어유의 Eicosapentaenoic Acid의 식이섭취가 성숙쥐의 혈청 및 간의 지질조성에 미치는 영향 (Effect of Dietary Eicosapentaenoic Acid on Serum and Liver Lipids Patterns of Male Rat)

  • The dietry effects of marine n-3, plant n-3 and plant n-6 fatty acid on serum lipids levels, liver phospholipid fatty acid composition in rat were investigated. Four groups of male Sprague-Dawley rats, 30 weeks old, were fed on one of 4 different experimental diets for 4 weeks. The diets were composed of 15% fat(w/w) of either concentrated EPA oil(20:5, n-3 : 65%), fish oil(20:5, n-3 : 19%, 22:6, n-3 : 18%), perilla oil(18:3, n-3 : 60%) or corn oil(18:2, n-6 : 49%). Blood was initially taken before experimental feeding and also taken after 2 weeks and 4 weeks feeding the diet respectively and then examined for the levels of serum lipids. Rats were sacrificed at 4 weeks after the diet for the analysis of liver phospholipid fatty acid. EPA feeding remarkably decreased the serum levels of triglyceride, total cholesterol, HDL-cholesterol and total phospholipid than any other oil feeding. Fish oil feeding decreased serum HDL-cholesterol level comparable to the effect of EPA feeding and decreased total cholesterol and phospholipid less than but close to the effect of EPA feeding. Perilla oil feeding did not change serum levels of triglyceride and phospholipid, but it decreased serum total cholesterol a lot and HDL-cholesterol a little. Corn oil feeding did not affect triglyceride and total cholesterol while it increased serum level of HDL-cholesterol and total phospholipid. Serum HDL-cholesterol level was increased only in corn oil group. But contrary to the result of serum total phospholipid, liver phospholipid level found to be higher in fish oil and EPA groups than in perilla oil and corn groups. The fatty acid composition of liver phospholipid, phosphatidylcholine(PC) and phosphatidyl ethanolamine(PE) turned out to be affected by dietary fatty acid. 18:2 of liver PC was the lowest in FO group following CO group. The ratio of 20:4/18:2 was lower in PO group than in EPA group in consequence of higher 18:2 and lower 20:4 in PO group and vise versa in EPA group. In the liver PC and PE, similar trends in the ratios of n-6/n-3 and 20:4/18 were found showing higher ratios with CO and EPA group over FO and PO group. EPA group showed the lowest level of 20:5 and lower level of 20:6 than group. Fish oil was more efficient than EPA oil and PO in lowering the ratio of n-6/n-3 in consequence of the highest 22:6, and the lowest 18:2 in liver phospholipid. But PO lowers the ratio or 20:4/18 more than FO. In conclusion, EPA oil was more effective in lowering serum lipids than FO and PO. Reviewing the dietary effect of fatty acid on eicosanoids composition in rats, it is considered that more possibility was with FO than PO in the effectiveness of atherosclerosis prevention and more with PO than with EPA oil. It was also found that FO showed more effective than EPA oil for atherosclerosis prevention. It was hardly found that CO had any effect on lowering serum lipids and on eicosanoids composition in liver phospholipid for the prevention of atherosclerosis.

Study on the fatty acid profile of phospholipid and neutral lipid in Hanwoo beef and their relationship to genetic variation

  • Maize which has very high omega-6 fatty acid content has been used as a main feed grain for Hanwoo beef production to increase marbling, and thus omega-6 to omega-3 fatty acids ratio in Hanwoo beef is expected to be biased. To elucidate the current status of omega fatty acids ratio in Hanwoo beef, fatty acid profiles of neutral lipid and phospholipid fraction were analyzed separately using 55 Hanwoo steers' longissimus dorsi muscle slaughtered at Pyeongchang, Korea from Oct. to Nov. 2015. In addition, an association study was conducted to evaluate associations between single nucleotide polymorphism (SNP) markers from references and omega fatty acid profiles in phospholipid of Hanwoo beef samples using analysis of variance (ANOVA). In neutral lipid fraction, composition of saturated and monounsaturated fatty acids was higher and polyunsaturated fatty acids was lower compared to those in phospholipid fraction. The mean n-6/n-3 ratios of Hanwoo were $56.059{\pm}16.180$ and $26.811{\pm}6.668$ in phospholipid and neutral lipid, respectively. There were three SNPs showing statistically significant associations with omega fatty acid content. GA type of rs41919985 in fatty acid synthase (FASN) was significantly associated with the highest amount of C20:5 n-3 (p = 0.031). CC type of rs41729173 in fatty acid-binding protein 4 (FABP4) was significantly associated with the lowest amount of C22:2n-6 (p = 0.047). AG type of rs42187261 in FADS1 was significantly linked to the lowest concentration of C20:4 n-6 (p = 0.044). The total n-6/n-3 ratio of the steer which has all four SNP types in above loci (27.905) was much lower than the mean value of the total n-6/n-3 ratio in phospholipid of the 55 Hanwoo steers ($56.059{\pm}16.180$). It was found that phospholipid and neutral lipid of Hanwoo have very high n-6/n-3 ratios compared to the reported data from different cow breeds. Four SNPs in genes related with fatty acid metabolism showed significant associations with the fatty acid profile of phospholipid and may have potential as SNP markers to select Hanwoo steers in terms of n-6/n-3 balance in the future.

사태의 가열시간 및 냉동저장에 따른 지방산 조성 변화 (Changes of Fatty Acid Composition in Shank During Heating Time and Frozen Storage)

  • This study was carried out to investigate changes of the lipid contents and the fatty acid composition in shank during heating time and frozen storage. 1. The total lipid contents of raw shank were about 3.57% and decreased stepwise during heating time 30, 60, 90 min and frozen storage(24hrs) The contents of neutral lipid, glycolipid and phospholipid were 70.71%, 6.36%, and 22.93% in raw shank, and neutral lipid contents were decreased, whereas Phospholipid contents were increased according to heating tide. In frozen storage, neutral lipid and glycolipid contents were increased, while phospholipid contents were decreased. 2. Lipids of shank possessed about 8 kinds of fatty acid as the constituent by gas-liquid chromatography analysis. The main fatty acids were oleic acid, palmitic acistearic acid and linoleic acid: the fatty acids of total lipids in raw shank were 43.48% of oleic acid, 23.13% of palmitic acid,12.00% of stearic acid and 6.75% of linoleic acid. Also the fatty acids were 43.32% of oleic acid, 23.26% of palmitic acid, 9.30% of stearic acid 2.15% of linoleic acid in neutral lipid, 22.63% of oleic acid, 8.44% of palmitic acid, 11.98% of stearic acid, 27.01% of linoleic acidin glycolipid, 39.38% of oleic acid, 15.89% of palmitic acid, 15.55% of stearic acid, 17.49% of linoleic acid in phospholipid. 3. The fatty acid pattern of total lipid, neutral lipid, glycolipid and phospholipid was not any changes, whereas there was a difference in the fatty acid contents: palmitic acid and stearic acid of total lipid were decreased in the 30 min and 60 min heating but increased in 90min heating, and linoleic acid of neutral lipid was increased stepwise during heating time and frozen storage. Also palmiict acid of glycolipid was increased gradually and linoleic acid in heating time 30, 60 min was higher than 90 min and frozen storage. Among fatty acids in phoapholipid, oleic acid was increased during heating time, while decreased in frozen storage, and linoleic acid was not any change but linolanic acid was increased. UFA/SFA of phospholipid was the highest when heating time was 60 min. From above results, it was found that when heating time was 60 min beneficial nutritionally, comparing with changes of fatty acid composition according to the heating time aid frozen storage.

밤 지질(脂質) 성분(成分)의 분석(分析) -제2(二)보. 구성 지질(脂質) 및 구성 지방산(脂肪酸)의 조성(組成)- (Analysis of the Lipid Components in Chestnut(Castanea crenata) -II. Lipid and Fatty Acid Composition of Neutral Lipid , Glycolipid and Phospholipid-)

  • 은기(銀寄), 녹조(鹿鳥), 상피(霜被)의 3가지 밤 품종(品種)의 지질(脂質)성분으로 부터 분획한 중성지질, 당지질 및 인지질의 구성지질(脂質)과 구성지방산(脂肪酸)을 thin-layer chromatography 및 gas chromatography에 의하여 분리 정량하였다. 유리지질(遊離脂質) 중의 중성지질(中性脂質)로는 glyceride, free sterol, esterified sterol 주성분이었으나 결합지질(結合脂質)에는 triglyceride는 거의 없었고 free fatty acid와 monoglyceride가 소량 함유되어 있었다. 외과육(外果肉)의 유리지질(遊離脂質)에는 triglyceride가 25.7%로 훨씬 많았으나 결합지질에는 거의 함유되지 않은 것이 특이하였고 기타 성분은 미량 존재하였다. 유리지질(遊離脂質)중의 당지질(糖脂質)은 digalactosyl diglyceride, esterified steryl glycoside가 주성분이었으나 결합지질(結合脂質)은 digalactosyl diglyceride가 주성분이었다. trigalactosyl diglyceride가 유리지질에는 극히 적으나 결합지질에는 2.0% 함유되어 있다. 유리지질 중의 인지질(燐脂質)은 lysophosphatidyl choline, phosphatidyl inositol, phosphatidyl choline, phosphatidyl glycerol, phosphatidyl ethanolamine이 비교적 고른 분포를 보이고 있으며 결합지질(結合脂質) 중에는 phosphatidyl choline, phosphatidyl inositol이 주성분이었다. 중성지질(中性脂質)의 지방산조성(脂肪酸組成)은 linoleic acid가 유리지질과 결합지질에 각각 49.3%, 45.1%로 제일 많았고 linolenic acid, palmitic acid가 역시 주요 지방산(脂肪酸)이었다. 당지질(糖脂質)과 인지질(燐脂質)에 있어서도 그 경향(傾向)은 비슷하였으나 palmitic acid의 함량이 상대적으로 많아 포화지방산 함량이 중성지질보다 훨씬 많았으며 전체적으로 결합지질에 포화지방산 함량이 많았다.

사람주나무 종실유의 화학적 조성 (Chemical Components of Seed Oil of Sapium japonicum Pax. et Hoffm.)

  • 사람주나무 (Sapium japonicum) 종실유의 물리 화학적 특성과 구성성분을 다양한 분석법으로 구명하였다. 사람주나무의 종실유를 구성하는 지질은 중성지질이 93% 로 가장 높고 , glycolipid 가 4.9%, phospholipid가 1.3%로 나타났다. GC 분석 결과 사람주나무의 종실에는 9종의 지방산이 존재하였다. 종실유에서 지방산의 함량은 저장기간 등에 따라 약간의 차이를 보였다. 종실유 중 3종의 지질은 silica open column을 통해 분획하고, 이들의 지방산 성분을 조사하였다. 3종의 지질 중 주요 지방산은 linoleic acid와 linolenic acid로 나타났다. 채취직후의 종실유에서 지방산의 함량은 저장된 종실유의 지방산에 비해 전반적으로높았다. 이상의 결과로 보아 사람주나무의 종실유는 화장품, 세정제, 의약품 등으로의 이용이 기대된다.

Linkage Between Biodegradation of Polycyclic Aromatic Hydrocarbons and Phospholipid Profiles in Soil Isolates

  • A bacterial consortium capable of utilizing a variety of polycyclic aromatic hydrocarbons has been isolated from a former manufactured gas plant site. The consortium consisted of four members including Arthrobacter sp., Burkholderia sp., Ochrobacterium sp., and Alcaligenes sp., which were identified and characterized by the patterns of fatty acid methyl esters (FAME analysis) and carbon source utilization (BIOLOG system). With the individual members, the biodegradation characteristics of aromatic hydrocarbons depending on different growth substrates were determined. FAME analyses demonstrated that microbial fatty acid profiles changed to significant extents in response to different carbon sources, and hence, such shift profiles may be informative to characterize the biodegradation potential of a bacterium or microbial community.

개량(改良)메주의 숙성과정(熟成過程) 중 지질(脂質)조성의 변화(變化)에 관(關)한 연구(硏究) (Studies on Change of Lipids Improvement-Meju during the Fermentation)

  • Aspergillus oryzae에 의한 개량메주의 숙성과정에 있어서 지질의 조성변화를 체계적으로 규명하고저 silicic acid column chromatography에 의한 당지질, 중성지질, 인지질의 분획정량 및 gas liquid chromatography에 의해서 중성지질의 성분 변화를 실험한 결과는 다음과 같다. 1. 대두 및 삶은 콩의 총 지질을 분획한 결과 중성지질 93~94%로 대부분이었고 인지질 4~5%, 당지질 2.0~2.1%였다. 개량메주 발효과정 중에 있어서 중성지질의 함량은 차차 감소하였으며 상대적으로 당지질 및 인지질은 증가하였다. 2. 대두 및 삶은 콩의 비극성 지질은 triglyceride가 88-89%로 주요 구성 성분이었고 sterol ester, 유리지방산, diglyceride 및 sterol은 삶은 콩보다 대두에서 함량이 높았다. 또한 메주의 숙성과정에 있어서는 triglyceride의 함량은 현저히 감소하는 경향을 나타내었으며 sterol ester, 유리지방산 및 diglyceride는 차차 증가하였다. 3. 조지질, 중성지질, 당지질 및 안지질의 지방산조성에 있어서 삶은 콩 및 대두에 있어서 linoleic acid 54~70%, oleic acid 20.0~22.6%, palmitic acid 11.0~12.4% 순이었으며 myristic acid는 trace로 검출되었다. palmitic acid는 당지질과 인지질에서 조지질 및 중성지질 보다 그 함량이 조금 높았다. 4. 메주의발효과정에 있어서 조지질, 중성지질, 당지질 및 인지질의 지방산의 변화는 대체로 linoleic acid 및 linolenic acid가 차차 감소하였고 palmitic acid, stearic acid 및 oleic acid는 차차 증가하여 대체로 4 일째에 최대였다. 5. 당지질과 인지질의 지방산의 변화에 있어서 다같이 미확인 지방산 3 종류를 포함하여 9종류가 검출되었으며 2 종류의 미확인 지방산은 발효과정에서 생긴 것으로 앞으로 계속 검토할 흥미있는 과제라 하겠다.

한국산 마의 지질 성분의 분리 및 분석 (Fractionation and Quantitative Analysis of Lipid Components in Korean Yam (Dioscorea) Tubers)

  • 한국산 마의 주요 품종인 Dioscorea batatas, D. aimadoimo and D. japonica 3가지 품종의 시료들을 동결건조하여 분말로 만든 다음 지질의 추출, silicic acid column chromatography에 의한 지질의 분획, thin layer chromatography에 의한 지질의 분리 및 gas chromatography에 의한 지방산 조성 등을 행하였다. 총지질의 함량은 D. japonica가 다른 마에 비해 적은 편이었으며, 지질을 분획한 결과 3가지 품종 모두 중성지질이 가장 많았고 그 다음 당지질, 인지질 순으로 나타났다. 분획한 각 지방의 구성 지질로는 중성지질이 sterolester, triglyceride, 1,3-diglyceride, 1,2-diglyceride 및 monoglycerid였고, 당지질에서 acylsterylglycoside, sterylglycoside, digalactosyldiglyceride 및 sulfolipid였고, 인지질에서 phosphatidylethanolamine, phosphatidylcholine 및 phosphatidylinositol 이었다. GC에 의한 총지질의 지방산 조성은 팔미트산과 리놀레산이 가장 많은 부분을 차지하였고 불포화지방산 함량이 포화지방산 함량보다 많았다. 또한 중성지질, 당지질 및 인지질의 지방산 조성도 총 지질을 구성하는 지방산 조성과 같은 양상을 보여 주었다.

Variation in Microbial Biomass and Community Structure in Sediments of Peter the Great Bay (Sea of Japan/East Sea), as Estimated from Fatty Acid Biomarkers

  • Variation in the microbial biomass and community structure found in sediment of heavily polluted bays and the adjacent unpolluted areas were examined using phospholipid fatty acid analysis. Total microbial biomass and microbial community structure were responding to environmental determinants, sediment grain size, depth of sediment, and pollution due to petroleum hydrocarbons. The marker fatty acids of microeukaryotes and prokaryotes - aerobic, anaerobic, and sulfate-reducing bacteria - were detected in sediments of the areas studied. Analysis of the fatty acid profiles revealed wide variations in the community structure in sediments, depending on the extent of pollution, sediment depth, and sediment grain size. The abundance of specific bacterial fatty acids points to the dominance of prokaryotic organisms, whose composition differed among the stations. Fatty acid distributions in sediments suggest the high contribution of aerobic bacteria. Sediments of polluted sites were significantly enriched with anaerobic bacteria in comparison with clean areas. The contribution of this bacterial group increased with the depth of sediments. Anaerobic bacteria were predominantly present in muddy sediments, as evidenced from the fatty acid profiles. Relatively high concentrations of marker fatty acids of sulfate-reducing bacteria were associated with organic pollution in this site. Specific fatty acids of microeukaryotes were more abundant in surface sediments than in deeper sediment layers. Among the microeukaryotes, diatoms were an important component. Significant amounts of bacterial biomass, the predominance of bacterial biomarker fatty acids with abundance of anaerobic and sulfate-reducing bacteria are indicative of a prokaryotic consortium responsive to organic pollution.

아마인유와 해바라기 종자유의 혼합급이가 식이성 고지혈증 흰쥐의 간장 지질성분 및 지방산 조성에 미치는 영향 (Effects of Feeding the Mixture of Lineseed and Sunflower Seed Oil on the Lipid Components and Fatty Acid Compositions of Liver in Dietary Hyperlipidemic Rats)

  • This study was carried out to investigate the effect of the feeding mixture of linesed oil, rich in n-3 PUFA and the sunflower seed oil, rich in n-6 PUFA on the lipid metabolism in the dietary hypprlidemic rats. After male Sprague-Dawley rats were induced hyperlipidemia by feeding the diet containing lard, butter, and cholesterol for 3 weeks, then they were fed with the diet containing lard 3.0% and butter 12.0% for control, the mixture in different proportion of both linseed oil and sunflower seed oil, and antihyperlipidemic durgs for 2 weeks. Analysis of the lipid component and the fatty acid composition of the liver showed following results. Concentration s of the total cholesterol and phospholipid in liver were significantly higher in group 2 (olive oil 12.0%) and lower in the other groups than in the control group, especially lower in groups 3 (cholestyramine 2.0%) and 9 (sunflower seed oil 12.0%) . Concentration of triglyceride was lower in the other groups except group 4 (liparoid), especially lowe rin group 9 than in the control group. In the fatty acid composition of liver lipids, C18:2 was the major fatty acid. Contents of n-6 PUFA increased , while those of n-3 PUFA decreased in groups composition of the test lipids. From the data on concentration s of total cholesterol. Phospholipid and triglyceride in liver, we concluded that the feeding mixed with 3.0% lard and 12.0 % sunflower seed oil were most effective for the improvement of the live lipids. The fatty acid composition in liver lipids were affected by the fatty acid composition of the test lipids.
