Soil Acclimatization of Regenerated Plants by Gibberellic Acid Treatments of Panax ginseng C. A. Meyer (GA3 처리에 의한 인삼 재분화 식물체의 토양 순화)

  • We established that regenerated plants of Panax ginseng were directly transplanted into the soil without the acclimatization process. When regenerated plants were treated at the concentration of $0.4mg\;l^{-1}$ for 9 hr before transplantation into the soil, our results showed that the survival rate of them was the highest. The optimal size of the root was above 4 cm in length and 0.4 g by weight before transplantation into the soil. The highest survival rate increased to 59.6%. After 4 years of cultivation in the fields, our results showed that there were no significant morphological differences between regenerated plants and wild-type ginseng. Also, we obtained healthy seeds from regenerated plants. This study provides the basis for a relatively rapid multiplication of selected genotypes and will allow a much more rapid evaluation of the germplasm of the unexplored and unstudied Korean ginseng.

Immunomodulating Effects of Red Ginseng on the Regulation of Cytokine Release in vivo (홍삼의 생체 내 사이토카인 분비에 대한 면역조절효과)

  • Cytokines released from innate immune cells play key roles in the regulation of the immune response. Red Ginseng (RG, steamed and dried root of Panax ginseng C.A. Meyer) is known to show different pharmacological effects by changed composition of saponins compared with Panax Ginseng. In this study, we examined the immunomodulatory effects of RG on the regulation of cytokine release in mice. RG was injected i.p at doses of 0.5, 5 and 50 mg/kg for 6 weeks. We assessed that the weight index of immune organs such as thymus, and spleen, and the mitogen blastogenesis of splenocytes. We also determined the levels of circulating cytokines in serum from RG-treated mice using ELISA assay. The weight index of thymus and spleen, and proliferation of mitogen response of splenocytes have increased in the RG-injected groups. In addition, the levels of IFN-${\gamma}$, TNF-${\alpha}$, IL-6, IL-12 and IL-2 concentrations have significantly increased in the serum of RG-treated mice, but that of IL-10 has not. These results suggest that RG has immune stimulating effects and could be useful as a immunoregulator of circulating cytokine release in vivo.

Effect of Soil Fumigation and Maize Cultivation on Reduction of Replant Failure in Ginseng (토양 훈증 및 녹비작물 재배가 인삼의 연작장해 경감에 미치는 영향)

  • Background: Dazomet are widely used as soil fumigant to solve soilborne problems, and the degradation intermediates are toxic to nematodes, fungi, bacteria, insects and weeds. Methods and Results: The effects of cultivation of green manure crop, maize before and after soil fumigation on the control of ginseng root rot disease were compared using soil where 6-years-old ginseng was harvested. Fumigant (dazomet) were used for soil fumigation in May and September, respectively. Maize was grown for soil management before and after soil fumigation. After May fumigation, the sowing date of maize was delayed by 15 days and thus its dry weight was decreased significantly. Maize cultivation after May fumigation increased pH but decreased EC, $NO_3$, $P_2O_5$, and K significantly. Maize cultivation after May fumigation decreased fungi population and the ratio of fungi and bacteria. Growth of 2-years-old ginseng was improved and the incidence of ginseng root rot was significantly decreased by maize cultivation after May fumigation. After harvesting 2-years-old ginseng, the population of Cylindrocarpon destructans was not different between treatment of May and September, but Fusarium solani showed a significant increase in September fumigation after maize cultivation. Conclusions: Maize cultivation after soil fumigation was effective in inhibiting ginseng root rot by the amendment of mineral composition and microorganism in fumigated soil.

Effects of light intensity and temperature on photosynthesis and respiration of Panax ginseng leaves (인삼엽(人蔘葉)의 광합성(光合成)과 호흡(呼吸)에 미치는 광도(光度) 및 온도(溫度)의 영향(影響))

  • Effects of temperature and light intensity on photosynthesis, respiration and chlorophyll content of ginseng (Panax ginseng C. A. Meyer) were as follow. 1. Optimum light intensity for apparent photosynthesis at $25^{\circ}C$ was 25Klux($1.35mgCO_2/dm^2{\cdot}hr$) for two years old ginseng grown in pot and 30Klux($1.94mgCO_2/dm^2{\cdot}hr$) for the six years old palmate cut leaves. Optimum temperature at 25Klux was $15^{\circ}C$ ($1.40mgCO_2$) for the 2 years old and 20 to $22^{\circ}C$ ($2.03mgCO_2$) for the 6 years old. 2. Dark respiration increased almost linearly with the increase of air temperature till $25^{\circ}C$ (2.6times between $16^{\circ}C$ to $25^{\circ}C$ for the 6 years old and 1.8 times between $15^{\circ}C$ to $25^{\circ}C$ for the 2 years old). Dark respiration was 11.1 % of net photosynthesis at $16^{\circ}C$, 17.8% at $25^{\circ}C$ for the 6 years old and 40% at $15^{\circ}C$, 64.7% at $25^{\circ}C$ for 2 years old. 3. Stomata appeared only in abaxial surface (lower epidermis) and stomatal frequency was $37per\;mm^2$. 4. Above results together with other informations quoted here strongly suggest that air temperature is much better criteria than light intensity for the improvement of shading roof material and shading construction. That is to promise maximum light intensity unless air temperature is above $25^{\circ}C$.

  • In this paper, new methods for the determination of the total and the individual saponin glucosides were proposed. One of them was a colorimetric method following Two-dimensional Thin layer chromatography. And the method employing Thinchrograph TFG-10 or Densitorol DMU-33C followed the separation of the saponins by means of a preparative thin layer chromatography. In accordance with the density of the chromatogram of each saponin, Thinchrogram or Densitogram of the individual saponin fraction was plotted and determined.

  • Hypolipidemic effect of Insamsansa-eum (ISE, Ren-Shen-Shan-Zah-Yin in Chinese) consisted of Red ginseng (RG; the steamed root of Panax ginseng C.A. Meyer) and Crataegii fructus (CF, the fruit of Crataegus pinnatifida BGE) is used frequently in China and Korea as a herbal medicine to treat arteriosclerosis, was investigated. Treatment of ISE significantly reduced blood triglyceride (TG) and total cholesterol (TC) levels in Triton WR-1339-induced hyperlipidemic mice and serum TG levels in corn oil-induced hypertriglyceridemic mice. ISE significantly lowered the high blood TG and TC levels as well as body and epididymal mass weights in hyperlipidemic mice induced by long-term feeding of a high-fat diet and increased blood HDL cholesterol level. ISE and its ingredients, RG and CF, inhibited pancreatic lipase and HMG-CoA reductase activities. Of its ingredients, RG reduced the blood TC level and HMG-CoA reductase activity more than CF. CF reduced blood TG level and pancreatic lipase activity more than RG. These finding suggest that the antihyperlipidemic effect of ISE may be due to synergistic inhibition of HMG-CoA reductase and pancreatic lipase by RG and CF, and that ISE may be effective hypolipidemic agents.

Crop Rotation in Paddy Soil Exhibiting Crop Failure Following Replanting: Effect on Soil Chemical Properties, Soil Microbial Community and Growth Characteristics of 2-Year-Old Ginseng (인삼 논재배 연작지에서 윤작물 재배가 토양화학성, 토양 미생물상 및 2년생 인삼의 생육에 미치는 영향)

  • Background: Crop rotation plays an important role in improving soil chemical properties, minimizing the presence of disease pathogens, and assists in neutralizing autotoxic effects associated with allelochemicals. Methods and Results: Five rotation crops of sudan grass, soybean, peanut, sweet potato, and perilla were cultivated for one year with an aim to reduce yield losses caused by repeated cropping of ginseng. In 2-year-old ginseng grown in the same soil as a previous ginseng crop, stem length and leaf area were reduced by 30%, and root weight per plant was reduced by 56%. Crop rotation resulted in a significant decrease in electrical conductivity, $NO_3$, and $P_2O_5$ content of the soil, whereas organic matter, Ca, Mg, Fe, Cu, and Zn content remained-unchanged. Soil K content was increased following crop rotation with sudan grass and peanut only. Rotation with all alternate crops increased subsequent ginseng aerial plant biomass, whereas root weight per plant significantly increased following crop rotation with perilla only. A significant positive correlation was observed between root rot ration and soil K content, and a significant negative correlation was observed between ginseng root yield and the abundance of actinomycetes. Crop rotation affected the soil microbial community by increasing gram negative microbes, the ratio of aerobic microbes, and total microbial biomass whereas decreases were observed in actinomycetes and the ration of saturated fatty acids. Conclusions: In soil exhibiting crop failure following replanting, crop rotation for one year promoted both soil microbial activity and subsequent ginseng aerial plant biomass, but did not ameliorate the occurrence of root rot disease.

Compound K, a Metabolite of Ginsenoside Rb1, Inhibits Passive Cutaneous Anaphylaxis Reaction in Mice

  • To understand the anti-allergic mechanism of compound K, which is a metabolite of ginsenoside Rb1, a main constituent of the root of Panax ginseng C.A. Meyer (family Araliaceae), its inhibitory effect against IgE-antigen complex IAC)-induced passive cutaneous anaphylaxis (PCA) reaction in mice and mRNA and protein expressions of allergic cytokines in lAC-stimulated RBL-2H3 cells were investigated. Orally administered ginsenoside Rb1 more potently inhibited PCA reaction when administered at 5 h prior to the lAC treatment than when administered at I h before. However, compound K orally administered 1 h before lAC treatment showed a more potent anti-PCA reaction effect than when treated at 5 h before. Orally administered ginsenoside Rb1 more potently inhibited PCA reaction induced by lAC in mice than intraperitoneally treated one, apart from orally administered its metabolite, compound K, which was more potent than the orally administered one. The compound K, a metabolite of ginsenoside Rb1, inhibited mRNA and protein expressions of IL-4 and TNF-${\alpha}$ and the activation of their transcription factor NF-$\kappa$B and MAPK in lAC-stimulated RBL-2H3 cells. These findings suggest that orally administered ginsenoside Rb1 may be dependent on its metabolism by intestinal microflora in the intestine and the compound K may improve allergic diseases by the inhibition of IL-4 and TNF-${\alpha}$ expresseion.

Comparison of Growth Characteristics and Quality of Korean Ginseng (Panax ginseng C.A. Meyer) by Different Shade Materials (해가림자재 종류별 고려인삼의 생육과 품질특성 비교)

  • This research investigated growth of ginseng regard to sunshade materials and micro metrological phenomena under the sunshade material to product high quality ginseng. Followings are results of investigation of active ingredient from raw ginseng producted under the sunshade materials. The highest temperature under the sunshade material from June to August was measured from a Three-layered blue and one-layered black polyethylene net (TBOBFN) followed by a aluminium-coated polyethylene sheet (ACPS) and blue polyethylene sheet (BPS). The highest light penetration of $381.7{\mu}mol/s/m^2$ was obtained from the BPS at August, also the temperature was highest at the ginseng field. Growth of above ground part of 2~3 year old ginseng under the ACPS was the highest, followed by the ACPS and the BPS. In case of 4 year old ginseng, the ACPS and the BPS was the same, followed by the TBOBPN. Root length and diameter of 2~3 year old ginseng showed difference among treatments, but 4 year old ginseng was not showed difference. Meanwhile, the highest amount of root of 4 year old ginseng of $896g/m^2$ was obtained from ACPS. The total amount of ginsenocide of 4 year old ginseng under the ACPS was highest, followed by the BPS and the TBOBPN. Chromaticity of ginseng root under the TBOBPN was highest, followed by the ACPS and the BPS.

Beneficial effect of Combination with Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori on Cholesterol and Erectile Dysfunction in Hyperlipidemia rats (홍삼, 천마, 적하수오 병용투여에 의한 고지혈증 랫드에서의 콜레스테롤 및 발기부전 개선효과)

  • Objectives : This study was designed to investigate effects of the combination with Korean Red Ginseng (Panax ginseng C.A. Meyer), Gastrodia Rhizoma (Gastrodia elata Blume) and Polygoni Multiflori Radix (Polygonum multiflorum Thunberg) on metabolic disorders including cholesterol and erectile dysfunction in hyperlipidemia rats.Methods : Animals were divided into six groups; Control with normal diet, high fat/cholesterol-diet (HFCD), fluvastatin, Korean Red Ginseng treated (KRG), and the combination treated (Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori Radix; 1:1:1 for KGP1 and 2:1:1 for KGP2). The experimental groups initially received HFCD for 10 weeks and then treated orally with fluvastatin, KRG, KGP1 and KGP2 during the final 6 weeks. Erectile function was determined by the measurements of intracavernosal pressure (ICP) and maximal arterial pressure (MAP) after electrical stimulation of the cavernosal nerve.Results : KGP2 decreased the level of total cholesterol and LDL cholesterol in the sera of HFCD rats without no changes of body weights. KRG, KGP1 and KGP2 decreased the level of C-reactive protein (CRP) levels except of fluvastatin, synthetic HMG-CoA reductase inhibitor. KRG, KGP1 and KGP2 significantly increased the ICP, ICP/MAP ratio, area under the curve (AUC) compared with those of normal rat. Morphometric analyses showed that KRG, KGP1 and KGP2 increased the volume of smooth muscle and the regular arrangement of collagen fibers in corpus cavernosum of HFCD rats. The penile expression of eNOS was increased by KRG, KGP1 and KGP2.Conclusions : Based on these results, we suggest that the combination with Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori may improve hyperlipidemia through regulating the lipid profiles and erectile dysfunction in rats.
