Analysis of Ginsenoside Composition of Ginseng Berry and Seed

  • This study was performed to provide basic information that can be used to differentiate Korean ginseng (Panax ginseng CA. Meyer) berry and seed from American ginseng (Panax quinquefolium L.) seed. Total ginsenoside contents of Korean ginseng berry, Korean ginseng seed, and American ginseng seed were 9.09, 3.30, and 4.06%, respectively. Total ginsenoside content of Korean ginseng berry was about 2.2 to 2.7 times higher than those of Korean ginseng seed and American ginseng seed. Particularly ginsenoside Re content of 4-year cultivated Korean ginseng berry (5.99%) was about 3.6 to 5.4 times higher than that of 4-year cultivated Korean ginseng seed (1.65%) and 4-year cultivated American ginseng seed (1.10%). The contents of total ginsenoside and ginsenoside Re of Korean ginseng berry were about 4.8 and 28 times higher, respectively, than those of 4-year cultivated Korean ginseng root. In general the contents of total ginsenoside and ginsenoside Re of Korean ginseng berry were significantly higher than those of Korean ginseng seed and American ginseng seed.

  • A new dammarane glycoside named ginsenoside $Rf_{2}$ has been isolated from Korean red ginseng (Panax ginseng) and its chemical structure has been elucidated as $6-O-[{\alpha}-L-rham-nopyranosyl (1{\rightarrow}2){\beta}-D-glucopyranosyl]$$dammarane-3{\beta}, 6{\alpa}, 12{\beta}$, 20(R), 25-pentol by chemical and spectral methods.

Adventitious Root Development and Ginsenoside Production in Panax ginseng, Panax quinquefolium and Panax japonicum

  • This work was carried out to establish adventitious root culture system in three Panax species (wild-grown P. ginseng, P. quinquefolium, and P. japonicum) to analyze their ginsenoside productivity. Adventitious roots were induced directly from segments of seedlings after cultured on MS(Murashige andSkoog 1962) solid medium containing 3.0 mg/l IBA. Omission of $NH_4NO_3$ from the medium greatly enhanced both the frequency of adventitious root formation and number of roots per explants in all the three Panax species. However, elongation of post-induced adventitious roots was enhanced on medium with $NH_4NO_3$. Two-step culture protocol: $NH_4NO_3$-free medium for first two weeks of culture, followed by $NH_4NO_3$ containing medium for further 4 weeks, greatly enhanced the fresh weight increase of adventitious roots in all the three ginseng species. The fresh weight of adventitious roots was high in P. quinquefolium and low in P. ginseng, followed by P. japonioum regardless of the composition of medium. Pattern and content of ginsenosides in adventitious roots differed among the three Panax species. Total ginsenoside content of adventitious roots in P. quinquefolium, P. ginseng, and p. japonicum was 8.03, 15.7 and 1.2 mg/g dry weight, respectively. Among the three speices, adventitious roots in P. quinquefolium produced hig-hamount of ginsenosides. The pattern of ginsenoside fractions between P. ginseng and P. quinquefolium was similar but the amount of ginsenoside differed between the two, While, in P japonicum, total ginsenoside content was very low and some ginsenosides such as ginsenoside Rb2 and Rf were not detected. Conclusively, we demonstrate that same culture condition was required for induction and elongation of adventitious roots of three ginseng species but growth of adventitious roots and their ginsenoside production were different among them.

Comparison of Ginsenoside Contents in Different Parts of Korean Ginseng (Panax ginseng C.A. Meyer)

  • The present study was conducted to investigate the ginsenoside profiles of the main root, root hair, and leaf of ginseng in order to demonstrate their possible application in medicine. The total ginsenoside content of the leaf was up to 12 times than that in the main root, and the content of protopanaxadiol groups was higher than that of protopanaxatriol groups in all the samples. The leaf was shown to contain high amounts of ginsenosides Rb3 and Rh1, whereas the main root contained large amounts of ginsenosides Rb1 and Rc. Moreover, Rb2, Rb3, and Rg1 were only detected in the root hair, leaf, and main root, respectively. The ginsenoside Re content of Panax ginseng leaf and root hair was 2.6~4 times higher than that of the main root. Therefore, the results indicate that the ginsenoside content of Panax ginseng is higher in the leaf and root hair, and lower in the main root.

A Study on the Effect of Ginseng Saponin on Rat Intestinal Mucosal $Na^+,K^+$-ATPase (인삼 사포닌이 백서 장점막 $Na^+,K^+$-ATPase에 미치는 영향에 관한 연구)

  • We have studied the effect of ouabain, tool ginseng saponin, panax saponin C (protopanaxatriol derivative) and ginsenoside $Rb_{1}$ (protopanaxadiol derivative) on $Na^+,K^+$-ATPase and $Mg^{++}$-ATPase activities were determined by the method of Robinson and ATPase activities were determined by the method of King. The $Na^+,K^+$-ATPase activities were inhibitied by 90.1% and 51.1% respectively at the concentration of $10^{-3}M$ and $10^{-4}M$ ouabain. The results are consistent with those of Robinson. The $Na^+,K^+$-ATPase activities were increased by 14.3% and 10.0% respectively at the concentration of $10^{-4}$g/ml and $10^{-5}$g/ml total ginseng saponin. Panax saponin C obtained by the method of Han and ginsenoside $Rb_{1}$ obtained by the method of Shibata were used. The $Na^+,K^+$-ATPase activities were increased in the presence of panax saponin C and the increased activity with panax saponin C was greater than that with total ginseng saponin. On the other hand ginsenoside $Rb_{1}$ showed an inhibitory effect on $Na^+,K^+$-ATPase. Total ginseng saponin, panax saponin C and ginsenoside $Rb_{1}$ had no effect on $Mg^{++}$-ATPase. Therefore, it may be concluded that total ginseng saponin has dual effects on microsomal $Na^+,K^+$-ATPase, that is, panax saponin C exhibits stimulatory action, whereas ginsenoside $Rb_{1}$ shows inhibitory action.

Quantitative Comparison of Ginsenosides and Nitrogen Compounds in Korean Ginsengs and Related Origin (고려인삼류와 유사생약중 ginsenoside 및 질소화합물의 함량 비교)

  • To differentiate the quality of Korean ginseng from those of other habitats, the quantitative analysis of free amino acids(FAA) and total amino acids(TAA) in addition to ginsenoside Rb1 and Rg1 was carried out using amino acid analyzer and HPLC, respectively. FAA pattern in Korean ginseng was much different from that of Panax notoginseng. The difference in total content of FAA was also found that Korean ginseng contained 26.3-39.8mg/g while Panax notoginseng contained 6.5mg/g. This FAA content had a tendency to increase with the age of radix. The contents of FAA and TAA in Korean ginseng(6 years old) from Kumsan were shown to be the highest than other ginseng origins tested. The content in the 6 years Panax ginseng from China was about same with that of 4 years Korean ginseng of Kumsan. However, regarding to gisenoside Rbl and Rg1, which have been accepted as the characteristic components of Panax ginseng-Panax notoginseng showed considerably higher content than those of any other ginseng origin.

  • Studies were made to examine the various effects of jasmonic acid and benzoic acid on ginsenoside production in suspension cultures of Panax ginseng C. A. Meyer. Jasmonic acid increased the ginsenoside production when it was dosed at the concentration of 50 $\mu$M or higher. The cell growth, however, was reduced with jasmonic acid. When benzoic acid was dosed simultaneously with iasmonic acid, the ginsenoside production increased 9.6 folds. It was 2.2 times higher than the result of single dose of jasmonic acid.

Saponin Contents of Root and Aerial Parts in Panax ginseng and Panax quinquefolium (고려인삼과 미국삼의 부위별 Saponin 함량)

  • In order to obtain the basic information for the development of ginseng varieties with high saponin contents. saponin contents and ginsenosides of Panax ginseng (Korean ginseng) and Panax quinquefolium (American ginseng) grown under the same environmental conditions were analysed. Crude saponin contents of root and aerial parts were more in Panax quinquefolium than in Panax ginseng, and aerial parts had more saponin contents in comparison with a root. Protopanaxatriol saponin was greatly more in the aerial parts of ginseng while more amount of protopanaxadiol saponins were detected in the root. As for the ginsenosides, the patterns of ginsenosides detected in total saponin of the aerial parts were not different between two species, Panax ginseng and Panax quinquefolium, but the root ginsenoside patterns were quite different. Ginsenosides such as Rg$_2$, R$_{f}$. R$_{a}$ and R$_{o}$ were not detected in the root of Panax quinquefolium (American ginseng).).).).

  • This study was carried out to compare the root yields, root characters, saponin and ginsenosides contents of 6-year-old p. ginseng and p. quinquefolium. These two ginseng species showed difference in the diameter and ten비h of mainroot. The main root length or p. quinquefolium was shorter than that of p. ginseng, whereas Jakyung-jong and Hwangsook-jong of P. ginseng showed similar root length. Proximate composition were similar between the two species, however, crude fibercontent was significantly higher in main and lateral root of Jakyung-jong and Hwang sook-jong of P ginseng than P quinquefolium. In regard to mineral contents of root, P ginseng contained more Ca and Mn and less Fe and Al than P. quinquefolium. P. quinquefolium contained more of Rbl and Rd of protopanaxadiol saponin, and less or Re, $Rg_1$ and $Rg_2$ of protopanaxatriol saponin than P ginseng. However, no Rf was detected in the p. quinquefolium. Key words Panax ginseng, Panax quinquefolium, ginseng character, ginsenoside.

Inhibitory Effect of Elastase and Tyrosinase of Ginsenoside $F_1$ Isolated from Panax ginseng Leaves (인삼잎으로부터 분리된 진세노사이드 $F_1$의 Elastase 및 Tyrosinase 억제 효과)

  • This study was carried to establish a simple isolation and purification method of ginsenoside $F_1$ from leaves of Panax ginseng and was to evaluate the inhibitory effect of purified ginsenoside $F_1$ on the activities of elastase and tyrosinase. The content of ginsenoside $F_1$ was 90-fold higher in leaves than in root of ginseng. Ginsenoside $F_1$ was isolated from EtOAc fraction between EtOAc and alkalized water of 80% EtOH extract after remove of hydrophobic components. The 50% inhibitory concentration ($IC_{50}$) of ginsenoside $F_1$ on elastase activity and tyrosinase activity was 1.07 mM and 1.81 mM, respectively. Especially, inhibitory effect of ginsenoside $F_1$ on tyrosinase activity was higher than that of arbutin ($IC_{50}$; 2.20 mM). These results indicate that ginsenoside $F_1$ have a potential for industrial cosmetic materials.
