground parts of the plant by steam distillation, and its main components, namely, Z-ligustilide, butylidene phthalide, and p-cresol were isolated by column chromatography. The antifungal activities of the essential oils were evaluated by the broth dilution method. Both the total essential oil fraction of L. chuanxiong and its components showed significant anti-Aspergillus activity against all five tested strains with MICs between 62.5 and 250 ${\mu}g$/ml, respectively. In a checkerboard microtiter assay, the combination of antibiotics, itraconazole with the essential oil fraction of L. chuanxiong or its main components exhibited synergistic or additive, and in some cases indifferent, effects against the tested Aspergillus species, resulting in FICIs (fractional inhibiting concentration indices) ranging from 0.12 to 2, while the combination of antibiotics, amphothericin B with L. chuanxiong essential oils mostly showed antagonistic effects.

천궁(Cnidium officinale)의 휘발성 향기성분 및 유리기 소거활성 (Volatile Flavor Components and Free Radical Scavenging Activity of Cnidium officinale)

  • 건조방법이 다른 신선, 열풍건조, 동결건조 천궁(Cnidium officinale)을 대상으로 전자코를 이용한 휘발성 향기성분 패턴 분석 및 GC/MS를 이용한 휘발성 향기성분 분석을 실시하고, 메탄올 및 에탄올을 용매로 하여 상온 및 환류 추출로 얻어진 추출물의 유리기 소거활성을 DPPH를 이용하여 측정하였다. 건조방법이 다른 천궁의 휘발성 향기 패턴을 전자코로 분석한 결과 주성분 분석에서 제1 주성분값에 의해서 신선한 천궁과 건조시료간에 판별이 가능하였다. GC/MS에 의한 휘발성 향기성분 분석 결과 신선한 천궁이 총 85종의 성분으로 가장 많은 성분이 확인되었으며 열풍건조 시료가 64종, 동결건조 시료가 73종으로 신선, 동결건조, 열풍건조 시료순으로 확인 성분의 수가 많았던 반면, 총 휘발성 향기성분의 함량은 동결건조 시료가 784.15 ppm으로 가장 높았고, 신선한 시료가 643.64 ppm, 열풍건조 시료가 218.15 ppm으로 열풍건조 시료의 휘발성 향기성분 함량이 다른 시료에 비해 낮았다. 천궁의 주요한 휘발성 향기성분인 탄화수소류 및 phthalide류의 함량도 동결건조(383.70 ppm, 235.98 ppm), 신선(376.21 ppm, 134.05 ppm), 열풍건조(122.22 ppm, 69.00 ppm) 시료 순으로 확인되었다. 각 처리군에서 butyl phthalide, neocnidilide, 3-n-butyl phthalide 등의 phthalide류 및 sabinene, ${\beta}-selinene$, myrcene, ${\alpha}-pinene$, $trans-{\beta}-farnesens$, ${\gamma}-terpinen$ 등의 terpene류가 천궁의 주요한 휘발성 향기성분으로 확인되었으며 이들 향기성분은 서양요리에 쓰이는 셀러리의 주요 향기성분과도 유사하였다. 천궁 추출문의 유리기 소거활성을 DPPH를 이용하여 측정한 결과, 동결건조 천궁의 methanol 상온추출물 500 ppm이 49.79%로 가장 높은 유리기 소거활성을 보였으며, 동결건조 시료의 ethanol과 methanol 상온 추출물은 250 ppm 이상의 농도에서 $25\;{\mu}M$농도의 ${\alpha}-tocopherol$보다 유의적으로 높은 유리기 소거활성을 지니는 것으로 나타났다. 환류 추출의 경우는 상온추출물보다 유리기 소거활성이 낮았으며 동결건조 천궁의 ethanol 환류 추출물 500 ppm은 34.71%의 유리기소거 활성을 보였다.

Variation of the Essential oil and Main Component from Different Origin of Levisticulum officinale Koch

  • The essential oils were isolated by hydrodistillation and their composition determined capillary GC method with standards. The essential oil content showed significant differences between the two populations on the vegetative organs. The essential oil level of the leaves and roots was considerably higher in the Korean population at full flowering and waxy ripening stage but essential oil content of the roots was significantly higher in the Hungarian taxon at leaf rosette stage. We observed the essential oil accumulation tendency was mianly dependent on plant organs and intra-specific taxon during the vegeation period. Butylidene-phthalide was proved to be the main component of the oil in both population roots (50.9-73.3%), while dimethyl-acetate was showed as a major compound on the over-ground parts (56.7-62.0%). The qualitative composition of the essential oil in the reproductive organs concerning the identified compounds was the same as the vegetative parts with the main component ${\alpha}-phellandrene$ (4.8-28.1%) and butylidene-phtalide (9.7-16.1%), The quantitative composition showed some changes during the ontogenesis phases. Most characteristic ones are the decreasing proportion of dimethyl-acetate (from 7.3% to 1.1%) and the appearance of ${\alpha}-pinene$ (from 0.5% to 1.5%) only after fruit setting in both population.
