Solid-Phase Extraction of Curcuminoid from Turmeric Using Physical Process Method (물리적 가공법을 이용한 강황으로부터 Curcuminoid의 고체상추출)

  • In order to extract the curcuminoid such as curcumin, demethoxycurcumin (DMC), and bisdemethoxycurcumin (BDMC) in turmeric (Curcuma longa), solvent extraction methods (dipping and ultrasonic extraction method) and solid-phase extraction (SPE) were used. RP-HPLC (reverse-phase high-performance liquid chromatography) and TLC (thin-layer chromatography) were used for identification and analysis the three curcuminoid. From the experimental results, it is evident that the percentage of curcuminoid extracted from turmeric by ultrasonic extraction method was higher than dipping method. The percentage of curcumin extracted from turmeric by pure methanol was higher than any aqueous methanolic composition. Moreover, the total peak area of three curcuminoid was above 92% in RP-HPLC using solid-phase extraction. These results will form a database for investigating the constituents of natural products and the resources of pharmaceutical, nutrition, and cosmetic products.

  • Test of the optimum condition of solid phase micro extraction(SPME) was performed by use of 5 vol-atile components in dilute aqueous solution. Volatile components of Sancho(Zanthoxylum schinifolium) were isolated by SPME method and were analyzed by GC/MSD and compared with volatile compone-nts isolated by simultaneous distillation extraction (SDE) method. Total 31 components were identified by comparing gas chromatography retention time and mass spectral data. The major compounds were limonene geranyl acetate $\beta$-phellandrene phellandral mycene linalool rose oxide caproic acid and caprylic acid SPME sampling procedure was found to be a good method for qualitative analysis of the volatile components.

  • Deformation phase can be obtained by using Least-Square Fitting. In extraction of phase values, Least-Square Fitting is superior to usual method like as 2, 3, 4-Bucket Algorithm. That can extract almost noise-free phase and retain 2$\pi$discontinuities. But more fringe in phase map, 2$\pi$ discontinuities is destroyed when that is filtered and reconstruction of deformation is not reliable. So, we adapted Least-Square Fitting using an isotropic window in dense fringe. using Sine-Cosine filter give us perfect 2$\pi$discontinuities information. We showed the process and result of extraction of phase map and filtering in this paper.

  • A power distribution facility map is drawn on cadastral map. Besides, grid lines are added on the map for sectionalization. For automatic recognition of the map, we first extract recognizing regions. In this paper, we propose an extraction method of recognizing regions by partially extending thinned image. The proposed method is consist of three phases, binarization phase, thinning phase and partial extending phase. The first phase generate a binary image using threshold value which is obtained by histogram analysis. The binary image contains many part of recognizing regions, but not all. The second phase generate thinned image which is generated by appling thinning operator to the binary image. And the third phase extends thinned image from terminal point until satisfying termination condition. The proposed method is tested on several power distribution facility maps, and the results are presented.

  • A multisample extraction device was newly designed for efficient extraction of dissolved lipophillic organic compounds from sea water sample. This device allowed extraction of organic compounds from up to 96 sample at a time using 96 multifolder on the principle of solid phase extraction with commercially available octadecyl silane (ODS) cartridges. The recovery yield of the new divice was higher than 90 % while that of conventional liquid-liquid extraction process are only 60 - 70 %. The amount of solvent required for the new device could be reduced to less than 20㎖ per 1ℓ of sample while 1 - 2 ℓ of solvent were used in the conventional liquid-liquid extraction process. The usefulness of this novel method was demonstrated with sea water samples collected from Yellow sea, and the qualitative and quantitative analyses results of the dissolved hydrocarbon showed this method was superior to that of conventional liquid-liquid extraction process in efficiency and reliability.

  • The Korean government has regulated emission of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) in waste water of manufacturing facilities producing chlorinated compounds since 2009. As this regulation is expected to be reinforced in 2013 to 50 pg I-TEQ/L, a large sample volume is required for the analysis of trace amounts of PCDD/Fs in waste water. Liquid-liquid extraction (LLE) is used to extract PCDD/Fs from aqueous samples; however, its low efficiency makes it inadequate for analyzing large sample volumes. Herein, we present a disk-type solid-phase extraction (SPE) method for the analysis of dioxin at a part per quadrillion level in waste water. This SPE system contains airtight glass covers with a decompression pump, which enables continuous semi-automated extraction. Small (0.5 L) and large (7 L) samples were extracted using LLE and SPE methods, respectively. The method detection limits (MDLs) were 0.001.0.25 and 0.015.4.1 pg I-TEQ/L for the SPE and LLE methods, respectively. The concentrations of detected congeners with both methods were similar. However, the concentrations of several congeners that were not detected with the LLE method were quantified using the SPE method.

Study on of Extraction Methods of Saponin in Ginseng Products (인삼제품의 Saponin 추출방법에 관한 연구)

  • In order to shorten the extraction time of saponin in ginseng products, election with Extrelut column and phase-separation methods were compared. The results obtained are as follows. 1. Saponin of ginseng products was extracted completely within 3 hours by election method with Extrelut column, and the rate of removing glucose by the column was increased with increasing glucose content in ginseng products. 2. Stirring method was superior to refluxing method for removing sugars from ginseng products, and removing rate was deceased in the order of lactose, sucrose and glucose. 3. Extraction rate of ginsenoside from ginseng extracts by the elution method was nearly same as that of phase-separation method; however, the former was much higher than the latter in the case of ginseng teas. Therefore, the elution by Extrelut column is to be improper for extraction of saponin in ginseng tea which contains much sugar. It was necessary to remove lipophilic compounds for extraction of saponin from ginseng extracts by elution with Extrelut column.

Integration of Headspace Solid Phase Micro-Extraction with Gas Chromatography for Quantitative Analysis of Formaldehyde

  • A study was carried out to evaluate the solid phase micro-extraction (SPME) for formaldehyde emission analysis of uncoated plywood. In SPME, formaldehyde was on-fiber derivatized through headspace extraction and analyzed by gas chromatography coupled with mass spectrometry (GC/MS). The SPME was compared with desiccators (DC-JAS 233), small-scale chamber (SSC-ASTM D6007) and liquid-liquid extraction (LLE-EPA 556) methods which were performed in accordance with their respective standards. Compared to SSC (RSD 4.3%) and LLE (RSD 5.0%), the SPME method showed better repeatability (RSD 1.8%) and not much difference from DC (RSD 1.4%). The SPME has proven to be highly precise (at 95% confidence level) with better recovery (REC 102%). Validation of the SPME method for formaldehyde quantitative analysis was evidenced. In addition, the SPME by air sampling directly from plywood specimens (SPME-W) correlated best with DC ($r^2$ = 0.983), followed by LLE ($r^2$ = 0.950) and SSC ($r^2$ = 0.935).

A Study on Analytical Method of Phthalate Esters in Water by SPE(Solid-Phase-Extraction) (고체상 추출법을 이용한 Phthalate Esters의 분석방법 연구)

  • The study was carried out to evaluate the new analytical method of phthalate esters(diethylphthalate, di-n-butylphthalate, butylbenzylphthalate, bis(2-ethylhexyl)phthalate), one of the endocrine disruptors, which were performed by GC/MS-SIM(selected ion monitoring). The phthalate esters were extracted from water samples using solid-phase extraction on $C_{18}$ columns. It investigated that the extraction recovery rate of phthalate esters with different solvents and solvent volume. The optimal solvent was dichloromethane and proper volume of dichloromethane for recovery of phthalate esters was 4 mL. There were good linearities(above $R^2$=0.9975) in the range 0.01~0.50mg/L, and the detection limits were below 0.01~0.03$\mu\textrm{g}$/L. The recovery rates, RSD and MDLs for phthalate esters were 80~114%, 5.0~8.1% and 0.03~0.11$\mu\textrm{g}$/L, respectively. This method shows a good precision of phthalate esters.
