A pesticide mix solution containing difenoconazole, lambda-cyhalothrin, and lufenuron was applied 3 times on field grown chili pepper at a fivefold overdose dilution concentration of the spray solution at a pre-harvest interval of 7 day. Difenoconazole, lambda-cyhalothrin, and lufenuron were detected at 4.43, 0.334, and 1.56 mg/kg, respectively, in raw chili pepper. Washing with water reduced the residue levels to 91.4, 94.3, and 85.3%, respectively. In dried chili pepper, the residues of difenoconazole, lambda-cyhalothrin, and lufenuron were 22.2 mg/kg (processing factor, Pf =5.01), 1.65 mg/kg (Pf =4.94), and 6.54 mg/kg (Pf =4.19). In the seeds, difenoconazole and lambda-cyhalothrin were not detected, and lufenuron was detected at 0.0075 mg/kg (n=1) and <0.005 mg/kg (n=2). Thus the pesticide residues in the seeds was negligible. In the seed oil, difenoconazole and lufenuron residues were 0.0263 and 0.0295 mg/kg, respectively (concentration factors=5.26 and 4.72). These concentration factors supported the theoretical concentration factor of 6.8, assuming that all of compound present in the seed are transferred into the oil.
Efforts to control viral diseases in crop production include several types of physical or chemical treatments; antiviral extracts of a number of plants have also been examined to inhibit plant viral infection. However, treatments utilizing naturally selected microorganisms with activity against plant viruses are poorly documented. Here we report isolation of a soil inhabiting bacterium, Pseudomonas oleovorans strain KBPF-004 (developmental code KNF2016) which showed antiviral activity against mechanical transmission of tobamoviruses. Antiviral activity was also evaluated in seed transmission of two tobamoviruses, Pepper mild mottle virus (PMMoV) and Cucumber green mottle mosaic virus (CGMMV), by treatment of seed collected from infected pepper and watermelon, respectively. Pepper and watermelon seeds were treated with culture supernatant of P. oleovorans strain KBPF-004 or control strain ATCC 8062 before planting. Seeds germinated after treatment with water or ATCC 8062 yielded about 60% CGMMV or PMMoV positive plants, whereas < 20% of KBPF-004-treated seeds were virus-infected, a significantly reduced seed transmission rate. Furthermore, supernatant of P. oleovorans strain KBPF-004 remodeled aggregation of PMMoV 126 kDa protein and subcellular localization of movement protein in Nicotiana benthamiana, diminishing aggregation of the 126 kDa protein and essentially abolishing association of the movement protein with the microtubule network. In leaves agroinfiltrated with constructs expressing the coat protein (CP) of either PMMoV or CGMMV, less full-size CP was detected in the presence of supernatant of P. oleovorans strain KBPF-004. These changes may contribute to the antiviral effects of P. oleovorans strain KBPF-004.
Sugars and organic acids in Korean hot pepper fruit (C, annuum L.) according to its portions, pericarp, seeds and placenta, were determined by gas liquid chromatography (GLC). ${\alpha}-\;and\;{\beta}-fructoses,\;{\alpha}-\;and\;{\beta}-glucoses$, galactose, sucrose and maltose were identified. Major sugar in pericarp was fructose, while those in seeds and placenta were surose and fructose. Amount of free sugars in seeds was higher than that in pericarp or placenta. Succinic, fumaric, malic, tartaric, citric and quinic acids were found. Citric acid was a major organic acid found in pericarp and placenta. The amount of each identified organic acid, however, was similar in seeds. Quantity of total organic acids identified in pericarp or placenta was higher than that in seeds.
Neutral lipids of hot pepper seeds were analyzed by employing TLC and GLC methods. This seeds contained TG, SE, FFA, S, DG, and MG, i.e., the main component. TG was found 75. 81%. From the analysis of fatty acids it was found that linolenic (37. 6%). linoleic (27. 0%) and palmitic acid (16. 5%) were presented as the main fatty acids. In the case of SE, linolenic, palmitic and linoleic acids were 38. 1%, 22. 0%, and 20. 9%, respectively, and other acids were presented in small quantities.
Bacteria from the Methylobacterium genus, called pink-pigmented facultative methylotrophic bacteria (PPFMs), are common inhabitants of plants, potentially dominating the phyllosphere population, and are also encountered in the rhizosphere, seeds, and other parts of plants, being versatile in nature. The consistent success of the Methylobacterium plant association relies on methylotrophy, the ability to utilize the one-carbon compound methanol emitted by plants. However, the efficiency of Methylobacterium in plant growth promotion could be better exploited and thus has attracted increasing interest in recent years. Accordingly, the present study investigated the inoculation effects of Methylobacterium sp. strains CBMB20 and CBMB 110 on seed imbibition to tomato and red pepper on the growth and accumulation of phytohormone levels under gnotobiotic conditions. Seeds treated with the Methylobacterium strains showed a significant increase in root length when compared with either the uninoculated control or Methylobacterium extorquens $miaA^-$ knockout mutanttreated seeds. Extracts of the plant samples were used for indole-3-acetic acid (IAA), trans-zeatin riboside (t-ZR), and dihydrozeatin riboside (DHZR) assays by immunoanalysis. The treatment with Methylobacterium sp. CBMB20 or CBMB 110 produced significant increases in the accumulation of IAA and the cytokinins t-ZR and DHZR in the red pepper extracts, whereas no IAA was detected in the tomato extracts, although the cytokinin concentrations were significantly increased. Therefore, this study proved that the versatility of Methylobacterium as a plant-growth promoting bacteria could be better exploited.
An automatic drum seeder was developed to improve the seeding operation. It consisted of a conveyor to transfer seedling trays, a seed-hopper to supply seeds, a drum to drop seeds on the tray, and an air blower to remove extra seeds. A photo sensor was used to detect the transfer of seedling trays, and its signal was fed into microcomputer which operated a stepping motor driving the drum. The seeds were adhered to the surface of drum by vacuum pressure, and were dropped into tray cells by compressed air. An air connection unit was devised to alternate between vacuum pressure and compressed air. A control program for the system, written in C language, could operate the drum at the given number of revolutions and revolutions per minute. The results showed that the air connection unit could operate well and the seeds were dropped satisfactorily into tray cells. In case of cabbage and perilla seeds, which are regular and spherical shape, the missing rate was low and the single seeding rate was more than 97%. Low missing rate and high multiple seeding rate were observed in lettuce seeds which have narrow ends with tight weight. The missing rate of pepper seed was very high because of heavy weight and irregular shape. To improve the performance of the seeder, adjustment of vacuum pressure based upon shape and weight of the seeds, careful selection of the material of drum, maintenance of consistent air blower pressure, and replacement of stepping motor to DC motor are recommended.
This studies were conducted to find out the possibility of utilizing red pepper seed as resources of food fats, the research method was designed to make a comparison between crude and refined oil, and the results of the studies are as follows : The red pepper seed contained 28% of crude fat and 21% of crude protein. The main fatty acids of red pepper seed oil were linoleic acid(72.10~72.31%), palmitic acid(12.81~13.28%) and oleic acid(9.47~10.48%). The linolenic acid content was so small that is will not influence the autoxidation of the red pepper seed oil. The major triglyceride type of crude and refined oil of red pepper seeds were C52 and C54. The other types were found in a small quantity. The sterol composition of crude oil was $\beta$-sitosterol, campasterol, stigmasteral and brassicasterol,in the quantity order. after refining, brassicasterol was not detected, and the content was decreased by one six and one eight. The toropherol composition of crude and refined oil, tocopherol analog was composed of three kinds $\alpha$-, ${\gamma}$-, $\delta$-, but no $\beta$-form. the quantity of ${\gamma}$-, $\alpha$- and $\delta$-tocopherol were 162.91, 83.72, 43.98mg% respectively. The Quantity of and capsaicin in crude oil was 1,296 ppm, and it was reduced consicerably by refining and removed completely after the process of redeodorization.
Quantity of protein, amino acid, and sugar leaked from seeds was greater as the viability of seeds was dropped by the time elapsed of seed aging treatment. In the seeds with the artificial aging treatment for 20 days, 35.8 mg of protein was leaked on the 4th day after soaking, which was 6.9 times higher than that of control. Leakage of amino acid was also higher from low quality seeds treated with the aging treatment. According to .the quantity of leakage, water soluble compounds, which can be used for the assessment of seed quality without any destruction, were protein and potassium. Germination rate and percentage of seeds were dropped with the seed aging treatment, and the seed viability could be recovered by priming treatment. This phenomenon was very clear when the low quality seeds were germinated at low temperature.
The questionnaire survey was conducted on 225 farmers in Gyeonggi-do, Jeollanam-do and Jeollabuk-do. A total of 189 (84%) farmers responded. 72% of the respondents were males, 50.3% were aged 60 or older, and 51.3% had less than 5 years of farming experience. 78.8% of the respondents are pesticide-free, and 44.4% of respondents have less than 0.5 ha of farming scale. 61.4% of the cultivated crops were vegetable crops. In this study, the questionnaire was conducted for three major organic farming regions, but many of the respondents were judged to have a legal position on the mandatory use of organic seeds. Therefore, the results of this study can be used as a basic data for reviewing the legislation on the organic seed production and distribution suitable for the situation of Korean organic farming.
