Intragastrically Applicated CCl4-Thiopental Sodium Enhanced Lipid Peroxidation and Liver Fibrosis (Cirrhosis) in Rat: Malonedialdehyde as a Parameter of Lipid Peroxidation Correlated with Hydroxyproline as a Parameter of Collagen Synthesis (Deposition)

  • We investigated the pathogenesis of liver tissue damage during the lipid peroxidation and fibrogenesis with the observation of correlations between the parameters of collagen synthesis (and deposition) and lipid peroxidation in liver fibrosis (cirrhosis) rats. Rats were randomly divided into two groups, normal and $CCl_4$-thiopental sod. intoxicated group. And the one group was treated intragastrically with the mixture of $CCl_4$-thiopental sod. 3 times per week for 3 weeks. The liver tissue and sera were used for the measurement of hydroxyproline (HYP), malonedialdehyde (MDA) and superoxide dismutase (SOD). Biochemical parameters such as aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), total-bilirubin and blood urea nitrogen (BUN) were measured. Additionally, the expression of collagen ${\alpha}1$(III) and $\beta$-actin mRNA was observed by RTPCR. The histological change in liver tissue was also observed by Masson's trichrome and H&E staining. Correlation analysis was carried by Spearman's rho method. All biochemical parameters except total-bilirubin were significantly higher in the $CCl_4$-thiopental sod. treated group than that of the normal group (p < 0.01). In the $CCl_4$-thiopental sod. treated group, Hyp as a parameter of collagen synthesis (deposition) and MDA as a metabolite of lipid peroxidation, were significantly elevated by 1.98 and 2.11 times higher than that of the normal group (p < 0.001) respectively. The activity of SOD in the $CCl_4$-thiopental sod. treated group is decreased significantly by 44.8% (p < 0.001). And collagen ${\alpha}1$(III) mRNA was more expressed in the $CCl_4$-thiopental sod. treated group than that of the normal group. However, the expression of $\beta$-actin mRNA is showed similar in both of groups. A good correlation was observed between the content of hyp and MDA concentration (r = 0.70, n = 40) in the two groups. And the correlation between the levels of hyp and SOD (r = -0.71, n = 25) is also reliable. However, no correlation were observed between MDA concentration and SOD (r = -0.40, n = 25) in the two groups. Elevated levels of MDA in $CCl_4$-thiopental sod. treated rats indicated enhancement of lipid peroxidation, which is accompanied by a decrease in SOD activity. Moreover, we could confirm that the parameters of collagen synthesis (and deposition) is in good correlation with the metabolite of lipid peroxidation (MDA) and the lipid peroxidation antagonizing enzyme (SOD). Hence, we propose that (1) lipid peroxidation and collagen synthesis (and deposition) could be enhanced by intragastrically application of $CCl_4$-thiopental sod. during a short terms. And (2) the intoxication of $CCl_4$-thiopental sod. could be used for monitoring of lipid peroxidation and collagen synthesis (and deposition) for test of antioxidant and antifibrotic agent.

  • This study was undertaken to determine Juglandis Semen extraction (JS) has a protective effect against the cell injury caused by oxidants, t-butylhydroperoxide (t-BHP) and $H_{2}O_2$ in rabbit lung slices. Cell injury was estimated by measuring tissue water content and peroxidation of membrane lipids was assessed by measuring malondialdehyde (MDA), an end-product of lipid peroxidation. t-BHP significantly increased water content in lung tissues over concentrations of 2-10 mM, and such effects were prevented by 5% JS. JS exerted the beneficial effect in a dose-dependent manner. $H_{2}O_2$ (100 mM) also increased water content in tissues, which was almost completely prevented by 5% JS. t-BHP induced lipid peroxidation in a dose-dependent fashion in lung tissues over concentrations of 0.5-10 mM. JS significantly reduced t-BHP induced lipid peroxidation and oxidant-independent endogenous lipid peroxidation, and such effects were dose-dependent at concentration of 0.5-10%. JS prevented $H_{2}O_2$ (100 mM)-dependent lipid peroxidation. These results suggest that JS prevents ceil injury induced by oxidants in the lung, and such effects may be attributed to inhibition of lipid peroxidation. The precise mechanisms remains to be explored.

The Secondary Effects of Pencycuron on the Formation of Giant Protoplasts and the Lipid Peroxidation of Rhizoctonia solani AG4

  • The secondary effects of pencycuron on cell membrane of Rhizoctonia solani AG4 were investigated by the observation of giant protoplast formation and lipid peroxidation. Compared to protoplasts of R. solani R-C (sensitive strain) and Rh-131 (non-sensitive strain) increased in their size by 2.0-3.5 times 12 h after incubation in potato-dextrose broth containing novozyme (7 mg/$m\ell$) and $\beta$-glucuronidase ($60\mu\textrm{g}/$\textrm{ml}) with 0.6 M mannitol (pH 5.2). The increase of protoplast size in R-C was slightly inhibited from $13.8\textrm{mg}/\textrm{ml}$ without pencycuron to 10.3 ${\mu}{\textrm}{m}$ with 1.0$\mu\textrm{g}$/$m\ell$ of pencycuron. However, the size of giant protoplast of Rh-131 was not affected by the pencycuron treatment. Both strains R-C and Rh-131 did not exhibit the lipid peroxidation 12 h after the application of 1.0 $\mu\textrm{g}$/$m\ell$ pencycuron. The remarkable peroxidation of membrane lipid was observed only in R-C 24 h after pencycuron application, but not in Rh-131. Althought the inhibition of giant protoplast formation and the membrane lipid peroxidation were observed only in the sensitive strain R-C by pencycuron, it is difficult to conclude that these are the primary mechanism of pencycuron. The mild activity of pencycuron on the inhibition of giant protoplast formation and late membrane lipid peroxidation in the fungicide-sensitive strain did not noincid with the dramatic activity of pencycuron in R. solani. Therefore, our results suggest that inhibition of giant protoplast formation and membrane lipid peroxidation is the secondary effect of pencycuron.

Lipid Peroxidation and Antioxidant Vitamin Status in Oral Cavity and Oropharyngeal Cancer Patients

  • Background: This study was conducted to determine levels of lipid peroxidation and antioxidant vitamin status in patients with oral cavity and oropharyngeal cancer. Methods: The study group consisted of a total number of 80 subjects between the age 40-68 years, 40 with clinically and histopathologically proved cases of oral cavity and oropharyngeal cancer and 40 normal healthy, age and sex matched volunteers as controls. Levels of lipid peroxidation products as malondialdehyde (MDA) and antioxidant vitamins as vitamin A and vitamin C were estimated and compared between the two groups. Results: There was a statistical significant difference in the mean MDA, plasma vitamin A and vitamin C in the oral and oropharyngeal cancer patients compared with the healthy controls (p<0.0001). Conclusions: Lipid peroxidation (MDA) is higher and plasma antioxidant vitamins like vitamin A and vitamin C were lower in oral cavity and oropharyngeal cancer patients than healthy controls.

마우스에서 dibutyl phthalate 급성 투여가 간 지질과산화와 gamma-glutamyl transferase 활성에 미치는 효과 (Effects of acute dibutyl phthalate administration on hepatic lipid peroxidation and gamma-glutamyl transferase activity in mice)

  • Dibutyl phthalate (DBP) is used extensively in the plastic industry and has been known as an endocrine disruptor. Present study was undertaken to examine whether DBP can induce oxidative stress in mice. In this study, oxidative stress was measured in terms of the modification of lipid peroxidation and gamma-glutamyl transferase (GGT) activity. The serum toxicity index, level of lipid peroxidation and triglyceride (TG), and activity of GGT were measured in male ICR mice after a single administration of DBP (5 g/kg, po). DBP did not alter serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatinine, glucose and cholesterol level. However, the treatment with DBP was found to significantly increase the level of lipid peroxidation in liver and lung. The TG content and activity of GGT in the liver of DBP-exposed animals was also increased. These results indicate that DBP can induce mild oxidative stress in mice. The GGT activity is considered to be increased as one of the adaptive defense mechanisms to oxidative stress induced by DBP.

Schisandra Chinensis Inhibits Oxidative DNA Damage and Lipid Peroxidation Via Antioxidant Activity

  • Schisandra chinensis have been traditionally used in Asia for the treatment of dyspnea, cough, mouth dryness, spontaneous diaphoresis, nocturnal diaphoresis, nocturnal emission, dysentery, insomnia and amnesia. The purpose of this study is to evaluate the protective effects of Schisandra chinensis on oxidative DNA damage and lipid peroxidation induced by ROS in non cellular and cellular system. DPPH radical, hydroxyl radical and hydrogen peroxide scavenging assay were used to measure the antioxidant activities. Phi X-174RF I plasmid DNA cleavage assay and intracellular DNA migration assay were used to evaluate the protective effect on oxidative DNA damage. MTT assay and lipid peroxidation assay were used for evaluating the protective effect on oxidative cell damage. It was found to scavenge DPPH radical, hydrogen peroxide and hydroxyl radical and it inhibited oxidative DNA damage, lipid peroxidation and cell death induced by hydroxyl radical. These data indicate that Schisandra chinensis possesses a spectrum of antioxidant and DNA-protective properties

Lipid Peroxidation and Its Toxicological Implications

  • Lipid peroxidation is a free radical oxidation of polyunsaturated fatty acids such as linoleic acid or arachidonic acid. This process has been related with various pathologies and disease status mainly because of the oxidation products formed during the process. The oxidation products include reactive aldehydes such as malondialdehyde and 4-hydroxynonenal. These reactive aldehydes can form adducts with DNAs and proteins, leading to the alterations in their functions to cause various diseases. This review will provide a short summary on the implication of lipid peroxidation on cancer, atherosclerosis, and neurodegeneration as well as chemical and biochemical mechanisms by which these adducts affect the pathological conditions. In addition, select examples will be presented where antioxidants were used to counteract oxidative damage caused by lipid peroxidation. At the end, isoprostanes are discussed as a gold standard for the assessment of oxidative damages.

  • Polygonum aviculare L. was extracted by methanol and then fractionated systematically with solvents and column chromatographic method in order to isolate ingredients with anti-lipid peroxidation and liver protectective effects. Among these fractions, ethylacetate soluble part(MWE) showed the strongest in the anti-lipid peroxidation. Furthermore, the crude subtraction(MWE 4) with Rf value near 0.42, which is separated from MWE by column chromatography using a solvent system, inhibited the lipid peroxidation of rat liver in vitro. Moreover, MWE 4 decreased GOT, GPT and TBA value compared with control and suggested high protective effects against hepatotoxicity induced by carbon tetrachloride in mice. The active compounds in MWE 4 were assumed to be flavonoid glucosides.

Effect of Sex Hormones on Lipid Peroxidation in Rat Liver

  • The role of sex homones in hepatic lipid peroxidation, and in hepatic adehyde odidase and xanthine oxidase activites were investigated using rat liver homogenates. It was observed that male rt had a significantly greater content of malondialdehyde in liver than female. Among the sex hormones tested, estradiol, one of female hormones, markedly inhibited the formation of lipid peroxides in liver tissues in vitro. Especially, the inhibitory effect of estradiol appeared more remarkably in Fe-induced lipid peroxidation. The hepatic xanthine oxidase activity was decreased about 15% by $10\;^6\;M$ estradiol, wherease, the adehyde oxidase activity was almost completely disappeared at the same concentration of estradiol. It implies that sex differences in lipid peroxidation is attributed to the suppression of radical generating system by estradiol.

Antioxidant Activity of Vegetables and Their Blends in Iron -Catalyzed Model Systems

  • Vegetables are known to contain high amounts of natural antioxidants such as ascorbate, $\alpha$-tocopherol, $\beta$-carotene, and flavinoids. The antioxidant activities of several vegetables including broccoli, carrot , green pepper, spinach and tomato, and their blends were investigated using various iron-catalyzed lipid peroxidation systems. In linoleic acid micelles, carrot and spinach significantly inhibited lipid peroxidation by 29.0% and 35.8% , respectively (p<0.05).Blends of two, three , or four vegetables indluding spinach increased the inhibitory effect on lipid peroxidation, mainly due to high level of antioxidants in spinach. In beef homogenates, tomato significantly inhibited lipid peroxidation by 19.9%(p<0.05), whereas spinach and broccoli significantly stimulated lipid peroxidation by 67.3% and 11.5%, respectively (p<0.05). In the presence of 100$\mu$M ferrous ions, all vegetables inhibited degradation of deoxy-ribose by 43.6~77.6%(p<0.05). In the presence of 100$\mu$M ferric ions , broccoli and spinach stimulated deoxyribose degradation by 39.8% and 55.8%, respectively. These results indicate that the antioxidant activity of vegetables varied with the different model systems and depended on the provided environment such as iron content and substrates. The activity of the various combinations (blends) of vegetables was strongly related to that of the individual vegetable.
