Casein 효소 가수분해물의 항균 활성과 그 응용 (Application and Antimicrobial Activities of Casein Hydrolysates Treated with Asp.oryzae Protease)

  • 단백 효소 가수분해물의 천연 항균제로서의 응용성을 검토하기 위하여 casein에 5종의 단백질 가수분해 효소를 작용시켜 얻어진 가수 분해물의 항균활성을 측정하고 활성이 가장 높은 가수분해물을 부분 정제하여 그 응용성을 검토하였다. Casein에 5종 단백질 분해효소를 작용시켜 얻은 가수분해물의 항균활성은 Aspergillus oryzae protease에 의한 것이 가장 높았다. 효소처리에 의하여 얻어진 가수분해 물을 30,000, 10,000, 3,000 membrane filter로 한외여과 하였을 때 항균활성은 3,000이하 분획물에 대부분 함유되어 있었으며 공시균주에 대한 최소저해농도는 $1.0\~1.5\;mg/mL$이었다. Aspergillus oryzae pretense로 작용시킨 casein 가수분해물은 $121^{\circ}C$에서 10분간 가열했을 때도 그 항균 활성을 유지하는 것으로 보아 열에 안정하였다. 가수분해물을 HPLC로 220 nm 280 nm에서 검출된 peak 별로 수집하여 항균활성을 측정 한 결과 retention time 12.6, 13.2 분에서 분취된 peptide가 활성이 있었다. 가수분해 동결건조물을 된장에 첨가하였을 때 미생물의 생육이 저해되었으며 실용성이 있었다.

  • This study aimed to determine the degree of hydrolysis and angiotensin-I-converting enzyme (ACE)-inhibitory activity of Giant Jellyfish Nemopilema nomurai (jellyfish) hydrolysates. The degree of hydrolysis using six proteolytic enzymes (Alcalase, Flavozyme, Neutrase, papain, Protamex, and trypsin) ranged from 13.1-36.8% and the inhibitory activities from 20.46-79.58%. Using papain hydrolysate, we newly isolated and characterized ACE-inhibitory peptides with a molecular weight of 3,000-5,000 Da that originated from jellyfish collagen. The purified peptide (FII-b) was predicted to be produced from an alpha-2 fragment of the type IV collagen of jellyfish. The N-terminal sequence of FII-b was Asp-Pro-Gly-Leu-Glu-Gly-Ala-His-Gly- and showed 87% identity to the collagen type IV alpha-2 fragment of Rattus norvegicus and a predicted protein from Nematostella vectensis, indicating that the ACE-inhibitory peptide originated from the collagen hydrolysate and had an $IC_{50}$ value of 3.8 ${\mu}g$/mL. The primary structure of the fragment is now being studied; this peptide represents an interesting new type of ACE inhibitor and will provide knowledge of the potential applications of jellyfish components as therapies for hypertension.

말쥐치 단백의 효소 가수분해물의 특성에 관한 연구 (Studies on the Properties of Enzymatic Hydrolysates from File-fish)

  • Pronase 등 8개의 단백분해효소를 사용하여 말쥐치를 가수분해시 생성되는 펩타이드의 양을 측정한 결과, bromelain과 neutrase에 의한 가수분해물의 펩타이드 생성량은 4시간 가수분해시 6.13 mg/ml와 6.01 mg/ml로 높은 생성량을 보였으며, 유리아미노산의 생성량은 4시간 가수분해시 alcalase에 의한 가수분해물이 4.20 mg/ml로 높은 유리 아미노산의 생성량을 보였다. 가수분해도를 측정한 결과, 4시간 가수분해시 esp/sav와 alcalase의 가수분해물은 88.9%와 86.2%의 비교적 높은 가수분해도를 보였다. 또한 핵산관련 물질은 5'-GMP가 다른 핵산 관련물질에 비해 높은 생성량을 보였다. 가수분해물의 펩타이드의 평균길이는 $8.5{\sim}14.5$로 bromelain에 의한 가수분해물이 가장 짧은 펩타이드 길이를 가진 반면, neutrase에 의한 가수분해물은 14.5로 가장 긴 펩타이드로 구성되었다.

Peptic Hydrolysate of Porcine Crude Myosin Has Many Active Fractions Inhibiting Angiotensin I-converting Enzyme

  • In order to clarify one of the biological functions of pork, we investigated whether a peptic hydrolysate of denatured porcine crude myosin showed inhibitory activity against angiotensin I-converting enzyme (ACE), which contributed to hypertension. Our results indicated that this hydrolysate showed relatively strong activity, and we therefore attempted to separate the involved peptides, which were considered to be active substances. To isolate these active peptides, the hydrolysate was separated using a solidphase separation, gel filtration high-performance liquid chromatography (HPLC), and two kinds of reverse phase HPLC. In each stage of separation, many fractions were detected, almost all of which showed ACE inhibitory activity. Thus, we suggested that the activity of the hydrolysate as a whole was a result of the activities of the many individual peptides. Six peaks were distinguished, with yields from 34 to 596 ppm of original crude myosin. In addition to the six peaks, many other active fractions were found throughout the separation steps, strongly suggesting that whole porcine crude myosin itself had ACE inhibitory activity. Moreover, pork as food was considered to function as an ACE inhibitory material in vivo, because pork proteins consist primarily of crude myosin, which included almost all the myofibrillar structural proteins.

명태단백 가수분해물 제조 및 plastein의 합성 (Production of protein hydrolysate and plastein from alaska-pollack)

  • 명태육의 식품가공적성과 이용성을 높이기 위해 pronase에 의한 명태단백의 가수분해조건과 fruit-와 stem-bromelain에 의한 plastein의 합성 반응조건을 검토하였다. Pronase 가수분해 최적조건인 반응pH 7.0, 반응온도 $40^{\circ}C$, 기질 g당 효소첨가량 1,000units 및 반응시간 4시간에서 89% 분해도를 보이는 명태단백의 가수분해물을 제조하였다. Plastein은 pH가 각각 5.0(stem-bromelain)과 7.0(fruit-bromelain)으로 조정된 기질 30% 용액에 bromelain 1%를 가하여 $40^{\circ}C$, 24시간 진탕반응하여 합성하였다. 이 최적조건에서 합성된 plastein은 각각 22.6과 20.8 아미노산 잔기를 가진 펩타이드로 구성되어 있었으며 관능검사에 의해 반응초보다 크게 쓴맛이 제거되는 결과를 나타내었다.

해조류 효소가수분해물질로부터 정제한 저분자 Peptide의 기능성 (Functional Activities of Low Molecular Weight Peptides Purified from Enzymatic Hydrolysates of Seaweeds)

  • 동해안 특산 해조류의 식품 또는 의약품재료로의 이용가능성을 검토하기 위하여 소화효소(pepsin)로 가수분해한 다음 저분자peptide를 정제하여 여러 가지 기능성을 연구하였다. 쇠 미역, 파래 및 지누아리의 효소가수분해물은 Bio-Rad P2 gel chromatography 상에서 3개의 peptide peak를 나타내었으나 김은 2개의 peak를 나타내었다. 항산화활성은 김 peak 1이 가장 높았으며 그 다음으로 김 peak 2 및 쇠미역 peak 2 순으로 높았다. ACE 저해활성은 김 peak 1, 파래 peak 3 및 peak 2순으로 높았으며, 항갈변활성은 김 peak 1 및 2, 파래 peak 2가 가장 높았으며 그 다음으로 파래 peak 3이 높았다. 항암(종양)활성은 파래 Peak 1이 가장 높았으며 그 다음으로 쇠미역 peak 2, 파래 peak 3, 지누아리 peak 3 순으로 높았다. 전반적으로는 김의 기능성이 가장 뛰어났으며, 이는 가장 높은 단백질함량을 가지고 있는 것도 한 이유라고 판단되며, 앞으로 저분자 peptide의 구조분석 및 아미노산 sequence의 규명도 필요하다고 본다.

Nutritional Studies on the Growth of the Rapamycin-Producing Streptomyces hygroscopicus

  • During our previous studies on the relationship between nutrient requirements of S. hygroscopicus C9 and rapamycin biosynthesis, we developed chemically-defined media containing among other nutrients, aspartic acid, arginine, histidine, or ammonium sulfate. However, these media (“Cheng et al. medium” and “Lee et al. medium”) showed very slow growth characterized by a very long lag phase of growth. In an attempt to develop a chemically-defined or semi-defined medium to support more rapid growth and increased cell production, we have carried out studies to shorten the lag phase. Of the various additives tested, vitamin-free casein acid hydrolysate was the most significant by shortening the lag phase by 2-3 days. Mixtures of amino acids failed to replace casein acid hydrolysate. The active principle passed through an ultrafilter with a molecular weight cutoff of 1,000 and thus may be a peptide. The present work has yielded a semi-defined medium which should be useful for further growth studies on S. hygroscopicus C9.

Angiotensin I-Converting Enzyme (ACE) Inhibitory Activity of Elk (Cervus elaphus) Velvet Antler

  • Angiotensin I-converting enzyme (ACE) inhibitory activities of elk antler hydrolysates prepared with three kinds of proteases, pepsin, trypsin and $\alpha-chymotrypsin$, were investigated. The ACE inhibitory activity of the pepsinolytic hydrolysate was the highest with an $IC_{50}$ value of $9.3\mu g/mL.$ In addition, three kinds of pepsinolytic hydrolysates with relatively high molecular weights (over 10,000 Da), medium molecular weights (5,000 to 10,000 Da), and low molecular weights (below 5,000 Da) were fractionated using an ultrafiltration membrane system. The below 5,000 Da hydrolysate exhibited the highest ACE inhibitory activity. These results indicate that the pepsinolytic hydrolysates of elk velvet antler could be a good source of peptides with ACE inhibitory activity.

Effects of sea horse (Hippocampus abdominalis)-derived protein hydrolysate on skeletal muscle development

  • Hippocampus abdominalis, the big belly sea horse, is widely known for its medicinal value in Chinese folk medicine. In this study, extract obtained by proteolytic degradation of this species was investigated for its effects on skeletal muscle development, both in vitro and in vivo. Muscle cell lines ($C_2C_{12}$ and $L_6$) treated with the bioactive peptide did not have any detrimental effects on the cell viability, which was above 80%. Optical microscopy analysis on the morphology of the sea horse extract (SHE)-treated cells showed enhanced differentiating ability with myotube formation. Moreover, cells incubated with the hydrolysate displayed decreased proliferation rate, as recorded by the electric cell substrate impedance sensing system, thereby supporting enhanced differentiation. For a period of 12 weeks, mice models were fed with SHE and simultaneously subjected to treadmill exercise, which increased the expression of Myogenin, a key myogenic regulatory factor. In addition, there was an increase in the expression of AMPK- and Cytochrome C, both of which are important in mitochondrial biogenesis. Thus, the SHE from Hippocampus abdominalis can be a promising candidate as protein supplement aiding muscle development.

Isoflavone-Free 검은콩펩타이드의 항고혈압 및 ACE 활성 억제효과 (Antihypertensive and ACE Inhibitory Effects of Novel Isoflavone-free Black Soy Peptide Mixture)

  • Antiobesity, hypotriglyceridemic and antihypertensive activities of isoflavone-free peptide mixture (black soybean peptide, BSP) were reported in our previous experiments. In the present study, angiotensin converting enzyme inhibitory (ACEi) activity was decreased in the aorta tissues of spontaneously hypertensive rats (SHRs) treated with BSP (1% in drink water) for 4 weeks, but not in serum. BSP administration significantly decreased ACE activity by 17.5% (from $33.2{\pm}4.5$ to $27.4{\pm}1.96$ mUnit/mg, p=0.0013) in aorta tissue hydrolysate. BSP treatment also decreased significantly mean blood pressure (BP) (from $213.0{\pm}16.96$ to $184.0{\pm}6.53$ mmHg, p<0.0001) as expected. These results indicate that BSP has antihypertensive activity as well as ACEi activity.
