수중 비열 유전체장벽 방전 플라즈마를 이용한 양식어류의 병원성세균 3종 및 Tetracycline계 항생제 제거 (Remove of Three Pathogenic Bacteria in Cultured Fish and Tetracycline Antibiotics Using Underwater Non-Thermal Dielectric Barrier Discharge Plasma)

  • The purpose of this study is to evaluate the effect of underwater non-thermal dielectric barrier discharge plasma (DBD plasma) on the sterilization of three types of pathogenic bacteria that cause diseases in freshwater fish and the reduction of a tetracycline antibiotics. This experiment was conducted in the DBD plasma generator, and the voltages used to generate plasma were 11.6 kV and 23.1 kV. The measurement intervals were 0, 1, 5, 10 and 15 min. As a result of DBD plasma treatment, Aeromonas hydrophila, Edwardsiella tarda and Pseudomonas fluorescens were removed 93-99% after 5 min at 23.1 kV, and the tetracycline antibiotics were reduced 70-95% after 15 min at 23.1 kV. In this study, as a result of treating the effluent with DBD plasma at a fish farm where the medicinal bath was conducted with oxytetracycline-HCl (OTC-HCl) products, OTC-HCl decreased by 62% after 10 min at 23.1 kV.

  • 세균성 어류질병의 치료 및 예방을 위하여 어류와 인체에 안전한 수산용 probiotic의 개발이 요구되고 있다. 본 연구에서는 남해안 일대에서 양식되고 있는 참굴 (Crassostrea gigas), 바지락 (Ruditapes philippinarum), 피조개 (Scapharca broughtonii), 새조개 (Fulvia mutica)의 가식 부위로부터 17균주의 candidate probiotic bacteria (CPB)를 분리하였다. 나아가 다양한 연쇄상구균 (Lactococcus garvieae, L. piscium, Streptococcus sp., S. iniae, and S. parauberis)에 대하여 강한 생장 억제력을 보이는 균주(CPB-St)를 선별하여 어류의 연쇄구균증 관리를 위한 probiotic 균주로서의 개발 가능성을 알아보았다. CPB-St 균주를 double layer test를 통하여 다양한 연쇄상구균들에 대한 생장 억제 정도를 알아보았으며, 혼합 배양에서의 Streptococcus sp.에 대한 생장 억제 능력을 확인하였고, CPB-St의 어류에 대한 안전성 및 장내 생존 유무를 평가하였다. CPB-St는 대부분의 연쇄상구균에 대하여 18~26 mm의 생장 억제대를 형성함으로써 높은 생장 억제 능력을 보였다. CPB-St와의 혼합 배양에서 Streptococcus sp.는 6시간째부터 생장 억제 현상이 관찰되기 시작하여 12시간 전후로 8~55배 정도의 감소를 나타내었다. CPB-St의 어류에 대한 병원성 유 무를 알아본 결과, 2주일 동안 CPB-St로 인한 어류 폐사는 관찰되지 않았다. CPB-St의 1회 강제 경구 투여 후, 위와 장에서 CPB-St의 생존을 확인한 결과 투여 24시간 후에는 CPB-St가 모두 배출되는 것으로 나타났다. CPB-St를 probiotic bacteria로 개발하기 위해서는 어류의 장내 정착 가능성과 먹이를 통한 효과의 정확한 검증이 추후 이루어져야 할 것이다.

2016년도 국내 해양환경내 병원성 비브리오균의 분포 및 해양환경인자간의 상관성 분석 (Distribution of Pathogenic Vibrios in the Aquatic Environment Adjacent to Coastal Areas of South Korea and Analysis of the Environmental Factors Affecting Their Occurrence)

  • Objectives: The pathogenic Vibrios genus denotes halophilic bacteria that are distributed in aquatic environments, including both sea and freshwater. Vibrio cholerae, Vibrio vulnificus, and Vibrio parahaemolyticus are the most important species since they can be potent human pathogens and leading causes of septicemia, wound infections, and seafood borne gastroenteritis. The recent emergence of a potential pandemic clone, V. cholera serotype O1 and the cholera outbreak in South Korea in 2016 indicates the importance of consistent surveillance of pathogenic Vibrio genus within coastal areas. Methods: The present study was undertaken to determine where and how vibrios live in the aquatic environment adjacent to coastal areas of South Korea. For this survey, a total of 838 samples were obtained at 35 different sites in South Korean coastal areas during the period from January 2016 to December 2016. Pathogenic vibrios was determined using the real-time PCR method, and its clones were isolated using three selective plating media. We also monitored changes in seawater and atmospheric temperature, salinity, turbidity, and hydrogen ion concentration at the collection points. Results: The total isolation rates of V. vulnificus, V. cholera (non-pathogenic, non-O1, non-O139 serogroups), and V. parahaemolyticus from seawater specimens in 2016 were 14.2, 13.48, and 67.06%, respectively. Conclusions: The isolation rates of pathogenic vibrios genus showed a positive correlation with temperature of seawater and atmosphere but were negatively correlated with salinity and turbidity.

가금 유래 병원성 대장균의 항균제 내성 및 R plasmid 전달 양상 (Antimicrobial resistance and transfer of R plasmid of pathogenic Eseherichia coli isolated from poultry in Korea)

  • Antimicrobial drugs are widely used in poultry industry as growth promoters or to control infectious diseases. However, this practice is reported to have caused high resistance to antimicrobial drugs in normal chicken flora and pathogens. Antimicrobial resistance to Escherichia coli (E. coli) from chicken has been mainly reported in normal flora, but rare in pathogenic organism in Korea, recently. Therefore, this study was conducted to investigate prevalence of antimicrobials resistance, transfer of R plasmid, and association between antimicrobial drug resistance and O serotype of 203 pathogenic E. coli from poultry in Korea during the period from April 2003 to December 2005. These isolates showed a high resistance to tetracycline (Tc, 93.6%), nalidixic acid (Na, 92.6%), streptomycin (Sm, 81.8%), ampicillin (Ap, 77.3%), ciprofloxacin (Ci, 70.9%), sulfisoxazole (Su, 66.5%), and trimethoprim (Tp, 58.1%). Two hundred-one (99.0%) of the isolates were resistant to one or more drugs. They showed 57 different resistant patterns, and the most prevalent resistant pattern among them was Tc, Sin, Su, Ap, Tp, Ci, Na. Sixty-eight (33.8%) of the isolates transferred all or a part of their antimicrobial resistant pattern to the recipient strain by R plasmid. The most common antimicrobial resistant pattern was Tc, Sm, Su, Ap, Tp, Ci, Na in serotype O78, O88 and O15, respectively. These results exhibit high individual and multiple resistance to antimicrobials of pathogenic E. coli from poultry in Korea. They also suggest the needs for surveillance to monitor antimicrobial resistance in pathogenic bacteria that can be potentially transmitted to humans from food animals and to regulate the abuse of antimicrobials on food-producing animals in Korea.

인천 연안지역의 해양환경요인과 병원성 비브리오균의 분포와의 관련성 (Relationships of Pathogenic Vibrios and Environmental Factors Affecting their Occurrence in the Seawater of Incheon Coastal Area)

  • The aim of this study was to investigate the effects of environmental factors such as temperature, salinity, turbidity and pH on the growth of pathogenic Vibrios. In this study, we was obtained the samples from 2 different sites of the Incheon coastal area between January 2012 and December 2012. The water temperature in August and September was the high. the Incheon port changes the width of a small, wherease in the case of Hanjin harbor of changes of larger width. Salinity and turbidity showed significant differences, whereas temperature and hydrogen ion concentration was not significant. Pathogenic vibrios was determined using the real-time PCR method. Pathogenic vibrios in the Incheon port and Hanjin harbor were detected in 11 samples (91.67%) and 9 samples (75.0%) of Vibrio cholerae, 7 samples (58.3%) and 6 samples (50.0%) of V. vulnificus, 10 samples (83.3%) and 12 samples (100.0%) of V. parahaemolyticus, respectively. Pathogenic Vibrio bacteria were the highest at $26.8^{\circ}C$ of seawater in August. Quantitative results were the following: 102 $cell/m{\ell}$ in Vibrio cholerae, 7.876 $cell/m{\ell}$ in V. vulnificus, and 503.4 $cell/m{\ell}$ in V. parahaemolyticus, respectively. The enumeration of pathogenic vibrios showed a positive correlation with temperature and pH, but was negatively correlated with salinity and turbidity.

잔디 토양전염성병원진균에 대한 길항미생물의 분리 및 길항효과 (Isolation of Antibiotic-producing Microorganisms Antagonistic to Soilborne Pathogenic Fungi of Bentgrass and Their Antifungal Activity)

  • Recently, the importance of management and cultivation of grasses has been increased in Korea. Among these cultural practices, the appropriate control of diseases is considered more important than other cultivation techniques such as fertilization and irrigation. The damages of brown patch and large patch caused by Rhizoctonia spp. and Pythium blight caused by Pythium spp. are serious in the major cultivation area of turfgrass in Korea. Since these diseases are difficult to control by agrochemicals, the damages are very serious if these are occured. The periodic spray of agrochemicals, to protect and control these diseases could make some problems of toxicity and environmental pollution as well as rising of non-target diseases. Therefore, the biological methods to control diseases have been required to decrease problems resulted from overuse of agrochemicals, to conserve natural ecosystem, and to control effectively diseases of grasses in the long period. The number of studies about biological control using antagonistic microorganisms have been increased for last half century. However, the application of biological control method has been very limited. In this study, thirteen isolates of R. cerealis, 8 isolates of R. solani and 3 isolates of Phthyn spp. have been isolated from diseased turfgrass in golf course and grass-culture area that have patch and wilting symptoms of zoysia grass and creeping bentgrass. Isolation frequency of R. cerealis and R. solani was high in especially zoysiagrass, while Pythym spp. was isolated from bent grass at low frequency but showed high pathogenicity. Totally, 205 isolates of soil microorganisms were isolated in this study as primary antagonistic microorganism by Herr's triple agar layer plate and dual culture method using rhizosphere of grasses, soil of crop field as the source of antagonistic microorganisms. Among the 205 isolates, 23 isolates were actinomycetes and 182 isolates were bacteria. All of the actinomycetes were isolated by Herr's method. Antagonistic effect of primary isolated microorganisms was tested for in vitro mycelial growth inhibition against pathogenic fungi isolated from grasses and for inhibition of disease occurrence in 24 well tissue culture plate and pot experiment. Then, four isolated of bacteria which are BG23, BG74, BG136 and BG171 were selected as antagonistic microorganisms against soil-born pathogenic fungi of bentgrass.

온도변화에 따른 건조 스트레스 환경에서 고추 세균점무늬병 발생 영향 (Evaluation of Bacterial Spot Disease of Capsicum annuum L. in Drought Stress Environment by High Temperature)

  • The global warming by increased $CO_2$ will effect of plant pathogenic microorganisms and resistance of host plants, and it is expected to affect host-pathogen interactions. This study used Capsicum annuum L. and Xanthomonas euvesicatoria, a pathogenic bacteria of pepper, to investigate interactions between hosts and pathogens in a complex environment with increasedcultivation temperature and drought stress. As a result, the bacterial spot disease of C. annuum L. caused by X. euvesicatoria was $35^{\circ}C$ higher than $25^{\circ}C$. In addition, the effect on water potential on bacterial spot disease was much greater water potential -150 kPa than -30 kPa. The disease progress and severity higher than water potential -30 kPa. This result will useful for understanding interaction with red pepper and X. euvesicatoria under the complex environment with increased cultivation temperature and in water potential -150 kPa drought stress in the future.

Paenibacillus polymyxa JB115로부터 생산된 항균물질의 특성 (Characteristics of the Antibacterial Substance Produced by Paenibacillus polymyxa JB115)

  • 가축 질병의 제어를 위해 면역증강제인 베타글루칸 생성균으로 분리한 Paenibacillus polymyxa JB115의 배양액이 가축병원성 세균에 대한 항균성을 보임에 따라, 배양액의 동결건조 분말을 대상으로 항균물질의 특성을 조사하여 다기능 가축 사료첨가제로의 개발 가능성을 타진하였다. 그 결과, P. polymyxa JB115에서 생산된 항균물질은 E. coli, S. aureus, S. choleraesuis 및 P. aeruginosa 등 가축 병원성 세균들에 대해 살균 또는 정균 활성을 보였으며, 항균범위도 광범위하게 나타났다. 또한 이 물질은 높은 내열성과 pH 안정성을 지녀 배양액의 제형화에도 적합하였으며, 인공 담즙산 및 인공 위액에서도 안정하여 사료첨가제로 투여시 가축의 장내 병원세균에 대해서 항균 활성을 충분히 나타낼 수 있을 것으로 판단되었다. 상기의 결과로부터 P. polymyxa JB115의 배양액은 베타글루칸 이외에도 가축 병원성 세균에 대한 항균물질을 지니고 있어서, 다기능 사료첨가제로 개발 가능함을 확인하였다.

감마선(60Co) 조사에 의한 항진균 세균의 돌연변이체 유도 (Mutant Induction of Several Antifungal Bacteria by Gamma Radiation (60Co))

  • 항진균 세균의 특성 및 기능변화 가능성을 조사하기 위하여 버섯폐배지, 온천수, 해조류 및 삼림토양으로부터 식물병원성 진균에 대한 8종의 항진균 활성 균주를 분리하였고 감마선($^{60}Co$)을 이용하여 $LD_{95}$에서 돌연변이체를 유도하였다. Bacillus circulans K1, Burkholderia gladioli K4와 Bacillus subtilis YS1은 12 종의 식물병원성 진균에 대해 항진균 활성을 보였다. 이들 균주의 방사선감수성 조사결과 B. gladioli K4는 감마선에 대한 높은 감수성을 보였으며, $D_{10}$ 값은 0.11 kGy 였다. 감마선에 의해 유도된 K1-1004와 YS1-1009는 Botryosphaeria dothidea에 대해 항진균 활성이 증가되었다. B. subtilis YS1의 돌연변이체인 YS1-1006과 YS1-1009는 tebuconazol과 copper hydroxide에 대해 농약 저항성을 나타냈다. SAR535, SAR5108 과 SAR5l18 돌연변이체는 야생형 균주인 Streptomyces sp. SAR01에 비해 5 종의 식물병원성 진균에 대해 항진균 활성이 없었다. 연구결과, 방사선을 이용하여 다양한 기능의 돌연변이체 유도가 가능하였다. 이를 이용하여 항진균 활성 관련 유전자 연구 및 균주개량이 가능할 것으로 사료된다.

생육 시기에 따른 자두류 중 후무사의 식중독균에 대한 저해효과 (Antimicrobial Activities of 'Formosa' Plum at Different Growth Stages against Pathogenic Bacteria)

  • 자두류중 후무사를 6월 말경에 처음으로 수확하여 5일 간격으로 6차례 채취한 다음 최종 수확 10∼25일전 까지의 후무사 1-4를 미숙과로, 최종 수확 0∼5일전까지의 후무사 5, 6은 완숙과로 구분한 후 각각 메탄을 추출물을 제조하여 대표적인 식중독 세균 4종에 대한 항균력을 조사하였다. 각 균에 대한 후무사의 항균력은 시료 농도가 증가할수록 높은 저해를 보였고, 특히 5.0 mg/disc의 농도에서는 이들 4종의 균 모두에서 가장 높은 항균활성을 나타내었다. 또한 완숙과에 비해 미숙과에서 높은 항균활성을 보였으며, 후무사 1, 2의 경우 가장 강한 항균활성을 보였다. E. coli O157:H7와 S. tyhimurium에 대한 후무사의 항균력도 완숙과에 비해 미숙과에서 항균력이 높았고, 특히 5.0 mg/disc 농도에서는 E. coli에 비해 S. typhimurium에서 항균활성이 감소되었다. 그람양성균인 고 L. monocytogenes와 S. aureus에 대한 후무사의 항균력도 완숙과에 비해 미숙과에서 높은 항균활성을 보이고, 시료 농도가 증가할수록 더 큰 억제 효과를 보였다. 그리고 후무사 미숙과의 최소 저해 농도는 E coli의 경우 320 $\mu\textrm{g}$/mL, S. aureus에서 160 $\mu\textrm{g}$/mL, S. typhimurium과 L. monocytogenes은 640 $\mu\textrm{g}$/mL이었다. 이와 같이 후무사 메탄올추출물은 4종의 식중독균에 대해 우수한 항균활성을 보였고, 특히 완숙과에 비해 미숙과에서 높은 항균활성이 존재함을 확인하였다.
