Effects of Adding Super Dose Phytase to the Phosphorus-deficient Diets of Young Pigs on Growth Performance, Bone Quality, Minerals and Amino Acids Digestibilities

  • Two experiments were conducted to evaluate the efficacy of feeding an Escherichia coli (E. coli) derived phytase to pigs fed P deficient, corn-soybean meal diets. In Exp. 1, one hundred and twenty crossbred piglets ($9.53{\pm}0.84$ kg) were allocated to one of five treatments which consisted of four low P diets (0.61% Ca, 0.46% total P and 0.24% non-phytate P) supplemented with 0, 500, 1,000, or 20,000 FTU/kg E. coli phytase as well as a positive control formulated to be adequate in all nutrients (0.77% Ca, 0.62% total P and 0.42% non-phytate P). The treatments were applied to six pens with four pigs per pen for 28 days. In Exp. 2, ten crossbred pigs ($19.66{\pm}1.16$ kg) fitted with ileal T-cannula were used in a nutrient balance study. The pigs were assigned to treatments similar to those used in Exp. 1 in a doubly replicated $5{\pm}4$ incomplete Latin square design (5 diets with 4 periods). Each period consisted of a 5-d adjustment period followed by a 3-d total collection of feces and urine and then a 2-d collection of ileal digesta. Supplementation with phytase linearly increased (p<0.05) weight gain, feed intake, feed efficiency, bone breaking strength and fat-free dry and ash bone weight. There were linear increases (p<0.01) in the apparent ileal digestibility (AID) of DM, GE, CP, Ca, total P, inositol hexaphosphate ($IP_6$) and some AA with increasing dose of E. coli phytase. Pigs fed 20,000 FTU/kg had a greater (p<0.05) AID of IP6 (80% vs 59% or 64%, respectively) than pigs fed diets with 500 or 1,000 FTU/kg phytase. There were linear increases (p<0.05) in the total tract digestibility of Ca, total P, Na, K, Mg, and Zn as well as in the retention of Mg and Zn with increased phytase dose. The retention and utilization of Cu, and the total tract digestibility of CP and Cu quadratic increased (p<0.05) with increased phytase dose. In conclusion, supplementation of 500 FTU of phytase/kg and above effectively hydrolyzed phytate in low-P corn-soybean diets for pigs. In addition, a super dose of phytase (20,000 FTU/kg) hydrolyzed most of the IP6 and consequently further improved mineral use, protein utilization and performance.

Effects of Italian ryegrass diet on the performance, carcass characteristics, and blood metabolites of Hanwoo steers

  • This study was conducted to develop a pasture feeding program for reducing the Hanwoo feeding cost. Italian ryegrass (IRG) is a major plant which is abundantly produced in the southern area of South Korea. Locally produced Italian ryegrass has a 30 - 50% reduced pasture cost compared to imported pasture grasses. We hypothesized that Italian ryegrass feed could reduce the feeding cost but not change the physiological characteristics of the Hanwoo steers in the feedlot. Four treatments (Control, short fed IRG silage, long fed IRG silage, and IRG hay) in a completely random design were used to feed 27 Hanwoo steers. Three or four steers were fed in the same pen, and 8 pens were used for the treatments. Blood was drawn from each steer on the first week of every month from 11 to 30 months. Overall, the average daily gain and feed efficiency were not different among the treatments (p < 0.05) except for the final fattening diets (p < 0.05). The long fed IRG silage group had a low ADG compared to the other groups (p < 0.05). The serum non-esterified fatty acid and glucose concentrations were reduced during the fattening periods (p < 0.05); however, the serum albumin, total proteins, triglycerides, and phosphorus were increased during the fattening periods (p < 0.05). The carcass weight, longissimus muscle area and back-fat thickness were low in the long fed IRG silage group (p < 0.05). The IRG diet induced saturated fatty acids in the longissimus dorsi (p < 0.05). These results show that the IRG diet increased the serum triglycerides but reduced the glucose level at the final fattening diet and decreased the meat yield score in Hanwoo steers.

  • This study was conducted to investigate both plant growth-promoting and plant disease- controlling activities of bacterial strains isolated from soil. All the isolated strains were able to grow at various temperatures. All the strains, except ANG40, showed antagonistic effects against various phytopathogenic fungi. This antagonism can be ascribed to the production of siderophores and antibiotic substances. In addition, all the strains showed abilities such as nitrogen fixation, phosphate solubilization, and siderophore production. These results suggest that nitrogen, phosphorus, and iron can be converted into forms that can be easily absorbed by the plants for their growth. Analysis of the growth-promoting properties revealed that ANG51 produced 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase and indole-3-acetic acid (IAA) both of which are related to ethylene production. In contrast, the other strains were found to have only IAA-producing ability. Therefore, this study suggests that Pseudomonas geniculata ANG3, Exiguobacterium acetylicum ANG40, and Burkholderia stabilis ANG51, which were selected through analysis of comparative advantages for both plant growth promotion and disease-controlling activity, may be used as biological agents.

SURFACE CHARACTERISTICS AND BIOLOGICAL RESPONSES OF HYDROXYAPATITE COATING ON TITANIUM BY HYDROTHERMAL METHOD: AN IN VITRO STUDY

  • Statement of problem. Hydroxyapatite(HA) coated titanium surfaces have not yet showed the reliable osseointegration in various conditions. Purpose. This study was aimed to investigate microstructures, chemical composition, and surface roughness of the surface coated by the hydrothermal method and to evaluate the effect of hydrothermal coating on the cell attachment, as well as cell proliferation. Material and Methods. Commercially pure(c.p.) titanium discs were used as substrates. The HA coating on c.p. titanium discs by hydrothermal method was performed in 0.12M HCl solution mixed with HA(group I) and 0.1M NaOH solution mixed with HA(group II). GroupⅠ was heated at 180 $^{\circ}C$ for 24, 48, and 72 hours. GroupⅡ was heated at 180 $^{\circ}C$ for 12, 24, and 36 hours. And the treated surfaces were evaluated by Scanning electron microscopy(SEM), Energy dispersive X-ray spectroscopy(EDS), X-ray photoelectron spectroscopy(XPS), X-ray diffraction method(XRD), Confocal laser scanning microscopy(CLSM). And SEM of fibroblast and 3-(4,5- dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) assay were used for cellular responses of the treated surfaces. Results. The color of surface changed in both groups after the hydrothermal process. SEM images showed that coating pattern was homogeneous in group II, while inhomogeneous in group I. H72 had rosette-like precipitates. The crystalline structure grew gradually in group II, according to extending treatment period. The long needle-like crystals were prominent in N36. Calcium(Ca) and phosphorus(P) were not detected in H24 and H48 in EDS. In all specimens of group II and H72, Ca was found. Ca and P were identified in all treated groups through the analysis of XPS, but they were amorphous. Surface roughness did not increase in both groups after hydrothermal treatment. The values of surface roughness were not significantly different between groups I and II. According to the SEM images of fibroblasts, cell attachments were oriented and spread well in both treated groups, while they were not in the control group. However, no substantial amount of difference was found between groups I and II. Conclusions. In this study during the hydrothermal process procedure, coating characteristics, including the HA precipitates, crystal growth, and crystalline phases, were more satisfactory in NaOH treated group than in HCl treated group. Still, the biological responses of the modified surface by this method were not fully understood for the two tested groups did not differ significantly. Therefore, more continuous research on the relationship between the surface features and cellular responses seems to be in need.

Effects of Dietary Lysine Levels on Apparent Nutrient Digestibility and Serum Amino Acid Absorption Mode in Growing Pigs

  • Two experiments were conducted to determine the effects of different dietary lysine levels on the apparent nutrient digestibility, the serum amino acid (AA) concentration, and the biochemical parameters of the precaval and portal vein blood in growing pigs. In Experiment 1, 15 noncannulated pigs received diets with different lysine densities (0.65%, 0.95%, and 1.25% lysine) for 13 d. A total collection digestion test was performed, and blood samples were collected from the precaval vein at the end of the experiment. In Experiment 2, four cannulated pigs were fed the same diets of Experiment 1. The experiment used a self-control experimental design and was divided into three periods. On d 5 of each period, at 0.5 h before feeding and hourly up to 8 h after feeding, single blood samples were collected from catheters placed in the portal vein. In Experiment 1, some serum AAs (including lysine), serum urinary nitrogen (SUN), and total protein (TP) concentrations were significantly affected by the dietary lysine levels (p<0.05). Moreover, the 0.65% lysine treatment showed a significant lower apparent digestibility of gross energy, dry matter, crude protein, and phosphorus than the other treatments (p<0.05). In Experiment 2, serum lysine, histidine, phenylalanine, threonine, valine, isoleucine (p = 0.0588), triglyceride, and SUN (p = 0.0572) concentrations were significantly affected by the dietary lysine levels (p<0.05). Additionally, almost all of the determined serum AA and total AA concentrations reached their lowest values at 0.5 h before feeding and their highest values at 2 h after feeding (p<0.05). These findings indicate that the greatest absorption of AA occurred at 2 h after feeding and that the dynamic profile of serum AA is affected by the dietary lysine levels. Moreover, when the dietary lysine content was 0.95%, the growing pigs achieved a better nutrient digestibility and serum metabolites levels.

The Effects of Organic Manure and Chemical Fertilizer Application Levels on the Growth and Nutrient Concentrations of Yellow Poplar (Liriodendron tulipifera Lin.) Seedlings (유기질 및 화학비료 처리수준이 어린 백합나무 생장 및 양분농도에 미치는 영향)

  • Soil nutrient management is important to maintain the constant productivity of seedling production in the nursery for successful forest restoration. This study investigated the effects of organic manure and chemical fertilizer application levels on the growth, soil properties, and nutrient concentrations of yellow poplar seedlings. One-year-old yellow poplar seedlings were treated with the combination of 3 level organic manures(0, 5 Mg/ha, 10 Mg/ha; mixture of poultry manure, cattle manure, swine manure, and sawdust) and 3 level nitrogen-phosphorus-potassium(NPK) chemical fertilizers(0, 1x(urea, $30g/m^2$; fused superphosphate, $70g/m^2$; potassium chloride, $15g/m^2$), 2x). Organic manure significantly increased the soil pH and the concentrations of nitrogen, available phosphorous, exchangeable potassium, calcium, and magnesium. In contrast, the NPK chemical fertilizer decreased the soil pH and exchangeable calcium concentration, did not affect the soil concentrations of nitrogen and magnesium, and increased the concentrations of available phosphorous and exchangeable potassium. Both organic manure and NPK chemical fertilizer treatments increased the seedling height, root collar diameter, and dry weight by 39% and 25%, respectively. The treatment with manure 5 Mg/ha and NPK 2x chemical fertilizer mostly increased seedling dry weight by 2.6 times more than that of the control. Compared to the effects of the fertilization treatments on the soil properties, the effects on nutrient concentrations in the leaves were relatively small. These findings indicate that organic manure that was derived from livestock byproducts and sawdust can be utilized with chemical fertilizer to improve seedling production as well as conserving soil quality.

Evaluation of Water Quality Characteristics on Tributaries of Mankyeong River Watershed (만경강 유역내 하천의 특성별 영향 평가)

  • A survey on four tributaries along with Mankyeong River was carried out to get the information far the water quality improvement and control. Typical paddy farming were major agricultural practices in Kosancheon and Soyangcheon. Iksancheon was livestock raising watershed. Chucheon was urban watershed. Water quality in six sites of main stream and four sites of tributaries in Mankyeong River were investigated from May to August in 2001. The concentration of nutrient in main stream of Mankyeong River were in the range of 3.78$\sim$12.68 mg/L for total nitrogen, 0.043$\sim$0.864 mg/L for total phosphorus, 2.59$\sim$13.29 mg/L for BOD and 12.9$\sim$119.5 mg/L for COD, respectively. Water quality of Mankyeong River mostly exceeded the standard water quality criteria of Korea. Major causes of water pollution were evaluated as sewage of swine and urban area. Among the four tributaries, water quality in agricultural practices, Kosancheon and Soyangcheon was relatively less polluted. While, the highest level of water pollution measured in Iksancheon was due to livestock. The water quality of Iksancheon and Chucheon was generally more polluted in the dry period than in rainy period.

Physiological Studies on Adaptation of Tilapia(Oreochromis miloticus) in the Various Salinities II. Serum Components Levels and Electrophoretic Patterns (틸라피아의 해수순치에 관한 생리학적 연구 II. 혈청성분과 전기영동상의 변화)

  • This study was taken to examine serum components concentrations and electrophoretic patterns of female tilapia(Oreochromis niloticus) living in 0$\textperthousand$, 10$\textperthousand$, 20$\textperthousand$, and 30$\textperthousand$ salt concentrations, respectively. The results obtained in these experiments were summarized as follows. The level of albumin and total protein showed changes in each salinity, but didn't significantly(P<0.05) change in Oreochromis niloticus. The level of BUN didn't significantly(P<0.05) change. When fish were adapted from 0$\textperthousand$ to 10$\textperthousand$, 20$\textperthousand$ and 30$\textperthousand$, each calcium level in every salinity groups showed less than that of control, and didn't significantly change in 10$\textperthousand$, 20$\textperthousand$, 30$\textperthousand$ salinity. The level of calcium didn't significantly(P<0.05) change in each salinity. In 20$\textperthousand$ salinity, the level of cholesterol was at the highest peak. When fish were adapted from 0$\textperthousand$ to 10$\textperthousand$, 20$\textperthousand$ and 30$\textperthousand$, each glucose level gradually decreased. When fish were adapted from 0$\textperthousand$ to 10$\textperthousand$, 20$\textperthousand$ and 30$\textperthousand$, each glucose level gradually decreased. When fish were adapted from 0$\textperthousand$ to 10$\textperthousand$, 20$\textperthousand$ and 30$\textperthousand$. In 30$\textperthousand$ salinity, the level of alkaline phosphatase was at the highest peak. The level of serum enzyme such as SGOT and SGPT was higher in seawater-adapted group than in freshwater group. The level of phosphorus chnage significantly(P<0.05) in each salinity. Correlation coefficient between serum albumin and glucose in 0$\textperthousand$ was +0.924. Correlation coefficient between serum SGOT and SGPT of individuals in 0$\textperthousand$ was +0.917. Fraction 1 of transferrin patterns of tilapia(Oreochromis niloticus) adapted in seawater was much thicker than that of transferrin patterns of individuals adapted in freshwater. Also fraction No. a wasn't observed in some individuals adapted in freshwater. These results showed that transferrin adapted in seawater relatively increased. Slight differences, that is, showed to be observed in total iron binding capacityand iron saturatin rate between tilapia adapted in freshwater and in seawater. The increase in total iron binding capacity was attributed to a rise in transferrin pressent in the first fraction of serum protein adapted in seawater. Accordingly, the serum iron levles seemed to be related to salinity($\textperthousand$).

Evaluation of True Metabolizable Energy and the Effect of Corn Distillers Dried Grains with Solubles in the Diets on Broiler Performance and Nutrient Availability (육계에 있어서 옥수수 주정박의 진정 대사 에너지가 측정 및 사료내 첨가가 생산성과 영양소 이용률에 미치는 영향)

  • This study was conducted to investigate the effect of corn distillers dried grains with solubles (CDDGS) in broiler diet on ileal amino acid digestibility and broiler performance. In experiment 1, TMEn of CGGDS ranged from 2,863 to 2,976 kcal/kg and averaged 2,904 kcal/kg on an 86% DM basis. In experiment 2, 192 male Ross broilers (one-day-old) were assigned randomly to 4 dietary treatment groups (12 birds/pen, 4 pens/treatment) as CDDGS levels (0, 6, 12 and 18%) and fed experimental diets for 6 weeks (starter ($0{\sim}2wk$); grower ($2{\sim}4wk$); finisher ($4{\sim}6wk$)). There was no difference in BWG of the chicks. However, feed intake and feed efficiency of chicks fed with non CDDGS groups were better than those of chicks fed with CDDGS fed groups (P<0.05). The amino acid digestibilities of diets containing CDDGS were high for phosphorus but low for lysine, threonine, aspartic acid, cystine and glycine (P<0.05) in the treatments compared to those of the control diet. This study showed that CDDGS has a high ME value and the treatments were low for the ileal amino acid digestibility, but there was no effect on the broiler performances.

Salts Reduction Effect of Natural Zeolite in Plastic Film House Soil (천연 Zeolite를 이용한 시설재배지 토양의 염류제거 효과)

  • This study was performed to investigate the effect of zeolite on the reduction of soil EC level in the plastic film house. The EC level of experimental soil was 5.0 dS $m^{-1}$ and the zeolite was applied to the soil at seven levels (0.5, 1, 2, 5, 10, 15, 20%) with three replications. The reduction degree of soil EC level showed positive tendency to the mixing ratio of zeolite. Especially, the EC level reduced rapidly from 5.01 to 2.8 dS $m^{-1}$ in the plot where zeolite was mixed by 20% 10 days after treatment. The pH level of soil was in positive relation to the mixing ratio of zeolite, contrary to the negative relation to the concentration level of water soluble Ca, Mg and phosphorus (P). The water contents of soil mixed with 15% and 20% zeolite were 14% and 17.3% respectively but it was 12.7% for control soil. Therefore, we expect natural zeolite to salts reduction agent for exchangeable cation and phosphate which is difficult to reduce by watering and other methods.
