Characteristics of $\beta$-Glucosidase Immobilized on the Modified Chitin in Bioresctors (수식 Chitin에 고정된 $\beta$-Glucosidase의 동특성)

  • Partial hydrolysed and deacetylated chitin, CHITA and CHITB as supports of immobilized enzyme were obtained by treatment of acid and base respectively. Glutaraldehyde, bifunctional reagent, was employed for crosslinking between $\beta$-glucosidase and support. Immobilized enzyme activities of CHITA-Gase and CHITB-Gase were determined with the reaction of p-nitrophenol-$\beta$-D-glucopyranoside(PNG) in batch reactor, CSTR and PFR. Their optimum temperature, pH and enzymatic characteristics including Km and Vmax values were observed with variation of the flow rates. Mass transfer coefficient(h), effectiveness factor(η), deactivation rate(kd ) of two immobilized enzymes were also examined to compare efficiency of reactors.

Lean Combustion Characteristics with Hydrogen Addition in a LPG Fuelled Spark Ignition Engine (LPG엔진에서 수소연료 보조분사에 의한 희박연소특성 연구)

  • The basic effects of hydrogen addition for engine performance and emission were investigated in single cylinder research engine. Seven commercial injectors were tested to choose a suitable injector for hydrogen injection prior to its engine implementation. The hydrogen fuel leakage and flow rate were evaluated for each injector and KN3-1(Keihin, CO.) showed the best performance for hydrogen fuel. At the higher excess air ratio(${\lambda}=1.7$, 2.0), the better combustion stability was found with hydrogen addition even though its effect was small at lower excess air ratio (${\lambda}=1.0$, 1.3). Stable operation of the engine was even guaranteed at ${\lambda}=2.0$, if the amount of hydrogen gas was near 15% of total energy. In the lean region, ${\lambda}>1.3$, thermal efficiency was improved slightly while it was not clearly observed at ${\lambda}=1.0$, 1.3. It is considered that, in some cases, high temperature environment due to hydrogen combustion caused further heat loss to surroundings. Except for ${\lambda}=1.0$, with larger amount of hydrogen addition, CO was reduced drastically but it was emitted more at the leaner region. Nitric oxides(NOx) was increased a little more with hydrogen addition at ${\lambda}=1.0$, 1.3. However, at ${\lambda}>1.3$ its relative amount of emission was low. In addition, the amount of NOx was continuously decreased with hydrogen addition, but, at ${\lambda}=2.0$ the amount of NOx was lowered to 1/100 of that of ${\lambda}=1.0$. THC emission was significantly increased as air/fuel ratio was raised to leaner region due to misfire and partial burn.

  • [ $Yb_2O_3$ ] films were successfully deposited on a cube-textured Ni and(100) $SrTiO_3$(STO) single crystal substrates by metal organic chemical vapor deposition(MOCVD) method using $H_2O$ vapor as an oxidant. $H_2O$ vapor was used in order to avoid the oxidation of Ni substrate. The working pressure and Ar flow rate were 10 Ton and 600 sccm, respectively. $Yb_2O_3$ films on STO were formed at high temperatures above $900^{\circ}C$. While XRD peaks from $Yb_2O_3$ were hardly detected at $900^{\circ}C$, the $Yb_2O_3$(400) texture was developed fur the films grown at deposition temperatures above $950^{\circ}C$. The AEM surface roughness of $Yb_2O_3$ film, grown on STO, was in the range of $6{\sim}10nm$ for the film deposited at $950^{\circ}C$ with a $H_2O$ vapor partial pressure of 5.5 Ton and deposition times of 3 and 5 mins. For cube-textured Ni substrate, both $Yb_2O_3$(222) and $Yb_2O_3$ (400) textures were developed textures at deposition temperatures above $850^{\circ}C$.

Study on the Performance of Laser Welded Joint of Aluminum Alloys for Car Body

  • Considering the fuel consumption of car, a light structure of aluminum alloys is desired fer car body nowadays. However, fusion welding of aluminum alloys has some problems of reduction of joint efficiency, porosity formation and hot cracking. In the present work, investigation to improve the joint performance of laser welded joint has been carried out by addition of Cu, Ni, and Zr to A6NO 1 alloy welds. Aluminum alloy plate of 2.Omm in thickness with filler metal bar was welded by twin beam Nd: YAG laser facility (total power: 5kW). The filler metals were prepared by changing the chemical compositions for adding the elements into the weld metal. Thirteen filler metal bars were prepared and pre-placed into the base metal before welding. Ar gas shielding with a flow rate of 10 1/min was used. The defocusing distance is kept at 0 mm. At travel speeds off 3 to 9 and at laser power of 5kW (front beam 2kW rear beam 3kW), full penetration welds were obtained, whereas at travel speeds of 12 to 18 m/min and same power, partial penetration was observed. The joint efficiency of laser-welded joint was improved by the addition of Cu, Ni, and Zr due to the solid solution hardening, grain refining and precipitation hardening. The type of hardening has been further considered by metallurgical examination.

Isolation and Partial Characterization of Phytotoxic Mycotoxins Produced by Sclerotinia sp., a Potential Bioherbicide for the Control of White Clover(Trifoliorum repens)

  • Sclerotinia sp. (isolate BWC98-105) causes stem blight and root rot in Leghum sp., and is presently being evaluated as a potential mycoherbicide for the control of Trifoliorium repens. Bioassays have shown that Sclerotinia sp. produces phytotoxic substance which is biologically active against T. repens. Two biologically active compounds, designated as compoundsI and II, were produced in vitro from the culture filtrate of BWC98-105 isolate Sclerotium sp. Compounds I and II were purified by means of liquid-liquid extraction and $C_{18}$ open column chromatography (300 ${\times}$ 30 mm, i.d). To determine the purity, the purified compounds were analyzed by RP-HPLC. The analytical RP-HPLC column was a TOSOH ODS-120T (150 ${\times}$ 4.6 mm i.d, Japan), of which the flow rate was set at 0.7 mL/min using the linear gradient solvent system initiated with 15 % methanol to 85 % methanol for 50 min with monitoring at 254 nm. Under these RP-HPLC conditions, compounds I and II eluted at 3.49 and 4.13 min, respectively. Compound II was found to be most potent and host specific. However, compound I had a unique antibiotic activity against phytopathogenic bacteria like bacterial leaf blight (Xanthomonas oryzae) on rice, where it played a less important role in producing toxicity on T. repens. No toxin activity was detected in the water fraction after partitioning with several organic solvents. However, toxin activity was detected in the ethyl acetate and butanol fractions. In the leaf bioassay using compound II, the disease first appeared within 4-5 h as water soaked rot, which subsequently developed into well-defined blight affecting the whole plant.

An Experimental Study on Optimizing for Tandem Gas Metal Arc Welding Process (탄뎀 가스메탈아크 용접공정의 최적화에 관한 실험적 연구)

  • To enhance productivity and provide high quality production material in a GMA welding process, weld quality, productivity and cost reduction affects the number of process variables. In addition, a reliable welding process and conditions must be implemented to reduce weld structure failure. In various industries the welding process mathematical model is not fully formulated for the process parameter and on the welding conditions, therefore only partial variables can be predicted. The research investigates the interaction between the welding parameters (welding speed, distance between electrodes, and flow rate of shielding gas) and bead geometry for predicting the weld bead geometry (bead width, bead height). Taguchi techniques are applied to bead shape to develope curve equation for predicting the optimized process parameters and quality characteristics by analyzing the S/N ratio. The experimental results and measured error is within the range of 10% presenting satisfactory accuracy. The curve equation was developed in such a way that you can predict the bead geometry of constructed machinery that can be used for making tandem welding process.

A Study on Operation of Sand Filters Coated with Manganese (망간사화된 모래여과지 운영에 관한 연구)

  • Filtration experiments were conducted to determine the characteristics of manganese removal in filtration using 4 different filter media including sand and manganese sand(MS). Filtration velocity was 123 m/d and the flow rate was $3.9m^3/d$ per column. Duration of these experiments was about one year, and manganese dioxide accumulation, turbidity removal, manganese removal, and organic material removal were examined depending on filter media. When filter influent(residual chlorine 1.0 mg/L) with an average manganese concentration of 0.208 mg/L was fed through a filter column, the sand+MS and MS columns removed 98.9% and 99.2% of manganese respectively on an annual basis. When there is need to replace the sand filters with a MS filter to remove manganese, it was shown that the replacement of a partial sand filter with MS had adequate manganese removal.

Spalling Resistance of High Strength Concrete Using Non-Stripping Form (비탈형 거푸집에 의한 고강도 콘크리트의 폭렬방지)

  • As a part of a series of study, this study reviewed the fire proof characteristics of high performance concrete RC column members using non-stripping form that accompanied metal lath lateral confinement to prevent spalling of high performance concrete which is increasingly used recently, and the results are as follows. Flow and air amount both satisfied target range, and compressive strength, over 80MPa at age 28 days, showed high strength range. As for spalling characteristics, in the case of plain in which no fiber is mixed, severe spalling occurred, and in the case of 0.05% nylon("NY" hereinafter)+polypropylene("PP" hereinafter) fiber mixture, only surface area experienced partial spalling. Regarding non-stripping form changes, both non-stripping 25-20 and non-stripping 50-20 experienced spalling at finish material area, and non-stripping 50-20 showed better spalling proof performance than non-stripping 25-20. In the case of non-stripping 50-40, spalling was prevented, and while mass reduction rate was less than 10%, its temperature hysteresis showed the most excellent fire proof performance with base metal surface area maximum temperature $376.1^{\circ}C$.

Optimal Design of Fuel-Rich Gas Generator for Liquid Rocket Engine (액체로켓의 농후 가스발생기 최적설계)

  • An optimal design of the gas generator for Liquid Rocket Engine (LRE) was conducted. A fuel-rich gas generator in open cycle turbopump system was designed for 10ton in thrust with RP-1/LOx propellant. The optimal design was done for maximizing specific impulse of thrust chamber with constraints of combustion temperature and for matching the power requirement of turbopump system. Design variables are total mass flow rate to gas generator, O/F ratio in gas generator, turbine injection angle, partial admission ratio, and turbine rotational speed. Results of optimal design provide length, diameter, and contraction ratio of gas generator. And the operational condition predicted by design code with resulting configuration was found to maximize the objective function and to meet the design constraints. The results of optimal design will be tested and verified with combustion experiments.

  • With the world's fast expanding energy usage comes a slew of new issues. Because one-third of energy is lost in overcoming friction, tremendous effort is being directed into minimizing friction. Surface texturing is the latest surface treatment technology that uses grooves and dimples on the friction surface of the machine to significantly reduce friction and improve wear resistance. Despite the fact that many studies on this issue have been conducted, most of them focused on parallel surfaces, with relatively few cases of converging films, as in most sliding bearings. This study investigated the lubrication performance of surface-textured inclined slider bearings. We analyzed the continuity and Navier-Stokes equations using a commercial computational fluid dynamics code, FLUENT. The results show the pressure and velocity distributions and the lubrication performance according to the number and orientation of rectangular dimples. Partial texturing somewhat improves the lubrication performance of inclined slider bearings. The number of dimples with the maximum load-carrying capacity (LCC) and minimum friction is determined. When the major axis of the dimple is arranged in the sliding direction, the LCC and friction reduction are maximized. However, full texturing significantly reduces the LCC of the slider bearing and increases the flow rate. The results have the potential to improve the lubrication performance of various sliding bearings, but further research is required.
