A Premature Newborn with Congenital Syphilis (미숙아에 발생한 선천성 매독 1 예)

  • A preterm newborn affected by congenital syphilis, born to mother not treated during pregnancy is described. The clinical picture was characterized by respiratory distress, cutaneous manifestations, massive hepatosplenomegaly, severe anemia, thrombocytopenia, disseminated intravascular coagulation syndrome and hypoalbuminemia. The patient was treated with daily injections of 190,500 units of crystalline penicillin G for 14 days. Premature infants with these symptoms and signs should be evaluated for congenital syphilis.

Studies on the Resistance to Antibiotics (세균의 약제 감수성에 관한 연구 II)

  • A total of 130 of Staphylococcus strains isolated from various clinical specimens of admitted patients of a university hospital with systemic or severe cases of infection. All of these were tested for the antimicrobial susceptibility to 11 drugs of common use. The hospital strains isolated showed higher frequency of resistance against four drugs including gentamicin, penicillin, erythromycin, kanamycin but amikacin, cephalothin, streptomycin were effective. And also 47.7% of methicillin-resistant Staphylococcus were isolated from the clinical patients. However, isolated Escherichia coli strains showed higher frequency of resistance, but two drugs, tobramycin and gentamicin were effective to them.

  • In order to establish the structural requirements for designing new ${\beta}$-lactam antibiotics it is necessary to build the molecular model of a penicillin binding protein. D-alanyl-D-alanine carboxypeptidase/transpeptidase (DD-peptidase) is a good model for PBPs. The X -ray crystallographic structure of DD-peptidase has been reported at the 1.6${\AA}$ resolution.   • 아미노산은 식품이나 사료의 첨가물로, 의약품으로 그리고 화학합성에 있어서의 단위구조체(building block)로써 널리 이용되어 왔는데, 위에서 광학적으로 순수한 single enantiomer로서 D- 혹은 L-form의 .alpha.-아미노산은 의약품의 합성에 있어서 그 산업적 중요성이 날로 증대되고 있다. 이중 D-form의 아미노산은 .betha.-lactam계 항생제, peptide hormone, 살충제 등의 합성에서 중간 물질로 사용되고 있는데, 특히 D-p-hydroxyphenylglycine(이하 D-HPG)의 경우 amoxicillin, cefadroxil, cefatrizine, cefaparole, cefaperazone등의 penicillin이나 cephalosporin계의 반합성항생제의 전구체로서 전 세계적으로 널리 사용되고 있다.

Structure-Activity Relationship Study on Cephalosporins with Mechanism-based Descriptors

  • The polarizability and the transition state energy of a cephalosporin are assumed to be theoretical indices of the permeability through the outer membrane and of reactivity of ${\beta}$ -lactam ring with penicillin binding proteins, respectively, in Gram-negative bacteria. They are computed by AM1 method and used as variables of quantitative structure-activity relationship study. The results justify quadratic dependence of the activity on the variables. The intersection of difference volumes between $\beta-lactamase$ stable cephalosporins and unstable ones manifests that the steric hindrance of 7-side chain is responsible for the ${\beta}$ -lactamase stability.

  • In Korea, antibiotic fermentation industries have just started budding out now. Tetracycline derivative (Pfizer, CKD), Kanamycin (Dongmyung) are produced by fermentation processes. Penicillin, erythromycin, gentamicin, rifamycin, and a few others are being planned for production within a few years by major pharmaceutical and chemical firms in Korea. Reviewing past and present activities in the antibiotic fermentation industries in Korea suggests that the prospects for antibiotics will be very bright.

Antibiotics Sensitivity to Isolated Staphylococcus sp. from the Air (공기중에서 분리된 포도구균의 항생제 감수성)

  • This investigation was performed to isolate and identify Staphylococcus sp. from air at the 25 sites in one hospital in Daegu. Drug sensitivity tests bacteriologically were also studied. The results are summarized as follows: 1. In gram staining of isolated colonies, the number of staphylococci strains was 959(70.2%) among 1367 collected in the morning, and 653(66.2%) among 987 collected in the afternoon. 2. The number of isolates was slightly higher in samples collected from the outpatient treatment rooms and wards in the main building than in samples from the other sites. 3. In biochemical tests of 1612 strains of isolates gram positive cocci, the number of coagulase positive strains was 584(36.2%) and coagulase negative 1028(63.8%). In experiments on growth on mannitol agar containing 7.5% NaCl and mannitol fermentation tests, the number of mannitol fermentation strains was 423(30.8%) and nonfermenter was 951(59.2%). 4. In antibiotic sensitivity tests of 746 identified strains of staphylococci, coagulase positive strains showed resistance in. higher portions to penicillin(89.7%), chloramphenicol(64.6%), gentamycin (52.1%) and tetracycline(45.2%), whereas in lower portions to kanamycin(39.2%) and apmicillin(33.1%). 5. Among coagulase negative and mannitol fermenting strains, higher portions showed resistance to chloramphenicol(76.5%), penicillin(69.8%) and ampicillin(63.6%), whereas lower portions showed resistance to kanamycin(48.8%), tetracycline(39.5%) and gentamycin(34.0%). 6. The number of strains showing multiple resistance to above 6 antibiotics were 15(52(19.8%) among coagulase positive and mannitol non-fermenter and 36(22.2%) among coagulase negative and mannitol fermenter.

  • The susceptibility of 536 isolates of Staphylococcus and 313 isolates of Streptococcus to a number of chemotherapeutics were studied. These organisms were isolated from bovine mastitis during 1973 and 1974. In addition to this, the rate of multiple resistance of 425 isolates of Staphylococcus and 164 isolates of Streptococcus, isolated in 1974, to the chemotherapeutics was analysed. The results obtained in this work were summerized as follows: 1. Staphylococcus and Streptococcus isolated in 1974 showed a higher resistance, with 3 exceptions of chemotherapeutics, than the isolates of 1973. 2. Staphylococcus isolated in 1973 and 1974 showed a higher susceptibility than Streptococcus. 3. The strains of Staphylococcus resistant to colistin were 39 strains (9.2%), to colistin and sulfisoxazole 33 (7.8%), to streptomycin, kanamycin, colistin and sulfisoxazole 20 (4.7%), and to penicillin, colistin and sulfisoxazole 18 (4.2%). 4. The strains of Streptococcus resistant to colistin were 17 strains (10.4%), to streptomycin, kanamycin, colistin and sulfisoxazole 13 (7.9%), to colistin and sulfisoxazole 11 (6.7%) and to penicillin, colistin and sulfisoxazole 11 (6.7%).

Isolation of Enteropathogenic Escherichia coli, Thermophilic Campylobacter and Salmonellae from Scouring Piglets (설사자돈으로부터 병원성대장균, 캠필로박터속균 및 살모넬라속균의 분리동정)

  • This study was conducted to isolate etiological agents from the 103 scouring piglets in Gyeongnam area and also carried out antimicrobial drug susceptibility test and epidemiogical served. The incidence of scouring piglet was most prevalent as 81.6% in the age of 2 to 4 weeks after birth, while the rate was less than 10% in the age of 5 to 6 weeks and under 1 week after birch. When compared the isolation frequency of the each etiological agent, enteropathogenic E. coli was most prevalent as 46.6%, thermophilic Campylobacter 26.2% and Salmonellae was 8.7% in order. In the OK serotyping for 117 isolates of enteropathogenic E. coli, type 0141 : K85 (20.5%), 0157:K88ac(14.5%), 0138:K81 and 0149:K91 (13.3%) were encountered most frequently. In the biotyping for 27 isolates of thermophilic Campylobacter, most strains of C. jejuni were belong to type I (50.0%) and II (25.0%), and most strains of C. coli were belong to biotype I (78.9%). In the serotyping for 9 strains of Salmonellae, 3 strains were grouped as D, 2 strains as C. and each 1 strain was group B and E. The other 2 strains were untypable. The 117 isolates of enteropathogenic E. coli were resistant more than 90% to erythromycin, penicillin, tetracycline and streptomycin, wherease about 90% of the isolates were sensitive to kanamycin and gentamicin. In the case of Salmonellae, all of the isolates were resistant to penicillin, but about 89% of the isolates were sensitive to gentamicin and colistin. All of C. jejuni and C. coli isolates were resistant to cephalothin, but more than 89% of C.jejuni and C. coli were sensitive to kanamycin and gentamicin.

SYBR Green I-based Real-time PCR Assay and Melting Curve Analysis for Rapid Detection of Staphylococcus aureus from Raw Milks Samples (Real-time PCR을 이용한 원유시료 유래 황색포도상구균의 신속 검출)

  • The aim of this study was to develop a LightCycler-based real time PCR (LC-PCR) assay and to evaluate its diagnostic use for the detection of Staphylococcus aureus in raw milk samples. Following amplification of 113 bp of coa gene encoding an coagulase precursor specific for Staphylococcus aureus, melting curve and DNA sequencing analysis was performed to verify the specificity of the PCR products. Amplification of 209 bp gene encoding an altered penicillin-binding protein, PBP2a (mecA), melting curve analysis and DNA sequencing analysis was performed to verify methicillin resistance Staphylococcus aureus (MRSA). According to this study, 6 of 647 raw milk samples showed S. aureus positive and 2 of them showed a mecA positive and the detection limit was 10 fg of DNA. And we also isolated Staphylococcus chromogenes a causative agent of exudative epidermitis in pigs and cattle from 3 samples.
