The present investigation was undertaken to study status of erythrocytic antioxidant enzymes in normal cycling and $\alpha$- tocopherol supplemented anestrus buffalo heifers. The pre-supplementation erythrocytic activities of superoxide dismutase (U/mg Hb), glutathione peroxidase (U/mg Hb) and glucose-6-phosphate dehydrogenase (U/g Hb) upregulated significantly (p<0.05) in anestrus heifers ($10.08{\pm}0.09$, $14.09{\pm}0.54$, $9.25{\pm}0.29$) when compared to normal cycling ones ($6.93{\pm}0.04$, $11.61{\pm}0.19$, $5.58{\pm}0.26$). The oral supplementation of $\alpha$-tocopherol (a) 3,000 mg per week per animal in anestrus heifers declined erythrocytic superoxide dismutase and glucose-6-phosphate dehydrogenase activities significantly (p<0.01) but led to non-significant increase in erythrocytic glutathione peroxidase activity. Results indicated that supplementation of $\alpha$-tocopherol to anestrus buffalo heifers mitigated the effects of oxidative stress to improve their antioxidant status.
Metallothioneins(MTs) belong to the class of low molecular weight proteins. Recently, it has been suggested that MTs may playa direct role in cellular defense against oxidative stress by functioning as antioxidants. Oxidative damage to different cellular components makes a major contribution to many pathogenenesses. Several studies have demonstrated that MT is able to quench a wide range of reactive oxygen species at a higher efficiency than other well known antioxidants such as superoxide dismutate(SOD). The present study was designed to evaluate the effect of MT on the activities of the reactive oxygen species removal system. MT showed the scavenging of superoxide in the SOD assay system in the presence or absence of SOD. When MT was added to nicotinamide adenine dinucleotide phosphate(NADPH) oxidation system in presence of fixed amount of SOD increase the breakdown rate of superoxide. When MT was added to the system that form nitrite from hydroxylammonium chloride, the formation of nitrite was inhibit. We concluded that the function of MT as antioxidant might have an effect on the level of superoxide scavenging.
The detoxifying properties of cruciferous vegetables components have been the subject of several recent investigations. Evidences from many biochemical and pharmacological studies indicated that higher consumption of cruciferous vegetables is associated with lower incidence of harmful actions such as hepatotoxicity and oxidative stress in animal and human populations. Recently, it has been reported that drug metabolizing and detoxifying enzyme activities were increased by cruciferous vegetable extract in which sinigrin is known to be a main active component, accounting for about 2 to 3 percents of total extract. The detoxifying effect of sinigrin has been well reported in several literatures. The metabolism of sinigrin in animal, however, has not been reported yet. That led us to study the metabolism of sinigrin in rat. Sinigrin is nown to be metabolized into three compounds, i.e., allyl isothiocyanate, glucose and potassium phosphate in cruciferous vegetables. Allyl isothiocyanate was formed in rat hepatic mitochondrial fraction in dose and incubation time dependent manner, that was confirmed by HPLC. Glucose formation was came up with results similar to that of allyl isothiocyanate. Three hours after i.p. administration of sinigrin to rat, allyl isothiocyanate appeared in rat liver, and five hours later it was detected in liver and blood. The above results suggested that sinigrin might be metabolized into allyl isothiocyanate, glucose and potassium phosphate in rat.
Effect of partial oxygen pressure on the cell growth and the activities of oxidative defense enzymes were measured in the continuous culture of Streptomyces coelicolor. Both the wild type and the mutant strain resistant to hydrogen peroxide were cultured and the dry cell weight of the two cultures were measured at different oxygen tensions. Growth of the wild type was inhibited by oxygen at above 0.5 vvm. Growth of the hydrogen peroxide resistant mutant was stimulated by pure oxygen at 0.5 vvm but was inhibited by oxygen at 1.0 vvm. Therefore, growth of the hydrogen peroxide resistant mutant was less affected by the deleterious oxidative stress of oxygen. Activities of the several defense enzymes were also measured at different oxygen tensions. Activities of catalase and glucose-6-phosphate dehydrogenase increased significantly as oxygen pressure increased in the wild type culture. In the mutant, however, increase in those enzyme activities was not obvious whereas the uninduced levels of the above enzymes were higher than those of wild type. As judged by Western blotting, the amount of the major catalase increased as the oxygen pressure increased. This indicates that the induction of the catalase activity by oxygen pressure is mostly due to the increase in the expression level for the major catalase.

Fumonisin B1 (FB1) structurally resembles sphingolipids and interferes with their metabolism leading to sphingolipid dysregulation. We questioned if FB1 could exacerbate liver or kidney toxicities in glutathione peroxidase 1 (Gpx1) and catalase (Cat) knockout mice. While higher serum levels of thiobarbituric acid reactive substances (TBARS) and sphinganine (Sa) were measured in Gpx1/Cat knockout mice (Gpx1/Cat KO) than wild type mice after 5 days of FB1 treatment, serum levels of alanine aminotransferase (ALT), sphingosine-1 phosphate (So-1-P), and sphinganine-1 phosphate (Sa-1-P) were found to be relatively low. Although Sa was highly elevated in Gpx1/Cat KO mice and wild mice, lower levels of So and Sa were found in both the kidney and liver tissues of Gpx/Cat KO mice than wild type mice after FB1 treatment. Paradoxically, FB1-induced cellular apoptosis and necrosis were hastened under oxidative stress in Gpx1/Cat KO mice.

Tissue engineering is an emerging research field that has the potential to restore, regenerate and repair damaged bone tissue and organs. Tricalcium phosphate and hydroxyapatite biomaterials-based calcium phosphate are excellent materials that have both osteoconduction and biocompatibility for bone tissue regeneration. In this study, solution structures were successfully fabricated using a fused deposition modeling system based on deposition and heating devices. The morphology characteristics of the bioceramic scaffolds sintered at a temperature of $1,300^{\circ}C$ were analyzed by scanning electron microscopy. The effects of various blended TCP/HA ratio on the microstructure and shrinkage were studied. The mechanical properties of the scaffolds were measured using a compression testing machine from stress-strain curves on the crosshead velocity of 1 mm/min. The fabricated scaffolds were evaluated by cell proliferation tests of MG-63 cells. The results of this study suggest that the blended TCP(75 wt%)/HA(25 wt%) scaffold is an appropriate scaffold for bone tissue regeneration.
Rheological properties of 5% starch suspension were investigated using a capillarity tube viscometer. Nonglutinous and glutinous rice starch suspensions at $30-60^{\circ}C$, and $30-55^{\circ}C$, respectively, had no yield stress and showed dilatant flow behavior in the absence or presence of phosphate. However, flow behavior of nonglutinuous starches at $65^{\circ}C$ and glutinous starch at $60^{\circ}C$ was pseudopalastic. Flow activation energies for rice starch suspensions were 0.88-1.45 kcal/mole at $30-50^{\circ}C$ and about 45-73 kcal/mole $60-65^{\circ}C$. Flow activation energy in the presence of phosphate was 90-182 kcal/mole at $60-65^{\circ}C$. Akibare starch had the highest activation energy, and glutinous starch the lowest.
Glucose-6-phosphate dehydrogenase (G6PD) deficiency is caused by X-linked recessive disorderliness. It induces severe anemia when a patient with G6PD deficiency is exposed to oxidative stress that occurs with administration of an antimalarial drug, primaquine. The distribution of G6PD deficiency remains unknown while primaquine has been used for malaria treatment in Myanmar. This study aimed to investigate the prevalence of G6PD deficiency and its variants in Chin State, Myanmar. Among 322 participants, 18 (11 males and 7 females) demonstrated a G6PD deficiency. Orissa variant was dominant in the molecular analysis. This would be related to neighboring Indian and Bangladeshi population, in which Orissa variant was also reported as the main mutation type. The screening test for G6PD deficiency before primaquine treatment appears to be important in Myanmar.

[Purpose] Recent studies have shown that glucose-6-phosphate isomerase (GPI)-which is a glycolysis interconversion enzyme-reduces oxidative stress. However, these studies are limited to tumors such as fibrosarcoma, and there are no studies that have examined the effects of exercise on GPI expression in mice skeletal muscle. Furthermore, GPI acts in an autocrine manner thorough its receptor, autocrine motility factor receptor (AMFR); therefore, we investigated expression level changes of secreted GPI from skeletal muscle in in vitro study to examine the potential role of GPI on skeletal muscle. [Methods] First, we performed an in vitro study, to identify the condition that upregulates GPI levels in skeletal muscle cells; we treated C2C12 muscle cells with an exercise-mimicking chemical, AICAR. AICAR treatment upregulated GPI expression level in C2C12 cell and its secretomes. To confirm the direct effect of GPI on skeletal muscle cells, we treated C2C12 cells with GPI recombinant protein. [Results] We found that GPI improved the viability of C2C12 cells. In the in vivo study, the exercise-treated mice group showed upregulated GPI expression in skeletal muscle. Based on the in vitro study results, we speculated that expression level of GPI in skeletal muscle might be associated with muscle function. We analyzed the association between GPI expression level and the grip strength of the all mice group. The mice group's grip strengths were upregulated after 2 weeks of treadmill exercise, and GPI expression level positively correlated with the grip strength. [Conclusion] These results suggested that the exercise-induced GPI expression in skeletal muscle might have a positive effect on skeletal muscle function.
We have investigated the effects of abiotic and biotic stresses on leaf senescence using transgenic tobacco plants, in which cellular contents of polyamines were increased by introducing the genes of polyamine and ethylene biosynthesis in sense or antisense orientation. These transgenic plants showed accumulations of polyamines at higher levels than were found in wild-type. Stress-induced senescence was attenuated in transgenic plants cpmpared with wild-type plants, in terms of total chlorphyll loss and phenotypic changes after oxidative stress of hydrogen peroxide($H_2O_2$), high salinity, acid stress (pH3.0), ABA and fungal pathogen(phytophothora parasitica pv.Nicotianae). Transcripts for antioxidant enzyme, glutathionine-S-transferase and catalase, were also more abundant in transgenic plants than wild-type plants. These result suggested that higher expression of those genes caused a broad-spectrum resistance to abiotic stress/biotic stress. These phenomena indicate that polyamines may play an important role in contributing to the antioxidant defense function in plants. Our findings suggest that facilitate the improvement of stress tolerance of crop plants.
