Microbiological Quality and Antibiotic Susceptibility of E. coli Isolated from Agricultural Water in Gyeonggi and Gangwon Provinces (경기, 강원 지역 농업용수의 미생물학적 특성 및 농업용수 분리 대장균의 항생제 내성)

  • BACKGROUND: Irrigation water is known to be one of the major sources of bacterial contamination in agricultural products. In addition, anti-microbial resistance (AMR) bacteria in food products possess serious threat to humans. This study was aimed at investigating the prevalence of foodborne bacteria in irrigation water and evaluating their anti-microbial susceptibility. METHODS AND RESULTS: Surface water (n = 66 sites) and groundwater (n = 40 sites) samples were collected from the Gyeongi and Gangwon provinces of South Korea during April, July, and October 2019. To evaluate the safety of water, fecal indicators (Escherichia coli) and foodborne pathogens (E. coli O157:H7, Salmonella spp., and Listeria monocytogenes) were examined. E. coli isolates from water were further tested for antimicrobial susceptibility using VITEK2 system. Overall, detection rate of foodborne pathogens in July was highest among three months. The prevalence of pathogenic E. coli (24%), Salmonella (3%), and L. monocytogenes (3%) was higher in surface water, while only one ground water site was contained with pathogenic E. coli (2.5%). Of the 343 E. coli isolates, 22.7% isolates were resistant to one or more antimicrobials (ampicillin (18.7%), trimethoprim-sulfamethoxazole (7.0%), and ciprofloxacin (6.7%)). CONCLUSION: To enhance the safety of agricultural products, it is necessary to frequently monitor the microbial quality of water.

  • Antimicrobial peptides (AMPs) play an important role in innate immunity against pathogenic infections. AMPs exterminate pathogenic bacteria by disrupting cell membranes or inhibiting intracellular molecules. NK-2, first identified in pigs and derived from NK-lysin, has antimicrobial effects against bacteria and parasites. In this study, chimeric peptides (cpNK) of chicken and pig NK-2 and cpNK-derived peptides (cpNK-a1 and cpNK-a2) were synthesized, and their antimicrobial effects against various pathogenic bacteria such as Escherichia coli, Salmonella spp., Listeria monocytogenes, Staphylococcus aureus, and methicillin-resistant Staphylococcus aureus (MRSA) were investigated. The structure of chimeric peptides from chicken and pig NK-2, cpNK, include α-helix like NK-2 and peptide net charge was +9 like porcine NK-2. The cpNK peptide showed powerful bactericidal effects against most bacterial species, including MRSA, especially against gram-negative bacteria. Furthermore, cpNK-derived short peptides, cpNK-a1 and a2 also showed bactericidal activity, but the effects were weaker than those of cpNK. Therefore, we conclude that cpNK- and cpNK-derived short peptides have the potential to be used as antibiotic alternatives.

Characterization of Lactobacillus cellobiosus D37 Isolated from Soybean Paste as a Probiotic with Anti-Cancer and Antimicrobial Properties

  • The probiotic characteristics of a total of 137 lactic acid bacterial strains isolated from soybean paste were investigated. Among those tested, the D37 strain was selected as a probiotic bacteria due to its acid and bile tolerance, and its strong anti-cancer and antibacterial activities. The D37 strain showed highly stable viability at acidic pH for 2 hr, and was very stable in 10% bovine bile. The viability of human colon cancer HT-29 cells was inhibited more than 60% at a $200\;{\mu}/mL$ concentration of D37 cell-free culture supernatant, and the degree of inhibition was concentration-dependent. The D37 strain showed a wide range of antibacterial activities against food-borne pathogenic bacteria such as Escherichia coli O157, Listeria spp., Vibrio spp., Salmonella spp., and Staphylococcus aureus. According to phenotypic characteristics and the utilization of various sugars, the D37 strain was identified as Lactobacillus cellobiosus.

Septicemia and Navel III Associated with Escherichia coli Infection in a Thoroughbred Foal (더러브렛 자마의 Escherichia coli 제대 감염에 의한 전신 패혈증)

  • Septicemic polyserositis and navel ill associated with Escherichia coli were reported in a 14-day-old male thoroughbred foal. The horse died after showing 12-day history of anorexia, lethargy, lameness and endophthalmus. Grossly, milky yellow abscesses were occupied in umbilicus, umbilical vein and artery. Large amounts of turbid pale yellow fluids were seen in pericardial sac, thoracic and abdominal cavity. Yellowish fibrinous materials were also presented in thoracic and abdominal cavity. Sticky pale yellow fluid and fibrinous materials were filled in stifle joint cavities of both hind limbs. Histologically, fibrino-purulent polyserositis and arthritis were observed. Severe omphalophlebitis with intra-lesional Gram negative bacterial colonies were noted in umbilical vein. Most of mesothelial cells in serosal cavities were severely hypertrophied. Pathogenic E. coli was purely isolated from ascites, thoracic and synovial fluids. Based on the results, the septicemic polyserositis may be originated from the umbilical cord infected with E. coli in this foal.

Feasibility Study of Producing an Organic Fertilizer using Sediments from Coastal Farming Areas (연안 양식장 퇴적물을 이용한 비료화 가능성 연구)

  • In an attempt to evaluate the possibility of producing an organic fertilizer using sediments from coastal farming areas, the chemical composition, bacteriological quality and heavy metals in the sediments alkalized by conditions : a 1:4 mixture of dry sediment to food wastes and the addition of 30% quicklime to the mixture. According to the classification standard for compost constituent by Higgins, all composts had a low or intermediate grade in T-N and $K_2O$ content, a low grade in $P_2O_5$ and a high grade in CaO and MgO content. Stabilization by quicklime and magnesium hydroxide is likely to inhibit th bacterial decomposition of organic matter and the actigity of pathogenic organic. Raising the pH of stabilized sediment to 12 for 2 hours(PSRP criteria of EPA) allowed 99.99% of the coliform group, fecal group and viable cell count to be reduced. the results suggested that the crude fertilizer produced by alkaline stabilization method was innoxious and thereby the sediments from coastal farming areas could be used as organic fertilizer.

Isolation of Bacillus atrophaeus MPL-01 from A Wild Boar and Characterization of Its Antifungal Activity (멧돼지 대장으로부터 Bacillus atrophaeus MPL-01의 분리 및 항진균 활성의 특성)

  • A bacterial strain MPL-01 was isolated from the large intestine of a wild boar. The strain was shown to have morphological, physiological and biochemical characteristics, fatty acids composition typical of Bacillus. The 16S rRNA gene sequence showed that the isolate formed distinct phyletic line that was most closely related to this of Bacillus atrophaeus (99.99%). It was proposed that the strain is classified as B. atrophaeus MPL-01. The strain MPL-01 exhibited the strongest antifungal activity against Colletotrichum acutatum, the pathogen of anthracnose of chili peppers. The ethyl acetate extract of culture filtrate possessed not only the antifungal activity but also the bio-surfactant activity. Therefore, the strain MPL-01 could be a useful bacterium in the development of bio-control process against the pathogenic fungi.

Comparative Analysis of Pathogenic Organisms in Cockroaches from Different Community Settings in Edo State, Nigeria

  • Cockroaches are abundant in Nigeria and are seen to harbour an array of pathogens. Environmental and sanitary conditions associated with demographic/socio-economic settings of an area could contribute to the prevalence of disease pathogens in cockroaches. A total of 246 cockroaches (Periplaneta americana) in urban (Benin, n=91), semi-urban (Ekpoma, n=75) and rural (Emuhi, n=70) settings in Edo State, Nigeria were collected within and around households. The external body surfaces and alimentary canal of these cockroaches were screened for bacterial, fungal, and parasitological infections. Bacillus sp. and Escherichia coli were the most common bacteria in cockroaches. However, Enterococcus faecalis could not be isolated in cockroaches trapped from Ekpoma and Emuhi. Aspergillus niger was the most prevalent fungus in Benin and Ekpoma, while Mucor sp. was predominant in Emuhi. Parasitological investigations revealed the preponderance of Ascaris lumbricoides in Benin and Emuhi, while Trichuris trichura was the most predominant in Ekpoma. The prevalence and burden of infection in cockroaches is likely to be a reflection of the sanitary conditions of these areas. Also, cockroaches in these areas making incursions in homes may increase the risk of human infections with these disease agents.

Detection of Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus and Listeria monocytogenes in Kimchi by Multiplex Polymerase Chain Reaction (mPCR)

  • We developed an mPCR assay for the simultaneous detection, in one tube, of Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus and Listeria monocytogenes using species-specific primers. The mPCR employed the E. coli O157:H7 specific primer Stx2A, Salmonella spp. specific primer Its, S. aureus specific primer Cap8A-B and L. monocytogenes specific primer Hly. Amplification with these primers produced products of 553, 312, 405 and 210 bp, respectively. All PCR products were easily detected by agarose gel electrophoresis, and the sequences of the specific amplicons assessed. Potential pathogenic bacteria, in laboratory-prepared and four commercially available kimchi products, were using this mPCR assay, and the amplicons cloned and sequenced. The results correlated exactly with sequences derived for amplicons obtained during preliminry tests with known organisms. The sensitivity of the assay was determined for the purified pathogen DNAs from four strains. The mPCR detected pathogen DNA at concentrations ranging from approximately 0.45 to $0.05\;pM/{\mu}l$. Thus, this mPCR assay may allow for the rapid, reliable and cost-effective identification of four potentially pathogens present in the mixed bacterial communities of commercially available kimchi.

Production of Monoclonal Antibodies Specific to FimA of Porphyromonas gingivalis and Their Inhibitory Activity on Bacterial Binding

  • Background: The FimA of Porphyromonas gingivalis is a crucial pathogenic component of the bacteria and has been implicated as a target for vaccine development against the periodontal diseases. Methods: In this study, the purified fimbriae (FimA subunit polymers) protein was used for immunization in their native form and B hybridoma clones producing antibodies specific to FimA were established. Results: The monoclonal antibodies prepared from selected two clones, designated #123 (IgG2b/ kappa) and #265 (IgG1/kappa), displayed different patterns of binding activity against the cognate antigen. Both antibodies reacted with conformational epitopes expressed by partially dissociated oligomers, but not with monomer as elucidated by Western blot analysis. Ascites fluid containing the monoclonal antibodies showed the inhibitory activity against P. gingivalis to saliva-coated hydroxyapatite beads, an in vitro model for the pellicle-coated tooth surface. Conclusion: These results suggest that the monoclonal antibodies could be used as vaccine material against the periodontal diseases through passive immunization.

Screening and Identification of an Antifungal Pseudomonas sp. That Suppresses Balloon Flower Root Rot Caused by Rhizoctonia solani

  • A pathogenic fungus causing balloon flower root rot (Platycodon grandiflorum) was isolated from naturally infected roots. The microbial characteristics of the isolated microorganism were similar to those of Rhizoctonia solani. About 500 bacterial species from field soils were screened for a biological agent against the above-mentioned putative pathogen, and several bacteria with the antifungal activity were isolated. Among them, the isolated JS2 was identified as Pseudomonas aeruginosa. This strain showed a broad spectrum of antifungal activity potentially. When the antifungal substance was purified from a broth culture of JS2, it was identified as 2,4-diacetylphloroglucinol (Phl).
