INHIBITORY ACTION OF PROCESSED HERBAL MEDICINES ON THE PRODUCTION OF ADVANCED GLYCATION ENDPRODUCTS(AGEs)

  • Diabetic nephropathy is major chronic complication of diabetes mellitus. Advanced glycation endproducts(AGEs) are largely involved in the pathogenesis of diabetic nephropathy. The irreversibly formed AGEs do not return to normal even if hyperglycemia is corrected and continue to accumulate over the lifetime of protein. The AGEs inhibitor. aminoguanidine(AG), is the only protein glycation inhibitor currently under development. its safety however is desirable. To find possible AGEs inhibitor in herbal medicines, bovine serum albumin was added to a mixture of sugars and some of processed. unprocessed herbal medicines or AG. Cyperi rhizoma was processed in four different methods according to chinese pharmacopoeia and traditional literatures. In comparision to the negative control with no inhibitor and positive control with AG. alcoholic extracts of these processed cyperi rhizoma proved to have more potent inhibitory activities than that of unprocessed cyperi rhizoma. These results revealed that some processed herbal medicines have a more potent in vitro inhibitory action on AGEs formation than AG. suggesting the possible candidate for diabetic nephropathy from the processed herbal medicines.

Development of official assay method for loperamide hydrochloride capsules by HPLC

  • Currently, the potentiometric titration and the high pressure liquid chromatography (HPLC) method were utilized in Korean Pharmacopoeia XII (KP XII) as well as other pharmacopoeias (USP, EP, BP) for determination of loperamide hydrochloride in raw materials and capsules, respectively. The research objective is to overcome the remaining drawbacks from current methods such as solubility of mobile phase (KP XII), less scientific approach (USP 43) or using paired-ion chromatography reagent which shows some limitations (BP2017 and other formulation monographs). The proposed method was optimized by Design of Experiment (DoE) tool to obtain the satisfied method for determination of loperamide hydrochloride. The optimal condition was performed on the common C18 column (150 mm × 4.6 mm; 5 ㎛) using isocratic elution with the mobile phase containing 40 mM of potassium phosphate monobasic (pH 3.0) and acetonitrile (56:44), at a flow rate of 0.7 mL/min. The optimized method was validated and met the requirements of the International Conference on Harmonization. The developed method was applied to determine loperamide hydrochloride in capsules and can be used to update the current monograph in KP XII.

Comparative Study of Terminology for Identification of the Chinese Herbal Materia Medica (초본류 한약재 감별을 위한 용어의 비교연구)

  • Objectives : To investigate terminology of Herbal drug discrimination commonly used in China, and it was translated into Korean. It also has been modified to fit the circumstances of the Korean for easier understand for Korean student. Methods : First,Terminology for Identification of the Chinese Herbal Materia Medica (TICHMM) was classified by the appearance, cut surface, material, processing and storage of Herb. Chinese commentary of TICHMM was translated into Korean. Comparative Study was performed based on the Pharmacopoeia of Korea and China. Depending on the meaning of the word, TICHMM was modified to revised Korean expression or remain chinese expression or suggest new Korean expression Result : In the 28 TICHMM terms that describes the shape properties of herbal medicine, 5 was remained in Chinese expression, 22 was modified to revised Korean expression and 1 was suggested in new Korean expression. 8 terms that describes the Section properties of herbal medicine, 6 was modified to revised Korean expression and 2 was suggested in new Korean expression. 3 terms that describes the Material properties of herbal medicine, 1 was remained in Chinese expression, 2 was modified to revised Korean expression. 3 terms that describes the processing and storage properties of herbal medicine, 2 was modified to revised Korean expression and 1 was suggested in new Korean expression. Conclusion : By comparative Study of TICHMM, commonly used in China, it will contribute new guideline on the Visual Examination of Herbal Medicine for Korean student.

Comparative Study of Extracts from Rhubarb on Inflammatory Activity in Raw 264.7 Cells (마우스 Raw 264.7 세포에 대한 4종 대황류 추출물의 항염증 활성 비교)

  • Rhubarb is the well-known and frequently used herbal medicine for the treatment of constipation, inflammation, and cancer. As described in the Korea and Chinese Pharmacopoeia, rhubarb consists of the roots and rhizomes of Rheum palmatum, R. officinale, and R. tanguticum. However, the pharmacological differences among rhubarb have not been scientifically established. In the present study, we investigated and compared the inhibitory effects of 70% ethanolic extracts from R. palmatum (RPE), R. officinale (ROE), R. nobile (RNE), and R. franzenbachii (RFE) on the production of inflammatory mediators, nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), interlukin-1beta ($IL-1{\beta}$), and tumor necrosis factor-alpha ($TNF-{\alpha}$) in RAW 264.7 macrophage cells. ROE, RNE, and RFE significantly inhibited the release of NO, $PGE_2$, $IL-1{\beta}$, and $TNF-{\alpha}$. RPE significantly reduced the release of $IL-1{\beta}$, but not NO, $PGE_2$, and $TNF-{\alpha}$. Overall, RFE was found to inhibit the release of $PGE_2$ and $IL-1{\beta}$, to a far greater degree than RPE, ROE, and RNE. Our results indicate that RFE possess the strongest anti-inflammatory activity among 4 tested rhubarb.

Antioxidant activity of ginseng cultivated under mountainous forest with different growing years

  • Ginseng cultivated and grown naturally under mountainous forest is formally called "Lin-Xia-Shan-Shen" (LXSS) and grown in manual condition is called garden ginseng (GG) according to Chinese pharmacopoeia (2010 edition). Usually the growing condition of LXSS is similar to wild ginseng and mostly used in Chinese folks in ancient times. The antioxidant properties of LXSS with different growing years were evaluated by their inhibitions of thiobarbituric acid-reactive substance (TBA-RS) formation in liver homogenate and 2, 2-diphenyl-1-picrylhydrazyl (DPPH)-radical scavenging activity comparing with those of GG. The inhibitions of different polar extracts (n-butanol and water) of LXSS and GG on TBA-RS formation were also evaluated. The results showed that the antioxidant effects of LXSS were higher than those of GG and the TBA-RS formation inhibition of LXSS with longer growing years were stronger than those with shorter growing years, while the DPPH-radical scavenging activity of LXSS did not show significant difference with the change of the growing year. The results indicated that the inhibitory effect of TBA-RS formation and the DPPH-radical scavenging of LXSS were correlated with the contents of ginsenosides. In adddition, the starch contents of LXSS and GG were determined by micro-amount method with spectrophotometer. It showed that the starch content in GG was higher than that of LXSS whose starch decreased gradually with the growing year.

Identification of Marker Nucleotides for the Molecular Authentication of Arisaematis Rhizoma Based on the DNA Barcode Sequences (천남성(天南星) 유전자 감별을 위한 DNA 바코드 분석 및 Marker Nucleotide 발굴)

  • Objectives : Official Arisaematis Rhizoma is described only three species, Arisaema amurnse, Arisaema erubescens, and Arisaema heterophyllum, in national Pharmacopoeia. However, other Arisaema species, Arisaema ringens, Arisaema takesimense and Arisaema serratum, also have been distributed as an inauthentic Arisaematis Rhizoma in the herbal market. To develop a reliable molecular authentication method for Arisaematis Rhizoma in species level, we analyzed DNA barcode regions using six Arisaema species. Methods : Thirty-eight samples of six Arisaema plants species (A. amurense, A. amurense f. serratum, A. heterophyllum, A. takesimense, and A. serratum) were collected from different habitate and nucleotide sequences of DNA barcode regions (rDNA-ITS, matK, and rbcL gene) were analyzed after PCR amplification. The species-specific sequences and phylogenetic relations were estimated using entire sequences of three DNA barcodes based on the analysis of ClastalW and UPGMA, respectively. Results : The comparative analysis of DNA barcode sequences were revealed inter-species specific nucleotides to distinguish the medicinal plant of Arisaema Rhizoma in species levels excluding between A. amurense and its subspecies (A. amurense f. serratum) and A. takesimense and A. serratum, respectively. However, we obtained sequence differences enough to discriminate authentic and inauthentic Arisaematis Rhizoma. Therefore, we suggest that these SNP type molecular genetic markers were an reliable method avaliable to identify official herbal medicines. Conclusions : These marker nucleotides could be useful to identify the official herbal medicines by providing definitive information that can identify original medicinal plant and distinguish from inauthentic adulterants and substitutes.

  • Clay minerals have been used in pharmaceutical industries as active ingredients and excipients without pharmacological activity such as diluents, emulsifying agents, viscosity-increasing agents, and lubricants. For example, bentonite, kaolin, magnesium aluminum silicate, and talc are generally and extensively used pharmaceutical ingredients, which are restrictedly regulated by Pharmacopoeias. We discuss the physicochemical and biopharmaceutical properties of clay minerals. In addition, we introduce the cases of pharmaceutical applications of clay minerals. From this review, pharmaceutical applications of clay minerals can be one of strategies for the development of high value-added products from clay minerals.

  • Artemisia Herb is the dry aboveground part of Artemisia asiatica (Compositae). This is listed officially in the Korean Herbal Pharmacopoeia and used mainly in stopping all kinds of bleeding, regulating menses and curing menstrual disorders as well as in alleviating pain. Eupatilin and jaceosidin were isolated from Sazabalsuk and Sajuarisuk. and identified by means of spectral methods $(UV,\;^1H\;and\;^{13}C-NMR,\;MS) $ and quantitative analysis using HPLC. Regression equations for two flavonoid component standards obtained from Artemisia asiatica were $y=76069_x\;+\;107329\;(r=0.9999^{**})$ for eupatilin, $y=76651_x\;+\;77399\;(r=0.9996^{**})$ for jaceosidin. The eupatilin content of Sazabalsuk was 240mg, jaceosidin was 10mg in 100 g dry leaves. The Sajuarisuk were 330mg, 123mg in 100g dry leaves, respectively.

Physical and Chemical Analysis of Epimedii Herba by the Packaging Methods and Storage Periods (포장방법 및 보관기간에 따른 음양곽의 이화학적 분석)

  • In order to obtain basic data on the shelf life of Epimedii Herba (EH), a physicochemical characteristics including weight loss rate, loss on drying, ash, acid-insoluble ash, and quantitative determination were evaluated to compare the data by packaging methods including general (G), general + silica gel (GS), alumium (AL), and alumium + silica gel (ALS) packaging and storage periods (0-66 months). All physicochemical analyzes were conducted using the method of "The Korean Pharmacopoeia (KP) 9th edition". As a result, weight loss rate (%), loss on drying (%), ash (%), acid-insoluble ash (%) of EH according to the packaging methods and storage periods were -0.27-4.73%, 3.06-9.35%, 5.48-7.37%, and 0.45-0.99, respectively. In the quantitative analysis of icariin, the marker compound of EH using high-performance liquid chromatography, the initial content was 0.44% at 0 month in all packaging methods. After that, the result of analyzing the content of icariin every 6 months was detected 0.44-1.18%. All of the above results satisfied the acceptance criteria for loss on drying, ash, acid-insoluble ash, and quantification contained in the KP. This study can be used as a basic data for establishing the packaging method and shelf life of EH.

Evaluation of the Antimicrobial Activity of Seven Gabonese Medicinal Plants against Methicillin-Resistant Staphylococcus aureus and Salmonella

  • The plant species reported here are used by traditional healers in Gabon for different ailments such as wounds, malaria, fever, gonorrhea or diarrhea. The aim of this study was to evaluate the antimicrobial activities of 7 plants (Strombosiopsis tetrandra, Tetraberlinia bifoliolata, Dichapetalum barbatum, Guibourtia demeusii, Dacryodes normandii, Manniophytum fulvum, Paropsia grewoides) against different strains of both Methicillin-Resistant Staphylococcus aureus (MRSA) and Salmonella. Disc diffusion was first used to determine the antimicrobial effectiveness of the plants' ethanolic extracts. Then the minimum inhibitory concentrations of the crude extracts of either leaves or stem barks of the 7 plants were determined using broth micro-dilution. The ethanolic plant extracts showed very good activity against both MRSA and Salmonella strains where the MICs ranged from $250\;{\mu}g/ml$ to $1000\;{\mu}g/ml$. The study shows that many of the tested plants used by Gabonese traditional healers have antimicrobial activities and give support to their traditional use.
