A tyrosinase-immobilized biosensor was developed based on various amine-modified multi-walled carbon nanotube (MWNT) supports for the detection of phenolic compounds. MWNTs with various amine groups were prepared by radiation-induced graft polymerization of glycidyl methacrylate (GMA) onto MWNT supports and the subsequent amination of poly(GMA) graft chains. The physical and chemical properties of the poly(GMA)-grafted MWNT supports and the aminated MWNT supports were investigated by SEM, XPS, and TGA. Furthermore, the electrochemical properties of the prepared tyrosinase-modified biosensor based on MWNT supports with amine groups were also investigated. The response of the enzymatic biosensor was in the range of 0.1-0.9 mM for the concentration of phenol in a phosphate buffer solution. Various parameters influencing biosensor performance have been optimized: binder effects, pH, temperature, and the response to various phenolic compounds. The biosensor was tested on phenolic compounds contained in two different commercial red wines.
Lipase-producing fungi have been isolated from environments containing lipids. The non-dairy creamer industrial waste has a high amount of lipids so it is a potential source for the isolation of lipase-producing fungi. However, the study of fungi that secrete lipase from this industrial waste has not been reported. The purpose of this study was to obtain lipase-producing filamentous fungi from non-dairy creamer industrial waste. Mineral salt and potato dextrose agar were used as media for the isolation process. The qualitative screening was conducted using phenol red agar medium and the quantitative screening using broth medium containing glucose and olive oil. Isolates producing the highest amounts of lipase were identified with molecular methods. We found that 5 out of 19 isolated filamentous fungi are lipase producers. Further analysis showed that isolate Ms.11 produced the highest amount of lipase compared to others. Based on ITS sequence Ms.11 was identified as Aspergillus aculeatus. The lipase activity in medium containing 1% glucose + 1% olive oil at pH 7.0 and 30℃ after 96 and 120 h of incubation was 5.13 ± 0.30 U/ml and 5.22 ± 0.59 U/ml, respectively. The optimum lipase activity was found at pH 7.0, 30℃ and using methanol or ethanol in the reaction tube. Lipase was more stable at 20-30℃ and maintained 85% of its activity. It was concluded that isolate Ms.11 is a potential source of lipase that catalyzes transesterification reactions. Further studies are required to optimize lipase production to make the strain suitable for industry purposes.
A multi-walled carbon nanotube (MWNT) support with dual properties, an ionic property via tetra-amine and unpaired electrons via tri-amine, was prepared by radiation-induced graft polymerization of glycidyl methacrylate (GMA) and the subsequent amination of its epoxy group. The electrochemical microbial biosensor (EMB) was then fabricated by immobilization of a microbe (Alkaligenes spp.) onto the dual property-modified electrode, which was prepared with the mixture of the MWNT support and a $Nafion^{(R)}$ solution on a glass carbon (GC) electrode surface by a hand-casting method. The sensing range of the prepared EMB for phenol in a phosphate buffer solution was 0.005~7.0 mM. The total concentration of phenolic compounds in a commercial red wine was also determined using the EMB.
The objective of this study was to investigate the contribution of various smoke constituents to the toxicological activity of total particulate matter(TPM) or the gas/vapor phase(GVP). These components included phenol compounds, aromatic amines, polycyclic aromatic hydrocarbons, heterocyclic amines, and carbonyl compounds. The mutagenic and cytotoxic potencies were assessed using the Salmonella mutagenicity assay with S. typimurium TA98 strain and the neutral red uptake cytotoxicity assay(NRU) with BALB/c 3T3 fibroblast cells, respectively. The Salmonella mutagenicity test showed that heterocyclic amines exhibited significantly higher levels of toxicity compared to other smoke constituents. Among them, 2-amino-3,4-dimethylimidazo[4,5-f]quinoline(MeIQ) was shown the most mutagenic compound with a specific mutagenicity of $7.9{\times}10^5\;revertants/{\mu}g$. An analysis of the possible contribution revealed that MeIQ account for only 0.85% of the 2R4F-TPM mutagenicity in TA98. NRU data demonstrated that high cytotoxic activity was obtained for hydroquinone, formaldehyde, and acrolein. Based on the results of the present study, the contribution of acrolein to the cytotoxicity of the GVP fraction was calculated as 61%. Thus, a large proportion of the cytotoxic activity of this complex mixture, cigarette smoke gas phase, can be attributed to the acrolein.

Red pepper (Capsicum annuum L.) is one of the most consumed vegetable worldwide. In this study, we tried to suggest the possibility of Cheongyang pepper as a functional cosmetic material by identifying the physiological activity, especially antioxidant and whitening effects of Cheongyang pepper through bibliometric analysis and experimental studies. A bibliometric analysis was performed through co-word analysis of 8,892 papers retrieved from SCOPUS. 4 research fields were obtained by cluster mapping from VOSviewer software, among which we noted the antioxidant activity of extracts from Capsicum annum L.. Phenol as a useful ingredient of Cheongyang pepper was analyzed using HPLC (high-performance liquid chromatography). Antioxidant and whitening effects were evaluated by measuring DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, hydrogen peroxide scavenging activity, and tyrosinase inhibitory activity. Cheongyang pepper extract showed contents of 0.106 ± 0.01 and 0.105 ± 0.02 mg/g, respectively, in the order of gallic acid and protocatechuic acid. The extract exhibited 56.95% DPPH scavenging activity and 43.97% hydrogen peroxide scavenging activity at a concentration of 1,000 ㎍/ml. In addition, 1,000 ㎍/ml of the extract inhibited tyrosinase activity by 52.44% and 42.61%, respectively in a whitening efficacy test using L-tyrosine and L-DOPA (L-3,4- dihydroxyphenylalanine) as substrates. These results suggest that Cheongyang pepper extract and its active ingredients have antioxidant and whitening effects, and the possibility of future development as a whitening functional cosmetic material.
To evaluate the adhesive characteristics of mixed cross-laminated timber (CLT) using domestic softwoods structural lumber proposed by KS F 3020 and yellow poplar, penetration depth of adhesive and thickness of bonding line were analyzed based on the results of boiling water soaking delamination. 3 Types of adhesives and 2 types of major layer were divided into a 5 ply CLT using yellow popular as minor layer. The bonding performance of the mixed CLT as structural members was evaluated based on the KS F 2081. The thickness of bonding line between layers of the mixed CLT was measured with a scanning electron microscope, and the adhesive penetration depth in the layer members was measured with an optical microscope. As a result of boiling water soaking delamination test of mixed CLT, the CLT specimens using PRF and PUR adhesives met the requirements of KS F 2081. It was verified that the penetration path of the adhesive in the layes was mainly through the tracheid cell in the case of Japanese larch and Korean red pine layers, and through the vessel and radial tissue in yellow popular layers. The penetration depth of the adhesive was the highest for the PRF adhesive under the same pressing time conditions, and the thickness of the bonding line was in inverse proportion to the penetration depth in the case of the PUR adhesive.
The domestic cultured Campbell's Early and Geubong grapes were fermented far the production of red wines with the isolated wild yeast Saccharomyces cerevisiae IJ850. For the isolation of wild yeast, Geubong and Campbell's Early grapejuices were naturally fermented at room temperature for 6 days without adding stater culture. The strain isolated from Geubong which has 1.8 times higher fermentative ability than the strains isolated Campbell Early was selected. The selected strain was identified by using 26S rDNA sequencing. The strain showed 99.7% of similarity with Saccharomyces cerevisiae and thus identified as Saccharomyces cerevisiae IJ850. It was investigated the fermentative ability as the start culture. For the production of grapewine, the final sugar concentrations of grapejuices were adjusted to the $25^{\circ}Brix$ with anhydrous glucose. The grapejuices were fermented at room temperature for 10 days in the air-locked bottles filled with $CO_2$ gas. The final yield and alcohol concentration of Campbell's Early and Geubong grapewines fermented with the isolated wild yeast were 80.8%, 11.0% and 87.8%, 13.0%, respectively. Between the isolated wild yeast S. cerevisiae IJ850 and the commercial yeast S. cerevisiae EC1118, total acidities of grapewines produced with wild yeast were lower than those produced with the commercial yeast. The pH values and the values of color analysis of grapewines produced with both strains were similar. The total phenol contents of campbell's Early and Geubong wines produced with the isolated yeast and the commercial yeast were obtained in the range of 75 to 125mg/L. In conclusion, S. cerevesiae IJ850 isolated from the domestic cultured Geubong grape is able to use to produce grapewines as stater culture.
A study was conducted to examine the effects of light quality on the growth and phytochemical contents of ice plant in a closed-type plant production system. Seeds were sown in a 128-cell plug tray using rockwool. The seedlings were then transplanted into a deep floating technique system with recirculating nutrient solution (EC $1.5dS{\cdot}m^{-1}$, pH 6.5) in a closed-type plant production system. The nutrient solution was supplied at two weeks after transplanting with 2.0 mM NaCl concentration in all treatments for the development of the bladder cells. The three light sources with different light qualities used were as followed; FL (fluorescent lamps), combined RW LED (red:white = 7:3), and combined RBW LED (red:blue:white = 8:1:1) at $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD with a photoperiod of 14/10 hours (light/dark). The results showed that the FL treatment had the greatest growth enhancement effects on the leaf area and the fresh and dry weights of the shoots and roots. The SPAD values were significantly higher under the FL and RBW LED treatments, at 29.8 and 30.6, respectively. No significant difference was observed in salinity under all treatments. Chlorophyll fluorescence was significantly higher under the FL treatment. The total phenol content and antioxidant activity were the highest under the RBW LED treatment. The total flavonoid content was significantly higher under the RBW LED and FL treatments. Hence, the results indicate that the growth of ice plant was maximized under the FL treatment. The phytochemical contents were maximized under the RBW LED treatment.

The food functional properties of Pleurotus eryngii cultivated with different wavelengths of light-emitting diode (LED) light were analyzed in this study. The levels of total soluble solids of Pleurotus eryngii increased with all the LED lights, except mixed LED light ($B^*R$, $B^*G$ and $R^*G$). Thirty one kinds of components were detected by analyzing the free amino acids. The total free amino acid contents had a slightly higher tendency in the control group, but the ratios of eight essential amino acids among the total amino acids of Pleurotus eryngii cultivated under all the LED lights, including the blue light (blue, $B^*R$, $B^*G$ and $B^*R^*G^*U$) and the green light, were higher than that in the control group. K was the highest-content of mineral, and Mn was the lowest-content. The Ca content increased through cultivation under all the LED lights, except the blue light, but the Fe content (under the green and $B^*R$ lights), K content (under the red and UV-A lights) and Mg content (under the $R^*G$, UV-A and red lights) increased under the exclusive LED light. The total phenolic compounds increased by cultivation under all the LED lights, except the UV-A and mixed lights ($R^*G$ and $B^*R^*G^*U$). The DPPH radical scavenging activity was also improved by all the LED lights, except the mixed light ($R^*G$).

Objectives: This study is to confirm the effect of combined extract and individual extract of Samjunghwan (SJH) in anti-oxidative and anti-obesity effect. Methods: Combined ethanol extract of readily made SJH and individual ethanol extract of Atractylodes japonica, Cortex lycii radicis, and Morus alba Linne was combined after the extraction. To evaluate the anti-oxidative effect of SJH, total phenol compound and 2,2-Diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging ability were conducted. Real-time quantitative-polymerase chain reaction analysis of transcription factor peroxisome proliferator-activated receptror ${\gamma}$ ($PPAR{\gamma}$), adenosine monophosphate-activated protein kinase (AMPK)-${\alpha}1$, tumor necrosis factor-${\alpha}$ ($TNF{\alpha}$) and 3-hydroxy-3-methylglutaryl CoA reducatase (HMG-CoA reductase) were done with 3T3-L1 cells to investigate the ant-obesity effect. Also, cell viability analysis were done to see to toxicity of SJH. Results: Individual extract of SJH showed significant decrease in $TNF{\alpha}$ and AMPK transcription while $PPAR{\gamma}$ showed significant increase. Combined extract and individual extract of SJH both showed decrease in HMG-CoA reductase. DPPH free radical scavenging ability and total phenol compound was analogous between two groups. Conclusions: Individual extract of SJH appears to be more effective in anti-oxidation and anti-obesity effect compared to combined extract of SJH.
