We investigated the effect of two nonlamellar-prone lipids, phosphatidylethanolamine (PE) and dioleoylglycerol (DOG), on the efficiency of protein encapsulation in liposomes. When the phosphatidylcholine (PC) matrix was replaced with PE or DOG during liposome formulation, the amounts of glutathione S-transferase and bovine serum albumin entrapped in the vesicles increased with increasing PE or DOG concentration. The presence of PE and DOG synergistically affected protein entrapment. These results suggest that protein encapsulation can be enhanced by the presence of nonlamellar lipids and/or lipid-induced membrane properties.
We investigated the responses of six types of lipid-coated AT-cut quartz crystal resonators to chemical vapour sensors. The responses of quartz crystal at 9 MHz coated with phosphatidylglycerol (PG), phosphatidylinositol(PI), phosphatidylethanolamine(PE), phosphatidylserine(PS), and lipid A(LA) were obtained for amyl acetate, acetoin, menthane and other organic gases which showed different affinities for each lipid. The identification of odorants depending on the species of lipid used for coating is discussed in terms of the normalized resonant frequency shift pattern.
Two innovative methods to prepare target-sensitive immunoliposomes containing doxorubicin by coupling monoclonal antibodies (mAb DH2, SH1) specific to cancer cell surface antigens ($G_{M3}$, $Le^X$) have been developed and are described here. Firstly, liposomes containing N-glutaryl phosphatidylethanolamine (NGPE) were prepared, followed by the encapsulation of doxorubicin, DH2 or SH1 antibodies were conjugated to NGPE in the liposomes (direct coupling). Secondly, liposomes were prepared with NGPE/mAb conjugates by the detergent dialysis method (conjugate insertion), and then doxorubicin was encapsulated by proton gradient. The immunoliposomes prepared by both methods were able to specifically bind to the surface of the tumor cells - B16BL6 mouse melanoma cells. The efficiencies of doxorubicin-entrapping into liposomes prepared by direct coupling and conjugate insertion was about 98% and 25%, respectively. These types of liposomal formulation are sensitive to target cells, which can be useful for various clinical applications.

The purpose of this study was to investigate phospholipase $A_{2}$ activity and lipid peroxidation I streptozotocin induced diabetic rats. Sprague-Dawley male rats weighting-Dawley male rats weighting 300$\pm$10gm were randomly assigned to normal and STZ-induced diabetic group. Diabetes was induced by intravenous injection of 55mg/kg of STZ in sodium citrate buffer(pH 4.3). Animals were sacrificed at the 6th day of diabetic states. Body weight gains were lower in DM group. Phosphatidylcholine hydrolysis in liver was not significantly different between two groups, whereas phosphatidylethanolamine hydrolysis in liver was increased by 69% in DM group comparing with that of normal group. Liver microsomal phospholipase $A_{2}$ activity and level of TBARS was increased by 91%, 109% in DM group compared with that of normal group, respectively. The present results indicate that phospholipase $A_{2}$ activity is specific to PE hydrolysis, leading to lipid peroxidation process in STZ induced diabetic rats.
A lipidomic study on extensive plasma lipids in bacterial peritonitis (cecal ligation and puncture, CLP)-induced sepsis in mice was done at 24 h post-CLP. The effects of administration of lysophosphatidylcholine (LPC) and lysophosphatidic acid (LPA), compounds known to have beneficial effects in CLP, on the sepsis-induced plasma lipid changes were also examined. Among the 147 plasma lipid species from 13 lipid subgroups (fatty acid [FA], LPA, LPC, lysophosphatidylethanolamine [LPE], phosphatidic acid [PA], phosphatidylcholine [PC], phosphatidylethanolamine [PE], phosphatidylinositol [PI], monoacylglyceride [MG], diacylglyceride [DG], triacylglyceride [TG], sphingomyelin [SM], and ceramide [Cer]) analyzed in this study, 40 and 70 species were increased, and decreased, respectively, in the CLP mice. Treatments with LPC and LPA affected 14 species from 7 subgroups, and 25 species from 9 subgroups, respectively. These results could contribute to finding the much needed reliable biomarkers of sepsis.
The action of phospholipid on the rhodotorucine A(RH.A) acceptance by heterbasidiomyceteous yeast Rhodosporidium toruloides mating type a cells and the effect of medium composition during sexual differentiation were investigated. Activation of trigger peptidase(TPase)was very sensitive to the originated phospholipid from R. toruloides and was more sensitive to phospholipid liposome made up of phospholipi. Phospholopod present on the membrance of mating type a cells consists of phospatidylglycerol(PG), phosphatidylethanolamine(PE), phospatidylcholine(PC), phospatidylinositol(PI), and phosphatidylserine(PS) of 12.9, suprisingly 45.4, 11.0, and 13.9%, respectively. As the result of using C-1 and N-1 mediums which limited C and N sources capable of inhibiting the synthesis of phospholipid, it resulted inhibiting sexual dlfferentiation and production of Rh.A from mating type Acells.
The influence of dietary cholesterol on phospolipid metabolism in rat liver microsmes was studied in rats fed a diet containing fish oil(FO) or beef tallow (BT). The hepatic phospholipid content decreased wherease gepatic triglyceride and cholesterol increased significantly in both groups after cholestered supplementation. Plasma concentrations of phospholipid and traiglyceride increased with cholesterol supplement in both groups while cholesterol decreased only moderately in the FO group. Dietary cholesterol affected microsomal phosphiolpids in liver ; the proportation of phosphatidylcholine decreased in the FO group, an d it also slightly decreased in the BT group at the expense of phosphatidylethanolamine. The activity of CTP : phospocholine cytidylytransferase , the rate-limiting enzyme of phosphatidylcholine synthesis, increased inhepatic mocrosomes whreas it decreased in hepatic cytosol of both groups by cholesterol supplementation. In conclusion, these indicated that the dietary cholesterol profoundly influences phospholipid metabolism in the rat liver.
Platelet-activating factor (PAF) acetylhydrolase, which removes the acetyl moiety at the sn-2 position, has been found in human amniotic fluid. We purified this enzyme by ammonium sulfate precipitation, and sequential use of DEAE-Sepharose CL-6B, hydroxyapatite, chelating-Sepharose, and Mono Q column chromatographies. This enzyme exhibited broad pH optima and was unaffected by EDTA. Partially purified enzyme had a molecular weight of approximately 34 kDa on SDS-PAGE. In addition, the enzyme activity was inhibited by either diisopropylfluorophosphate(DFP) or p-bromophenacylbromide (p-BPB), suggesting that this enzyme possesses active serine and histidine residues. The enzyme showed similar activity towards PAF and oxidatively modified phosphatidylcholine, but didn't hydrolyze phosphatidylcholine or phosphatidylethanolamine with a long chain fatty acyl group at sn-2 position.
Effect of sodium deoxycholate and sodium dodecyl sulfate on Rhizopus oryzae were investigated. Morphological change was obtained by supplement of these surfactants into culture media during the sumerged culture. In accordance with morphological changes, composition of phospholipid was changed. In case of surfactant-free culture, phosphatidylcholine, phosphatidylethanolamine, and phosphatidylserine were measured more than 95% of total phospholipid. But cardiolipin and phosphatidylinositol were conspicuously increased by treatment of both sufactants. Presence of phospolipase A, C, and D were detected from mycelium. Phospholipase A and D were activated by supplement of sodium deoxycholate and C was activated by sodium dodecyl sulfate. These results were interpreted in respect of polymorphism of phospholipid and membrane stability against solubilization effect of surfactants.
Cytochrome c induces fusion of phosphatidylserine /phosphatidylethanolamine vesicles while apocytochrome c does not have a fusogenic capability despite the fact that the apoprotein binds to the vesicles more extensively. In order to see whether the difference in the fusogenic behavior comes from the topological variation in membrane bound proteins, the holoprotein and apoprotein were labeled with phenylisothiocyanate, a hydrophobic label, in the presence of its hydrophilic analogue p-sulfophenylisothiocyanate. Apocytochrome c was labeled with the hydrophobic probe more extensively than the cytochrome c, indicating that the apoprotein penetrates deeper into the bilayer than cytochrome c does. The translocation experiments of these proteins by trypsin entrapped vesicles further supported this conclusion.
