Sixteen adult Barbari bucks were divided into 4 homogeneous groups, each group was offered wheat straw (WS) at levels 75, 100, 125 and 150 per cent of ad libitum intake in a factorial randomized complete block design. All goats were uniformly fed a supplement ($13.5g\;DM/kg\;W^{0.75}$) to fulfill their requirement for protein at maintenance level. The experimental feeding of goats at different levels for 21 days was followed by a digestion trial of 6 days and determination of rate of passage of digesta for 144 h duration, respectively. Intake ($g/kg\;W^{0.75}$) of dry matter (DM), organic matter (OM), digestible DM and digestible OM increased with increase in amount of WS offered. The levels of WS refusals for obtaining upper limit of intake was found to be above 30% of DMO. Concentration of ADF and lignin was significantly lower and CP was higher (p>0.05) in food ingested relative to food offered or refused in response to increasing allowance of WS. Dietary means of particulate rate of passage from rumen, transit time and total mean retention time ranged from 0.032 to 0.036/h, 19.10 to 21.72/h and 58.61 to 61.53/h respectively and did not show significant differences (p>0.05) irrespective of dietary level of WS. The results suggest that DMI and quality of ingested WS would improve with higher rates of offer and refusals (${\geq}30%$ of DMO) without any pronounced effect on passage kinetics of digesta, because of the greater opportunity afforded to goats for selective feeding.
Mesenchymal stem cells (MSCs) are often known to have a therapeutic potential in the cell-mediated repair for fatal or incurable diseases. In this study, canine umbilical cord MSCs (cUC-MSCs) were isolated from umbilical cord matrix (n = 3) and subjected to proliferative culture for 5 consecutive passages. The cells at each passage were characterized for multipotent MSC properties such as proliferation kinetics, expression patterns of MSC surface markers and self-renewal associated markers, and chondrogenic differentiation. In results, the proliferation of the cells as determined by the cumulative population doubling level was observed at its peak on passage 3 and stopped after passage 5, whereas cell doubling time dramatically increased after passage 4. Expression of MSC surface markers (CD44, CD54, CD61, CD80, CD90 and Flk-1), molecule (HMGA2) and pluripotent markers (sox2, nanog) associated with self-renewal was negatively correlated with the number of passages. However, MSC surface marker (CD105) and pluripotent marker (Oct3/4) decreased with increasing the number of subpassage. cUC-MSCs at passage 1 to 5 underwent chondrogenesis under specific culture conditions, but percentage of chondrogenic differentiation decreased with increasing the number of subpassage. Collectively, the present study suggested that sequential subpassage could affect multipotent properties of cUC-MSCs and needs to be addressed before clinical applications.

The differences in rumen particle pool size, passage rate and rumen degradability in sheep receiving three varieties of orchardgrass hay harvested at pre-heading (H1), early-bloom (H2) and late-bloom (H3) were investigated using four ruminal-cannulated wethers (68 kg) fed 1,300 g of the hay once a day. Representative samples of whole rumen contents were collected at different times after feeding and the quantities of rumen particle pools [large particle pool (LPP), retained on a $1,180{\mu}m$ sieve; small particle pool (SPP), retained on a 47 but passed a $1,180{\mu}m$ sieve; and soluble fraction (SOL), passed a $47{\mu}m$ sieve (SOL)] were determined by a wet-sieving technique. The fullowing results were obtained: 1) The dry weight of whole rumen contents were significantly lower (p < 0.05) for HI than for H2 or H3. The reduction rate of whole rumen contents was slightly but significantly greater for HI that, the other hay varieties. 2) The LPP disappearance rates were 26.2, 25.3 and 21.7 g DM/h for H1, H2 and H3, respectively, and no statistical differences were found among the hay varieties. Appreciable changes were not observed with SPP and SOL throughout measurements for all hay varieties; however the SPP was markedly greater (p <0.05) for H2 and ill than for HI, while SOL did not differ among hay varieties. 3) The SPP passage rate (g DM/h) and effective rumen degradability (%) for HI, H2 and ill were, respectively, 9.7, 56.6; 16.9, 42.3; and 18.0, 28.9. The ruminal tum-over rate for SPP appeared to be higher for HI than for the other hay varieties.

This paper is aiming at calculating NOx concentration, which is one of the harmhul components of emission from the gasoline engine, formed in the combustor through the presess of combustion. Instantaneous temperature and concentration of each components for each division can be determined by the solution of simulatneous equation of reaction equation and equation of energy conservation, inputting the estimated temperature with a considerably wide rage of temperature. After determining instantaneous temperature and instantanous concentration of each components, the nonequilibrium calculation is performed based on the reaction kinetics in order to determine NOx concentration. To summarize the result abtained from the above method ;through the passage of NO concentration, NO concentration is the highest in the first division and it is gradually decreasing through the following divisions In the final division, NO concentation is the lowest.
Normal somatic cells proliferate for a limited number of doublings in culture and then enter an irreversible growth-arrest state called replicative senescence. Replicative senescence has been believed a reason for the limited cellular turnover and deterioration of tissue function in aged animals. However, there is no experimental evidence supporting this assumption. Furthermore, cells from aged person have been poorly characterized with an exception of the cases of T cells. In this study, we examined cell biological changes occurring in replicative senescence of fibroblast strains originated from a new-born (NHF-NB) and a 87 year old man (NHF-87). NHF-87 (and the cells from a 75-year old) proliferated to smaller population doublings and with longer doubling times than NHF-NB did. At early passages, NHF-87 exhibited a low senescence-associated ${\beta}-Gal$ (SA ${\beta}-Gal$) activity and lipofuscin level, typical markers for cellular senescence. Furthermore, they maintained low levels of lysosome and reactive oxygen species (ROS). All of these levels increased dramatically in the late passage NHF-87 quite similarly as those in the late passaged NHF-NB did. These results indicate that most cells originated from the aged maintain a phenotype of the cells originated from new-born donors and undergo replicative senescence with the same kinetics as that of the cells from new-born. It is also indicated that not SA ${\beta}-gal$ activity but cell proliferation rate may be qualified as a biomarker for cells aged in vivo.
We have exmained the re-establishment of HIMRE mediated silencing function on the transcriptional activity of yeast heast shock gene HSP82. To test whether the onset of SIR repression can occur in growing cells in the rpesence of a potent inhibitor of DNA replication, HMRa/HSP82 strains with SIR4- and SIR4S$^{+}$ genetic backgrounds were arrested in S phase by incubation of a culture in 200 mM hydroxyurea for 120 min. It was clear that following a 20 minute heat shock, silencing of the HMRa/HSP82 allele in cells pretreated with hydroxyurea does occur in a SIR4-dependen fashion, even though the kinetics of repression appears to be substantially delayed. We also have tested whether re- establishement of silencing at the HMR/hsp82 locus can occur in G1-arrested cells. Cell cycle arrest at G1 phase was achieved by treatment of early log a cell cultures with .alpha.-factor mating pheromone, which induces G1 arrest. The result suggests that passage through S phase (and therefore DNA replication) is nor required for re-establishing silencer-mediated repression at the HMNRa/HSP82 locus. Finally, to test whether de nono protein synthesis is required for re-establishment of silencer-mediated repression, cells were pretreated with cycloheximide (500 /.mu.g/ml) 120 min. It was apparent that inhibiting protein synthesis delays, but does not prevent, re-establishment of silencer-mediated repression. Altogether, these results indicate that re-establishment of silencer-mediated repression is not dependent on the DNA replication and has no requirement for protein synthesis.s.
Growing interest in the nasal route as a drug delivery system calls for a reliable in vitro model which is crucial for efficiently evaluating drug transport through the nasal cells. Various in vitro cell culture systems has thus been developed to displace the ex vivo excised nasal tissue and in vivo animal models. Due to species difference, results from animal studies are not sufficient for estimating the drug absorption kinetics in humans. However, the difficulty in obtaining reliable human tissue source limits the use of primary culture of human nasal epithelial cells. This shortage of human nasal tissue has therefore prompted studies on the "passage" culture of nasal epithelial cells. A serially passaged primary human nasal epithelial cell monolayer system developed by the air-liquid interface (ALI) culture is known to promote the differentiation of cilia and mucin gene and maintain high TEER values. Recent studies on the in vitro nasal cell culture systems for drug transport studies are reviewed in this article.

Multilayered WC-Ti/suv $1-x/Al_{x}$ N coatings were deposited on AISI D2 steel using cathodic arc deposition (CAD) method. These coatings contain structural defects such as pores or droplets. Thus, the substrate is not completely isolated from the corrosive environment. The growth defects (pores, pinholes) in the coatings are detrimental to corrosion resistance of the coatings used in severe corrosion environments. The localized corrosion behavior of the coatings was studied in deaerated 3.5 wt.% NaCl solution using electrochemical techniques (potentiodynamic polarization test) and surface analyses (GDOES, SEM, AES, TEM). The porosity was calculated from the result of potentiodynamic polarization test of the uncoated and coated specimens. The calculated porosity is higher in the $WC-Ti_{0.6}$$Al_{0.4}$ N than others, which is closely related to the packing factor. The positive effects of greater packing factor act on inhibiting the passage of the corrosive electrolyte to the substrate and lowering the localized corrosion kinetics. From the electrochemical tests and surface analyses, the major corrosion mechanisms can be classified into two basic categories: localized corrosion and galvanic corrosion.

In the framework of the Generation IV research and development project, in which the French Commission of Alternative and Atomic Energies (CEA) is involved, a main objective for the design of Sodium-cooled Fast Reactor (SFR) is to meet the safety goals for severe accidents. Among the severe ones, the Unprotected Transient OverPower (UTOP) accidents can lead very quickly to a global melting of the core. UTOP accidents can be considered either as slow during a Control Rod Withdrawal (CRW) or as fast. The paper focuses on fast UTOP accidents, which occur in a few milliseconds, and three different scenarios are considered: rupture of the core support plate, uncontrolled passage of a gas bubble inside the core and core mechanical distortion such as a core flowering/compaction during an earthquake. Several levels and rates of reactivity insertions are also considered and the thermal-mechanical behavior of an ASTRID fuel pin from the ASTRID CFV core is simulated with the GERMINAL code. Two types of fuel pins are simulated, inner and outer core pins, and three different burn-up are considered. Moreover, the feedback from the CABRI programs on these type of transients is used in order to evaluate the failure mechanism in terms of kinetics of energy injection and fuel melting. The CABRI experiments complete the analysis made with GERMINAL calculations and have shown that three dominant mechanisms can be considered as responsible for pin failure or onset of pin degradation during ULOF/UTOP accident: molten cavity pressure loading, fuel-cladding mechanical interaction (FCMI) and fuel break-up. The study is one of the first step in fast UTOP accidents modelling with GERMINAL and it has shown that the code can already succeed in modelling these type of scenarios up to the sodium boiling point. The modeling of the radial propagation of the melting front, validated by comparison with CABRI tests, is already very efficient.

The insulin-like growth factors(IGFs) are bound to several binding proteins(IGFBPs) that appear to regulate IGF transport, receptor binding, and its action. The concentration of these peptides are altered by catabolic conditions. To determine IGF-I and IGFBP levels in noninsulin-dependent diabetes mellitus (NIDDM), sera was obtained from 5 patients and 7 controls. Serum levels of IGF-I in NIDDM were lower than those in either of the controls. By western immunoblot analysis, especially IGFBP-1 levels are increased, whereas IGFBP-3 levels decreased and their fragments was increased in NIDDM serum. IGFBP-3 proteolytic activity in NIDDM sera was inhibited by phenylmethylsulfonylfluoride (PMSF), aprotinin, and ethylenediaminetetraacetic acid(EDTA). This pattern of inhibition was consistent with a metal-dependent serine protease. By gelatin zymography, these proteolytic enzymes were identified as the size of 97 and 69 kDa. IGFBP-1, which is primarily insulin regulated, was increased in NIDDM and may modulate circulating IGF-I levels by regulating capillary passage of IGF-I. IGFBP-3 proteolysis markedly reduces its affinity for the IGFs, particularly for IGF-I. This accelerates their kinetics of dissociation, thereby increasing the proportions of IGF-I in free form and its availability to the cells.
