We investigated the leaf demography of a temperate woody liana, Akebia trifoliata, in a temperate forest in Japan, Akebia is semi-evergreen: some leaves are shed before winter, while others remain through the winter. Previous studies of semi-evergreen species found that variation in leaf life span was caused by variation in the timing of leaf emergence, Leaves that appeared just before winter over-wintered, while leaves appearing earlier were shed, However, it is unclear whether leaves of the same cohort (i.e., leaves that appear at the same time within a single site) show variation in life span under the effect of strong seasonality. To separate variation in life span among the leaves in each cohort from variation among cohorts, we propose a new method - the single leaf diagram, which shows the emergence and death of each leaf. Using single leaf diagrams, our study revealed that Akebia leaves within a cohort showed substantial variation in life span, with some over-wintering and some not. In addition, leaves on small ramets in the understory showed great variation in life span, while leaves on large ramets, which typically reach higher positions in the forest canopy, have shorter lives, As a result, small ramets were semi-evergreen, whereas large ramets were deciduous, The longer lives of leaves on small ramets can be interpreted as a shade-adaptive strategy in understory plants.

Many freshwater snail taxa are difficult to identify using morphological traits due to phenotypic plasticity. However, using of molecular DNA marker in combination with morphological traits can provide a reliable means for discriminating among freshwater snail taxa including cryptic species. To discriminate among Korean freshwater snail taxa and resolve their systematic relationships, wesequenced a fragment of mtDNA cytochrome oxidase I (COI) gene from 82 specimens collected from ten different sites distributed along the Korean peninsula. We identified more than seven freshwater snail taxa including cryptic species in Korea. Whereas traditional shell morphology of freshwater snails offers only weak discriminatory power for recognizing 'good' taxa, DNA sequence data provided positive and reliable identification. In addition, a major Semisulcospira clade was clearly separated from the remaining lineages observed including cryptic species. However, a phylogenetic tree inferred from the COI gene data did not fully resolve systematic relationships among pleurocerid taxa in Korea. Establishing more robust shell characteristics for identifying taxa unambiguously and hence improving traditional key shell morphology characters for freshwater snail species is an urgent requirement and will require more rigorous examination of all nominal taxa. While molecular data generated here will be useful for species identification and for describing the systematic relationships among Korean freshwater snails, further analysis will be required.

Green microalgae from the class Chlorophyceae represent a major biodiversity component of eukaryotic algae in continental water. Identification and classification of this group through morphology is a hard task, since it may present cryptic species and phenotypic plasticity. Despite the increasing use of molecular methods for identification of microorganisms, no single standard barcode marker is yet established for this important group of green microalgae. Some available studies present results with a limited number of chlorophycean genera or using markers that require many different primers for different groups within the class. Thus, we aimed to find a single marker easily amplified and with wide coverage within Chlorophyceae using only one pair of primers. Here, we tested the universality of primers for different genes (tufA, ITS, rbcL, and UCP4) in 22 strains, comprising 18 different species from different orders of Chlorophyceae. The ITS primers sequenced only 3 strains and the UCP primer failed to amplify any strain. We tested two pairs of primers for rbcL and the best pair provided sequences for 10 strains whereas the second one provided sequences for only 7 strains. The pair of primers for the tufA gene presented good results for Chlorophyceae, successfully sequencing 21 strains and recovering the expected phylogeny relationships within the class. Thus, the tufA marker stands out as a good choice to be used as molecular marker for the class.

Distinguishing individual Russula species has been difficult due to extensive phenotypic plasticity and obscure morphological and anatomical discontinuities. Due to highly similar macroscopic features, such as the presence of a red-cap, species identification within the Russula subgenus Amoenula is particularly difficult. Three species of the subgenus Amoneula have been reported in Korea. We used a combination of morphology and three molecular markers, the internal transcribed spacer (ITS), 28S nuclear ribosomal large subunit (LSU), and RNA polymerase II gene (RPB2), for identification and study of the genetic diversity of Russula subgenus Amoenula in Korea. We identified only two species in Korea (R. mariae and R. violeipes); these two species were indistinguishable according to morphology and LSU, but were found to be reciprocally monophyletic species using ITS and RPB2. The markers, ITS, LSU, and RPB2, have been tested in the past for use as DNA barcoding markers, and findings of our study suggest that ITS and RPB2 had the best performance for the Russula subgenus Amoneula.

Background and objective: Native to the limestone zones of the Korean Peninsula, Zabelia tyaihyonii is a popular plant for landscaping. As it is now classified as a rare species, the conservation of its genetic resources is necessary. Methods: In this study, which aimed to understand the morphological variation of Z. tyaihyonii, 18 characteristics of Z. tyaihyonii from five habitats were examined. Results: Of these 18 characteristics, 16 characteristics showed significant differences among sites, and the coefficient of variation ranged from 5.4% (for corolla lobe number) to 31.3% (for flower number). Notable variations were observed in the size of flower and calyx lobe. When the corolla length and calyx lobe length were used as the classification key of Z. tyaihyonii, the sites were divided into those with small, intermediate, and large values. Hair was observed on the filament of all samples, a finding which conflicts with an earlier report. Rather than classifying Z. tyaihyonii into different species on the basis of corolla length (COL) and calyx lobe length (CALL) values, we recommend modifying the species description to incorporate the variation in these characteristics of interest. Principal component analysis results showed that the first main component was highly correlated with the traits related to the size of the calyx lobe (length: 0.819, width: 0.758), and the second main component was highly correlated with the traits related with the size of the inflorescence (length: 0.790, width: 0.626). Conclusion: Several notable variations were identified among the characteristics related to inflorescence and calyx lobe. There is little genetic exchange among groups, or each group is influenced by micro environmental factors, because sites that are located nearby. In addition, the difference between COL and CALL, which is used as the classification key for Z. tyaihyonii, was divided into small group, large group, and intermediate group, regardless of the sites' geographical distance.

The genera Carolibrandtia and Chlorella have been described as small green algae with spherical cell shapes that inhabit various environments. Species of these genera are often difficult to identify because of their simple morphology and high phenotypic plasticity. We investigated two small coccoid strains from Antarctica based on morphology, molecular phylogeny by two alignment methods which have been applied to previous phylogenetic studies of the genus Chlorella, and comparison of the secondary structures of nuclear small subunit (SSU) and internal transcribed spacer (ITS) rDNA sequences. Light microscopy of two strains revealed spherical cells containing chloroplasts with pyrenoids, and the morphological characteristics of the strains were nearly identical to those of other Chlorella species. However, based on the phylogenetic analyses of nuclear SSU and ITS rDNA sequences, it was determined that the Antarctic microalgal strains belonged to two genera, as the Chlorella and Carolibrandtia. In addition, the secondary structures of the SSU and ITS2 sequences were analyzed to detect compensatory base changes (CBCs) that were used to identify and describe the two strains. A unique CBC in the SSU rDNA gene was decisive for distinguishing strain CCAP 211/45. The ITS2 rDNA sequences for each strain were compared to those obtained previously from other closely related species. Following the comparison of morphological and molecular characteristics, we propose KSF0092 as a new species, Chlorella terrestris sp. nov., and the reassignment of the strain Chlorella antarctica CCAP 211/45 into Carolibrandtia antarctica comb. nov.

Identification of Helicotylenchus species is very challenging due to phenotypic plasticity and existence of cryptic species complexes. Recently, the use of rDNA barcodes has proven to be useful for identification of Helicotylenchus. Molecular markers are a quick diagnostic tool and are crucial for discriminating related species and resolving cryptic species complexes within this speciose genus. However, DNA barcoding is not an error-free approach. The public databases appear to be marred by incorrect sequences, arising from sequencing errors, mislabeling, and misidentifications. Herein, we provide a comprehensive analysis of the newly obtained, and published DNA sequences of Helicotylenchus, revealing the potential faults in the available DNA barcodes. A total of 97 sequences (25 nearly full-length 18S-rRNA, 12 partial 28S-rRNA, 16 partial internal transcribed spacer [ITS]-rRNA, and 44 partial cytochrome c oxidase subunit I [COI] gene sequences) were newly obtained in the present study. Phylogenetic relationships between species are given as inferred from the analyses of 103 sequences of 18S-rRNA, 469 sequences of 28S-rRNA, 183 sequences of ITS-rRNA, and 63 sequences of COI. Remarks on suggested corrections of published accessions in GenBank database are given. Additionally, COI gene sequences of H. dihystera, H. asiaticus and the contentious H. microlobus are provided herein for the first time. Similar to rDNA gene analyses, the COI sequences support the genetic distinctness and validity of H. microlobus. DNA barcodes from type material are needed for resolving the taxonomic status of the unresolved taxonomic groups within the genus.
Cymbidium kanran Makino is being threatened in its own habitats due illegal collecting and habitat changes by vegetation growth along historical landuse change. In this study, we established habitat restoration model for conservation of C. kanran based on ecological diagnosis. Through exploration to Jeju Island in 2014 and 2015, we identified 27 unknown habitats of C. kanran and in there, abiotic variables and vegetation structure and composition were quantified. Altitudinal distribution of C. kanran was between 200 m~700 m a.s.l. and compared to distribution in 2004, Area of Occupation (AOO) decreased at 82%. Specific habitat affinity was not observed by evenly found in mountain slope and valley and summergreen and evergreen broadleaved forests, but likely more abundant in valley habitats with higher soil and ambient moisture. Total of 96 individual of C. kanran was observed with an average density of $942.6individuals\;ha^{-1}$. The plants showed relatively short leaf length (average=$10.7cm{\pm}1.1cm$) and small number of pseudo bulbs ($1.2{\pm}0.2$). Flowering and fruiting individuals were not observed in field. C. kanran was classified into endangered plant species as CR (Critically Endangered) category by IUCN criteria. Phenotypic plasticity of C. kanran was likely support to sustain in more shaded habitat environment and recent habatat changes to closed canopy and low light availability may exhibit negatively effects to C. kanran's life history. Restoring C. kanran habitat should create open environment as grassland and low woody species density.

In recent years, macroalgal bloom occurs frequently in coastal oceans worldwide. It might be attributed to accelerating climate change. "Green tide" events caused by proliferation of green macroalgae (Ulva spp.) not only damage the local economy, but also harm coastal environments. These nuisance events have become common across several coastal regions of continents. In Korea, green tide incidences are readily seen throughout the year along the coastlines of Jeju Island, particularly the northeastern coast, since the 2000s. Ulva species are notorious to be difficult for morphology-based species identification due to their high degrees of phenotypic plasticity. In this study, to investigate temporal variation in Ulva community structure on Jeju Island between 2015 and 2020, chloroplast barcode tufA gene was sequenced and phylogenetically analyzed for 152 specimens from 24 sites. We found that Ulva ohnoi and Ulva pertusa known to be originated from subtropical regions were the most predominant all year round, suggesting that these two species contributed the most to local green tides in this region. While U. pertusa was relatively stable in frequency during 2015 to 2020, U. ohnoi increased 16% in frequency in 2020 (36.84%), which might be associated with rising sea surface temperature from which U. ohnoi could benefit. Two species (Ulva flexuosa, Ulva procera) of origins of Europe should be continuously monitored. The findings of this study provide valuable information and molecular genetic data of genus Ulva occurring in southern coasts of Korea, which will help mitigate negative influences of green tide events on Korea coast.
Studies on molecular plasticity of Bermann glia (BG) after harmaline-induced Purkinje cell (PC) degeneration in the rat cerebellum. The intimate structural relationship between BG and PC, evidenced by the sheathing of the PC dendrites by veil-like process from the BG has been suggestive of the close functional relationship between these two cell types. However, little is known about metabolic couplings between these cells. This study designed to investigate molecular plasticity of BG in the rat cerebellum in which PCs were chemically ablated by harmaline treatment. Immunohistochemical examination reveals that harmaline induced PC degeneration causes a marked glial reaction in the cerebellum with activated BG and microglia aligned in parasagittal stripes within the vermis. In these strips, activated BG were associated with upregulaion of metallotheionein, while GLAST and was down regulated, as compared with nearby intact area where both BG are in contact with PCs. The data from this study demonstrate that BG can change their phenotypic expression when BG loose their contact with PCs. It is conceivable that activated BG may upregulate structural proteins, metallothionein expression to use for their proliferation and hypertrophy; metallothionein expression to cope with oxidative stress induced by PC degeneration and microglial activation. On the contrary, BG may down regulated expression of GLAST because sustained loss of contact with PCs would eliminate the necessity for the cellular machinery involved glutamate metabolism. In conclusion, BG might respond man to death of PCs by undergoing a change in metabolic state. It seems possible that signaling molecules released from PCs regulates the phenotype expression of BG. Also ultrastructures in the organelles of normal PC and BG are distinguished by mitochondrial appearance, and distributed vesicles at the synaptic area in the cytoplasm.
