This document was prepared to review and summarize the analytical methods for airborne and bulk asbestos. Basic principles, shortcomings and advantages for asbestos analytical instruments using phase contrast microscopy(PCM), polarized light microscopy(PLM), X-ray diffractometer (XRD), transmission electron microscopy(TEM), scanning electron microscopy(SEM) were reviewed. Both PCM and PLM are principal instrument for airborne and bulk asbestos analysis, respectively. If needed, analytical electron microscopy is employed to confirm asbestos identification. PCM is used originally for workplace airborne asbestos fiber and its application has been expanded to measure airborne fiber. Shortcoming of PCM is that it cannot differentiate true asbestos from non asbestos fiber form and its low resolution limit ($0.2{\sim}0.25{\mu}m$). The measurement of airborne asbestos fiber can be performed by EPA's Asbestos Hazard Emergency Response Act (AHERA) method, World Health Organization (WHO) method, International Standard Organization (ISO) 10312 method, Japan's Environmental Asbestos Monitoring method, and Standard method of Indoor Air Quality of Korea. The measurement of airborne asbestos fiber in workplace can be performed by National Institute for Occupational Safety and Health (NIOSH) 7400 method, NIOSH 7402 method, Occupational Safety and Health Administration (OSHA) ID-160 method, UK's Health and Safety Executive(HSE) Methods for the determination of hazardous substances (MDHS) 39/4 method and Korea Occupational Safety and Health Agency (KOSHA) CODE-A-1-2004 method of Korea. To analyze the bulk asbestos, stereo microscope (SM) and PLM is required by EPA -600/R-93/116 method. Most bulk asbestos can be identified by SM and PLM but one limitation of PLM is that it can not see very thin fiber (i.e., < $0.25{\mu}m$). Bulk asbestos analytical methods, including EPA-600/M4-82-020, EPA-600/R-93/116, OSHA ID-191, Laboratory approval program of New York were reviewed. Also, analytical methods for asbestos in soil, dust, water were briefly discussed. Analytical electron microscope, a transmission electron microscope equipped with selected area electron diffraction (SAED) and energy dispersive X-ray analyser(EDXA), has been known to be better to identify asbestiform than scanning electron microscope(SEM). Though there is no standard SEM procedures, SEM is known to be more suitable to analyze long, thin fiber and more cost-effective. Field emission scanning electron microscope (FE-SEM) imaging protocol was developed to identify asbestos fiber. Although many asbestos analytical methods are available, there is no method that can be applied to all type of samples. In order to detect asbestos with confidence, all advantages and disadvantages of each instrument and method for given sample should be considered.

The separated quasi-one-dimensional ($7{\times}7$) and ($5{\times}5$) phases on vicinal Si(557) surfaces were successfully realized by changing the crystallographic orientation and thermal treatment conditions. A small change in the crystallographic orientation of the Si(557) surface stabilized the quasi-one-dimensional ($5{\times}5$) phase of a (111) facet on vicinal Si(557) surfaces and made it coexist with a quasi-one-dimensional ($7{\times}7$) phase after an optimal thermal treatment, whereas only the quasi-one-dimensional ($7{\times}7$) phase was stable on the Si(557) surface. Interestingly, this causes the (111) terraces with different widths (L) to prefer only one of the $5{\times}5$ (L=12) and $7{\times}7$ (L=9) phases resulting in long-range order of both phases along the step edge direction, which was observed by scanning tunneling microscopy (STM) and was supported by first principle calculations. In contrast, the quasi-one-dimensional ($5{\times}5$ and ($7{\times}7$) phases were arranged randomly across the step edge direction. The change of surface morphology of vicinal Si(557) surfaces will be discussed with STM images and theoretical calculations by changing crystallographic cutting angles and thermal treatment conditions.
PURPOSE. This study evaluated the relationship among translucency, crystalline phase, grain size, and fracture toughness of zirconia. MATERIALS AND METHODS. Four commercial zirconia - Prettau®Anterior® (PA), Prettau® (P), InCorisZI (ZI), and InCorisTZI (TZI)- were selected for this study. The bar specimens were prepared to determine fracture toughness by using chevron notched beam method with four-point bending test. The grain size was evaluated by a mean linear intercept method using a scanning electron microscope. X-ray diffraction and Rietveld refinement were performed to evaluate the amount of tetragonal and cubic phases of zirconia. Contrast ratio (CR) was measured to investigate the level of translucency. RESULTS. PA had the lowest fracture toughness among other groups (P < .05). In addition, the mean fracture toughness of P was significantly less than that of ZI, but there was no difference compared with TZI. Regarding grain size measurement, PA had the largest average grain size among the groups. P obtained larger grain size than ZI and TZI (P < .05). However, there was no significant difference between ZI and TZI. Moreover, PA had the lowest CR value compared with the other groups (P < .05). This means PA was the most translucent material in this study. Rietveld refinement found that PA presented the greatest percentage of cubic phase, followed by TZI, ZI, and P, respectively. CONCLUSION. The different approaches are used by manufacturers to fabricate various types of translucent zirconia with different levels of translucency and mechanical properties, which should be concerned for material selection for successful clinical outcome.
In this study, five series of novel polyurethanes was synthesized by the polyaddition reaction of diisocyanates such as 2,6-tolylene diisocyanate(2,6-TDI), 2,5-tolylene diisocyanate(2,5-TDI), 2,4-tolylene diisocyanate(2,4-TDI), and 1,4-phenylene diisocyanate(1,4-PDI), hexamethylene diisocyanate(HDI) with 4,4'-Bis(3-hydroxypropoxy)biphenyl (BP3), 4,4'-bis(3-hydroxypropoxy)biphenyl exhibited a smectic type mesophase. Monotropic mesophase was found for all synthesized liquid crystalline polyurethanes except HDI/BP3. In contrast, 1,4-PDI/BP3 without a methyl substituent in the phenylene unit exhibited no explicit mesomorphic behavior, which was confirmed by DSC and polarizing microscopy experiments. Structures of the compound were identified by FT-IR and $^1H$-NMR spectroscopies. Their phase transition temperatures and thermal stability were also investigated by differential scanning calorimetry(DSC), polarized optical microscopy(POM) and x-ray diffraction analysis.
This commentary presents the regulatory backgrounds and development of the national proficiency testing (PT) scheme on asbestos analysis in the Republic of Korea. Since 2009, under the amended Occupational Safety and Health Act, the survey of asbestos in buildings and clearance test of asbestos removal works have been mandated to be carried out by the laboratories designated by the Ministry of Employment and Labor (MOEL) in the Republic of Korea. To assess the performance of asbestos laboratories, a PT scheme on asbestos analysis was launched by the Korea Occupational Safety and Health Agency (KOSHA) on behalf of the MOEL in 2007. Participating laboratories are evaluated once a year for fiber counting and bulk asbestos analysis by phase contrast microscopy and polarized light microscopy, respectively. Currently, the number of laboratory enrollments is > 200, and the percentage of passed laboratories is > 90. The current status and several significant changes in operation, sample preparations, and statistics of assigning the reference values of the KOSHA PT scheme on asbestos analysis are presented. Critical retrospect based on the experiences of operating the KOSHA PT scheme suggests considerations for developing a new national PT scheme for asbestos analysis.

Purpose: There are some reports presenting that peripheral blood contain circulating hematopoietic cells as well as, in significantly smaller quantities, mesenchymal stem cells. The purposes of this study is to isolate and characterize circulating mesenchymal progenitor cells with osteogenic potential from human peripheral blood. Methods: Human buffycoat containing mononuclear cells was harvested from peripheral blood of normal persons and isolated using a density gradient centrifugation and serially subcultured in osteogenic media for 1-4 weeks. The proliferation capability, phase-contrast microscopy, transmission electron microscopy, immunophenotype FACS analysis, Alizarin red staining and RT-PCR assays for osteogenic differentiation potential were performed. Results: The phenotype of cultured cells changed from small round or cuboidal cells at passage 1 into large spindle-shaped fibroblastic morphology cells at passage 4. Surface marker expressed CD14, but did not express CD34, CD80, CD83. Strong positive staining was observed for Alizarin reds in osteogenic medium on day 14, Using RT-PCR, the mRNA levels of bone- specific genes, such as ALP, c-bfa-1 and osteocalcin were detected. Conclusion: A new subset of peripheral blood derived progenitor cells described here has the ability to proliferate and differentiate into osteogenic cell lineages in vitro, and to be candidate for regenerative therapy.

Purpose: The aim of this study is to determine whether certain biomaterials have the potential to support cell attachment. After seeding bone marrow stromal cells onto the biomaterials, we investigated their responses to each material in vitro. Methods: Rat bone marrow derived stromal cells were used. The biomaterials were deproteinized bovine bone mineral (DBBM), DBBM coated with fibronectin (FN), synthetic hydroxyapatite (HA), HA coated with FN, HA coated with $\beta$-tricalcium phosphate (TCP), and pure $\beta$-TCP. With confocal laser scanning microscopy, actin filaments and vinculin were observed after 6, 12, and 24 hours of cell seeding. The morphological features of cells on each biomaterial were observed using scanning electron microscopy at day 1 and 7. Results: The cells on HA/FN and HA spread widely and showed better defined actin cytoskeletons than those on the other biomaterials. At the initial phase, FN seemed to have a favorable effect on cell adhesion. In DBBM, very few cells adhered to the surface. Conclusions: Within the limitations of this study, we can conclude that in contrast with DBBM not supporting cell attachment, HA provided a more favorable environment with respect to cell attachment.
Objectives: The purpose of the study was to investigate the bacterial morphology attached on ultrasonic scaler tips using no cleansing solution, alcohol cotton, liquid chemical disinfecting agent, and autoclave method. Methods: Scaling tip was applied to the mouth and the ultrasonic scaler tips were assigned to four groups. Group 1 was control group with no cleansing solution. Group 2 was treated with alcohol cotton. Group 3 was treated with 2% green Y-Na solution in liquid chemical disinfecting agent, and Group 4 was sterilized by autoclave method. Live bacteria were observed by phase contrast microscopy. The scanning electron microscopy(SEM) revealed the morphological characteristics of scaler surface. The type of attached bacteria were analyzed using SPSS 21.0 program. The data were analyzed by one-way analysis of variance(ANOVA) and Tukey's post-hoc test. Results: The types of sterilization methods had influences on the bacterial viability. The numbers of cocci, bacilli, spiral form bacteria, and filamentous bacteria was observed in $89.00{\pm}3.60%$, $29.67{\pm}3.51%$, $3.33{\pm}0.57%$ and $1.67{\pm}0.57%$ in control group, $31.67{\pm}3.51%$, $63.33{\pm}4.04%$, $2.00{\pm}1.00%$ and $1.67{\pm}0.57%$ in alcohol cotton group, $69.67{\pm}4.50%$, $12.33{\pm}2.51%$, 0% and 0% in liquid chemical disinfecting agent group, and 0.0%, 0.0%, 0.0% and 0.0% in autoclave method group. The clean surface of ultrasonic scaler tip was shown on SEM by autoclave method. Conclusions: The most effective sterilization method of ultrasonic scaler tip was the autoclave method. Autoclave method is the most effective sterilization method and can reduce the cross-infection in the dental clinic.
Background: In tumor cells, aberrant differentiation programs have been described. Several neuronal proteins have been found associated with morphological neuronal-glial changes in breast cancer (BCa). These neuronal proteins have been related to mechanisms that are involved in carcinogenesis; however, this regulation is not well understood. Microtubule-associated protein-tau (MAP-Tau) has been describing in BCa but not its variants. This finding could partly explain the neuronal-glial morphology of BCa cells. Our aim was to determine mRNA expression of MAP-tau variants 2, 4 and 6 in breast cancer cell lines. Materials and Methods: Cultured cell lines MCF-10A, MDA-MB-231, SKBR3 and T47D were observed under phase-contrast microscopy for neural morphology and analyzed for gene expression of MAP-Tau transcript variants 2, 4 and 6 by real-time PCR. Results: Regarding morphology like neural/glial cells, T47D line shown more cells with these features than MDA-MB-231 and SKBR. In another hand, we found much greater mRNA expression of MAP-Tau transcript variants 2, and to a lesser extent 4 and 6, in T47D cells than the other lines. In conclusion, regulation of MAP-Tau could bring about changes in cytoskeleton, cell morphology and motility; these findings cast further light on neuronal transdifferentiation in BCa.
The phagocytic petencies and birefringences of peritoneal mast cells of rats treated with particular dyes (neutral red, toluidine blue, pyronin, methylene blue, alcian blue, trypan blue, carmine, orange G, aniline blue, Janus green B, and India ink) were analyzed by means of phase contrast microscopy and polarizing microscopy. In addition, cytomorphic effects of the dyes on the peritoneal mast cells were also discussed. Phagocytic activities or ingestion of the dye particles were not observed in most cases, except for the India ink group. Hardly a macrophage appeared without some dark particles which were ingested or phagocytosed. Trypan blue and aniline blue produced very weak birefringence in the cytoplasm of mast cells but the rest did not produce even in the acid medium (neutral red and toluidine blue). The short and slender ectoplasmic processes of the mast cells and the leucocytes were also found in certain groups. The cytomorphic effects of the dyes on the mast cell were slight and variable.
