가지과 작물에서 분리한 Alternaria 속 균의 형태적, 분자생물학적 특징 (Morphological and Molecular Characterization of Alternaria Isolates from Solanaceous Crops)

  • 국내의 가지과 작물에서 분리한 Alternaria 25 균주를 공시하여 분생포자의 형태적 특징을 조사하였고 A. solani와 A. tomaotphilad의 대표균주와 비교하였다. 분리균주 중 형태적으로 구별되는 몇 균주들과 대표균주를 공시하여 감자, 토마토, 가지, 고추에 대한 병원성 검정을 실시하였다. 대표균주를 포함한 Alternaria 17개 균주를 공시하여 ITS 영역과 histone h3 유전자의 염기서열분석, URP (universal rice primer)에 의한 핵산지문분석을 실시하였다. Alternaria 균주들은 분생포자의 형태적 특징에 의하여 A. tomaotophila(ATO), A. solani(ASO) 및 미동정의 Alternaria sp.(ASP)의 group으로 나눌 수 있었고 A. solani(ASO)는 분생포자 부리(beak)의 특징에 의하여 다시 ASO(1)과 ASO(2)의 2 type으로 나눌 수 있었다. ATO와 ASO 균주들은 실험에 사용한 감자, 토마토, 가지, 고추에 모두 병원성이 있었고 ATO 균주는 특히 토마토에 강한 병원성이 있었으나, ASP 균주는 감자에만 병원성이 있었다. 이 연구에서 사용한 molecular marker중 ITS와 histon H3 유전자의 염기서열 분석은 4개의 형태적 group을 명확히 구분할 수 없었지만, URP-PCR 핵산지문분석법은 이들 group을 명확히 구분할 수 있었다. 분생포자의 형태, 병원성검정, 분자생물학적 특징을 종합할 때 토마토 겹무늬병에 관여하는 A. tomatophila는 감자겹둥근무늬병에 관여하는 A. solani와 뚜렷이 구분할 수 있었다. 또한 감자에서 분리한 Alternaria sp.(ASP)는 A. solani(ASO)와 형태적으로 유사하였지만 분생포자의 폭이 두껍고 부리(beak)가 작다는 점에서 ASO와 구별되었으며 가지과 작물에서 보고된 바 없는 신종으로 추정되었다.

우리나라 잣나무 털녹병균(病菌)의 기주선택성(寄主選擇性)과 병원성조사(病原性調査) (Studies on the Host Selectivity and Pathogenicity of White Pine Blister Rust Fungus (Cronartium ribicola J. C. Fischer ex Rabenhorst) in Korea)

  • 우리나라 잣나무털녹병균(病菌)의 동포자기주(冬胞子寄主)는 송이풀류(類)이나 인공접종(人工接種)을 통(通)하여 까치밥나무류(類)도 중간기주(中間寄主)임이 이미 밝혀져 있다. 그러나 최근(最近)에는 병원균(病原菌)의 분포지역(分布地域)에 따라 기주(寄主)의 선택(選擇)과 병원성(病原性)을 달리하는 새로운 생태형(生態型)이 있는 것으로 추정(推定)되고 있어, 본(本) 시험(試驗)에서는 지역적(地域的)으로 격리(隔離)된 강원도(江原道) 평창(平昌)과 전북(全北) 남원지역산(南原地域産) 털녹병균(病菌)의 수포자(銹胞子)를 까치밥나무류(類)와 송이풀에 접종(接種)하였다. 그 결과(結果) 양지역(兩地域)의 털녹병균(病菌)은 공(共)히 Ribes montigenum 과 송이풀에 하포자퇴(夏胞子堆)를 형성(形成)함으로서, 동일(同一)한 병원성(病原性)을 나타냈다. 한편, 우리나라에 분포(分布)하는 잣나무 3종(種)을 털녹병피해임지내(病被害林地內)에서 자연감염(自然感染) 시킨 결과(結果), 4년후(年後)의 발병율(發病率)은 잣나무가 17.7%, 스트로브잣나무가 26.3%이였으나 섬잣나무는 전혀 발병(發病)되지 않았다. 따라서 우리나라의 털녹병균(病菌)은 전국적(全國的)으로 동일(同一)한 생태형(生態型)으로, 잣나무와 스트로브잣나무를 수포자기주(銹胞子寄主)로 하며, 송이풀류(類)와 까치밥나무류(類)를 동포자기주(冬胞子寄主)로 하고 있다.

  • 잠두위조바이러스2 (BBWV2)는 경제적으로 중요한 원예, 관상작물에 심각한 피해를 주는 바이러스 중의 하나다. BBWV2의 넓은 기주 범위, 매개충 방제의 어려움 등 효과적인 치료제가 없기 때문에, 병을 예방하거나 저항성 품종의 개발 등을 위해서는 BBWV2의 새로운 분리주들의 특성 조사와 연구가 필요하다. 본 연구에서는 2019년 금산지역의 잎들깨 재배농가에서 분리된 BBWV2 분리주(BBWV2-GS-PF)의 유전학적·생물학적 특성을 구명하고 기존에 보고된 분리주들과의 특성을 비교하였다. 순수 분리된 BBWV2-GS-PF는 기존 분리주와 달리 들깨에서 심한 모자이크 증상과 고추에서 원형 반점 증상을 보였다. BBWV2-GS-PF의 유전자 계통분석 결과, 기존에 알려진 2개의 그룹과 약 76~80%의 비교적 낮은 유전자 상동성을 보이며 계통이 분화된 것을 확인할 수 있었다.

한우(韓牛)에 집단발생(集團發生)한 백선균증(白癬菌症)에 관한 연구(硏究) (Studies on Dermatophytosis of Korean Cattle)

  • This experiment was undertaken to determine the causative agent of dermatophytosis occurred in Korean native cattle. A total of 54 cases were examined, of which 52 were derived from a herd and the others from market. Direct microscopic examination, culture and pathogenicity test were performed for the samples (hairs and scales) obtained from the skin lesions of the affected cattle. The causative agent was identified as Trichophyton verrucosum exclusively in these cases.

  • A causal agent of bacterial soft rot occurring in soybean sprout cultivation in Korea was isolated and identified, and its incidence in several sprout-soybean cultivars was examined. Infected soybean seeds became light brown and whitish, and could not germinate until 3 days after seeding, accompanying rotting of soybean seeds and sprouts. The causal organism isolated from the rotten seeds and sprouts was identified as Erwinia carotovora subsp. carotovora on the basis of its pathogenicity, morphological and physiological characteristics and the results of the Biolog GN microplate test program. The bacterial soft rot by E. c. subsp. carotovora was firstly described in soybean sprout in Korea, and we name it“the bacterial soft rot of soybean sprout”. The disease occurred more frequently in Nam-hae and Fu-reun sprout-soybean cultivars than in Eun-ha, So-baek, and Ik-san cultivars.

First Report of Pitch Canker Disease on Pinus rigida in Korea

  • Pitch canker of Pinus rigida, caused by Fusarium subglutinans f. sp. pini, was first noticed in Inchon, Korea, and is now being spread into other plantations of pines. Typical symptoms re resin flows from canders on the shoots, branches, and trunks, and resin-soaking of the sap wood under the bark. Pathogenic fungi were isolated from the infected shoots, branches, and sees on Fusarium-selective medium. Mycelial growth and microscopic characteristics were examined. Pathogenicity test was carried out by inoculating four common species of pines (P. rigida, P. densiflora, P. thunbergii, and P. koraiensis) in Korea. P. rigida and P. thunbergii showed symptoms identical to those of naturally infected trees, while P. densiflora and P. koraiensis remained free symptomless.

Fusarium oxysporum과 Rhizoctonia solani에 의한 카네이션 줄기썩음병 (Stem Rot of Carnation Caused by Fusarium oxysporum and Rhizoctonia solani)

  • Incidence of stem rot on carnation (Dianthus caryophyllus L.) ranged 11 to 29% in Namwon and Chongup area during the growing seasons from 1993 to 1994. Among 129 isolates from carnations in Namwon, 77 isolates were identified as Rhizoctonia solani, 38 isolates were Fusarium oxysporum and 14 isolates were not identified. Among 169 isolates in Chongup, 19 isolates were identified as R. solani, 106 isolates as F. oxysporum and 44 isolates were not identified. Among 77 isolates of R. solani isolated from the specimens of Namwon, 52 isolates were classified as anastomosis group AG 2-2 by anastomosis test, 14 isolates as AG 2-1 and 11 isolates as AG 4. Among 19 isolates from specimens of Chongup, 14 isolates were classified as anastomosis group AG 2-2 and 5 isolates as AG 4. Pathogenicity tests revealed that isolates of F. oxysporum and R.solani AG 2-2 were highly virulent and isolates of R.solani AG 2-1 and AG 4 were mildly virulent.

Sclerotium rolfsii에의한 잇꽃 흰비단병 (Collar Rot of Safflower Caused by Sclerotium rolfsii)

  • A destructive collar rot of safflower occurred severely research farm of at Kyongsangnam-do Agricultural Research and Extension Services in 1999. Incidence of the disease at 3 fields in Chinju was ranged from 21.6 to 34.2% Upper parts of infected stems were mostly blighted and white mycelia were found on the lesions. The same fungus was isolated consistently from the infected tissues and confirmed its pathogenecity to safflower. The causal fungus of collar rot disease was identified as Sclerotium rolfsii by the examination of colony type sclerotium formation and pathogenicity test. This fungus also causes stem rot crown rot wilt or blight on the safflower. This is the first report on the collar rot of safflower caused by Sclerotium rolfsii in Korea.

제주도에서 처음으로 발생한 Phytophthora palmivora에 의한 심비디움 역병 (First Report of Phytophthora palmivora in Cheju Island as the Causal Pathogen of Phytophthora Crown Rot of Cymbidium)

  • Phytophthora crown rot of cymbidium was observed in Cheju island since June of 1996. The disease initiated at the basal portion of infected plant progressed upward to lower leaves. Soon after distinct water-soaking lesions appeared on lower leaves, the plant was wilted, blighted and died. Four orchid farms at Sogwipo out of 16 surveyed in the island were infected by the disease estimating 5~20% infection rates. The causal fungus was identified as P. palmivora based on following distinguishing characteristics. All isolates were heterothallic as A1 types and readily produced chlamydospores with cultural age. Sporangia were conspicuous papillate, ellipsoidal to ovoid, highly deciduous with short pedicels ca. 3~4 ${\mu}{\textrm}{m}$. Koch's rules were satisfied by a pathogenicity test and re-isolation of the fungus from inoculated plants. The pathogen has never been reported in Cheju island previously and its firstly recorded as the cause of Phytophthora crown rot of cymbidium in Korea.

  • A bacterial disease of pepper (Capsicum annuum L.) that rooted the stem nodes to black was found in pepper plants which cultivated in plastic house at Chungdo, Kyungpook, Korea in March, 1998. Bacterial isolates derived from the diseased peppers were pathogenic to potato, eggplant and Chinese cabbage but, was not pathogenic to chrysanthemum by artificial inoculation. On the basis of bacteriological characteristics and pathogenicity test on host plants, the causal organism of the node soft rot of pepper is identified as Erwinia carotovora subsp. carotovora and the name of disease is proposed as bacterial node soft rot of pepper.
