  • It was attempted to observe the effect of garlic on the hepatic glutathione s-transferase and glutathione peroxidase activity in this study. Glutathione s-transferase (EC 2.5.1.18) are thought to play a physiological role in initiating the detoxication of potential alkylating agents, inclnding pharmacologically active compounds. Glutathione peroxidase (EC 1. 11. 1. 9) might play an important role in the protection of cellular structures against oxidative challenge. The activities of glutathione s-transferase and glutathione peroxidase in rat liver were increased by the treatment of garlic juice. Allicin fraction, heat-treated allicin fraction and garlic butanol fraction markedly inhibited glutathione s-transferase activity in vitro, whereas glutathione peroxidase activity was significantly increased in heat-treated allicin fraction and garlic butanol fraction.

  • Peroxidase as one of the organic enzyme catalyst is useful for the oxidation treatment of various aromatic compounds such as phenols. The peroxidase content of Oenanthe javanica was 24.85 unit/g-fw in leaf, 5.74 unit/g-fw in stem, and 34.69 unit/g-fw in root respectively. The crude peroxidase extracted from Oenanthe javanka can be kept under low temperature (-70$\circ$C) condition for 6 months with the maximum 1% activity reduction. The optimum conditions of removal for 100 ppm phenol was pH 6, hydrogen peroxide 3.5 mM, peroxidase activity 8 unit/ml, temperature 20$\circ$C respectively. In the wide range of concentration from 50 ppm to 750 ppm phenol reveals average 54% removal rate under the same peroxidase activity (8 unit/ml) and different amount of hydrogen peroxide proportional to phenol concentration. Especially at the concentration of 100 ppm the maximum phenol removal rate was 72%.

  • This study was prepared to observe the change of enzyme activities in kidney treated with red ginseng extract in the gamma ray irradiated mice. Determine the activity of SOD, peroxidase, catalase in the kidney a period of 1 day, 2 day, 3 day, 4 day, 5 day after a saline injection or injection of red ginseng extract or gamma ray irradiatied group into four classify. The activity SOD and catalase showed a tendency to increase and recovery at the early state but pay no regard. Wherease, the activity of peroxide restored and increased pay regard. A physiological saline injection group after gamma ray irradiation showed a tendency to diminish after remakable increase of activity of SOD, peroxidase and catalase than control group. Injection group of red ginseng extract after gamma ray irradiation observed rapid recovery on activity of SOD, peroxidase, catalase than a saline injection group. Experimental result suggested that injection of red ginseng extract after irradiation have the recovery effect on the changed of activity of SOD, peroxidase and catalase against radiation injury.

  • The present study was carried out to investigate the change of peroxidase activity of soybean seed infected with Cerrospora kikurhii. The protein content, polyphenol oxidase activity and peroxidase isozyme pattern in health and infected soybean seed were also compared. 1. The peroxidase activity was substantially higher in the infected soybean seeds than that in the healthy seeds either cracked or not. No significant differences in protein content were recognized among the seeds tested. 2. No significant differences in peroxidase activities and protein contents were notified between healthy and infected seeds from the measurements on each parts of dissected seeds, cotyledon and seedcoat, however the peroxidase activity in the seed coat of the stained seed was 2.5 times to healthy seed. 3. The activities of polyphenel oxidase were undectable in both healthy and diseased seeds. 4. The electrophoretic Patterns of the Peroridase isozyme were the same between healthy and in footed seed. 5. Therefore, the increase of peroxidase activity in infected soybean seedcoat was mainly due to the biochemical reaction against the pathogen.

  • In early maturation stages of Paragonimus westermani (metacercariae, 4-, 8-, 12-week old worms), activities of antioxidant enzymes, such as superoxide dismutase, catalase, peroxidase and glutathione peroxidase, were examined. Specific activity of catalase was the highest in metacercariae and decreasing with age. That of superoxide dismutase was higher in metacercariae and 4-week worms. Specific activity of peroxidase was at its peak in 4-week worms while that of glutathione peroxidase was in 8-week worms. Specific activities of all these antioxidant enzymes were decreased to their lowest in 12-week old adults.

  • In order to study the antioxidative action of the effectient garlic components, peroxidase and superoxide dismutase activity were compared through the in vitro and in vivo experiments. RESULTS : 1. Observing the effects on peroxidase activity of efficient components in vitro, garlic oil, alliin and ethanol fraction showed effective, which was similar to the trend of TBA value, peroxide value and induction time for the first period of lipoperoxide formation in vitro. 2. In vivo experiment with peroxidase activity, the ethanol fraction and garlic oil were effective when intraperitoneally administered as well as orally administered. 3. Considering the superoxide disutase activity in vitro, the garlic oil, alliin and ethanol fraction were effective in efficient components. But non-kaolin fraction inhibited the activity on the contrary. 4. In terms of the efefcts on superoxide dismutase activity in vivo, alliin and garlic oil wee effective in intraperitoneal adminstraton and the ethanol fraction and alliin in oral administration.

Influence of Hyaluronic Acid on the Different Levels of Lysozyme and Peroxidase in the Aspects of Candidacidal Activities

  • Purpose: The purpose of the study was to investigate the influences of hyaluronic acid on the candidacidal activities of lysozyme, the peroxidase system, and the glucose oxidase-mediated peroxidase (GO-PO) system at different concentrations of antimicrobial enzymes. Methods: Hyaluronic acid was used at a final concentration of 0.5 mg/mL. Hen egg-white lysozyme (HEWL) was used at concentrations ranging from 10 to $100{\mu}g/mL$. The peroxidase system included bovine lactoperoxidase (bLPO), potassium thiocyanate (KSCN, 1 mM), and hydrogen peroxide ($100{\mu}M$). The GO-PO system included bLPO, KSCN (1 mM), glucose oxidase (10 units/mL), and glucose ($30{\mu}g/mL$). The final concentration of bLPO in the peroxidase and GO-PO systems ranged from 12.5 to $100{\mu}g/mL$. Candida albicans strains ATCC 10231, 11006, and 18804 were utilized. Candidacidal activities of antimicrobials and the influence of hyaluronic acid on their candidacidal activities were determined based on colony forming units. Results: Candidacidal activities of the peroxidase and GO-PO systems increased with increasing concentrations of bLPO. This tendency was the same in the presence or absence of hyaluronic acid. Candidacidal activity of HEWL was not significantly concentration-dependent. Candidacidal activities of the GO-PO system were higher than those of the corresponding peroxidase system. Candidacidal activity was inhibited in the presence of hyaluronic acid in the following order: HEWL, the peroxidase system, and the GO-PO system. Conclusions: Hyaluronic acid inhibited the candidacidal activities of HEWL, the peroxidase system, and the GO-PO system. The GO-PO system exhibited better candidacidal activity than HEWL and the peroxidase system both in the presence and absence of hyaluronic acid.

Effect of NADH-Dependent Enzymes Related to Oxygen Metabolism on Elimination of Oxygen-Stress of Bifidobacteria (NADH요구 산소대사관련 효소가 bifidobacteria의 산소스트레스 제거에 미치는 영향)

  • Selection of oxygen-tolerant strains and elucidation of their oxygen tolerance mechanism were crucial for effective use of bifidobacteria. Oxygen-tolerant bifidobacteria were able to significantly remove environmental oxygen (oxygen removal activity) as compared to oxygen-sensitive strains. Most oxygen removal activity was inhibited by heat treatment and exposure to extreme pH (2.0) of bifidobacterial cell. NADH oxidase was major enzyme related to oxygen removal activity. Oxygen-tolerant bifidobacteria possessed high NADH peroxidase activity level to detoxify $H_2O_2$ formed from reaction of NADH oxidase. Addition of oxygen to anaerobic culture broth significantly increased activities of HADH oxidase and NADH peroxidase within 1hr and rapid increment of oxygen concentration was prevented. Results showed NADH oxidase and NADH peroxidase of oxygen-tolerant bifidobacteria played important roles in elimination of oxygen and oxygen metabolite $(H_2O_2)$.

Effects of the Foliar Applied Acifluorfen on the Peroxidase Activity in Leaf Tissue of Soybean (Glycine max Merr.) and Crabgrass (Digitaria adscendens (L.) Scopol.) (Acifluorfen 의 경엽처리(莖葉處理)가 대두(大豆) 및 바랭이의 엽조직(葉組織)에서 Peroxidase 활성(活性)에 미치는 영향)

  • This study was carried out to investigate the specific activity fo peroxidase(POD) in conjunction with the phytotoxic effect of the tolerant soybean(Glycine max Merr.) and the susceptible crabgrass(Digitaria adscendens(L.) Scopol.) to acifluorfen. The POD activity increased during the growth of soybean and crabgrass seedlings. But after the foliar application of acifluorfen, the POD activity in the treated leaves was considerably higher than in controlled leaves, suggesting that soybean had higher tolerance to acifluorfen with growth. All the treated soybeans recovered from herbicidal injury in 14 days after treatment, while all the treated crabgrasses were dead with extreme necrosis and leaf burning. It was measured that the POD activity of crabgrass increased abruptly in 1 to 2 days after treatment and then significantly decreased in 5 days.

Effects of Diallyl Disulfide on the Hepatic Glutathione Peroxidase Activity in Rat (흰쥐 간 Glutathione peroxidase 활성에 미치는 Diallyl disulfide의 영향)

  • Glutathione peroxidase might play an important role in the protection of cellular structures against oxidative challange by hydrogen peroxide and several organic hydroperoxides. It is widely accepted that allicin is biological active component of garlic, and allicin is easily degraded to diallyl disulfide and other components. This study was attempted to elucidate the effect of diallyl disulfide on some biological activities. It was observed that the activity of serum transaminase and glutathione level in liver were not changed by the treatment of diallyl disulfide. The liver cytosolic glutathione peroxidase activity was significantly enhanced. Whereas, mitochondrial enzyme activity was slightly increased. In the presence of diallyl disulfide in vitro, $V_{max}$ value of glutathione peroxidase for hydrogen peroxide was increased. On the other hand, Km value was not changed.
