Recovery of High Unsaturated Fatty Acid from Squid Processing Wastes using Supercritical Carbon Dioxide Extraction Method (초임계 이산화탄소 추출법을 이용한 오징어 가공 부산물로부터 고도불포화 지방산 회수)

  • A squid viscera oil contains a high content of EPA, DHA, and other valuable polyunsaturated fatty acids. The extractions of squid viscera oil by supercritical carbon dioxide both with/without $3\%$ (v/v) ethanol were performed in a semicontinuous flow extractor at 8.3 to 13.8 MPa and 25 to $50^{\circ}C$. When ethanol was added to $SC-CO_2$, the extraction ratio of lipid increased. The extracts contained high content of unsaturated oils like DHA and EPA. The highest extraction yield of lipid from squid viscera oil extracted by supercritical carbon dioxide was obtained at 12.4 MPa and $40^{\circ}C$ with/without entrainer. The main fatty acids of squid viscera oil extracted by supercritical carbon dioxide were myristric acid (14:0), palmitic acid (16:0), palmitoleic acid (16:1), oleic acid (18:1), arachidic acid (20:0), eicosapentaenoic acid (20:5), and docosahaxaenoic acid (22:6).

Changes in Body Composition of Protaetia brevitarsis seulensis Larvae Fed Sulfur and Selenium (황 및 유황 급이에 따른 흰점박이꽃무지 유충의 체성분 변화)

  • This study examined changes in the body composition of Protaetia brevitarsis larvae fed sulfur and selenium. Among the general chemical components, an increase in protein was observed in the sulfur-fed group, whereas an increase in fatty acid was found in the groups fed selenium and sulfur/selenium. The structural amino acid showed some tendency to be increased compared to the control group, but this was especially true in the sulfur-fed group. The free amino acid were increased in the groups fed selenium and sulfur/selenium, with GABA, Glu, and Pro, in particular, being increased in the group fed sulfur/selenium. Among the essential amino acids, Met, Trp, and Val tended to be increased in selenium and sulfur/selenium groups. Saturated and polyunsaturated fatty acids showed a tendency to increase in the group fed sulfur/selenium, but monounsaturated fatty acid tended to be decreased in the group. In particular, no oleic acid was detected in the sulfur/selenium group, whereas palmitic acid, palmitoleic acid, and linoleic acid were increased. From observations of these body composition changes associated with the feeding of sulfur and selenium, it is estimated that the group fed sulfur/selenium would likely undergo a pronounced change in body composition. Therefore, we suggest that it is possible to manufacture a food that has enhanced functionality by appropriately adjusting the diet of Protaetia brevitarsis larvae.

A Study on the Lipid Components of Amorpha Fruticosa Seed (쪽제비싸리종자(種子)의 지질성분(脂質成分)에 관한 연구(硏究))

  • The lipid components, fatty acid compositions, physical and chemical characteristics of crude oil in Amorpha fruticosa seed were determined and proximate compositions of it were also analyzed. The results were summarized as follow: 1) The proximate composition showed moistrue to be 10.14%, crude protein to be 21.77%, crude fats to be 12.73%, carbohydrates to be 51.75% and crude ash to be 3.61%. 2) Specific gravity, refractive index, smoke point and titer of the crude oil were $0.925\;(15/15^{\circ}C)$, $1,477(15^{\circ}C)$, $175^{\circ}C$ and $14.8^{\circ}C$, respectively. 3) Iodine, saponification, acid, ester value and unsaponifiable content of the crude oil were 144, 182, 2.9, 179 and 5.17%, respectively. 4) Lipid components were separated by TLC and quantitatively determined by TLC scanner to give 75% triglycerides, 14% esterified sterols, 3.08% phospholipids, 4.4% free sterols and 3.77% free fatty acids. 5) Fatty acid compositions were quantitatively determined by GLC to give 76.21% linoleic acid, 9.92% palmitic acid, and 5.07% stearic acid as the main components, oleic, linolenic, palmitoleic and arachidic acid were presented in small quantities.

Fatty Acid Composition of Canned Fish and Shellfish Products on Korean Market (국내시판 수산물통조림의 지방산조성)

  • The fatty acid composition of lipids separated from 14 kinds of canned fish and shellfish products from the market were examined. The crude lipid contents of canned fish and shellfish products showed wide difference (1.3-15.2%) depending on raw materials and kinds of products. Fatty acid composition of products were mainly consisted of polyenoic acid(34.5-61.4%), and followed by saturated acid(24.5-47.8%), monoenoic acid(12.4-32.0%). The major fatty acids were palmitic acid, palmitoleic acid oleic acid, eicosapentaenoic acid and docosahexaenoic acid. In case of canned tuna in oil and canned smoked oyster in oil, linoleic acid was the most predominant component(48.2%, 34.6%). Judging from the results, canned fish and shellfish products were abundant of the polyunsaturated fatty acids in spite of the high temperature sterilization and lone storage periods.

Relationship between Monounsaturated Fatty Acid Composition and Stearoyl-CoA Desaturase mRNA Level in Hanwoo Liver and Loin Muscle (한우 간 및 등심 조직에서 불포화지방산의 조성비율과 Stearoyl-CoA Desaturase mRNA의 발현 양상)

  • The Stearoyl-CoA Desaturase(SCD) is a key enzyme, which converting palmitic acid(16:0) and stearic acid (18:0) to pahnitoleic acid(16:1) and oleic acid(l8:1), respectively. The concentration of oleic acid(18:1) in meat of beef cattle could influence both palatability and perception of meat. This experiment has conducted to determine relationship between the compositions of monounsaturated fatty acids and the SCD mRNA level in bovine liver and loin muscle tissue. The compositions of palmitoleic acid(16:1) and oleic acid(18:1) in loin muscle were 5% and 46% of total lipid and in liver were 2% and 20% of total lipid, respectively. On the other hand, the compositions of palmitic acid(16:0) and stearic acid(18:0) in loin muscle were 25% and 45% of total lipid and in liver were 14% and 43% of total lipid, respectively. The ratio of monounsaturated to saturated fatty acids(the desaturation index) was used as a measure of SCD activity in tissues. The average desaturation index in loin muscle was higher about 3.6-fold than that in liver. The desaturation index of oleate/stearate and palmitoleatelpalmitate in loin muscle were higher 8-fold and 1.8-fold than those in liver, respectively, showing that the substrate specificity of SCD enzyme was very different between liver and muscle tissues. To determine whether the composition of monounsaturated fatty acids in liver and muscle are dependent on SCD expression, SCD mRNA level was examined by RT-PCR analysis. The SCD mRNA level in loin muscle was higher about 3-fold than that in liver. Thus, the quantitative relationship between the desaturation index of fatty acid and SCD mRNA was observed in liver and muscle. The difference in the compositions of monounsaturated fatty acids between bovine liver and muscle tissues may be due to different level of Stearoyl-CoA Desaturase mRNA.

  • Sea-buckthorn fruits (Hippophae rhamnoides L.) are used in Mongolia as traditional medicine due to the health-benefits associated with its bioactive components. The purpose of this study was to investigate the chemical composition of Mongolian sea-buckthorn fruits. In terms of proximate composition, crude fat content was the highest, whereas its crude ash content was the lowest. In organic acid contents, malic acid content ($6.30{\pm}0.005$ mg/g) was the highest. Free sugars were composed of sucrose, xylose, glucose and mannitol. Vitamin C and carotenoid content were 320 mg/100 g and 715.25 mg/100 g, respectively. The major fatty acids were palmitic (C16:0), palmitoleic (C16:1), caprylic (C8:0) and linoleic (C18:2) acid.

  • This study was performed to determine chemical compositions of dried persimmons stored with plastic film at low temperature and the effect of film packaging on dried persimmons during storage. The moisture contents of dried persimmons were 30 to 36% and alcohol contents were 264 to 318 mg/100g and acetaldehyde contents were 25 to 40mg/100g. Total lipid contents of dried persimmon were 626 to 869mg/100g and oleic, palmitic, linolenic, and palmitoleic acid were major fatty acids in total lipids. Major amino acids were glutamic acid, arginine, cystine and aspartic acid. From the result of storage experiment, non-packed group was that hardness was high as decreased in moisture content. In case of film packed groups stored at 5$^{\circ}C$, self life was longer than non-packed and packed groups stored at room temperature. But, it need to develop available packing material and storage method.

  • Carotenoids liberated from the red carotenoprotein from the skin of ascidian(Halocynthia roretzi)were developed on TLC, and two bands were obtained with different Rfvalues. Their chemical reactivities and spectroscopic properties were studied and elucidated as astaxanthin diester and astaxanthin monoester. After saponification of carotenoid ester, GC analysis was performed. The carotenoid ester contained oleic acid, palmitic acid and palmitoleic acids as 50% of total amount, and had higher content unsaturated fatty acid including eicosapentanoic acid than saturated fatty acid.

Quality Properties of Conger Eel (Conger myriaster) Oils Extracted by Supercritical Carbon Dioxide and Conventional Methods (초임계 이산화탄소 및 유기용매를 이용하여 추출된 붕장어(Conger myriaster) 오일의 품질특성)

  • In this study, the extraction of Conger myriaster oil by using supercritical carbon dioxide (SC-CO2) and organic solvent was investigated. The extraction conditions conducted for SC-CO2 varied for pressure (25, 30 MPa) and temperature (45, 55 ℃), while the SC-CO2 flow rate was kept constant during the experiment (27 g min-1) and hexane was used as a conventional organic solvent. The extraction yield indicated that the best extraction condition would be SC-CO2 at 55 ℃ and 30 MPa, resulting in the highest yield of 37.73 ± 0.14%. The oils were characterized for their fatty acid (FAs) composition using gas chromatography, while it was revealed that the major FAs were mystric acid, palmitoleic acid, oleic acid, electroosapentaenoic acid (EPA), and docosahexaenoic acid (DHA). The oxidation stability of the extracted C. myriaster oil was evaluated by measuring the acid value, peroxide value, and free fatty acid. The best oxidative stability was obtained from SC-CO2 extracted oil at 30 MPa and 55 ℃. There was a significant difference in the color properties of the SC-CO2 and hexane extracted oils, with the SC-CO2 extracted oil showing better chromaticity than the oil extracted using hexane. Extracting oils from C. myriaster with SC-CO2 could bring better economic benefits than using organic solvents. When supercritical carbon dioxide was used, there was no post-treatment process; thus, it was confirmed that this is a more environmentally friendly oil extraction method.

Processing and Fermentation of Food Wastes with MS Microorganism Complex for Swine Feeds (MS 미생물복합군에 의한 음식폐기물의 양돈사료화를 위한 적정처리공정)

  • In order to investigate the proper processing of food wastes with miraculous soil-microorganisms (MS) for final use of swine feeds, calory, amino acid and fatty acid in food wastes were determined in relation with fermentation process with MS microorganism complex. Aflatoxin test was also performed to check safety of the fermented food wastes. Calory of food wastes was determined in average $7.60\;Kcal{\cdot}g^{-1}\;D.W.$ In finally processed food wastes, total content of amino acid was $93.0\;mg{\cdot}g^{-1}]\;D.W$, showing 18.5% of increase by the anaerobic fermentation. Essential and non-essential amino acids were measured at respectively 34.43 and $58.56\;mg{\cdot}g^{-1}\;D.W.$ Leucine, phenylalanine, isoleucine and threonine of essential amino acids and proline and glutamic acid of non-essential amino acids were highly composed as compared to others. The composition of fatty acid in food wastes was also increased by anaerobic fermentation for 3 weeks. Palmitic acid, oleic acid and palmitoleic acid were more important in quantity. Present results indicate that food wastes properly processed with MS have enough calory and are safe from aflatoxin, and that anaerobic fermentation with MS microorganism in an efficient process for hydrolyzing protein and lipids in food wastes.
