The variational characteristics of Water Quality and Chlorophyll a Concentration in Kogum-sudo, Southern Part of Korean Peninsula (거금수도 양식어장의 해양환경 특성 2. 수질과 엽록소 양의 변동특성)

  • Field survey on the variational characteristics of water quality and chlorophyll a concentraion were carried out at the 25 stations for four seasons in Kogum-sudo(Straits) southern coast of Korean Peninsula from Feburuary to October in 1993. We made an analysis on biological factor as chlorophyll a concentraion as well as physicio-chemical factors such as water temperature salinity sigma-t transparency dissolved oxygen(DO) chemical oxygen demand(COD) nutrients (ammonia, nitrite, nitrate, phosphate, and silicate) N/P ratio and suspended solid(SS). The waters in the Kogum-sudo were not stratified due to the tidal mixing. And the high productivity in photic layer were supported by high nutrients concentration from bottom waters. The high concen-trations of suspended solid in straits had a bad influence upon marine biology of nature and cultivations. In Kogum-sudo had a sufficient nutrients for primary productivity during a year. Especially phosphate and inorganic nitrogen were high the other side silicate was very low. The source of nutrients supply depend on rather mineralization of organic matters and input of seawater from outside than input of freshwaters from lands. Phytoplankton biomass as measured by chlorphyll a concentratiion was very high all the year round and it was controlled by the combination o f several environmental factors especially of phosphate in summer and dissolved nitrogen in other seasons.

Effects of Nitrogen Application on Growth and Bioactive Compounds of Chrysanthemum indicum L. (Gamgug) (질소시비가 감국의 생육 및 유효성분에 미치는 영향)

  • To fulfill the increasing demand for a high quality of flower, we investigated the effects of nitrogen application on plant growth, yield and bioactive compounds of Chrysanthemum indicum L.. C. indicum L. was cultivated in a pot scale, and nitrogen applied with the level of 0 (N0), 50 (N50), 100 (N100), 150 (N150), 200 (N200) and $300\;(N300)\;kg\;ha^{-1}$ to suggest optimum rate of nitrogen fertilization. Phosphate and potassium applied the same amount of $80-80\;kg\;ha^{-1}$ ($P_2O_5-K_2O$) in all treatments. Growth characteristics and yields of C. indicum L. were significantly affected by nitrogen application. Maximum yield achieved in 265 and $295\;kg\;ha^{-1}$ N treatment on the whole plant and the flower parts, respectively. The nitrogen content and uptake of whole plant significantly increased by the increase of nitrogen application. Five major components of essential oil, $\alpha$-pinene, 1,8-cineol, chrysanthenone, germacrene-D, and $\alpha$-curcumene in flowerheads of C. indicum L. occupied approximately 40% of peak area, germacrene-D decreased by the increase of nitrogen application among them. However, cumambrin A contents in the flower parts of C. indicum L. were affected negatively by the increase of nitrogen application, but total yields of cumambrin A in flower part significantly increased. Conclusively, nitrogen fertilization could increase the yield of flowerheads. The optimum application level of nitrogen fertilizer might be on the range of $265-295\;kg\;ha^{-1}$ in a mountainous soil.

  • 1. Kum Nam Poong which is highly susceptible to bacterial leaf blight was used as the host plant throughout this experiment. Xanthomonas oryzae 6526 was inoculated on the top of upper leaves by single needle inoculation method. After 14 days, the enlarged spots were examined in the experimental pots. Each of 3 levels of nitrogen, phosphate and potassium pots were arranged at random with three replications. 2. The amounts of nitrogen fertilizer applied and the lesion development of bacterial leaf blight were positively correlated regardless of application of phosphate and potassium fertilizers. 3. The effect of phosphate fertilizer on the lesion development was not significantly different from standard level. The lesion development was stimulated when the amount of phosphate fertilizer was increased as twice as standard level. 4. The inhibitory effect of potassium fertilizer on leaf blight was maximum by applying standard level. The stimulative effect of potassium fertilizer on the lesion development, however, was noticed. when potassium fertilizer applied was increased as twice as the standard level. 5. The heading date and spike number of rice plant were significantly correlated with the lesion development, and such phenomena were depended on the amount of nitrogen fertilizer applied regardless of the other fertilizers applied in this experiment.

Assessments of the Nutrient Losses in the Sloped Farm Land (경사지 밭토양에서의 양분유실량 평가)

  • Nutrient losses, especially nitrogen and phosphorus, in agricultural runoff can contaminate surface and ground water, leading to eutrophication. Thus, erosion control is crucial to minimizing nutrient losses from agricultural land. Assessments of various erosion control practices were carried out under various cropping system, soil management practices, and slope conditions by means of a lysimeter study and under artificial rainfall. Soil and nutrient losses were monitored in a small agricultural field to evaluate the soil conservation practices. Nutrient losses occur in runoff and leachate (dissolved nutrient) and in sediments (particulate nutrient). Dissolved nitrates accounted for the majority (about 90%) of nitrate transport within the soil. Particulate phosphate in sediments represented the majority (60% to 67%) of phosphate transport. Recently, engineering and agronomic erosion-control practices haver been used to reduce erosion problems in fields on slopes. These practices reduced soil loss, runoff, and nutrient loss to 1/6, 1/2,and 1/3 their original levels, respectively. Bioavailable particulate phosphate in sediments represents a variable but longterm source of phosphate for algae. Dissolved nitrate and phosphate are immediately available for algal uptake, so reducing fluxes of these nutrients should also reduce the risk of eutrophication.

Nitrogen and Phosphorus Runoff Loss during Summer Season in Sandy Loam Red Pepper Field as Affected by Different Surface Management Practices in Korea

  • A field study was conducted to determine the runoff loss of N and P in small scale of red pepper field plots (10% slope), consisting of three different plots with black polyethylene vinyl mulching (mulching), ridge without mulching (ridge), and flat without ridge and mulching (flat). Composted manure and urea as a basal application were applied at rates of $20MT\;ha^{-1}$ and $93kg\;N\;ha^{-1}$, respectively. Urea at $189kg\;N\;ha^{-1}$ and fused phosphate at $67kg\;P_2O_5\;ha^{-1}$ were additionally applied on June 25 with different fertilization methods, broadcast application in flat plot and hole injection in ridge and mulching plots. Plant uptake of N and P was positively correlated with their respective concentrations in surface soil: mulching > ridge > flat plots. The runoff loss by soil erosion was higher in flat plot than ridge and mulching plot with contour line. Nitrate loss by the runoff water had no significant differences among three surface management practices, but the higher average value in ridge and mulching plots than flat plot. Especially, the flat plot had no phosphate loss during summer season. This is probably due to low labile P content in surface soil of flat plot. In the summation of soil and water loss, flat plot was higher in N and P loss than ridge and mulching plot with contour line. Nevertheless, the nitrate and phosphate loss by runoff water could be more important for non-point source management because the water could meet the river easier than eroded soil because of re-deposition around slope land.

Feasibility Tests on Struvite Production from Liquid Fertilizer by Utilizing Ferronickel Slag and Sewage Sludge Ash (페로니켈슬래그와 하수슬러지소각재를 이용한 액비로부터 스트루바이트 생산 타당성 연구)

  • Liquid fertilizers made from livestock manure contain high concentrations of nitrogen and phosphorus and thus are used as a fertilizer. However, excessive use of liquid fertilizer causes eutrophication of agricultural land and nonpoint source pollution. In this study, as a means of lowering the nutrient concentrations, struvite ($MgNH_4PO_4{\cdot}6H_2O$) production from the liquid fertilizer was investigated. When liquid fertilizers produced in Gyeonggido were analyzed, its characteristics differed by region and season, but the phosphorus concentration was commonly lower than that of nitrogen. When $K_2HPO_4$ and $MgCl_2$ were added to the liquid fertilizers, the optimal pH for struvite formation was pH 9.5. For environmentally friendly sources of magnesium and phosphate, ferronickel slag (FNS) and sewage sludge ash (SSA) were suspended in deionized water and extracted by sulfuric acid with various mass ratios. The optimum conditions for extracting FNS and SSA were 4.0 M sulfuric acid and 0.35 mass ratio of sulfuric acid to sewage sludge ash, respectively. For forming struvite, 0.233 L of SSA leachate (SSAL) was added into 0.3 L of liquid fertilizer containing 2,586 mg/L of ammonia and 110 mg/L of phosphate, pH was then adjusted to pH 9.5 using 10 M of NaOH. Afterwards 0.333 L of FNS leachate (FNSL) was added to this mixed solution. After a reaction for 1 hr at room temperature, the remaining concentrations of magnesium, ammonium, and phosphate were less than 50 mg/L, 500 mg/L and 150 mg/L, respectively, and 30 g of precipitates were obtained, most of which were struvite.

Fertilization Efficiency of Livestock Manure Composts as Compared to Chemical Fertilizers for Paddy Rice Cultivation

  • To promote the practical use of livestock manure compost (LC) for paddy rice cultivation, the fertilization efficiency of nutrients in LCs was investigated compared to that of chemical fertilizer. This experiment was conducted at rice field in Hwaseong, Korea, with 6 treatments by each of 3 kinds of tested LCs, cattle manure compost (CaC), swine manure compost (SwC) and chicken manure compost (ChC). The treatments consisted of 3 application levels of LCs and 3 chemical fertilizer treatments having the same application levels with LCs. $NH_4$-N content in soil became higher according to the increase in the urea application rate, while it became lower in LC plots than in urea plots, and statistically had no significant difference among LC plots. There was a close relationship between phosphate fertilization rate and the increment of soil available phosphate content after experiment resulting y = 0.1788x - 6.169 ($R^2=0.9425$) when applied fused superphosphate fertilizer, and y = 0.0662x - 2.689 ($R^2=0.9315$) when applied LC at the equivalent rates to phosphate input (x: phosphate application rate, kg $ha^{-1}$, y: increment in soil available phosphate content, mg $kg^{-1}$). And from these two equations, the correlation on the phosphate application rate between fused superphosphate fertilizer and LC could be obtained as y = 2.7056x - 52.492 (x: $P_2O_5$ application rate of fused superphosphate, kg $ha^{-1}$, y: $P_2O_5$ application rate of LC, kg $ha^{-1}$). Plant height, number of tillers, nutrients uptake by rice, and rice yield showed higher levels in N 100% and N 150% application plots of chemical fertilizers, while every LC plots exhibited lower values and no significant difference among them. Relative nitrogen fertilization efficiencies of LCs compared to urea was 12.3% for CaC, 8.8 for SwC and 24.6 for ChC, respectively.

Effects of Amino Acids, Carbohydrates and Phosphorus Sources on Growth and Alkaline Phosphatase Activity of the Marine Cyanobacterium Anabaena sp. Strain CA

  • Alkaline phosphatase (APase) was found to be inducible in Anabaena sp. strain CA Growth was less than control in presence of most amino acids except glycine and serine, but most amino acids enhanced APase activity. Highest APase activity was recorded in tyrosine supplemented culture followed by hydroxyproline, cystein, valine and glutamic acid. Threonine supplemented material showed lowest APase level (1.8 nmol/mg protein/min). Lactose, glucose, sodium pyruvate and succinate stimulated growth but not APase activity. APase activity was high in the presence of sucrose, mellibiose, mannitol, arabinose, maltose and sorbose, even though the growth in these supplements was less than in control. Organic phosphate sources supported good growth of the organism. Best growth occurred in presence of inorganic phosphate, adenosine diphosphate, fructose 1,6-diphosphate or ribulose 1,5-diphosphate, followed by other phosphorus sources tested. APase activity in presence of any of the organic phosphate sources was 3 to 5 fold low as compared to phosphate limited culture. Also, there was no APase activity in cultures grown on inorganic phosphate. These data indicate that most amino acids and a few carbohydrates (sucrose, mellibiose, arabinose and sorbose) are suitable for APase production. Lactose, glucose, pyruvate or succinate may be used as a carbon source during photoheterotrophic growth of the cyanobacterium. Glycine and serine are preferred nitrogen sources for its growth. Phosphate repressible APase activity has been found in Anabaena sp. strain CA.

A Study on the Removal of Nitrogen and Phosphorus by Addition of Coagulant in the Sulfur Denitrification Process Coupled to the Membrane Bioreactor (MBR과 황탈질 공정에서 응집제 주입에 따른 질소.인 제거에 관한 연구)

  • A method for simultaneous removal of nitrogen and phosphate from sewage by elemental sulfur denitrification with membrane bioreactor was proposed, and capacity $10\;m^3$/day of pilot plant was operated for 350 days. This study was investigated to have the effect of denitrification rate and T-P removal with the addition of Alum in Sulfur denitrification Reactor (SDR). The addition of Alum and alkalinity ($NaHCO_3$) in the effluent of MBR was tried to remove simultaneous phosphate and nitrogen in SDR. Characteristics of total nitrogen (T-N) and total phosphate (T-P) removal was compared without and with the addition of Alum as a coagulant. T-N removal without and with the addition of Alum was 92.1% and 87.8%, respectively. And denitrification efficiency was 93.8% and 87.1%, respectively. T-P removal rate was increased to 75.6% in SDR by addition of Alum (2.6~4 mg/L as Al), but T-P removal rate was about 26.7% without the addition of Alum. Therefore, denitrification rate was 6.7% of reduction but T-P removal rate was increased by addition of Alum.

Effects of Dissolved Inorganic and Organic Nutrient (Nitrogen and Phosphorus) on the Growth of Dinoflagellate Alexandrium affine (와편모조류 Alexandrium affine의 생장에 미치는 용존태 무기 및 유기 영양염(질소와 인)의 영향)

  • In this study, we investigated the effects of dissolved inorganic and organic nutrient on the growth of dinoflagellate Alexandrium affine (LIMS-PS-2345). The maximum uptake rates (ρmax) and half saturation constants (Ks) calculated from the uptake kinetics experiment were 77.0 pmol/cell/hr, 17.6 μM for nitrate and 15.5 pmol/cell/hr, 3.88 μM for phosphate, respectively. These results suggested that this species has high inorganic nutrient demand and a low affinity for inorganic nutrients. During the utilization of organic nutrients for A. affine, growth rates of experimental groups added by organic nitrogen (urea and glycine) and phosphorus (adenosine triphosphate and glycerol phosphate) were above 70 %, compared to the experimental groups added by inorganic nutrients. Thus, A. affine may need to utilize organic nutrients to understand the dominant strategy and advantageous position in the interspecific competition within low inorganic nutrient environments.
