Studies on the structure and expression of penicillin G acylase gene I (Penicillin G acylase 유전자의 구조와 발현기작에 관한 연구 I)

  • The penicillin G acylase(pga) gene was cloned in the vector plasmid pKM $300(Ar^r,\;Tc^r,\;6.33kb)$ for the study of the structure and expression of the pga gene. This recombinant plasmid pPAKS-1 DNA(24.5 Kb) was cleaved into 2 fragments by restriction enzyme Eco R1.1fragment by BamH1, 4fragments by Hind III, and 2 fragments by Pst I. The pga gene was located on the Eco R1.Hind III-C fragement of pPAKS-1. The recombinant plasmids pPAKS-1 and pPAKS-2, in which the Hind III-B and Hind III-D fragments pPAKS-1 are deleted, are characterized. The results are summarized as follows : 1. Doubling times of bacterial strain bearing pPAKS-1 and pPAKS-2 are 90 and 60 minutes, respectively. 2. pPAKS-1 and pPAKS-2 are present at about 16-32 and 70 copies per cell, respectively, are 0.66 and 5.5 units, respectively, which represent 2-fold and 20-fold higher enzyme 4. pPAKS-1 is very unstable, but pPAKS-2 is stable.

  • In this study, we proposed a real-time ECG analytical method for predicting stress and dangerous heart condition using the ECG signal in playing AR/VR device. A real-time diagnosis is used as R-R interval based HRV(:Heart rate variability), BPM(:Beats Per Minitue) and autonomic nervous research with through mapping method of two-dimensional planes. The ECG data were analyzed every 5 minutes and derived from autonomic nervous system diagnosis.

  • In this study, th effective extraction of 5 vegetables, which have been shown to inhibit the growth of the aflatoxin production of Aspergillus parasiticus R-716, was investigated and their effects were examined. Radish, cabbage, garlic and zinger were effectively with water-chloroform, but crown daisy with n-hexane. Among them, water-chloroform extract of radish was remarkably effective, and garlic extract only inhibited the growth strongly. The growth and the aflatoxin production of the strain were showed 0.758g/25ml, 763ug/25ml with the addition of water-chloroform extract equivalent 15g of raw radish, and an increase in the level of radish extract resulted in a decrease both the growth and the aflatoxin production per mycerial weight.

  • The effect of allyisothiocyanate, the mahor compound of radish on the growth of Aspergillus parasiticus R-716 and aflatoxin production, was investigated. An increase in the level of allylisothiocyanate results in a decrease both growth and aflatoxin per myclial weight, and the addition of 125 ppm allylisothocynate completely inhibited the growth of the strain. The addition of allylisothiocyanate to the culture of R-716 strain the production of aflatoxin. The inhibition of aflatoxin was more B-group than G-group and M-group during cultural period. The growth of strain and aflatoxin production were greatly affected by the addition of allylisothiocyanate.

Market, Firm, and Project-level Effects on the Innovation Impact of FP RTD Projects

  • This paper explores the determinants of the innovation impact of publicly funded R&D projects along three broad dimensions, namely project, firm and market-related factors. In addition to these factors we examine the attributes of the research result per se and aspects of the commercialization process. The observations from empirical and qualitative analyses are based on R&D projects funded by the Fifth and Sixth Research Framework Programmes of the European Union. Firm size, prior experience, innovation culture, the nature of the project itself, explicit intension to commercialize, consortium management and strategy are the factors with the strongest effect on project success, defined in terms of product/process innovation and/or technical knowledge creation. The paper provides important implications for the organization, objectives, and management of public programmes that fund R&D and for project and participant selection.

  • Recently, diagnostic techniques have been investigated to defect a partial discharge in high voltage electrical equipment. We have studied the characteristics of the acoustic partial discharge originating from the electrical defects in cubicle GIS(C-GIS). An acoustic emission(AE) sensor is used on the enclosure to detect partial discharge source because the sensor is sensitive to stress waves in its frequency range that may not be from a partial discharge source. AE signal is analyzed with phase-magnitude-frequency number(${\Phi}$-V-n) and pulse per second(PPS). Experience result has shown that the omitted acoustic signal has phase dependency and phase shift characteristic according to increase with applied voltage. These result will be helpful to the pattern recognition of the acoustic partial discharge in a C-GIS.

Taxonomic characters of wild rats by isoenzyme analysis (조직내 효소형 분석에 의한 집쥐류의 분류학적 특성 조사)

  • Sixty wild rate (fifty eight of Rattus norvegicus and two R rattus) were caught from Seoul, Kyonggi, Kangwon, Honam, and Yongnam areas between August and October 1992. From liver homogenates of the wild rats, isoenzyme patterns were analysed by starch gel electrophoresis. Using 9 enzyme systems, eight electrophoretic types were identified among wild rats with genetic diversity per locus between 0.00 and 0.49 (Mean 0.15). R rattus from Kyonggi (Kanghwa) and Kangwon (Cholwon) were distinct from R norvegicus from nine regions with 0.581 in genetic divergence. Therefore genetic divergence was different not only in interspecies(0.581) but also in intraspecies(0.111~0.375). These data suggested that isoenzyme electrophoresis could be used as a potential application in taxonomic studies of wild rats.

  • First lactation records (174) of Jersey${\times}$Sahiwal and Holstein Friesian${\times}$Sahiwal half breds under 9 sires maintained at Chandra Shekher Azad University of Agriculture and Technology, Kanpur, Uttar Pradesh, India from 1975-1983, were used to estimate the genetic parameters and to predict herd life milk yield and average milk yield per day of herd life from first lactation traits. The traits included were: age at first calving, first service period, first lactation period, first calving interval, first lactation milk yield, milk yield per day of first calving interval, herd life milk yield, herd life and average milk yield per day of herd life. Most of the production and reproduction traits were found to have positive and significant correlations between them on genetic as well as phenotypic scales. Total twelve regression equations were fitted. The prediction equation of herd life milk yield in both the genetic groups showed linear relationship with AFC, FSP, FLP, FLMY and MY/DCI and was apparent and significant. Similarly, polynomials for milk yield per day of herd life for J${\times}$S and HF${\times}$S half breds also showed linear trend, which was found highly significant. The highest and lowest $R^2$ values were found for FCI and AFC, respectively.

SMALL SCALE DAIRYING IN THREE FARMING SYSTEMS IN EAST JAVA I. FARMER'S INCOME AND HOUSEHOLD CHARACTERISTICS

  • The annual income (gross margin) in 1989/90 of a sample of 274 farmers in seven milk cooperatives was analyzed in the sugar cane, cassava, and horticulture areas in East Java. On average dairying contributed 42%, crops 29% and off-farm revenue 29%. Dairy income was highest in the cassava area, where it compensated for the low crop income, and lowest in the sugar cane area. Farm area and average milk yield per day per cow correlated positively with farmer's income, whereas crop income increase significantly with farm area and with the number of cows. The level of total cost per cow had a negative impact on dairy and with the number of cows. The level of total cost per cow had a negative impact on dairy and on total income. Government officials and other professionals engaged in dairying had a significantly higher total income than those with their main occupation in dairying, cropping or working as farm labourers. Uneducated farmers obtained a significantly larger income through crops, whereas farmers with tertiary education obtained more income through off-farm work, This study suggests that more attention must be paid to the actual use of labour and the improvement of the dairy output/cost ratio.

The Effect of Tread-Pressure on the Growth of Plantago asiatica Linne (답압이 질경이(Plantago asiatica Linne)의 생장에 미치는 영향)

  • To investigate the distribution of Plantago asiatica Linne, almost, restricted on the road-side, the plant growth analyzed by experimental cultivation under the condition of tread-pressure; 10 times/day (7.33kg/$cm^2$ ), 20 times/day (12.27kg/$cm^2$), 30 times/day (20.05kg/$cm^2$ ) and the control plot (1.68kg/$cm^2$ ). The matter production in the 10-time-per-diem plot (7.33kg/$cm^2$ ) of tread-pressure howed more large value than that inthe control one (1.68kg/$cm^2$ ) and those in the 20-0time-per-diem plot (12.27kg/$cm^2$ ) and the 30-time-per-diem one (20.05kg/$cm^2$ ) small values than that of the control plot. Especially, the growth of roots and spikes was extremely repressed. The number of leaves, scapes and lateral buds tend to increase in the tread-pressure of 20 times/day (12.27kg/$cm^2$ ) and 30 times/day (20.05kg/$cm^2$ ). The water contents of roots along the increase of tread-pressure to continuosly decreased. By the increase of tread-pressure, T/R ratio and C/F ratio their group decreased. The plant growth (7.33kg/$cm^2$ ) in the 10-time-per-diem plot increased more than that in the control one is regarded as the effect of tread-pressure affection on the road-side distribution of this plant.
