Detection of Alternaria spp. in Carrot Seeds and Effect of the Fungi on Seed Germination and Seedling Growth of Carrot

  • Five seed samples of carrot were tested to detect Alternaria spp. by blotter method. A. alternata and A. radicina were detected from all the seed samples as high as $25.8-70.5\%$ and $37.5-63.5\%$, respectively. A. dauci was detected from four seed samples as low as $0.5-7.5\%$. The three Alternaria spp. were detected from the pericarp and the seed coat and endosperm of the carrot seeds but not from the embryo by component plating test. A. alternata and A. radicina were much more detected from the pericarp than the seed coat and endosperm. A. dauci was detected from the pericarp and the seed coat and endosperm at similar rate. The seed sample which was most severely infected with A. radicina showed the lowest rate of germination in the test on top of paper (TP). In the TP test, differences in total infection rate of A. radicina and A. dauci of the seed samples were very closely correlated with those in incidence of seedling rot on the seed samples. However, there was no correlation between infection rate of A. alternata and rate of germination or seedling rot of the seed samples. Soil test for seedling growth revealed that there was no correlation between differences in total infection rate of A. radicina and A. dauci and those in rate of normal seedlings of the seed samples.

Antioxidative Activity and Varietal Difference of Cyanidin 3-glucoside and Peonidin 3-glucoside Contents in Pigmented Rice

  • The content of anthocyanin, cyanidin 3-glucoside (C3G) and the peonidin 3-glucoside (P3G) from 591 rice cultivars and the antioxidative activities of MeOH extract from 8 rice cultivars were evaluated. Among them, C3G content of pigmented rice were ranged from 0 to 451.9mg in 100 g brown rice, while the P3G contents were in the range from 0 to 42.7mg in 100g brown rice. There was no correlation between C3G and P3G content. Total anthocyanin content ranged from 0 to 475.1mg in 100g brown rice. The antioxidative activity, the scavenging activity on DPPH (2,2-diphenyl-picryl-hydrazyl) radical of MeOH extracts from rice grain, were different according to cultivars. The activity of blockish purple pericarp rice cultivars was twice stronger than that of white pericarp cultivar. Especially, the antioxidative activity of Heugjinjubyeo was four times stronger than that of white pericarp cultivar. The scavenging effect on DPPH radical in rice extracts was related to the total anthocyanin contents of the extracts.

Studies on the Changes of Vitamin C content in the Hot Green Pepper Fruits during the Circulation Periods (풋고추의 유통과정중 Vitamin C 함량의 변화)

  • This study was carried out to observe the changes of Vitamin C content and to preserve the quality for the circulation periods of hot green pepper fruits wrapped with polyethylene film under the indoor temperature, as well as to find the proper time for harvest of hot green pepper fruits as a hot taste food and vegetable. the results obstained were as follows: 1. The consignment rate of hot green pepper fruits was highest, 60.2% on the 25th day after flowering, the nest 25.4% on the 20 th day, 10.3% on the 30th day and 4.1% on the 35th day. 2. the content of Vitamin C I each pepper fruits was highest o the 30th day, considerably high on the 25th day and 20th day. 3. The total content of Vitamin C in each part of hot green pepper fruits; the content in pericarp was much higher than in placenta and seed, and that of hydro type Vitamin C was almost the same. 4. The reduction rate of vitamin C during circulation periods was highest on the 3rd day; 42% in pericarp, 50% in placenta, 65% in seed. But in the package wrapped up with polyethylene film, the reduction rate in the same period was 5% in pericarp, which was very low in reduction, and those in placenta and seed were almost the same as in the unwrapped state.

XVIII. Study on Waxy Corn Characteristics of corn hybrids on second cropping of using green house (XVIII. 찰옥수수연구 시설하우스에서 2기작 찰옥수수 교잡종의 주요 생육특성 비교)

  • The major characteristics of the developed corn hybrids were compared at each different planting time; 1st time(April 28) and 2nd time(August 20), respectively. Among traits surveyed, stem height showed highly at 1st time than 2nd, while ear height, stem diameter and ear length were similar at two times. Sugar content of the used hybrids showed highly at 2nd planting time than 1st. Pericarp thickness was also high at 2nd planting time. Especially, among hybrids CNU99 was the highest in stem height and ear length, CNU11 and CNU56 were high in sugar content, and CNU51 and CNU124 were thiner than other hybrids in pericarp thickness. Accordingly, these hybrids will be planted and recognized in 2nd yield trial for superior corn hybrid selection.

Effects of polyphenol and anthocyanin contents of Punica granatum fruit parts on their antioxidant and anticancer activities (석류 과실 부위별 폴리페놀 및 안토시아닌 함량이 항산화 및 항암 활성에 미치는 영향)

  • The purpose of this study was to determine the content of phenolics and various anthocyanin compounds, and antioxidant and anticancer activities of the extracts from pomergranates, which were produced in two different regions including Goheung, Korea and California, USA. These pomergrantes were divided into juice, pericarp, and seed parts and each part was extracted with 95% methanol. Content of total phenolics [mg chlorogenic acid equivalents/kg DW] was highest in pericarp, followed by juice and seeds from pomergrantes in both regions. The anthocyanins identified in pomegranate fruit were cyanidin-3,5-diglucoside chloride (Cy3,5G), cyanidin-3-O-glucoside chloride (Cy3G), delphini din-3,5-di-O-glucoside chloride (Dp3,5G), delphinidin-3-O-glucoside chloride (Dp3G), pelargonidin-3,5-di-glucoside chloride (Pg3,5G), and pelargonidin-3-glucoside chloride (Pg3G). Among these, cyanidin-3,5-diglucoside chloride (Cy3,5G) was the major anthocyanin in California pomegranate fruit juice and Goheung pomegranate fruit pericarp. DPPH (1,1-diphenyl-2-picryl hydrazyl radical) free radical scavenging activity was dose-dependently increased, and was higher in pericarp part than juice or seed parts from pomergrantes in both regions. By MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay, pericarp extracts from pomergrantes in both regions showed the highest anticancer activity, which was higher on Calu-6 for human pulmonary carcinoma than SNU-601 for human gastric carcinoma. Correlation between polyphenols and anticancer activity on Calu-6 was determined to be in the range of $r^2=0.8904$ to 0.9706.

Genetic Analysis of Complementary Gene Interactions of Pb and Pp Genes for the Purple Pericarp Trait in Rice (흑미의 자색종자과피(Purple pericarp) 형질을 결정하는 상보적 유전자 Pb와 Pp 유전자들의 상호관계 분석)

  • The Purple pericarp (Prp) trait is a trait often bred for in black rice. Generally, the Prp trait is displayed in the color variations of seeds following the 9:3:4 purple, brown, and white ratio, respectively. The Prp trait is a recessive epistasis of two gene interactions; however, it is caused by the two complementation genes Pb and Pp. Here we present a study of the genetic characteristics of the Prp trait using an $F_1$ hybrid with a Pbpb Pppp genotype. This hybrid generated four seed colors with the following numbers: 3 dark purple, 6 medium purple, 3 brown, and 4 white (or 9 purple, 3 brown, and 4 white). However, further biochemical analysis of the all progenies divided them into two groups. One group had the Pb_ Pp_ allelic constitutions and contained cyanidin 3-O-glucoside (C3G) in both the dark purple or medium purple seeds. The other group, however, was absent of C3G in both the brown and white seeds, resulting in a ratio of 9:7, respectively. This segregation revealed the extended Mendelian 9:7 ratios of the complementary gene interactions with a good fitness in ${\chi}^2$ analysis. Further analysis revealed that brown seeds with the Pb_ pppp genotype corresponded with a null C3G, indicating that the Brown pericarp trait in rice is caused by a dominant allele of the Pb gene. Therefore, we conclude that the production of C3G is a main phenotype of the black and purple colored rice in the Prp trait, and it is governed by the complementary gene interactions between Pb and Pp genes.

Popping Mechanism and Shape Moulding Factor of Popcorn (튀김옥수수의 파열방향 및 튀김형태 결정요인)

  • Popped popcorn generally have a regular popping direction and typical shape. But the reason and mechanism are not clear yet. This experiment was carried out to investigate the shape moulding factor of popped popcorn. Pericarp thickness of tip-cap section of kernels is slightly thicker than that of top section and this fact provides the important information to the reason. Popping starts when the moisture pressure of heated popcorn is increased and reaches at the critical pressure. Therefore, in the same moisture pressure conditions, top sections are bursted first because their pericarp section is thinner than that of tip-cap section. At the very moment tip-cap sections pull down the top sections of peri carp as bi-metal does. So kernels which removed tip-cap section showed the irregular popping shape because they lost the tip-cap pericarp function. How-ever, kernels which removed embryo showed the typical popping shape but their popping volume was small due to emition and shortage of critical moisture pressure. But kernels which removed the whole pericarp and top pericarp were not popped at all because moisture was entirely emitting out of kernels. These results suggest that the shape moulding factor of popped popcorn is the pericarp thickness differences between the top and tip-cap section of kernels.

Anti-HIV and Antihepatotoxic Constituents from Medicinal Plant Resources

  • Medicinal plants were screened for the inhibitory effects on human immunodeficiency virus type 1 pretense. Of the extracts tested, the strong inhibitory effects were observed in the acetone extracts of the pericarp of Camellia japonica. Camelliatannin H from the pericarp of C. japonica showed a potent inhibitory activity on HIV-1 pretense. Effects of the extract and compound from leaves of Zanthoxylum piperitum on the enzyme activities were investigated in the liver of bromobenzene-treated rats. The methanol extract and protocatechuic acid isolated from Z. pipetitum reduced the activity of aniline hydroxylase that increased by bromobenzene, while did not affect the activities of aminopyrin N-demethylase and glutathione S-transferase. The extract and protocatechuic acid recovered significantly the activity of epoxide hydrolase decreased by bromobenzene.
