Multi-channel images of 11-MUA(11-Mercaptoundecanoic acid) and 11-MUOH(11-Mercaptoundecanol) self-assembled monolayers were obtained by using two-dimensional surface plasmon resonance (SPR) absorption. The patterning process was simplified by exploiting direct photo-oxidation of thiol bonding (photolysis) instead of conventional photolithography. Sharper images were resolved by using a white light source in combination with a narrow bandpass filter in the visible region, minimizing the diffraction patterns on the images. The line profile calibration of the image contrast caused by different resonance conditions at each point on the sensor surface (at a fixed incident angle) enables us to discriminate the monolayer thickness in nanometer scale. Furthermore, there is no signal degradation such as photo bleaching or quenching, which are common in the detection methods based on fluorescence.

Recent studies have suggested that integrin (CR3) participates in the signal transduction pathways of certain GPI-anchored phagocytic receptors including $Fc{\gamma}RIIIB$. One consequence of this functional linkage is an inducible association between CR3 and cortical microfilaments that is triggered by $Fc{\gamma}RIIIB$ binding to immobilized immune complexes (IC). That this signaling event requires the co-expression of $Fc{\gamma}RIIIB$ with CR3 was documented by the use of NIH 3T3 transfectants expressing both CR3 and $Fc{\gamma}RIIIB$ (clone 3-23), CR3 alone (clone 3-19), and $Fc{\gamma}RIIIB$ alone (clone 3-15). Pretreatment of 3-23 cells with protein kinase inhibitors such as staurosporine and methyl 2,5-dihydroxycinnamate (MDHC) blocked IC-stimulated CR3 microfilament proximity without affecting the extent to which $Fc{\gamma}RIIIB$ constrains the lateral membrane mobility of a subset of CR3 on the cell surface (as measured in fluorescence recovery after photobleaching experiments). These data support that CR3 and $Fc{\gamma}RIIIB$ molecules are physically and functionally associated and that ligation of FcgRIIIB triggers CR3-dependent signal transduction.
This study is to assess the photocatalytic degradation of PET and Nylon 6 films containing nano-sized $TiO_2$ powders of anatase and rutile types. The PET and Nylon 6 films containing six kinds of the nanoparticles were prepared by melt casting method using a heating press machine. Reflectance in visible region and water contact angles of the irradiated PET and Nylon 6 composite films decreased with increasing UV/$O_3$ irradiation. Also the enhanced hydrophilicity has a close relationship with the increase in the Lewis base parameter, which indicates more oxidized polymer surfaces. The photocatalytic degradation of the nanocomposite films increased with increasing $TiO_2$ content and UV energy, which is more significant with the anatase types rather than the rutile types. The amide linkages in the Nylon 6 seemed to be more susceptible to the UV light compared to the ester groups in the PET, particularly in the presence of the $TiO_2$ photocatalysts. The photoscission and photodegradation of the polymers in the composites produced more degraded structure assisted by the photocatalytic activity of the $TiO_2$ nanoparticles. Also the composite films can bleach the methylene blue dyes more easily under the UV/$O_3$ irradiation, suggesting the photobleaching activity of the $TiO_2$ nanoparticles.
The three-dimensional organization of chromatin and its time-dependent changes greatly affect virtually every cellular function, especially DNA replication, genome maintenance, transcription regulation, and cell differentiation. Sequencing-based techniques such as ChIP-seq, ATAC-seq, and Hi-C provide abundant information on how genomic elements are coupled with regulatory proteins and functionally organized into hierarchical domains through their interactions. However, visualizing the time-dependent changes of such organization in individual cells remains challenging. Recent developments of CRISPR systems for site-specific fluorescent labeling of genomic loci have provided promising strategies for visualizing chromatin dynamics in live cells. However, there are several limiting factors, including background signals, off-target binding of CRISPR, and rapid photobleaching of the fluorophores, requiring a large number of target-bound CRISPR complexes to reliably distinguish the target-specific foci from the background. Various modifications have been engineered into the CRISPR system to enhance the signal-to-background ratio and signal longevity to detect target foci more reliably and efficiently, and to reduce the required target size. In this review, we comprehensively compare the performances of recently developed CRISPR designs for improved visualization of genomic loci in terms of the reliability of target detection, the ability to detect small repeat loci, and the allowed time of live tracking. Longer observation of genomic loci allows the detailed identification of the dynamic characteristics of chromatin. The diffusion properties of chromatin found in recent studies are reviewed, which provide suggestions for the underlying biological processes.

Chromatin dynamics is essential for maintaining genomic integrity and regulating gene expression. Conserved bromodomain-containing AAA+ ATPases play important roles in nucleosome organization as histone chaperones. Recently, the high-resolution cryo-electron microscopy structures of Schizosaccharomyces pombe Abo1 revealed that it forms a hexameric ring and undergoes a conformational change upon ATP hydrolysis. In addition, single-molecule imaging demonstrated that Abo1 loads H3-H4 histones onto DNA in an ATP hydrolysis-dependent manner. However, the molecular mechanism by which Abo1 loads histones remains unknown. Here, we investigated the details concerning Abo1-mediated histone loading onto DNA and the Abo1-DNA interaction using single-molecule imaging techniques and biochemical assays. We show that Abo1 does not load H2A-H2B histones. Interestingly, Abo1 deposits multiple copies of H3-H4 histones as the DNA length increases and requires at least 80 bp DNA. Unexpectedly, Abo1 weakly binds DNA regardless of ATP, and neither histone nor DNA stimulates the ATP hydrolysis activity of Abo1. Based on our results, we propose an allosteric communication model in which the ATP hydrolysis of Abo1 changes the configuration of histones to facilitate their deposition onto DNA.
This study was conducted to find out the optimal solvent extraction method [Distilled water (DW), 70% ethanol, 99% ethanol] of mushrooms, including Pleurotus ostreatus (Jacq.) Que, Pleurotus eryngii and Flammulina velutipes and improve their usability as natural antioxidants. To analyze antioxidant activities in each mushroom, total polyphenol, flavonoid contents, 2,2-Diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) cation radical (ABTS+) and fluorescence recovery after photobleaching (FRAP) were measured. All mushrooms showed the highest total polyphenol contents in DW mushroom extract (p<0.001). Total flavonoid contents were the highest in P. eryngii and F. velutipes DW and 70% ethanol mushroom extracts (p<0.05). All mushrooms showed the highest activities using DPPH and FRAP assays in the DW extraction method (p<0.001). P. ostreatus (Jacq.) Que and P. eryngii showed the highest ABTS+ radical scavenging activity in the DW extraction method, and F. velutipes showed the highest activity in the 70% ethanol extraction method (p<0.001). As a result of comparing IC50 values of DPPH and ABTS+ radicals and FRAP EC50 values, the DW P. ostreatus (Jacq.) Que extract showed high antioxidant activities (p<0.001). Pearson's correlation between total polyphenol contents and antioxidant activities showed a positive correlation in all mushrooms (p<0.01). Therefore, extraction of the mushrooms with DW can enhance the extraction of effective bioactive substances and antioxidant activity.
In order to determine the relationships between the lea(-burning disease and the light harvesting chlorophyll-protein (LHCP) complex in Panax ginseng C. A. Meyer, we investigated the chlorophyll-protein (CP) complex of the thylakoid membrane and its characteristics. In P. ginseng four Cp-complex bands determined by non-denaturing SDS-PAGE were identified CP I'(containing reaction center of photosystem I and LHCP I antennae), CP I (reaction center of photosystem I) LHCP II** (oligoform of LHCP II), and LHCP II (photosystem II antennae, CP 26 and CP 29) by Bassis and Dunahay's procedures. Under our experimental condition, the CP I band was only observed in P. ginseng and the band intensity of LHCP II** in P ginseng was higher than in spinach and soybean. There were differences in the absorption and fluorescence spectra and chlorophyll a/b ratio of the CP-complex bands between P. ginseng and other Plants. The Polypeptidr content of P. ginseng thylakoid was lower than in spinach and soybean thylakoid, and the Polypeptide profiles of P. ginseng was low band intensity, especially about 29-35 kD, 55 kD, and 60 kD, compared to spinach and soybean. The inhibitory effects of 2,5-dimethylfuran, specific singlet oxygen ($^1O_2$) quencher, showed that singlet oxygen destroyed 60% of chl.a, 90% of chl.b and 70% of carotenoid in bleaching P. ginseng with leaf-burning disease.
The thermoluminescence(TL) glow curves and the optical absorption of $Al_2O_3$ irradiated with ${\gamma}$-ray, electron, and $Li^{+}$ ion followed by electron irradiation have been investigated to determine the relation of TL peak to its defect type. The TL glow curve of $Al_2O_3$ irradiated with ${\gamma}$-ray shows TL peaks at 380 K, 415 K, and 475 K. The UV photobleached TL glow curve of $Al_2O_3$ irradiated with ${\gamma}$-ray shows that the 380 K and 475 K TL peaks completely disappear while the 415 K TL peak still exists. The electron beam induced TL glow curve of $Al_2O_3$ after $Li^{+}$ ion implantation shows that the TL peak at 440 K is enhanced by a factor of 2 over the TL intensity of unimplanted $Al_2O_3$ while the TL peak at 380 K evidently disappears The implanted $Li^{+}$ ions are assumed to form singly charged interstitial cations and then recombine with electron trapped in F centers to produce F+ centers. The 380 K and 475 K TL peaks are proposed to be associated with F center, while the 415 K and 440 K TL peak are connected with F$^{+}$ center.

Chromophoric Dissolved Organic Matter (CDOM) plays a dominant role in absorbing UV-VIS light and is also important in the biogeochemical carbon cycle due to the production of carbon dioxide from photo-oxidation at the sea surface in marine environments. Since absorption by CDOM was recently found to be responsible for increasing the energy absorbed in the mixed layer by 40 % over pure seawater, the importance of CDOM absorption in seawater for increasing sea surface temperature has come to be well recognized. We measured aCDOM and the absorption characteristics of CDOM during spring 2012 and 2014 in the southwestern East Sea. Distribution of CDOM in spring 2012 and 2014 was compared and S value was used to find the source of CDOM in the study area. As a result, the average $a_{CDOM}$ was $0.237m^{-1}$ ($0.009{\sim}0.988m^{-1}$) and the average S value was $16{\mu}m^{-1}$,which shows coastal properties. Also a positive correlation between Chl a and CDOM was observed ($r^2=0.34$), with an especially strong correlation near coastal stations. aCDOM in 2014 was about 40 % higher than aCDOM in 2012 during spring in the study area. This difference in aCDOM concentration resulted not only from annual variation but also from stratification and photobleaching in late spring 2012. This observation implies the possibility of flux of carbon dioxide into the atmosphere as a result of photo-oxidation in the East Sea.
