Effects of Seeding Bed Media and Fungicide on Control of Clubroot Disease of Chinese cabbage Caused by Plasmodiophora brassicae (배추 뿌리혹병(Plasmodiophora brassicae) 방제를 위한 육묘용 상토와 농약처리 효과)

  • Clubroot disease of Chinese cabbage has extremely occurred in recent years. Conventional soils such as sandy loam soil (saprolite) and clay soil (yellow soil) as bed media combined with field soil application of fungicidal chemicals were tested for the control of clubroot disease. Using sandy loam soil and clay soil as plug seedling bed media efficiently reduced clubroot disease occurrence down to 21.7% and 14.1%, respectively compared to peatmoss (75.7%) and Baroco soil (36.6%) when seedling plants were transplanted into previously-infected Yoncheon field. Application of flusulfamide and azoxystrobin to previsouly-infected soil prior to transplanting also effectively reduced disease incidence, especially when combined with growing seedlings in sandy loam or clay soil media. In conclusion, flusulfamide application prior to trnasplanting as well as utiliz-ing sandy loam and clay soil as a plug bedding media may effectively reduce the occurrence of clubroot dis-ease of Chinese cabbage.

The Study on Native Plants and Planting Soil for Extensive Rooftop Greening (관리조방적 옥상녹화에 적합한 자생초화류와 식재토양에 관한 연구)

  • Green spaces in cities were insufficient and decreased as artificial structures increase. Accordingly, greening rooftop space contributed to urban ecosystems as green space. Especially, since 2002 Seoul supported rooftop greening and introduced 'Ecology-area rates'. Thus rooftop greening had the potentiality of extension. This prepared the extensive rooftop greenings within planting-base for extensive management rooftop greening system to be easily popularized. The extensive rooftop greening conducted from 1997-2005 reported that soil depth was 8cm and soil mixtures were 5 types. And plant were Crysanthemum zawaskii var. latilobum, Aster hayatae, Crysanthemum zawaskii herb. ssp. coreanum, Sedum sarmentosum, Sedum rotundifolium, Sedum oryzifolium, Sedum takemensis, Sedum middendorffianum, Sedum telerium var. purpureum, Sedum spectabile, Carypteris incana, Dianthus superbus, Hosta minor. As a result, for nine years after the modular type rooftop greening established, survival plants were Sedum takemensis, Sedum sarmentosum, Sedum spectabile, Sedum middendorffianum, Carypteris incana, Crysanthemum zawaskii var. latilobum, Sedum telerium var. purpureum, Sedum rotundifolium, Dianthus superbus. Also effect of greening was possibled soil depth 8cm. And growth of plants from 1997 to 2005 were fine on two soil mixtures of 'perlite+peatmoss+vermicompost+moisturizer' and 'perlite+moisturizer+bark(mulching)'. Invasive plants are Ixeris dentata Nakai, Ixeris chinensis var. strigosa, Youngia sonchifolia, Eragrostis ferruginea, Aster pilosus Willd., Ixeris japonica Nakai, Valpia myuros, and Setaria viridis. In conclusion, selection of suitable native plants was possible extensive management rooftop greening with effect of continuous greening. The extensive rooftop greening were lightweight and simple preparation without management and can popularize readily.

  • This experiment was aimed to investigate the growth and yield responses of hydroponically grown cucumber as affected by cultural containers and substrates using perlite and mixtures with perlite. Containers used in this experiment were bed and box made of styrofoam, PE film and Wagner pot and substrates used were perlite, peatmoss, rice hull and carbonized rice hull. The results obtained were as follows : Based on the growth and yield responses determined from the leaf area and total fruit weight, bed and box made of styrofoam was considered the best containers among tested for cucumber hydroponics. Recommendable substrates for hydroponically grown cucumber using perlite were evaluated in the order of perlite, perlite with rice hull and perlite with carbonized rice hull in styrofoam bed and box. Increase in marketable yield and decrease in abnormal fruits was observed in the mixed substrates with rice hull or carbonized rice hull when compared to perlite single media.

Somatic Embryogenesis and Plant Regeneration in Leaf Explant Cultures of Gentiana scabra var buergeri (용담(Gentina scabra var. buergeri)의 잎 절편 배양에서 체세포배발생에 의한 식물체 재분화)

  • Plant regeneration system via somatic embryogenesis in leaf explant cultures of Gentiana scabra var. buergeri has been established. Leaf segments formed calli when cultured on MS medium supplemented with 0.5 mg/L 2,4-D and 2 mg/L BAP After transferred to SH medium supplemented with 0.5 mg/L 2,4-D, 2 mg/L CPA and 0.5 mg/L kinetin, the callus became embryogenic. The embryogenic callus was subcultured every 3 to 4 weeks. Upon transfer onto SH basal medium the embryogenic callus gave rise to numerous somatic embryos, which subsequently developed into plantlets. The regenerated plants were potted in an artificial soil with mixture (peatmoss : pearlite : vermiculite : 2 : 1 : 1) and transplanted to the soil after kept under a high humidity for two weeks. A total of 78 plants out of 105 regenerated plants survived in the soil. Phenotypic variations in height, number of stems and the flowering time were observed in tile regenerated plants. Cytogenetical analyses showed no chromosomal variation.

Studies on Short Term Hardening Method of Tree Seedlings for Afforestation of Cut-Rock Slope (암반절개사면 녹화용 강건묘목의 속성육묘법에 관한 연구)

  • This study was carried out to develop a short term hardening method of tree seedlings of Rhus chinensis Mill., Evodia daniellii Hemsley and Parthenocissus tricuspidata(Sieb. et Zucc.) Planck for afforestation on a concave and a crack of cut-rock slope. The seedlings were grown in a cylinder shaped pot made of polyvinyl net with the soil media of peatmoss, vermiculite, clay, compost, fertilizer, and absorbant(40:25:19:15:1:0.1, v:v). They were cultivated in a greenhouse for four months and in field condition for two months. During the last three months of the growing period the seedlings were hardened by periodic desiccation and irrigation in 4 to 10 days interval. The hardened seedlings showed lower leaf water potential, higher leaf osmotic pressure, and lower T/R ratio than those before the hardening. The hardened seedlings survived well on the soil medium in the concave of cut-rock slope.

  • In this study, manufacture of kitchen garden soil and its application on the growth of Lettuce has been performed. The optimal ratio of various raw materials for pH, total nitrogen, phosphate, cations, and salt content was found to be 49 : 35 : 10 : 5 : 1(w/w) in peatmoss, livestock manure, cocopeat, zeolite, and microbial fertilizer, respectively. In the growth experiment using Lettuce, the manufactured soil obtained 21% & 20% increase in leaf length and width, 17% increase in chlorophyll, and 22% in biomass, compared to the control.

Shoot Regeneration of Mature Nodal Segments in Poplar (Populus deltoides) (미류나무 성숙 절간조직으로부터 식물체 증식)

  • Mature nodal segments of 2-year-old greenhouse stock plant were cultured on Murashige and Skoog basal medium supplemented with the different kinds and various concentrations of cytokinins to produce multiple shoots in in vitro condition. The most adventitious shoots were produced from excised ends of nodal segments. The highest average number $(24.6\;{\pm}\;4.6)$ of shoots was produced with the combination of BA 1.0mg/L and TDZ 0.1mg/L in MS medium. In addition, several shoots were formed from lenticels of bark cambium with the same treatment. These concentrations promoted high shooting capability upto $94.6\%$ and NAA was the best cytokinin among five different PGR sources.

Mass-production of Eleutherococcus seoulensis Seedlings Through Somatic Embryogenesis (체세포배 형성을 통한 서울오갈피(Eleutherococcus seoulensis) 묘목의 대량생산)

  • This study was conducted to establish the optimal condition for acclimatization from somatic embryos of Eleutherococcus seoulensis. Torpedo-shaped embryos of Eleutherococcus seoulensis were cultured on 1/3 MS and WPM media supplemented with $GA_3$ (3.0, 5.0 mg/L) for 4 weeks. Plentlets were transferred to 1/2 SH solid medium with 1.0 mg/L $GA_3$ and 0.2% activated charcoal for shoot and root elongation and them elongated plantlets further developed on 1/2 SH medium for 4 weeks. Developed plantlets further elongated into well-shaped leaf and root system on 1/3 SH medium under ventilation condition for 4 weeks. Plantlets grew normally on 1/3 SH basal medium, were acclimated on various soil. Survival frequency of plantlets was influenced by soil type(peatmoss+perlite, perlite, soil on Nam mountain). The highest survival rate to soil was more than 70% when plantlets were 1/3 SH medium under ventilation condition in Nam mountain soil. These results indicate that the systematic procedure of plant production in Eleutherococcus seoulensis could be practically applicable for mass propagation.

Stabilization of Rhizosphere pH during Tomato Cultivation Using Expanded Rice Hull Substrate (양액재배용 평연화 왕겨 배지의 근권 pH 안정화)

  • In countries that consumes rice as a main staple, rice hulls are natural resources composed of a large amount of organic compounds and high uniformity in size. Rice hulls are expanded to get rid of a defect in untreated rice hulls and to be used as a hydroponic substrate. Research on rice hulls is continuing for the agricultural application. This research was conducted to stabilize rhizosphere pH of the expanded rice hull substrates because of high pH caused by repeated use in ERH(expanded ride hull) substrates and without increasing the cost of developing new substrates. Sphagnum peatmoss (pH 3.0-4.0) wee mixed with the expanded rice hull substrate in the ratio of 10% (v/v), and this ratio kept the pH range of 6.0 to 6.5 in the root area of tomato plants during growth and at the time of harvest of tomato fruits. Also absorbtion of nutrients was highly increased. The yield increased from 1,051 to 1,266 kg per tomato plant which were harvested by two clusters.

Growth of Mandshurian Linden (Tilia mandshurica Pupr, et Max.) Seedlings as Affected by Container Types and Volumes (용기의 형태와 용적에 따른 찰피나무 유묘의 생장)

  • This study was carried out to investigate the growth characteristics and root development of Mandshurian Linden (Tilia mandshurica Rupr. et Max.) seedlings grown in plastic net container and plain plastic container. The seedlings were grown in the mixture of peatmoss perlite : vermiculite (1:1:1, v/v/v) for five months in the plastic greenhouse. The seedlings grown in plastic net container showed greater root collar diameter growth, better root development, and more increment of root dry mass than those grown in the plain plastic container. In particular, the seedlings grown in plastic net container had no spiraling roots which were always observed in the control . These results suggest that the net container is suitable for container culture of Tilia mandshurica seedlings by enhancing root development. Seedlings grown in the net container and transplanted to various environments showed vigorous rootage and growth.
