The pathogenicity of Streptococcus parauberis isolated from cultured olive flounder Paralichthys olivaceus (양식 넙치, Paralichthys olivaceus에서 분리한 연쇄구균증 원인균, Streptococcus parauberis의 병원성)

  • There is an increasing number of reports describing Streptococcus parauberis as an important pathogen of cultured olive flounder Paralichthys olivaceus and starry flounder Platichthys stellatus in Korea. We tried to determine the effects of water temperature (14${^{\circ}C}$ and 21${^{\circ}C}$) on the pathogenicity in Streptococcal disease caused by S. parauberis. We have challenged 180 olive flounder by i.p injection to $2.0{\times}10^{7}$ live cells/fish. Mortality was monitored for 21 days post challenge. And histopathological characterizations as infection degree, tissue degeneration and/or bacterial distribution were investigated with H&E stain and in situ hybridization technique. Fifty percent and 16.7% of mortality occurred within 21 days at 21${^{\circ}C}$ and 14${^{\circ}C}$ water temperature, respectively. In most cases, the typical symptoms of olive flounder infected with S. parauberis were darkness of the skin, lethargy, mild abdominal distension cause by ascites, splenomegaly, congested liver and internal organs paleness. The pericardial sac contained large amounts of cloudy fluid. Numerous whitish nodules, which were variable in size and often confluent, were randomly scattered throughout the myocardium. Especially, pericarditis and/or myocarditis was observed in all tested fishes after death. Positive in reaction with S. parauberis were found in all tissues in situ hybridization analysis. The relative numbers of S. parauberis in heart were much more than in liver, spleen, kidney and stomach. We evaluated that S. parauberis strain causes serious damage in the pericardium, shortness of breath and the blood disorder. Therefore, pericarditis and myocarditis caused by S. parauberis were closely related to mortality of olive flounder.

  • Industrial advancements have resulted in food culture development, followed by increased seafood consumption and large-scale seafood farming, which has been accompanied by an increased prevalence of fish disease. The antibiotic oxytetracycline (OTC) is commonly used to prevent and treat bacterial diseases in fish. However, overuse of OTC had led to negative aspects. In view of this, we conducted a research with regard to aspects of remnants on olive flounder skin, liver, and muscle through dipping treatment and oral feeding of OTC and analyzed the results with bioassay and HPLC quantitative analyses. The dipping treatment was carried out once with 25 g/ton/hr of OTC, and the oral treatment with 62.5 mg/kg body weight/7 days. The results underwent a bioassay analysis. The dipping group reacted only on the skin right after dipping, while the oral feeding group responded on the skin for 77 days after feeding and on the muscle for 14 days. In the dipping group, the HPLC quantitative analysis revealed remnants in the skin on the 37th day and on the 13th day in the liver group. No remnants were found in the muscle, even immediately after dipping. In the oral feeding group, there was a high concentration (1.07 mg/kg) of remnant in the skin, even on the 77th day. 0.56 mg/kg in the liver, even a small amount, and no remnant in the muscle on the 42nd day. To sum up, the results suggest that it will not be harmful to our body to observe the OTC withdrawal period of 40 days with the muscle because OTC will hardly remain on it. When using olive flounder for sashimi, the skin and liver should not be used for broth, as the quantity of OTC residue is several times higher than that found in muscle. As previous studies reported that the concentration of remnants gradually decreased with heating, so it was likely to lessen, depending on the cooking temperature.

  • This study was aimed to investigate the effects of injection of garlic, Allium sativum, extract and immersion in garlic juice on the nonspecific immunity and the resistance against the artificial infection of Streptococcus iniae and Edwardsiella tarda of the olive flounder, Paralichthys olivaceus. The nonspecific immune mechanisms were assessed in terms of lysozyme activity, nitroblue-tetrazolium (NBT) assay and superoxide dismutase (SOD) activity etc. Relative percent survival (RPS) was assessed by the challenge with S. iniae BS10 or E. tarda KE-1. Almost of the garlic extract injected groups showed the enhanced level of the tested nonspecific immune factors. In the challenge test with S. iniae and E. tarda, RPS of 5% garlic extract pre-injected group was much higher than that of any other tested groups, respectively. Almost of the garlic juice immersion tested groups exhibited strengthened nonspecific immune defence factors, lysozyme activity, the number of lymphocytes and neutrophils, NBT reduction and SOD activity in kidney. In the challenge with S. iniae and E. tarda, RPS in the 0.25 g/L of garlic juice immersed group was much higher than any other tested groups, respectively. The results suggest that the garlic extract and juice would be effective to enhance the nonspecific immunity and protective ability of olive flounder against fish disease such as S. iniae and E. tarda.

  • The Nile tilapia Oreochromis niloticus, Atlantic salmon Salmo salar, rainbow trout Oncorhynchus mykiss, olive flounder Paralichthys olivaceus, common carp Cyprinus carpio, grass carp Ctenopharyngodon idella and rohu carp Labeo rohita are farmed commercially worldwide. Production of these important finfishes is rapidly expanding, and intensive culture practices can lead to stress in fish, often reducing resistance to infectious diseases. Antibiotics and other drugs are routinely used for the treatment of diseases and sometimes applied preventatively to combat microbial pathogens. This strategy is responsible for the emergence and spread of antimicrobial resistance, mass killing of environmental/beneficial bacteria, and residual effects in humans. As an alternative, the administration of probiotics has gained acceptance for disease control in aquaculture. Probiotics have been found to improve growth, feed utilization, immunological status, disease resistance, and to promote transcriptomic profiles and internal microbial balance of host organisms. The present review discusses the effects of single and multi-strain probiotics on growth, immunity, heamato-biochemical parameters, and disease resistance of the above-mentioned finfishes. The application and outcome of probiotics in the field or open pond system, gaps in existing knowledge, and issues worthy of further research are also highlighted.

Dietary effect of Bacillus subtilis MD-02 on Innate Immune Response and Disease Resistance in Olive Flounder, Paralichthys olivaceus (넙치(Paralichthys olivaceus)의 비특이적 면역반응 및 병 저항성에 대한 Bacillus subtilis MD-02의 효과)

  • Among several marine-derived microorganisms isolated from the coast of Jeju Island that had antimicrobial activity against fish disease pathogens, Bacillus subtilis MD-02 was tested for its dietary effect on the innate immune response and disease resistance of olive flounder. Strain MD-02 was fed to the olive flounder at a concentration of $1.2{\times}10^4$, $1.2{\times}10^6$, or $1.2{\times}10^8CFU/100g$, respectively. Consequently, the hematocrit was higher in these three groups than that in the control group at 4 weeks, and the aspartate aminotransferase and alanine aminotransferase levels were decreased in the $1.2{\times}10^8$ and $1.2{\times}10^4CFU/100$ groups compared with the control group levels. The amylase activity and total protein were significantly increased in the $1.2{\times}10^4CFU/100g$ group at 3 weeks. The innate immune response, determined from the lysozyme and macrophage activities, was higher in the $1.2{\times}10^8CFU/100g$ group than in the control group. In addition, treatment of the olive flounders with Streptococcus parauberis at $1.2{\times}10^6CFU/ml$ confirmed the mortality rate, which was 100% in the control group and 40-60% in the groups fed B. subtilis MD-02, indicating that the fish had resistance to fish disease pathogens. Therefore, it was confirmed that when fed MD-02, olive flounder builds an innate immune response and acquires resistance to fish disease pathogens, indicating that B. subtilis MD-02 can be developed as a beneficial feed additive.

Residues of Fluoroquinolones in Olive Flounder (Paralichthys olivaceus) and Black Rockfish (Sebastes schlegell) Following Oral Administration (Fluoroquinolone계 항생제의 경구투여에 따른 양식넙치 및 조피볼락의 체내 잔류량의 변화)

  • The residues of fluoroquinolones in Olive flounder (Paralichthys olivaceus) and Black rockfish (Sebastes schlegeli) were investigated after oral doses of 20 mg/kg for 30 days. Blood samples were taken at 5 hours, 10 hours, 1 day, 2 days, 3 days, 5 days, 7 days, 9 days, 13 days, 20 days and 30 days after treatment. The concentrations of fluoroquinolones were detected by high performance liquid chromatography using a UV detector. The recovery rates of fluoroquinolones in fish samples ranged from 92.0-99.7%, 93.4-97.5% and 93.0-97.9% for treatment of 0.1, 0.5, and $1.0{\mu}g/g$, respectively. Ciprofloxacin, enrofloxacin, norfloxacin and sarafloxacin were detected in the blood of the olive flounder at 10 hours, 10 hours, 1 day and 10 hours after treatment with $6.43{\mu}g/g$, $6.07{\mu}g/g$, $11.83{\mu}g/g$ and $11.02{\mu}g/g$ as maximum level, respectively. Ciprofloxacin, enrofloxacin, norfloxacin and sarafloxacin were detected in the blood of black rockfish 1 day, 1 day, 1 day and 10 hours after treatment with $8.26{\mu}g/g$, $8.03{\mu}g/g$, $7.29{\mu}g/g$ and $9.38{\mu}g/g$ as maximum level, respectively.   • Probiotic principles can be applied in aquaculture for the purpose of growth and immunity stimulation, disease prevention and eventually better production performance. This study was to assess effects of combinations of microbes containing two Bacillus sp., plus one Lactobacillus sp. as the basal preparation (BSL-LAB), and additional Nitrosomonas sp. (nitrifying bacteria consortium, NBS) in olive flounder (Paralichthys olivaceus). The effects examined were growth parameters, hematologic parameters, innate immunity and pathogen challenge test. Fish were assigned to 4 treatments as Control (no probiotics), Group A (Bacillius and Lactobacillus to culture water), Group B (Bacillius and Lactobacillus both in water and feed), Group C (same as Group B with additional Nitrosomonas in feed). Fish were allocated to the above 4 groups, each group being composed of triplicate 30 fish, for a 7-week feeding in the laboratory. Positive effects were observed both in growth and pathogen sensitivity with all three probiotic combinations. Such effects were attributed to improved innate immune functions. This result indicates that the tested probiotic microbes are beneficial to olive flounder aquaculture.

  • The effects of dietary $\beta$-1,3/1,6-linked glucan administration on nonspecific immune responses, hematology and disease resistance against Edwardsiella tarda in olive flounder, Paralichthys olivaceus were evaluated. Fish were fed the experimental diets supplemented with 0 %, 0.01 %, 0.05 %, 0.1 %, and 0.5 % of $\beta$-glucan to a commercial diet for 7 weeks. The test was performed in two ways such as fed on each $\beta$-glucan concentrations daily and every other week alternately. The body weight gain from the fish fed on daily the 0.05 % supplemented diet and fed on alternately the 0.1 % supplemented diet of $\beta$-glucan were significantly higher than the control. Both lysozyme activity and intracellular superoxide anion production of kidney phagocytes were higher in the all experimental groups than in the control. But there was no large difference in hematology among each group. The relative percent survival rate (RPS) after an artificial challenge with $4{\times}10^6$ cells of E. tarda per fish was higher than the control, except for that fed on daily the 0.01 % supplemented diet.

Monitoring of Kudoa septempunctata in Cultured Olive Flounder and Wild Fish in Jeju Island during 2012 (2012년 제주의 양식 넙치 및 자연산 어류의 Kudoa septempunctata 감염 조사)

  • The study surveyed infection rate of Kudoa septempunctata parasitized in the trunk muscle of olive flounder, Paralichthys olivaceus, cultured in Jeju Island and wild fish species caught in the coastal area around Jeju Island during 2012. Among 143 olive flounder that were randomly sampled from 26 different culture farms, K. septempunctata was detected in 7 fish samples (4.9%) from 4 different culture farms, showing no typical Kudoa infestation. However, K. septempunctata was not detected in olive flounder fry sampled from hatcheries and 8 species of wild fish. In addition, we compared 3 different sampling sites on trunk muscle of 7 Kudoa positive fish that included head part, tail part and entire muscle. Among 7 fish, K. septempunctata was detected in 3 fish that were sampled from head part; while 4 fish from tail part of trunk muscle. However, all 7 fish were positive when sampled from entire muscle. Thus, we suggest that it will be more efficient to use entire muscle sample than sampling partial muscle parts for detection of K. septempunctata.

  • A study on efficacy and safety of Bio-Gammamix was performed to use for the preventive drug against bacterial diseases to mirror carp(Cyprinus carpio), rainbow trout(Salmo gairdneri), eel(Anguilla japonica) and flounder(Paralichthys olivaceus). The main ingredients of Bio-Gammamix were ferrous fumarate and mixed extrated powders of licorice root, ox bile and garlic. The test drug was proved to improve feeding habit and feed efficiency and decrease cumulative mortality remarkably. After administration, no side effects and histopathological changes were observed. We can prevent outbreaks of bacterial diseases of fish by the dosage of 1kg/ton of fish body weight perorally once or two times in a day for 7 days and Bio-Gammamix is thought to be used effectively an widely against most bacterial fish pathogens.
