Expression of orf8 (chlD) as Glucose-1-Phosphate Thymidylyltransferase Gene Involved in Olivose Biosynthesis from Streptomyces antibioticus Tü99 and Biochemical Properties of the Expressed Protein

  • The orf8(chlD) gene cloned from Streptomyces antibioticus T$\"{u}$99 was overexpressed using an E. coli system to confirm its biological function. Induction of the E. coli strain transformed with recombinant plasmid pRFJ 1031 containing orf8 resulted in the production of a 43,000 dalton protein. Glucose-1-phosphate thymidylyltransferase activity of the cell extract obtained from the transformed strain was 4-5 times higher than that of the control strain. The expressed protein was purified 18-fold from E. coli cell lysate using three chromatographic steps with a 17% overall recovery to near homogeneity. The N-terminal amino acid sequence of the purified protein agrees with the nucleotide sequence predicted from the orf8 gene. The SDS-PAGE estimated subunit mass of 43,000 dalton agrees well with that calculated from the amino acid composition deduced from the nucleotide sequence of the orf8 gene (43,000 Da). Also, the native enzyme has a monomeric structure with a molecular mass of 43,000 dalton. The purified protein showed glucose-1-phosphate thymidylyltransferase activity catalyzing a reversible bimolecular group transfer reaction, and was highly specific for dTTP and ${\alpha}$-D-glucose 1-phosphate as substrates in the forward reaction, and for dTDP-D-glucose and pyrophosphate in the reverse reaction.

  • The waste solution discharged form the LCD(Liquid Crystal Display) manufacturing process contains phosphoric acid, nitric acid, acetic acid and metal ions such Al and other impurities. In this study, vacuum evaporation and diffusion dialysis was developed to commercialize an efficient system for recovering the high-purity phosphoric acid and manufacturing monoammonium phosphate. By vacuum evaporation, almost 99% of nitric and acetic acid was removed. Also, by diffusion dialysis, about 97.5% of Al was removed. Monoammonium phosphate was manufactured from purified phosphoric acid and ammonium hydroxide. In order to get the optimum manufacturing condition, the molar ratio of ammonium hydroxide and phosphoric acid, pH and temperature was controlled. Using this optimum condition, we obtained the recovery rate of monoammonium phosphate of about 90%.

  • Milking parlor wastewater contains high concentration suspended solid (SS), nitrogen, and/or phosphate as well as organic compounds. A new biological wastewater process by magnetic separation, magnetic activated sludge (MAS) process, was applied to milking parlor wastewater treatment process. A three step wastewater treatment process of coagulation sedimentation / ammonia stripping (C/S), magnetic activated sludge process and contact oxidation (CO) was proposed for removal of these pollutants. First step, C/S process recovered 96% TN and 96% PO43--P as resource for fertilizer from the wastewater. 81% biochemical oxygen demand (BOD) in wastewater was removed after MAS process. As a results, all pollutant concentrations satisfied Japanese effluent standards. Most of residual BOD and SS were removed by the CO process. It was estimated that the proposed process could reduce the process space to 1/7.

  • Human Bence Jones Protein could be purified by DEAF-Sephadex A-50 column $(2{\times}37cm)$ with 0.02M phosphate Buffer (pH 8.0) and gradient increasing with NaCl concentration as in Fig. 2-4. Sample As (K-type Bence Jones Protein) had two component, F-I was major component and its dried weight was 350mg. of starting material of 500mg. Other Sample Im and Ik (${\lambda}$-type Bence Jones Protein) was purified by DEAE-Sephadex A-50 with 0.02M phosphate Buffer(pH 8.0)too. F-I (major component) of Im and F-I of Ik were 242mg and 146mg. its dried weight respectively. K-type of Bence Jones Protein's(As, Ko, Ta.) N-terminal amino acid residue was determined by method of DNP,. K-type of Bence Jones Protein's amino acid residue were either glutamic acid or aspartic acid. Sample Ta was confirmed as glutamic acid its N-Terminal. As and Ko were aspartic acid. Each yellowish spot (DNP-amino acids) were extracted with 4ml. of pH 8.05% $NaHCO_3$ solution and calculated its recovery by O.D. $(360m{\mu}$ using the ${\varepsilon}=18.1{\times}10^3DNP$ $Asp\;{\varepsilon}=17.41{\times}10^(3)\;DNP\;Glu$ considering 50% lose during; the acid (6N-HCI) hydrolysis. Recovery of ko and As were 54.3% and 65% of its starting materials (DNP-Protein). Sample Ta's recovery was 85% of its DNP-protein. ${\lambda}$-type of Bence Jones Protein was rot investigated its N-terminal amino acid residue by DNP-method, probably it was blocked its N-terminal residue with glutamic acid.

Enzyme Immunoassay for the Sulfamethazine Residues in Pork Tissue

  • To control the maximum residue level (MRL) for sulfamethazine (SMZ) residues in pork tissue, a microbial inhibition method is a regulatory screening assay method in Korea. Microwell plate-based competitive enzyme immunoassay (ELISA) kit is avalable for routine screening of SMZ residues in pork tissue. One ELISA kit is evaluated. Phosphate buffer extracts of samples fortified with SMZ at 0, 1, 5, and 10 ng/g were used in a recovery test of the kit. Market pork samples were assayed by the kit. Recovery of sulfamethazine was 104% at 10 ng/g. Intraassay variations and interassay variations for the kit were 7.70% and 5.76%, respectively. Concentration causing 50% inhibition of color development compared with blanks was 16.4ng. The violative pork samples with over MRL (0.1 $\mu\textrm{g}$/g) was 4 of 32 cases (12.5%) by used ELISA kit. This result indicates a possibility of the ELISA kit for screening test of SMZ residues in pork tissue, and still needs a comfirmatory assay for mandatory purposes.

  • Cyclodextrin Glucanotransferase(EC 2.4.1.19 : 1,4-${\alpha}$-glucano) transferase, cyclizing; CGTase) can be separated and recovered in an aqueous two-phase system composed of poly(ethylene glycol)(PEG)/dextran and PEG/salt. In an aqueous two-phase system consisting of PEG 35000 (5%) and dextran T2000 (7%), all cell and debris were collected at the interphase. CGTase partitioned to the denser dextran phase at an yield of 83.4%. On the other hand, in an aqueous two-phase system consisting of PEG 35000 (10%) and sodium phosphate (15%), CGTase partitioned to the denser salt phase at an yield of 95.5%. In order to recover CGTase using an aqueous two-phase system, the PEG/salt system proved to be more efficient than the PEG/dextran system in terms of yield and cost.

  • The present study to develop a process for extracting nitric acid and gold from aqua regia leach solution using TBP(tributyl phosphate) was conducted. The pure aqua regia was used to investigate the extractive behavior of nitric acid depending on the concentration of extractant, concentration ratio of nitric and hydrochloric acid. The extraction rate of nitric acid and gold from the gold bearing aqua regia was also examined. The theoretical extraction number was verified by counter current using the number of operations and the phase ratio obtained from McCabe-Thiele diagram. Stripping experiments were carried out for continuous recovery of nitric acid and gold in loaded organic. Considering the effect of extraction acid and gold, the simulation showed that greater than 99.9% extraction of $103.0mg{\cdot}L^{-1}$ gold and 98.0% of $151.2g{\cdot}L^{-1}$ nitric acid could be attained in a two and three-stage counter-current extraction at an O/A phase ratio of 1:0.85. Distilled water and sodium thiosulfate were used as the nitric acid and gold stripping solution. The stripping rates were 99.5% and 92.0%, respectively. The study revealed that the recovery of nitric acid and gold from gold bearing aqua regia was a plausible approach through simultaneous extraction and continuous stripping of nitric acid and gold.

Pre-leaching of Lithium and Individual Separation/Recovery of Phosphorus and Iron from Waste Lithium Iron Phosphate Cathode Materials (폐리튬인산철 양극재로부터 리튬의 선침출 및 인과 철의 개별적 분리 회수 연구)

  • As demand for electric vehicles increases, the market for lithium-ion batteries is also rapidly increasing. The battery life of lithium-ion batteries is limited, so waste lithium-ion batteries are inevitably generated. Accordingly, lithium was selectively preleached from waste lithium iron phosphate (LiFePO4, hereafter referred to as the LFP) cathode material powder among lithium ion batteries, and iron phosphate (FePO4) powder was recovered. The recovered iron phosphate powder was mixed with alkaline sodium carbonate (Na2CO3) powder and heat treated to confirm its crystalline phase. The heat treatment temperature was set as a variable, and then the leaching rate and powder characteristics of each ingredient were compared after water leaching using Di-water. In this study, lithium showed a leaching rate of approximately 100%, and in the case of powder heat-treated at 800 ℃, phosphorus was leached by approximately 99%, and the leaching residue was confirmed to be a single crystal phase of Fe2O3. Therefore, in this study, lithium, phosphorus, and iron components were individually separated and recovered from waste LFP powder.

Contractile Response of Lidocaine-Depressed Isolated Atria in the Absence of Glucose (Lidocaine에 의해 억제된 적출심방의 수축력에 대한 Glucose제거의 영향)

  • The contractility of isolated rat atria, suspended in Krebs-Ringer bicarbonate medium containing 5.5mM glucose, was depressed approximately 50% by 0.1 mM of lidocaine. Partial recovery of the lidocaine-depressed contractility was achieved by the metabolizable substrates pyruvate, acetate, and fructose, but not by addition of glucose. Glucose produced the dose-dependent increase in the force of contraction of normal atria, whereas pyruvate, acetate, and fructose produced no significant effect in the contractile activity of the normal atria. In the absence of exogenous glucose lidocaine produced more marked depression of atrial contractility than that in the presence of exogenous glucose. The results of this study may confirm that the utilization of cardiac glycogen is also inhibited by lidocaine at sites of the glucose phosphate isomerase step or step between glycogen to glucose-6-phosphate.

  • The hypoglycemic and metabolic effects of Commelina communis L. extract were investigated in alloxan-induced diabetic rats. The increased blood glucose level in the diabetic rats was sinificantly lowered with the treatments of the plant protein extract. Administration of the plant extract ellicited the significant increase of glucose-6-phosphate dehydrogenase (G6PD) activity in liver of alloxan-induced rats. Three isozyme patterns(band I, II & III : in order decreasing mobility) of G6PD were found when normal rat liver extract were subjected to electrophoresis on native polyacrylamide gel. On the other hand, G6PD band patterns of alloxan-induced rat liver extract were found band II isozyme missing. By treatment of plant extract in alloxan-induced rats has been showed pattern the recovery of missing band patterns. This indicates that changes of the G6PD isozyme might be related to the cellular process of diabetes.
