Fluctuation Characteristics of Seawater Temperature Identified by Time Series Analysis Off the Southern Coast of Korea (한국 남해안에서 시계열 분석을 통한 수온 변동 특성)

  • In order to understand the fluctuation characteristics of water temperature in August, water temperature, tide level, air temperature and wind data were analyzed using a power spectrum and coherence analysis for a region off the southern coast of Korea. Spectrum analysis showed that the water temperature and tide level in Wando, Goheung, Yeosu, Tongyeong and Masan have peaks over a semi-diurnal period. Coherence showed that water temperature was affected by tide in Wando, Goheung, Yeosu and Tongyeong where tidal range is relatively high. In Masan and Busan, where tidal range is relatively low, however, the tide did not affect water temperature significantly. In particular, wind was the most important factor in relation to water temperature in Masan. Time lags were calculated from phase. In Wando, water temperature decreased 1.5 hrs after a flood tide began. Water temperature decreased 0.3 hrs after a flood tide began in Goheung and Tongyeong. In Masan, water temperature increased 3 hrs after a south wind began. Water temperature in Yeosu was affected by tide, but air temperature, tide and wind in Busan affected water temperature very little.

Variation in Fatty Acid Composition, Caffeic and Rosmarinic Acid Content, and Antioxidant Activity of Perilla Accessions (들깨 유전자원의 지방산 변이 및 rosmarinic acid와 caffeic acid의 함량에 따른 항산화 활성 비교)

  • Background: The aim of this study was to compare the fatty acid composition, antioxidant activity, and the content of two major phenolic acids, caffeic and rosmarinic acids, of 45 Perilla accessions collected from Russia. Methods and Results: A total of 45 accessions of the genus Perilla were used in this study. The antioxidant activities of these accessions were analyzed using a spectrophotometer, and their caffeic and rosmarinic acid contents were determined using a reversed-phase ultraperformance liquid chromatography (UPLC) system. The Perilla seed oil was recovered using hexane in a soxhlet extraction method. The fatty acid compositions were analyzed using a Shimadzu QP2010 gas chromatography-mass spectoroscopy system. The results showed that accessions IT226732 and IT274300 had the highest content of caffeic and rosmarinic acid, respectively. A cluster analysis based on antioxidant assay results and concentration of phenolic acids led to the categorization of Perilla accessions into three major groups. The accessions in Group I were characterized by higher caffeic and rosmarinic acid content and antioxidant activity, compared to the accessions in the other two groups (p < 0.05). The total oil content ranged between 28.39 and 46.89%. The compositions of oleic, linoleic, and linolenic acid ranged from 11.83 to 19.55%, 11.92 to 16.71%, and 59.19 to 67.28%, respectively. Conclusions: The results of this study indicated that accessions IT274300, IT226732, IT274293, IT235818, and IT235820 could be used as a source of functional materials.

Development of Individual Residue Analysis Method for Cyanazine in Agricultural Commodities as an Unregistered Herbicide in Korea (국내 미등록 제초제 cyanazine의 농산물 중 개별 잔류분석법 개발)

  • Cyanazine is a member of the triazine family of herbicides. Cyanazine is used as a pre- and post-emergence herbicide for the control of annual grasses and broadleaf weeds. This experiment was conducted to establish a determination method for cyanazine, as domestic unregistered pesticide, residue in major agricultural commodities using HPLC-DAD/MS. Cyanazine was extracted with acetone from representative samples of five raw products which comprised apple, green pepper, Kimchi cabbage, hulled rice and soybean. The extract was diluted with saline water and partitioned to dichloromethane for remove polar extractive in the aqueous phase. For the hulled rice and soybean samples, n-hexane/acetonitrile partition was additionally employed to remove non-polar lipids. The extract was finally purified by optimized florisil column chromatography. On a $C_{18}$ column in HPLC, cyanazine was successfully separated from co-extractives of sample, and sensitively quantitated by diode array detection at 220 nm. Accuracy and precision of the proposed method was validated by the recovery experiment on every major agricultural commodity samples fortified with cyanazine at 3 concentration levels per agricultural commodity in each triplication. Mean recoveries were ranged from 83.6 to 93.3% in five major representative agricultural commodities. The coefficients of variation were all less than 10%, irrespective of sample types and fortification levels. Limit of quantitation(LOQ) of cyanazine was 0.02 mg/kg as verified by the recovery experiment. A confirmatory method using LC/MS with selected-ion monitoring(SIM) technique was also provided to clearly identify the suspected residue.

Development of Analytical methods for Chinomethionat in Livestock Products (축산물 중 살균제 Chinomethionat의 개별 잔류분석법 확립)

  • BACKGROUND: The analytical method was established for determination of fungicide chinomethionat in several animal commodities using gas chromatography (GC) coupled with electron capture detector (ECD). METHODS AND RESULTS: In order to verify the applicability, the method was optimized for determining chinomethonat in various livestock products including beef, pork, chicken, milk and egg. Chinomethionat residual was extracted using acetone/dichloromethane(9/1, v/v) with magnesium sulfate and sodium chloride (salting outassociated liquid-liquid extraction). The extract was diluted by direct partitioning into dichloromethane to remove polar co-extractives in the aqueous phase. The extract was finally purified with optimized silica gel 10 g. CONCLUSION: The method limit of quantitation (MLOQ) was 0.02 mg/kg, which was in accordance with the maximum residue level (MRL) of chinomathionate as 0.05 mg/kg in livestock product. Recovery tests were carried out at two levels of concentration (MLOQ, 10 MLOQ) and resulted in good recoveries (84.8~103.0%). Reproducibilities were obtained (Coefficient of variation <5.2%), and the linearity of calibration curves were reasonable (r2>0.995) in the range of 0.01-0.2 ㎍/mL. This established analytical method was fully validated and could be useful for quantification of chinomathionat in animal commodities as official analytical method.

Vulnerability Analyses of Wave Overtopping Inundation by Synthesized Typhoons with Sea-Level Rise (해수면 상승과 빈도 합성태풍이 고려된 월파범람 위험성 분석)

  • Storm surges caused by a typhoon occur during the summer season, when the sea-level is higher than the annual average due to steric effect. In this study, we analyzed the sea-level pressure and tidal data collected in 1 h intervals at Incheon, Kunsan, Mokpo, Seogwipo stations on the Yellow Sea coast to analyze the summer season storm surge and wave overtopping. According to our analyses, the summer mean sea-level rise on the west and south coasts is approximately 20 cm and 15 to 20 cm higher than the annual mean sea-level rise. Changes in sea-level rise are closely related to changes in seasonal sea-level pressure, within the range of 1.58 to 1.73 cm/hPa. These correlated mechanisms generates a phase difference of one month or more. The 18.6 year long period tidal constituents indicate that in 2090, the amplitude of the $M_2$ basin peaks on the southwest coast. Therefore, there is a need to analyze the target year for global warming and sea-level rise in 2090. Wave overtopping was simulated considering annual mean sea-level rise, summer sea level rise, the combined effect of nodal factor variation, and 100-year frequency storm surge. As a result, flooding by wave overtopping occurs in the area of Suyong Bay, Busan. In 2090, overtopping discharges are more than doubled than those in Marine City by the recent typhoon Chaba. Adequate coastal design is needed to prepare for flood vulnerability.

Mechanisms of Salt Transport in the Han River Estuary, Gyeonggi Bay (경기만 한강 하구에서의 염 수송 메커니즘)

  • A 3-D hydrodynamic model is applied in the Han River Estuary system, Gyeonggi Bay, to understand the mechanisms of salt transport. The model run is conducted for 245 days (January 20 to September 20, 2020), including dry and wet seasons. The reproducibility of the model about variation of current velocity and salinity is validated by comparing model results with observation data. The salt transport (FS) is calculated for the northern and southern part of Yeomha channel where salt exchange is active. To analyze the mechanisms of salt transport, FS is decomposed into three components, i.e. advective salt transport derived from river flow (QfS0), diffusive salt transport due to lateral and vertical shear velocity (FE), and tidal oscillatory salt transport due to phase lag between current velocity and salinity (FT). According to the monthly average salt transport, the salt in both dry and wet seasons enters through the southern channel of Ganghwa-do by FT. On the other hand, the salt exits through the eastern channel of Yeongjong-do by QfS0. The salt at Han River Estuary enters towards the upper Han River by FT in dry season, whereas that exits to the open sea by QfS0 in wet season. As a result, mechanisms of salt transport in the Han River Estuary depend on the interaction between QfS0 causing transport to open sea and FT causing transport to the upper Han River.

Design and output control technique of sonar transmitter considering impedance variation of underwater acoustic transducer (수중 음향 트랜스듀서의 임피던스 변화를 고려한 소나 송신기의 설계 및 출력 제어 기법)

  • The active sonar transmission system consists of a transmitter that outputs an electrical signal and an underwater acoustic transducer that converts the amplified electrical signal into an acoustic signal. In general, the transmitter output characteristics are dependent on load impedance, and an underwater acoustic transducer, which is a transmitter load, has a characteristic that the electrical impedance varies largely according to frequency when driven. In such a variable impedance condition, the output of the active sonar transmission system may become unstable. Hence, this paper proposes a design and control technique of a sonar transmitter for transmitting a stable transmission signal even under variable impedance conditions of an underwater acoustic transducer in an active sonar transmission system. The electrical impedance characteristics of the underwater acoustic transducer are experimentally analyzed, and the sonar transmitter is composed of a single-phase full-bridge inverter, an LC filter, and a matching circuit. In this paper, the design and output control method of the sonar transmitter is proposed to protect the transmitter and transducer. It can secure stable output voltage characteristics even if it transmits the Linear Frequency Modulation (LFM) signal. The validity is verified through the simulation and the experiment.

Active Front End Rectifier Control of DC Distribution System Using Neural Network (신경회로망을 적용한 직류배전시스템의 AFE 정류기 제어에 관한 연구)

  • As regulations of emissions from ships become more stringent, electric propulsion systems have been increasingly used to solve this problem in vessels ranging from large merchant ships to small and medium-sized ships. Methods for improving the efficiency of the electric propulsion system include the improvement of power sources; the use of a system linked to environmentally friendly power sources, such as batteries, fuel cells, and solar power; and the development of hardware and control methodology for rectifiers, power conversion devices, and propulsion motors. The method using a phase-shifting transformer with diodes has been widely used for rectification. Power semiconductor devices with grid connection to an environmentally friendly power source using DC distribution, a variable speed power source, and the application of small and medium-sized electric propulsion systems have been developed. Accordingly, the demand for active front-end (AFE) rectifiers is increasing. In this study, a method using a neural network rather than a conventional proportional-integral controller was proposed to control the AFE rectifier. Tested controller data were used to design a neural network controller trained through MATLAB/Simulink. The neural network controller was applied to a rectification system designed using PSIM software. The results indicated the effectiveness of improving the waveform and power factor DC output stage according to the load variation. The proposed system can be applied as a rectification system for small and medium-sized environmentally friendly ships.

Development of LC-MS/MS analytical methods for metalaxyl in Atractylodes macrocephala Koidzumi and Achyranthes japonica Nakai (LC-MS/MS를 이용한 생약 백출 및 우슬 중 Metalaxyl 잔류분석법 개발)

  • A new rapid and simple method for metalaxyl in Atractylodes macrocephala Koidzumi and Achyranthes japonica Nakai has been developed and validated. This study was conducted to develop a method for analyzing metalaxyl by a method based on QuEChERS using liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). Samples were extracted with acetonitrile and purified using amino-propyl (NH2) Solid Phase Extraction cartridge. The method limit of quantitation (MLOQ) was 0.01 mg/kg. The linearity of matrix-matched calibration curve (r2) was ≥0.99 at the calibration range of 0.001-0.05 mg/kg. For recovery test, Atractylodes macrocephala Koidzumi or Achyranthes japonica Nakai was treated with standard solutions at MLOQ and 10MLOQ levels. Recovery rates were in the range of 88.1-109.1% with <5.5% coefficient of variation. This established analytical method was fully validated. Based on these results, it can contribute to improving the safety of residual pesticides in Atractylodes macrocephala Koidzumi and Achyranthes japonica Nakai.

A Suggestion of Standard for Science Teacher Professional Development (과학 교사의 전문성 발달을 위한 기준 제안)

  • Science education researchers have discussed more effective STPD(Science Teacher Professional Development) based on a variety of perspectives, pointing out the limitations of traditional STPD. In this study, we selected eight major issues for more efficient STPD based on the literature review, summarized the literature review contents for each issue, and proposed a draft STPD standard based on the literature review. Using the draft STPD standard, a questionnaire for the Delphi survey was developed and a two-phase Delphi survey was conducted. The questionnaire consisted of 25 statements describing the STPD standard, and respondents were asked to indicate the validity and importance of the statement on a Likert scale, followed by an explanation and suggestions for revision. Twenty-one science educators and science teachers participated in the Delphi survey, and the results of the second round of the survey showed high levels of agreement on the validity and importance of all statements describing the STPD standard. For the content validity ratio and coefficient of variation, all statements met the criteria, and for the convergence and consensus, all but two statements met the criteria. Two statements were revised to reflect comments from the Delphi survey and were included in the final STPD standard. As a result, we developed STPD standard consisting of 23 statements in eight categories, and we discussed the disadvantages and advantages of the developed STPD standard and how to utilize them, as well as suggestions for future improvement and research.
