Bactericidal Activity of Sawa-wasabi (Wasabia japonica) Against the Fish Pathogenic Bacteria

  • In this study, the bactericidal activity of each extract from Sawa-wasabi (Wasabia japonica) root, stem and leaf against the fish pathogenic bacteria were examined. The main component related to bactericidal activity in Sawa-wasabi was well known to AlT. The Sawa-wasabi roots showed the highest AIT amount with 1.18 mg/g. Stems was 0.41 and leaves was 0.38 mg/g. All of them showed bactericidal activity against 2 strains of Vibrio hollisae, V. anguillarum, and 2 strains of Edwardsiella tarda, but weak effect against Staphylococcus capitis. The Sawa­wasabi leaves showed the strongest bactericidal activity with minimal bactericidal concentrations (MBCs) of 156.3mg of dry weight/mL against 2 strains of V. hollisae, V. anguillarum and 2 strains of E. tarda. The roots and stems showed a little weak bactericidal activities with 312-1,250mg of dry weight/mL against them. These results suggest that certain components besides AIT in Sawa-wasabi are affective in killing fish pathogenic bacteria.

A View of Soil Microbial Contamination on the Three Sandy Beaches in Busan (부산광역시 세 해수욕장 백사장에서 세균 오염도 조사)

  • Bacterial contamination of beach sand was examined in April, June, July, and August. Twenty four topsoil and subsoil samples were taken from the 1m and 5m points from the coastline in three Busan beaches (Haeundae, Gwanganli, and Songjeong). The 5m points from the coastline showed higher coliform contamination than 1 m points. July showed the highest bacterial contamination on beaches among surveyed months. Coliform contamination in the subsoil was higher than that of the topsoil. The bacterial contamination of 5m points of topsoil and subsoil in June except the Songjeong Beach was higher than those of July and August. We investigated Staphylococcus aureus, Vibrio parahaemolyticus, Vibrio vulnificus, Vibrio cholerae, and Bacillus cereus. Only B. cereus was detected at the beach in August. Although microbiological pollution of the Gwanganli Beach was the highest among three Busan beaches, the degree of contamination was not high compared to those of other countries. However, sandy soil management in public beach for pathogenic microorganisms is needed.

Distribution of Vibrio parahaemolyticus and Vibrio vulnificus in Seawater and Shellfish at Gomso Bay (곰소만의 해수 및 패류에 있어서 장염비브리오균(Vibrio parahaemolyticus)과 비브리오패혈증균(Vibrio vulnificus)의 분포)

  • In the case of being contaminated with pathogenic Vibrio, shellfish pose a serious threat for public health. This study was conducted to investigate the presence of Vibrio parahaemolyticus and V. vulnificus in the seawater, bottom deposit and shellfish samples collected from the Gomso Bay, west coast of Korea. V. parahaemolyticus (30-80 CFU/g) was detected from seawater Jul. to Oct. and highest at Sep. V. parahaemolyticus was detected less than 10 CFU/g from seawater at Nov. with $14.6^{\circ}C$ of seawater temperature. V. parahaemolyticus (180-1,850 CFU/g) was detected more than 100 CFU/g from 3 kinds of shellfish, Short-necked clam Ruditapes philippinarum, Corb shell Cyclina sinensis, and Surf clam Mactra veneriformis, Jul. to Oct. and was highest in Surf clam at Oct. V. parahaemolyticus was detected less than 20 CFU/g from all tested shellfish at Nov. V. vulnificus (1.8-2.7 MPN/100 mL) was detected from seawater Jul. to Oct. and was not detected at Nov. V. vulnificus (18-236.7 MPN/100 g) was also detected from 3 kinds of shellfish Jul. to Oct. and was highest in Corb shell at Jul. V. vulnificus was not detected from all tested shellfish at Nov.

Quantitative Microbial Risk Assessment of Pathogenic Vibrio through Sea Squirt Consumption in Korea (우렁쉥이에 대한 병원성 비브리오균 정량적 미생물 위해평가)

  • This study evalutated the risk of foodborne illness from Vibrio spp. (Vibrio vulnificus and Vibrio cholerae) through sea squirt consumption. The prevalence of V. vulnificus and V. cholerae in sea squirt was evaluated, and the predictive models to describe the kinetic behavior of the Vibrio in sea squirt were developed. Distribution temperatures and times were collected, and they were fitted to probabilistic distributions to determine the appropriate distributions. The raw data from the Korea National Health and Nutrition Examination Survey 2016 were used to estimate the consumption rates and amount of sea squirt. In the hazard characterization, the Beta-Poisson model for V. vulnificus and V. cholerae infection was used. With the collected data, a simulation model was prepared and it was run with @RISK to estimate probabilities of foodborne illness by pathogenic Vibrio spp. through sea squirt consumption. Among 101 sea squirt samples, there were no V. vulnificus positive samples, but V. cholerae was detected in one sample. The developed predictive models described the fates of Vibrio spp. in sea squirt during distribution and storage, appropriately shown as 0.815-0.907 of R2 and 0.28 of RMSE. The consumption rate of sea squirt was 0.26%, and the daily consumption amount was 68.84 g per person. The Beta-Poisson model [P=1-(1+Dose/β)] was selected as a dose-response model. With these data, a simulation model was developed, and the risks of V. vulnificus and V. cholerae foodborne illness from sea squirt consumption were 2.66×10-15, and 1.02×10-12, respectively. These results suggest that the risk of pathogenic Vibrio spp. in sea squirt could be considered low in Korea.

Effect of Spices on the Growth of Pathogenic Bacteria (향신료가 식중독세균의 증식에 미치는 영향)

  • The sensitivity of various pathogenic bacteria (Aeromonas hydrophila, Estherichia coli O157:H7, Listeria monocytogenes, Staphylococcus aureus 196E, Salmonella typhimurium and Vibrio parahaemolyticus) to the spices, allspice, clove, oregano, and thyme, was tested. Tryptic soy broth (TSB) containing 0∼2% (w/v) of spices was inoculated with 10sup 5/∼10$\^$6/ cells/$m\ell$ of each bacterium and incubated at 35$^{\circ}C$ for 24 hr. The growth of pathogenic bacteria was inhibited with increasing concentrations of spices in the culture broth. At 2% spice concentration, Gram positive bacteria were more sensitive than Gram negative bacteria with the exception of V. parahaemolyticus. Clove had the highest antibacterial activity, followed by allspice and oregano. At the concentration of 0.3%, clove inhibited the growth of all strains tested. Kanagawa-positive strain of V. parahaemolyticus displayed the highest sensitivity to clove and allspice. Thyme was the least effective for growth inhibition, while 1% clove killed all pathogens tested.

Altered expression of mud loach (Misgurnus mizolepis; Cypriniformes) hepcidin mRNA during experimental challenge with non-pathogenic or pathogenic bacterial species

  • Transcriptional response patterns of mud loach (Misgurnus mizolepis; Cypriniformes) hepcidin, a potential ortholog to human hamp1, in response to experimental challenges with non-pathogenic and pathogenic bacterial species were analyzed based on the semi-quantitative reverse transcription-PCR assay. Mud loach hepcidin transcripts were much more preferentially induced by pathogenic bacterial species (Edwardsiella tarda and Vibrio anguillarum) causing apparent pathological symptoms than by non-pathogenic species (Escherichia coli and Bacillus thuringiensis) displaying neither clinical signs nor mortality. However in overall, the induced amounts of hepcidin transcripts were positively related with the number of bacterial cells delivered in both pathogenic and non-pathogenic bacterial species. Inducibility of hepcidin transcripts were variable among three tissues examined (liver, kidney and spleen) in which kidney and spleen were more responsive to the bacterial challenge than liver. Time course expression patterns of hepcidin mRNAs after challenge were different between groups challenged with pathogenic and non-pathogenic species, although the overall pattern of hepcidin expression was in accordance with that generally observed in battery genes appeared during early phase of inflammation. Fish challenged with E. coli (non-pathogenic) showed the significant induction of hepcidin transcripts within 24 hr post injection (hpi) but the level was rapidly declined to the basal level either at 48 or 96 hpi. On the other hand, hepcidin transcript levels in E. tarda (pathogenic)-challenged fish were continuously elevated until 48 hpi, then downregulated at 96 hpi, although the level at 96 hpi was still significantly higher than control level observed in non-challenged fish. This expression pattern was consistent in all the three tissues examined. Taken together, our data indicate that hepcidin is tightly in relation with pathological and/or inflammation status during bacterial challenge, consequently providing useful basis to extend knowledge on the host defensive roles of hepcidin under infectious conditions in bony fish.

Purification and characterization of biochemical properties of hemolysin from Vibrio fluvialis (Vibrio fluvialis 유래의 hemolysin 정제와 생화학적 특성)

  • Hemolysin (VFH) of V. fluvialis, which is a pathogenic bacteria, causing watery diarrhea with vomiting, abdominal croup, was purified. V. fluvialis was cultivated in BHI medium and the culture supematant was precipitated by ammonium sulfate. The protein was purified by chromatographies on columns of DEAE-cellulose and Mono-Q. Molecular weight of the purified VFH was estimated as 79kDa by SDS-PAGE. The optimal temperature for a maximum hemolytic activity was at around 35$^{\circ}C$ and the activity was decreased at 4$0^{\circ}C$ Cytotoxicity of VFH was also investigated using RTG-2 cell line. LDH assay study showed that 50$\mu\textrm{g}$/m1 of VFH release 80% of total cellular LDH (lactate dehydrogenase) from RTG-2 cell and microscopic observation also showed the morphological change of cell.

Microbial Flora in Ascitic Cultivated Olive Flounder (Paralichthys olivaceus) in Koe-je Island in Korea During 2002-2003. (2002-2003년도 복수증 양식산 넙치로부터 동정된 미생물상)

  • A lot of cultured flounders died by ascitic disease blooming during the summer season in 2002, southern parts of Korea. This study was conducted to investigate the microbial flora for the ascitic cultivated olive flounder (Paralichthys olivaceus) which were collected in the Koe-je island in the southern of Korea from July 2002 to October 2003. Three Genera (Vibrio, Photobacterium and Edwardsiella), seven species of bacteria (V. harveyi, V. alginolyticus, V. parahaemolyticus, V. carcariae, V. metschenikovii, P. damsela and E. tarda) and a fish pathogenic birnavirus (marine birnavirus, MABV) were isolated from the liver, dropsy, spleen and identified by biochemical and molecular biological characterization.

Intraspecific Variation of Environmental and Clinical Vibrio vulnificus Isolates as Demonstrated by Restriction Endonuclease Digestion Profiles

  • Thirty-six environmental isolates of Vibrio vulnificus obtained from seawater, sediments, and raw seafoods, and 18 clinical isolates from Vibrio septicemia patients were typed by restriction endonuclease digestion profiles (REDP) of genomic DNA with SfiI. The results revealed a high-level of variation in REDPs, indicating a vast genomic diversity among V. vulnificus strains. Genetic relatedness of the strains showed similarities ranging from 10% to 100%. Different REDPs for isolates from various raw seafoods were obtained, and clustering of strains according to type of seafoods was not observed. In contrast, clinical isolates of V. vulnificus showed higher similarity to one another, and could be subdivided into one separate group. The difference in REDPs of the V. vulnificus isolates from clinical origin and from raw seafoods substantiates the previous observation that only a single type of pathogenic strain was involved in each human infection, despite the numerous genetically polymorphic strains found from implicated oysters.
