Inhibition of Proliferation of Cervical and Leukemic Cancer Cells by Penicillin G

  • Cancer, despite all the efforts, still causes one in five deaths worldwide. Surgery, chemotherapy and radiotherapy provide inadequate protection and instead affect normal cells along with cancer cells. The search for cancer cures from natural products (plants and animals) has been practice for over a decade and the use of purified chemical to treat cancer still continues. Several studies have been undertaken during last three decades to find the anti-cancerous property of various plant extract and toxins secreted by animals and micro-organism. These lead to the discovery of several promising molecule having anticancer activity, some of which are in clinical trial and may emerged to be a potential future drug in cancer therapy. In this study we have used penicillin to evaluate its anti-cancer activity. It shown significant effects at cellular and molecular levels against growth of HeLa and K562 cell lines.

Effect of Sub-Minimal Inhibitory Concentrations of Antibiotics on Biofilm Formation and Coaggregation of Streptococci and Actinomycetes

  • Minimal inhibitory concentration (MIC) is the lowest antibiotic concentration that inhibits the visible growth of bacteria. Sub-minimal inhibitory concentration (Sub-MIC) is defined as the concentration of an antimicrobial agent that does not have an effect on bacterial growth but can alter bacterial biochemistry, thus reducing bacterial virulence. Many studies have confirmed that sub-MICs of antibiotics can inhibit bacterial virulence factors. However, most studies were focused on Gram-negative bacteria, while few studies on the effect of sub-MICs of antibiotics on Gram-positive bacteria. In this study, we examined the influence of sub-MICs of doxycycline, tetracycline, penicillin and amoxicillin on biofilm formation and coaggregation of Streptococcus gordonii, Streptococcus mutans, Actinomyces naeslundii, and Actinomyces odontolyticus. In this study, incubation with sub-MIC of antibiotics had no effect on the biofilm formation of S. gordonii and A. naeslundii. However, S. mutans showed increased biofilm formation after incubation with sub-MIC amoxicillin and penicillin. Also, the biofilm formation of A. odontolyticus was increased after incubating with sub-MIC penicillin. Coaggregation of A. naeslundii with S. gordonii and A. odontolyticus was diminished by sub-MIC amoxicillin. These observations indicated that sub-MICs of antibiotics could affect variable virulence properties such as biofilm formation and coaggregation in Gram-positive oral bacteria.

AP-PCR을 이용한 다제내성 Staphylococcus aureus의 유전형 분석 (Genotypic Analysis of Multi-drug Resistant Staphylococcus aureus by Arbitrarily Primed Polymerase Chain Reaction)

  • Many strains of Staphylococcus aureus were isolated from pus samples from primary, secondary, and tertiary medical institutions and were subjected to an antibiotic sensitivity test. Ciprofloxacin, clindamycin, erythromycin, gentamicin, oxacillin penicillin, tetracycline, trimethoprim/sulfamethoxazole, vancomycin and teicoplanin were used for the antibiotic sensitivity test. The strains showed hightest resistance to penicillin(91%), but all of strains tested were susceptible to vancomycin and teicoplanin. The isolated multi-drug(penicillin-tetracycline-ciprofloxacin-clindamycin-erythromycin- oxacillin-gentamicin) resistant S. aureus were analyzed genotypically using an AP-PCR(Arbitrarily Primed polymerase chain reaction) with an arbitrary 3 primers. Based on the result for genotype analysis, the genotypes identified by S1 primer did not coincide with those of S2 or E2 primers. Genotypes identified by S2 primer did not coincide with those of S1 or E2 primers. Also genotypes identified by the E2 primer did not coincide with those of S1 or S2 primers. Therefore, an analysis of AP-PCR test with multiple primers will provide more sensitive identification. A strain from a secondary medical institution and a strain from a tertiary medical institution which showed the same genotype for S1, S2, and E2 primers are required for further epidemiological study.

  • 근래 천공된 고막에 Ear drop를 사용하는 것은 그 효과가 의간시되 오고, 또 그 안전성도 도전을 받아왔다. 전신적으로 투여하였을때는 이독성이 없는 많은 약제들도, 중이강에 국소적으로 투여했을 때는 이독성이 될수있다. Stupp등(1973)에 의하면, Neomycin, polymyxin-G, gentamycin, erythromycin, tetracycline penicillin중, 다만 penicillin만이 독작용이 없다고 알려져 있다. Ototoxic antibiotics는 round window막을 투과하여 내이, perilymph에 들어가고 또다시 Reissner's막을 통하여 Endolymph에 이르게 된다고 보고있다. 주병변은 Stria vascularis의 degeneration, Sensory epithelia의 degeneration 및 Ganglion cell의 degeneration을 이르킨다. 63세 여자의 양측천공성 만성 중이염 환자가 가정에서 중이강내에 Terramycin분말 한 Capsule을 양이에 나누어 넣고, 심한 Sensorineural Deafness를 초래, 6개월이 지난 현재, 병변의 호전을 보지못하고 있는 일증례를 문헌적고찰과 함께 보고하는 바이다.

  • This study was carried out the antimicrobial resistance of staphylococcus aureus isolated from bovine mastitic milk, chickens, korean native cattle, korean native goats, pigs, dog and mice in northern area of kyongbuk. The result were summarised as follows ; A total of 149 S aureus were isolated from bovine mastitic milk, chickens, korean native cattle, korean native goats, pigs, dog and mice. In 80 isolates of S aureus from bovine mastitic milk, 60% of isolates revealed resistance to penicillin and ampicillin, 19% to gentamicin, 6% to tetracycline. In 36 isolates of S aureus from chickens, 72% of isolates revealed resistance to tetracycline, 58% to penicillin and ampicillin, 42% to streptomycin, 31% to lincomycin, 25% to norfloxacin, 22% to gentamicin. In 17 isolates of S aureus from korean native cattle, 100% of isolates revealed resistance to penicillin and ampicillin, 88% to lincomycin, 76% to tetracycline. 2 MRSA were isolated from the isolates of S aureus from bovine mastitic milk and revealed multi-drug resistance.

유산균 발효에서 항생물질 불활화에 관한 연구

  • 발효유 원료유중에 항생물질이 함유되어있을 경우 이것이 발효유 제조에 미치는 영향을 검토하였다. 발효유의 원가유 살균과정, 배양기간, 보존기간중에 있어서 항생물질의 변화를 검토하기 위하여 Bacillus stearothermophilus var. calidolactis C 953을 사용한 cylinder plate법으로 penicillin의 역가를 측정하였다. 저온 장시간 $살균(60^{\circ}C,$ 30분) 조건에서는 조금도 불활성화되지 않았으나, 온도를 높이고, 시간을 길게함에 따라 점점 불활화률이 높아져 $고압멸균조건(121^{\circ}C,$ 15분)에서는 약 90% 이상이 불활화되었다. 또 현재 우리나라에서 발효유제조에 사용되고 있는 Lactobacillus casei, Hy3와 Lactoba-cillus bulgaricus Hy4A, Hy4B를 사용하여 $37^{\circ}C에서$ 배양할 경우 배양기간 중에 있어서 penicillin은 2일내에 95% 이상 불활화되었다. 그리고 $보존기간(5^{\circ}C)$ 중에는 phosphate buffer(pH 6.0)와 10% skim milk의 경우에 10일까지도 거의 불활화가 되지 않았으나, 발효유내에서는 5일만에 85% 이상이 불활화된다는 결과를 얻었다. 이와같은 발효유 배양기간과 보존기간 중의 penicillin 불활화의 원인을 규명하기 위하여 각종 유기산의 영향을 조사한 결과(조건 pH.3.30~3.45, 보존온도 $37^{\circ}C),$ 염산과 유산의 경우 24시간, 구연산의 경우 48시간, 초산의 경우 72시간내에 실험에 사용한 penicillin 농도의 99.99%가 불활화되었다. 이러한 결과로 볼 때 유산발효에서 penicillin이 불활화되는 주원인은 발효에 의하여 생성된 유기산에 의한 것으로 추정된다.방식이군이 중지방식이군과 고지방식이군에 비해 혈장내 LCAT 활성이 유의하게9p<0.001) 증가하였다. 3) 간의 콜레스테롤합성 능력은 정어리유군이 다른지방군보다(p<0.001), 무지방식이군이 식이지방첨가군보다(p<0.001), 동물성지방군의 식물성유지군보다 유의하게(p<0.001) 증가하였으나, 식이 지방의 수준에 의한 차이는 나타나지 않았다. 수용성 식이섬유소가 생리져 기능이 거의 비슷하고 무독성이 관찰됨으로써 신갈나무로부터 제조된 수용성 식이섬유소의 제조 방법이 우수하다고 볼 수 있다.있었다.세에 해당되는 중년 여성의 에너지, 단백질, 철분 섭취량을 권장량과 비교하여 보면, 각각 74.8$\pm$12.6%, 94.6$\pm$26.4% 및 64.5$\pm$14.1%로 당뇨군 (각각 112.8$\pm$28.5%, 157.8$\pm$68.2%, 92.8$\pm$21.7%)에 비해 유의하게 낮았고 정상군과는 유의한 차이를 보이지 않았다.상고나성이 있었다. 혈중 호모시스테인 농도는 질병의 위험요인으로서 뿐 아니라 대사적으로 밀접하게 연관된 비타민 영양상태의 biomarker로서도 그 영향력이 크다고 할 수 있다. 따라서 성별에 다른 다양한 연령집단에서 건강한 일반인과 심혈관계 질환자 등을 대상으로 호모시스테인과 비타민 영양상태에 대한 연구가 체계적으로 이루어 져야 할 것이다.태를 보다 효율적으로 증진시킬 수 있는 대안이 마련되어져야 한다고 사료된다.$\ulcorner$순응$\lrcorner$의 범위를 벗어나지 않는다. 그렇기 때문에도 $\ulcorner$순응$\lrcorner$$\ulcorner$표현$\lrcorner$의 성격과 형태를 외형상으로 더욱이 공간상에서는

일부 무의촌지역과 병원에서 분리한 Enterococci의 항균제 감수성에 관한 연구 (Studies on the Antimicrobial Susceptibility of Enterococci isolated at Doctorless areas and Seoul National University Hospital)

  • 28 strains of Enterococci isolated at doctorless areas in Korea from Oct. 1976 through Mar. 1977 and 30 strains of Enterococci isolated at Seoul National University Hospital from Sep. 1976 through Dec. 1976 were examined for susceptibilities to 14 antimicrobial agents by agar dilution method. The susceptibilities of two groups to each antimicrobial agent were compared and incidence of resistant strains in each antimicrobial agent, incidence of multiply resistant strains and correlations in the antimicrobial susceptibility of 58 strains of Enterococci among 14 antimicrobial agents were analyzed. 1. With Tetracycline, Oxytetracline, Doxycycline, Minocycline and Erythromycin, the mean MIC's of Enterococci isolated at Seoul National University Hospital were 1.9-2.5 times higher than those of Enterococci isolated at doctorless areas, though there were no statistical significance. 2. Frequency of resistant strains in Enterococci isolated at Seoul National University Hospital was the highest with Cephalexin(100%), Gentamicin(100%), and Streptomycin(100%), followed by Kanamycin(93.3%), Carbenicillin(90%), Tetracycline(26.7%), Oxytetracycline(26.7%), Doxycycline(26.7%), Minocycline(20%), Erythromycin(10%), Troleandomycin(10%), Penicillin(6.7%), Cotrimoxazole(6.7%) and Ampicillin(0%). 3. Frequency of resistant strains in Enterococci isolated at doctorless areas was the highest with Kanamycin(100%) and Streptomycin(100%), followed by Cephalexin(96.4%), Gentamicin(96.4%), Carbenicillin(92.9%), Tetracycline(10.7%), Oxytetracycline(10.7%), Doxycycline(10.7%), Minocycline(10.7%), Cotrimoxazole(10.7%), Penicillin(3.6%), Ampicillin(0%), Erythromycin(0%), and Troeandomycin(0%). 4. In study with Penicillin, Ampicillin, Tetracycline, Cotrimoxazole, Erythromycin and Troleandomycin, there were no multiply resistant strains in Enterococci isolated at doctorless areas, while 4 strains of Enterococci isolated at Seoul National University Hospital showed multiple resistance. 5. There were high correlation in susceptibility of 58 strains of Enterococci among Tetracycline, Oxytetracycline, Doxycycline and Minocycline(correlation coefficients 0.86-0.97). Correlation coefficients of susceptibility among penicillin analogues(Penicillin, Ampicillin, Carbenicillin and Cephalexin) ranged between 0.75-0.89. Correlation coefficients of susceptibility among aminoglycosides(Kanamycin, Gentamicin and Streptomycin) ranged between 0.09-0.51.

무의촌균주(無醫村菌株) 및 병원균주(病院菌株)의 항균제(抗菌劑) 내성(耐性) 양상(樣相)에 관(關)한 연구(硏究) (Drug Resistance Patterns of the Bacterial Strains Isolated from Rural Areas and an Urban General Hospital)

  • Besides the benefits of antimicrobial agents in the control of various infectious diseases, widespread and prolonged use of particular antimicrobial agents has brought about the increase of drug-resistant strains in a community and the profound changes in the pattern of infectious diseases. In Korea, there are some remote villages where no clinics and drug stores are available and the residents in those areas are assumed to have fewer chances to contact with antimicrobial agents. In the present study, the differences in susceptibilities to 14 antimicrobial agents between the isolates from rural areas(R) and Seoul National University Hospital(SNUH, H) were studied. The isolates and their numbers were Staphylococcus aureus, R;55, H;68), Enterococci(R;28, H;30), Escherichia coli(R;40, H;40), Enterobacter aerogenes(R;25, H;21) and Klebsiella pneunoniae(R;58, H;67). Minimal inhibitory concentrations(MIC's) of penicillin, ampicillin, carbenicillin, cephalexin, tetracycline, oxytetracyline, doxycycline, minocycline, gentamicin, kanamycin, streptomycin, erythromycin, troleandomycin and co-trimoxazole were determined by agar dilution method. I. Comparison of MIC's and resistant strain proportions between isolates from SNUH and rural areas. MIC's and/or resistant strain proportions of the isolates from SNUH were significantly higher than those of the isolates from rural areas in the cases of 1. S. aureus to doxycycline, streptomycin and kanamycin. 2. E. coli to penicillin, ampicillin, carbenicillin, tetracycline, oxytetracycline, doxycycline, minocycline, streptomycin, kanamycin, erythromycin and co-trimoxazole. 3. E. aerogences to carbenicillin, tetracycline, oxytetracycline, doxycycline, minocycline, streptomycin, kanamycin, genaamicin and co-trimoxazole. 4. K pneunoniae to penicillin, ampicillin, tetracycline, oxytetracycline, doxycycline, monocycline, streptomycin, kanamycine, gentamicin and co-trimoxazole. However, the mean MIC and resistant strain proportion of S. aureus to tetracycline were higher in isolates from rural areas than in those from SNUH and Enterococci showed no differences in susceptibilities to the antimicrobial agents between isolates from rural areas and from SNUH. Therefore, in general, differenes in susceptibility to these antimicrobial agents between the isolates from rural areas and SNUH were remarkably greater and broader in gram negative enteric bacteria. II. Multiple drug resistance pattern. Patterns and incidences of multiple drug resistance were studied with penicillin, ampicillin, tetracycline, cephalexin, gentamicin, streptomcin, kanamycin and co-trimoxazole in Enterococci, E. coli, E. aeroges and K. pneumoniae. There appeared significant differences in the incidence of multiply drug-resistant strains and multiple drug resistance patterns between the isolates from SNUH and rural areas in Enterococci, E. coli, E. aerogenes and K. pneumoniae. However, there was no difference in the incidence of multiply drug-resistant strains between isolates of S. aureus from SNUH and rural areas but the pattern of multiple resistance of the SNUH strains of S. aureus was diverse, while that of the rural strains was predominantly confined to penicillin-tetracycline combination. The incidence of multigly drug-resistant strains and diversity of their patterns were the highest in E. coli strains isolated from SNUH and there were no multiply drug resistant strrains in Enterococci and K. pneumoniae strains isolated from rural areas. The number of drug-resistance determinants was also different between the isolates from rural areas and SNUH. Most of the multiply drug-resistant strains of E. coli, E. aerogenes and K. pneumoniae isolated from SNUH were resistant to more than 3 kinds of antimicrobial agents, most frequently to ampicillin, tetracycline and streptomycin, while multiply drug-resistant strains from rural areas were resistant to 2 kinds of antimicrobial agents among ampicillin, tetracycline and streptomycin. With drug-resistant E. coli strains, resistance to tetracycline which was used most widely since 1951 was most frequently involved as a part of mutliple drug-resistance, followed by resistance to ampicillin and streptomycin. This strongly suggests that emergence of drug-restant strains in a community is directly dependent on the selective pressure exerted by the antimicrobial agent used. III. Cross resistance. Cross resistance of bacteria was studied among tetracycline penicillin, aminoglycoside and macrolide derivatives by analyzing correlation coefficients of sucseptibilities using the least square method. In this study, there were high correlations among the susceptibilities to related derivatives. It appears that the relatively low correlations in susceptibilities present in some cases are due to intrinsic resistance of E. aerogenes to penicillin, Enterococci to aminoglycoside and E. coli E. aerogenes and K. pneumoniae to macrolide derivatives.

소아 화상의 화상부위 세균 집락화와 항균제 감수성 변화(1999년 1월~2002년 12월) (The Bacterial Colonization of Burn Wound and the Changes of Antibiotic Susceptibility in Childhood Burn (January, 1999~December, 2002))

  • 목 적 : 화상의 병태 생리에 대한 지식 증가와 효과적인 항균제의 이용 및 상처의 괴사조직을 조기에 제거하는 수술 방법의 도입으로 최근 사망률이 감소하고 있으나, 아직도 세균 감염으로 인한 패혈증과 폐렴 등의 합병증이 사망의 가장 큰 원인이 되고 있다. 최근 내성균주의 증가로 적절하고도 제한적인 항균제 선택의 중요성이 증대되고 있다. 특히 Acinetobacter의 multi-drug resistant 균주는 최근 들어 관심의 대상이다. 중환자실의 MRSA, VRE 등의 균주가 증가되고 있는 우리나라의 실정에서 소아 연령의 화상부위 세균 감염 양상을 파악하고 각 균주에 따른 항균제 내성의 추이를 파악하여 적절한 항균제 선택으로 소아 화상 부위의 치료에 도움이 되고자 한다. 방 법 : 1999년 1월부터 2002년 12월까지 4년간 한림대학교 의료원 한강성심병원 화상 센터에 입원한 만 15세 이하 환아 중, 화상 부위 세균 배양 검사 양성으로 나타난 213명, 406검체를 대상으로, 의무기록의 후향적 고찰을 통하여 감염세균 및 항균제 감수성 양상을 조사, 분석하였다. 결 과 : 1) 총 213명의 환아 중 남아 127명(59.6%), 여아 86명(40.4%)이었다. 나이별로는 1~5세까지의 학령 전기에서 132명(62.0%)으로 가장 많았고, 1개월~1세의 영아기 45명(21.1%), 6~10세의 학령기 24명(11.3%) 순이었다. 2) 화상의 종류로는 열탕 화상이 167명(78.4%)으로 가장 많았고, 다음으로 화염 화상이 35명(16.4%), 접촉 열화상 6명(2.8%) 순이었다. 3) 총 213명 406검체 중 64검체(15.8%)에서 두 종류 이상의 균주가 분리되었다. P. aeruginosa가 236검체(58.1%)에서 분리되어 가장 흔한 원인균이었다. Enterococcus species가 73검체(18.0%), S. aureus가 51검체(12.6%), S. epidermidis가 48검체(11.8%)에서 분리되었다. 그 다음으로 Streptococcus species, Acinetobacter species, E. coli 순이었다. 4) P. aeruginosa의 경우 항균제에 대한 내성률은 trimethoprim-sulfamethoxazole 100%, cephalothin 98.1%, ampicillin-sulbactam 96.2%, ampicillin 95.3%, ceftriaxone 95.2%, tobramycin 93.7%, cefoperazone 68.9%, ceftazidime 67.7%이었다. 5) Enterococcus species는 tetracycline 63.9%, streptomycin 45.5%, gentamicin 36.1%, penicillin G 13.7%의 내성을 보였다. 6) S. aureus는 gentamicin 89.7%, tetracycline 86.2%, ciprofloxacin 86.2%, penicillin G 84.3%, oxacillin 78.4%, erythromycin 76.5%의 내성을 보였다. 7) S. epidermidis는 oxacillin 81.2%, erythromycin 79.2%, gentamicin 76.2%, penicillin G 70.8%, fusidic acid 70.3%, tetracycline 61.9%의 내성을 보였다. 8) Acinetobacter species는 ampicillin-sulbactam 100%, gentamicin 85.7%, ampicillin 83.3%, piperacillin 61.5%, aztreonam 53.3%의 내성을 보였다. 결 론 : P. aeruginosa의 선택적 항생제 중 cefoperazone 68.9%, ceftazidime 67.7% 등 높은 내성을 보였고, imipenem 역시 25.9%의 내성을 나타냈다. 현재 P. aeruginosa에 대한 백신이 개발 중이나 효과에 대해선 더 많은 연구가 필요할 것이다. S. aureus는 penicillin G 84.3%, oxacillin 78.4% 등의 높은 내성을 보였으나, vancomycin 0%, teicoplanin 2.2%로 아직 낮은 내성률을 보였다. Acinetobacter species는 최근 계속 증가하는 multi-resistant drug 양상을 보이고 있어 향후 항균제 선택에 있어 신중한 주의가 필요하다.
