Breeding of 'Seeberry' F1 Hybrid Strawberry (F1 종자 딸기 '씨베리' 육성)

  • 'Seeberry' is a short-day $F_1$ hybrid strawberry for seed propagation, that was released by the Protected Horticulture Experiment Station (RDA, Republic of Korea) in 2013. Inbred lines derived from domestic germplasms and selected varieties were used to develop this cultivar. 'Seeberry' originated from a cross of two inbred lines: a male parent 'Wongyo 3115' with high firmness and a female parent 'Wongyo 3116' with excellent fruit shape and high yield. 'Seeberry' cultivar should be grown with a crown diameter of over 10 mm to reveal its unique characteristics. 'Seeberry' has an upright plant shape with elliptic leaves, 12-15 flowers per cluster, vigorless growth habit, and average flower bud differentiation compared to vegetatively propagated cultivars. Fruits of 'Seeberry' are conical, having light skin color and approximately 15-16 g in average weight. Yield of this cultivar is about 92% lower than that of 'Akihime', but 'Seeberry' has excellent taste, sugar content of $9.7^{\circ}Bx$, acidity of 0.6%, and good texture. With regard to disease and pest resistance, 'Seeberry' is sensitive to powdery mildew, aphids, and the two-spotted spider mite, and is resistant to anthracnose.

Characterization of a new variety Auricularia polytricha (Mont.) Sacc.'Geoni' (털목이 신품종 '건이')

  • Geoni, a new variety of Auricularia polytricha, was bred in JARES(Jeollanamdo Agricultural Research and Extension Services) in 2012. Geoni was selected through the monospore hybridization with JNM21008 and JNM21014 in 2010. Based on a performance test conducted from 2011 to 2012, the Geoni was selected from a line showing an excellent light brown pileus and strong pest resistance. Geoni has a favorable chewiness, light brown and smooth pileus. In addition, Geoni was rich in dietary fiber. MCM(mushroom complete medium), Malt and PDA(Potato Dextrose Agar) mediums were suitable for cultivating the Geoni. The number of effective stipes was 39 ea/0.9 kg and minor axis and major axis of pileus were 6.9 cm and 8.7 cm respectively. The yield of Geoni was 291 g/0.9 kg in plastic bag. Geoni was required 40~54 days for culture at $20^{\circ}C$ and 24days for the primordia and growth period, which is longer than that of the control(Pung-un). Somatic incompatibility was formed between parental strains and Geoni. Analysis of the genetic diversity of the new variety "Geoni"revealed a different profile from the parental strains when RAPD(random Amplified Polymorphic DNA) primers were used.

Insecticidal Effect of an Entomopathogenic Fungus, Beauveria bassiana ANU1 to Spodoptera exigua and Plutella xylostella by Different Temperature and Humidity Conditions (파밤나방과 배추좀나방에 대한 곤충병원성 곰팡이 Beauveria bassiana ANU1의 온도와 습도조건에 따른 살충효과)

  • Entomopathogenic fungi have been studied to develop for biological control agents as an alternative to chemical control agents in insect pest management. Two Lepidopteran insects, Spodoptera exigua and Plutella xylostella, are serious insect pests infested various crops, but not effectively controlled by commercial chemical pesticides due to its high insecticide resistance. A fungal isolate was isolated from S. exigua larvae collected from green onion field in Andong, Korea. To identify the fungal isolate, 18srRNA sequence for internal transcribed spacer (ITS) and ${\beta}$-tubulin regions were sequenced. The ITS and ${\beta}$-tubulin sequence were highly matched to Beauveria bassiana and morphological characteristics also was fit to known B. bassiana. Finally, isolated fungus has identified as B. bassiana and named B. bassiana ANU1. The result of bioassay, median lethal concentrations were $2.7{\times}10^3$ and $0.9{\times}10^3conidia/ml$ and medial lethal times were 65.6 and 60.8 h to S. exigua and P. xylostella, respectively. B. bassiana ANU1 showed high pathogenicity to two insect pests from $20^{\circ}C$ to $30^{\circ}C$ at 50% relative humidity (RH) and more than 40% RH at $25^{\circ}C$ with $10^7conidia/ml$ of concentration.

Combination effects of Organic Materials and Bacillus thuringiensis on Spodoptera exigua (유기농자재와 Bacillus thuringiensis의 혼합처리에 의한 파밤나방의 방제효과)

  • This study is conduced to investigate control efficacy of mixture of plant extracts which has been reported to control pest and Bacillus thuringiensis or alone. Control efficacy of 0.025% matrine or mixture of 0.016% matrine and BT ($5{\times}10^4cfu$) against $3^{rd}$ instar larvae of beet armyworm were best as 98.7% and 93.3%. Treatment of mixture of 0.01% matrine and BT ($5{\times}10^4cfu$) showed the mortality of 69.3%. In the case of neem, when the mixture of 0.1% neem and BT was applied to larvae of S. exigua, their mortality was 50.0% and weight of larvae were significantly different compare with control and only neem treatment. Therefore For effective control reducing population of beet armyworm and possibility to develop resistance against BT and eco-friendly agents, mixture of 0.016% Matrine and BT ($5{\times}10^4cfu$) could be used.

Analysis of Research Trend on Zoysiagrass (Zoysia spp.) (한국 잔디류의 연구동향 분석)

  • Korean lawn grass, which belongs to Zoysia genus in Gramineae family, is one of the major turfgrass which is widely adapted to its native niches in the temperate region of the north east Asia through tropical regions including Philippine, Thailand and Australia. The Turfgrass Information Center' database provides 5,340 descriptive records including 638 referred papers and 1,370 technical reports concerning 'Zoysia'. The database focused on researches on golf courses and lawn care industries. The researches provide informations on seed technology and use of plant protectant for pest management in turf management industries. The purpose of this study is to analyze and classify the research contents of zoysiagrasses which have been published in Korean journals. The total number of research papers published in Korean journals were 274 including 102, 38, and 134 in the 'Botanical', 'Environmental' and 'Cultural' researches, respectively. Publication in foreign journals by Korean investigators were not counted in this research. Research fields concerning golf course maintenance and plant protectant are believed to be necessary when compared to international trends. Moreover, advanced research efforts for the development of new cultivars with various environmental and disease resistance should be accomplished.

Insect Resistance of Tobacco Plant Expressing CpBV-ELP1 Derived from a Polydnavirus (폴리드나바이러스 유래 CpBV-ELP1 발현 담배의 내충성)

  • Polydnaviruses (PDVs) are a group of double-stranded DNA viruses symbiotic to some endoparasitoid wasps. Cotesia plutellae bracovirus (CpBV) is a PDV symbiotic to an endoparasitoid wasp, C. plutellae, parasitizing young larvae of Plutella xylostella. An early expressed gene, CpBV-ELP1, plays an important role in the parasitism by suppressing host cellular immunity by its cytotoxic activity against hemocytes. This study aimed to test its oral toxicity against insect pest by expressing it in a recombinant tobacco plant. A recombinant CpBV-ELP1 protein was produced using a baculovirus expression system and secreted to cell culture medium. The cell cultured media were used to purify CpBV-ELP1 by a sequential array of purification steps: ammonium sulfate fractionation, size exclusion chromatography, and ion exchange chromatography. Purified rCpBV-ELP1 exhibited a significant cytotoxicity against Spodoptera exigua hemocytes. CpBV-ELP1 was highly toxic to the fifth instar larvae of S. exigua by injection to hemocoel. It also showed a significant oral toxicity to fifth instar larvae of S. exigua by a leaf-dipping assay. CpBV-ELP1 was cloned into pBI121 vector under CaMV 35S promoter with opaline synthase terminator. Resulting recombinant vector (pBI121-ELP1) was used to transform Agrobacterium tumefaciens LBA4404. The recombinant bacteria were then used to induce callus of a tobacco (Nicotiana tabacum Xanthi) leaves and subsequent generation (T1) plants were selected. T1 generation tobacco plants expressing CpBV-ELP1 gave significant insecticidal activities against S. exigua larvae. These results suggest that CpBV-ELP1 gene can be used to control insect pests by constructing transgenic crops.

Variety and phosphate fertilizer dose effect on nutrient composition, in vitro digestibility and feeding value of cowpea haulm

  • Background: Cowpea (Vigna unguiculata [L.]) is a legume cultivated throughout most tropical countries and is valued as food and feed for human and livestock respectively. The search for an improved cowpea variety has been on-going with the aim of improving traits such as grain yield, drought and pest resistance. But no information exist on the feeding value of these improved varieties. Phosphate (P) fertilizer application is recommended to augment grain yield in grain legumes but data on the effect of P fertilizer on haulm quality is limited. Results: Two separate experiments were conducted to determine the effect of P fertilizer dose on the nutritive value of haulms from different cowpea varieties (V). In experiment 1, effect of three P doses (30, 60 and 90 kg $P_2O_5/ha$) on in vitro gas production (IVGP) characteristics, concentrations of digestible organic matter (DOM), crude protein (CP), acid detergent fiber (ADF) and neutral detergent fiber (NDF) of haulms from five cowpea varieties (Zaayura-SARC 4-75, Songotra-IT97K-499-35, Hewale-IT93K-192-4, IT99K 573-1-1 and Asomdwe-IT94K-410-2) were investigated using the $3(P){\times}5(V)$ factorial treatment arrangements in a completely randomized design. In experiment 2, the effects of two P doses (30 and 90 kg $P_2O_5/ha$) and two varieties (Zaayura-SARC 4-75 and Hewale-IT93K-192-4) on the voluntary feed intake, live weight, haematology and carcass characteristics of Djallonke lambs were also assessed using a $2(P){\times}2(V)$ factorial treatment arrangement. The $V{\times}P$ interaction significantly affected CP, NDF and ADF with CP concentration increasing with increase in P doses in Zaayura-SARC 4-75 and Asomdwe-IT94K-410-2. Whilst an increase (P < 0.05) in NDF was observed in Songotra-IT97K-499-35and Asomdwe-IT94K-410-2 as P doses increased, the other V only increased from P dose 30 to 60 kg/ha and declined at P dose 90 kg/ha. The ADF decreased (P < 0.05) with increase in P dose for all V with the exception of Songotra-IT97K-499-35. There was a significant V effect on DOM with the highest reported in Zaayura-SARC 4-75 (43.44 %). Daily DM intake, carcass length and blood urea nitrogen of the lambs were significantly affected by the V x P interaction. There was a significant V effect on globulin and P effect on live weight at slaughter, dressed weight, chuck, leg, loin, rib and flank and liver and lungs. Conclusion: It can be concluded that nutrient concentrations of cowpea haulms were positively influenced by different P dose and varieties with favorable effects on growth, haematology and carcass composition of lambs. Varieties Zaayura-SARC 4-75 and Hewale-IT93K-192-4 at P dose at 90 kg/ha are recommended to enhance growth performance and carcass yield of Djallonke lambs.

Breeding of 'Daewang' Strawberry for Forcing Culture with Good Taste and Fragrance (맛과 향이 좋은 촉성재배용 딸기 '대왕' 품종의 육성)

  • A new cultivar 'Daewang' strawberry was developed by the National Institute of Horticultural & Herbal Science for forcing culture in 2010. The cultivar 'Daewang' was originated from the cross between 'Maehyang', a high firmness cultivar and 'Wongyo 3111', a high sugar content in 2006. The cultivar shows erect plant type, vigorous growth habit, early flower bud differentiation with 12-15 flowers per cluster from planting on healthy nursery. Fruits of 'Daewang' are conical type having a bright red skin color, and 16-17 g in an average weight. 'Daewang' is suitable for forcing culture as time required for dormancy breaking ranged between 50 and 100 hours. This cultivar has excellent taste for high sugar/acid ratio as sugar content of $11.1^{\circ}Brix$, acidity of 0.39% with abundant texture and can be harvested by late spring because the fruit firmness of 'Daewang' was $18.2g{\cdot}mm^{-2}$ that was about $7.9g{\cdot}mm^{-2}$ higher than $10.3g{\cdot}mm^{-2}$ of 'Akihime' cultivar. But although total yield is not significantly different from 'Akihime' cultivar, its marketable yield is remarkably higher than that of 'Akihime' cultivar. Disease and pest resistance of 'Daewang' have a tendency to sensitive powdery mildew, anthracnose and spotted spider.

Effect of Brown Planthopper Resistance Gene, Bph18 to Yield Components in Rice (벼멸구 저항성 Bph18 유전자가 벼의 수량구성요소에 미치는 영향)

  • Brown planthopper(BPH) is one of the most destructive insect pests of rice. The use of genetically resistant cultivars has proven to be a more economical, efficient and environment friendly means to combat this pest. This study was carried out to investigate the relationship between BPH resistant gene, Bph18 and yield components of rice using DH(doubled haploid) lines derived from 'Saenuri'/SR30071-3-7-23-6-1-1-1. SR30071-3-7-23-6-1-1-1 line has Bph18 gene derived from wild species, Oryza australiensis. BPH resistant gene, Bph18 shortened heading days, enlarged culm length and panicle length and reduced ratio of ripened grains of rice. The results indicate that backcrossing breeding is necessary to develop elite cultivars carrying Bph18 gene.

Responses of Growth Characteristics of Soybean [Glycine max (L.) Merr.] Cultivars to Riptortus clavatus Thunberg (Hemiptera: Alydidae) (콩 톱다리개미허리노린재 피해와 관련형질의 품종간 차이)

  • In recent years, an increasing amount of research has been conducted on the yield loss of soybean crop caused by Riptortus clavatus, a serious pest of legumes. The objective of this study were to examine crop damage caused by various population densities of R. clavatus and determine the association of unique growth and seed characteristics with crop damage rate in conventional soybean fields. Major growth responses to R. clavatus were examined. The cultivars that were not as seriously damaged by R. clavatus were Pungsannamulkong, Sobaegknamulkong, Duyoukong, and Sinpaldal 2, but Myeongjunamulkong, Mallikong, and Hwangkeumkong were significantly (P<0.05) damaged. The rate of seed damage caused by R. clavatus was 8.2% in a group of brown colored pubescence, while the rate was 13.0% in a group of the gray colored. Crop damage by R. clavatus clearly more reduced in cultivars with dark hilum colors including dark brown, brown, and grayish brown than those with yellow hilum colors. Concerning pod colors, crop damage was less significant in cultivars with dark brown pod color than with the light brown. Cultivars with short pod length (4.0-4.5 cm) at full seed (R6) stage showed lower seed damage rates by R. clavatus than those with long pod length (>5.6 cm). Therefore, characteristics of hilum, pod, pubescence colors, and pod length were significantly related and linked to the cultivar resistance to R. clavatus.
