Hepatitis C Virus Core Protein Is Efficiently Released into the Culture Medium in Insect Cells

  • Hepatitis C virus (HCV) is a causal agent of the chronic liver infection. To understand HCV morphogenesis, we studied the assembly of HCV structural proteins in insect cells. We constructed recombinant baculovirus expression vectors consisting of either HCV core alone, core-E1, or core-E1-E2. These structural proteins were expressed in insect cells and were examined to assemble into particles. Neither core-E1 nor core-E1-E2 was capable of assembling into virus-like particles (VLPs). It was surprising that the core protein alone was assembled into core-like particles. These particles were released into the culture medium as early as 2 days after infection. In our system, HCV structural proteins including envelope proteins did not assemble into VLPs. Instead, the core protein itself has the intrinsic capacity to assemble into amorphous core-like particles. Furthermore, released core particles were associated with HCV RNA, indicating that core proteins were assembled into nucleocapsids. These results suggest that HCV may utilize a unique core release mechanism to evade the hosts defense mechanism, thus contributing to the persistence of HCV infection.

  • Understanding the cellular and molecular mechanisms involved in the development and progression of pulmonary hypertension (PH) remains imperative if we are to successfully improve the quality of life and life span of patients with the disease. A whole plethora of mechanisms are associated with the development and progression of PH. Such complexity makes it difficult to isolate one particular pathway to target clinically. Changes in intracellular free calcium concentration, the most common intracellular second messenger, can have significant impact in defining the pathogenic mechanisms leading to its development and persistence. Signaling pathways leading to the elevation of $[Ca^{2+}]_{cyt}$ contribute to pulmonary vasoconstriction, excessive proliferation of smooth muscle cells and ultimately pulmonary vascular remodeling. This current review serves to summarize the some of the most recent advances in the regulation of calcium during pulmonary hypertension.

  • Urinary tract infections (UTIs) represent one of the most prevalent bacterial infections globally, manifesting in diverse clinical phenotypes with varying degrees of severity and complications. The mechanisms underlying UTIs are gradually being elucidated, leading to an enhanced understanding of the immune responses involved. Innate immune cells play a crucial defensive role against uropathogenic bacteria through various mechanisms. Despite their significant contributions to host defense, these cells often fail to achieve complete clearance of uropathogens, necessitating the frequent prescription of antibiotics for UTI patients. However, the persistence of infections and related pathological symptoms in the absence of innate immune cells in animal models underscore the importance of innate immunity in UTIs. Therefore, the host protective functions of innate immune cells, including neutrophils, macrophages, mast cells, NK cells, innate lymphoid cells, and γδ T cells, are delicately coordinated and timely regulated by a variety of cytokines to ensure successful pathogen clearance.

  • The persistence of jaundice beyond the first 2 weeks of life require further investigation and this can be determined if the conjugated bilirubin levels are greater than 1.5 mg/dL or greater than 20% of the total bilirubin level. There is a diverse differential diagnosis for the cause of neonatal cholestasis due to hepatobiliary disease including biliary atresia, which eventually leads to liver cirrhosis if uncorrected before 60~80 days of life. Long-established initial studies include abdominal ultrasonography, hepatobiliary scintigraphy and liver biopsy, but better diagnostic methods are needed. Promising new options are described including MRCP (magnetic resonance cholangiography), ERCP (endoscopic retrograde cholangiography), and PCC (percutaneous cholecysto-cholangiography). Though no single test can differentiate biliary atresia from other neonatal cholestasis with confidence, a combination of diagnostic methods is usually consistently beneficial. By excluding biliary atresia as early as possible, the risk of unnecessary explolaparotomy with intraoperative cholangiography is decreased. Further evaluation would be required for the diagnosis of neonatal cholestasis after excluding biliary atresia.

Relevant Variables of Effective Parental Disciplinary Practices (부모의 효율적 훈육태도에 관련된 변인 분석)

  • The purpose of this study was to identify how characteristics of children, parents, and the family context relate to parental disciplinary practices and to examine the effects of cumulative positive variables on effective parental disciplinary practices. Subjects were 220 each mothers and fathers of 6-year-old children. Data were analyzed with correlation, multiple regression, and chi-square. Both Maternal and paternal disciplinary practices were correlated with child birth order, child emotionality, attention span and persistence, perceived social support and quality of life, and retrospective punitive parenting. Perceived quality of life and social support were predictive of maternal disciplinary practices and child emotionality was predictive of paternal disciplinary practices. Both mothers and fathers exposed to several positive variables were much more likely to exhibit effective disciplinary practices than parents exposed to no positive variables. Findings were generally consistent with Belsky's(1984) process model of parenting.

  • Hanok with a long tradition in our country, but a significant period of modernization in the process of being cut off did not have continuity. Many of Hanok aging, according to the rapid residential development and life-changing was the subject of a complaint. Conversely, the benefits of Hanok was given to the advantages that compare to a monotonous mass housing of apartments. Despite these changes and the potential of these social needs and demands Hanok quickly did not respond: First, for the life of contemporary and traditional Hanok with a form of gap is a matter of space and style. Economy and lifestyle $20,000 for the era of Hanok was to develop a model for spatial configurations. Second, Hanok in a low density, is evaluated to aging, because increasing the economic utilization of land and tailored to their needs as a Hanok housing requires the development of a model, but this did not present a layered model. The purpose of this study is a modern residential Hanok persistence of this set to have 1) the spatial characteristics of traditional and modern urban life Hanok living space that meets the requirements of the degree of each other, to find sustainable elements, and 2) these demands the modern residential area type, combined with a set of Hanok 3) Korea Hanok cultural characteristics which set is created to residential housing types is to develop a basic research.

Mechanism of T cell exhaustion in a chronic environment

  • T cell exhaustion develops under conditions of antigen-persistence caused by infection with various chronic pathogens, such as human immunodeficiency virus (HIV) and myco-bacterium tuberculosis (TB), or by the development of cancer. T cell exhaustion is characterized by stepwise and progressive loss of T cell function, which is probably the main reason for the failed immunological control of chronic pathogens and cancers. Recent observations have detailed some of the intrinsic and extrinsic factors that influence the severity of T cell exhaustion. Duration and magnitude of antigenic activation of T cells might be associated with up-regulation of inhibitory receptors, which is a major intrinsic factor of T cell exhaustion. Extrinsic factors might include the production of suppressive cytokines, T cell priming by either non-professional antigenpresenting cells (APCs) or tolerogenic dendritic cells (DCs), and alteration of regulatory T (Treg) cells. Further investigation of the cellular and molecular processes behind the development of T cell exhaustion can reveal therapeutic targets and strategies for the treatment of chronic infections and cancers. Here, we report the properties and the mechanisms of T cell exhaustion in a chronic environment.

Simulation and Measurement of Degradation and Movement of Insecticide Ethoprophos in Soil (토양(土壤)중 살충제(殺蟲劑) ethoprophos의 분해성(分解性) 및 이동성(移動性)의 측정(測定)과 예측(豫測)에 관한 모델 연구(硏究))

  • The behaviour of insectcide ethoprophos (O-ethyl S,S-propyl phosphorodithioate) in soil was investigated. In a laboratory study, the degradation of ethoprophos in soil followed first-order reaction kinetics. The half-life of the insecticide in the soil incubated with 10, 18 and $25^{\circ}C$ was 12.4, 5.5 and 2.5 days, respectively. Arrhenius activation energy was 73.8 KJ/mole. The half-life was 46.4, 17.6 and 6.9 day in the soil with 7, 14 and 19% of soil water content, respectively. The moisture dependence B value in empirical equation was 1.67. The adsorption isotherm for ethoprophos in the soil agreed with freundlich equation. The adsorption distribution coefficient (Kd) was 0.27. In a field study prepared in autumn with undisturbed soil column in a mini-lysimeter system, ethoprophos residues were largely distributed in the top $0{\sim}2cm$ soil layer and moved down to the top 6cm soil layer. Persistence of ethoprophos in field soil was correlated with variation in weather pattern during the period of experiments. The half-life of ethoprophos treated at March and October was about 17 and 5 days, respectively. The ethoprophos woil was degraded up to 90% at 37day after the both treatment. In persistence and mobility of ethoprophos in field soil, the observed data were reasonably corresponded with predicted data by some computer model of pesticide behaviour.

Effect of Water, Organic Matter, and Lime on Degradation of Herbicide in Soil (토양중(土壞中) 제초제분해(除草劑分解)에 미치는 수분(水分), 유기물(有機物), 석회(石灰)의 영향(影響))

  • The persistence of butachlor and nitrofen in different soil conditions applied organic matter, lime, and other pesticides was studied under submerged and field moisture capacity. Degradation of the herbicides in soil was significantly retarded by autoclaving the soil and half-life of nitrofen was much longer than that of butachlor under this condition. Submerging the soil enhanced degradation of the herbicides, in particular that of nitrofen. On the other hand, half-life of nitrofen under field moisture capacity was twice longer than that of butachlor. Increased amendment of rice straw to the soil shortened the half-life of nitrofen under submerged soil, however it prolonged that of butachlor when the amendment was exceeded 1000kg/10a level. Liming the soil stimulated herbicide decomposition in the soil, which appears to be pH independent. Butachlor degradation in submerged soil was slightly stimulated by simultaneous application of fungicides and insecticides, but nitrofen persistence was not influenced.

  • The goal of this study is to analyze the problem and performance of life sympathy policy and to present development plan. For this, literature, and empirical research were performed. The analysis showed the problem of life sympathy policy in the following manner. That is the confusion of the mix of department, the lack of publicity, the uncertain of persistence and the lack identity of the monitor, the absence a unified operating instructions on the monitor, the insufficient to activities and operations housewife monitors etc. To vitalize life sympathy policy and operational needs of aspects of the institutional and the activation. First, The task of activation of the institutional challenge is to modify the related law, strengthen the organization, strengthen the personnel, expand the scope of participation monitors. The improvement of the operational aspects of the policy include strengthening integration and connectivity, the construction of policy promotion environment that reinforce feedback and outcome of life sympathy policy, And the energization of on-line activities needs the reinforcement of the operating system, etc. Through the activation of the sector policy, the application of the idea of housewives, and the participation of government policies will be a lot of contribution to the development of South Korea in the 21st century.
