Removal of Cadmium Ion (Cd2+) by Pseudomonas aeruginosa Immobilized in Ca-Alginate Gel Beads in Packed-Bed Column Reactor (충전층 반응기내에서 고정된 Pseudomonas aeruginosa에 의한 Cd2+의 제거)

  • The effects of initial cadmium ion concentrations (50, 100, 200, 300ppm), and feeding velocities (30, 45, 60mL/hr) on the removal ratio of cadmium ion by Pseudomonas aeruginosa ATCC 27853 immobilized in Ca-alginate gel beads in a packed-bed column reactor were investigated at operating temperature $37^{\circ}C$. The removal ratio of cadmium ion with variable initial concentration was decreased in the following order : 50ppm > 100ppm > 200ppm > 300ppm. The optimum removal conditions of cadmium ion by Pseudomonas aeruginosa ATCC 27853 were initial concentration 50ppm, feeding velocity 30mL/hr.

NUMERICAL STUDY ON THE OPTIMAL DESIGN OF SPRAY SYSTEM IN PACKED BED SCRUBBER (충진층식 스크러버의 스프레이 시스템 최적 설계에 대한 수치해석적 연구)

  • This study evaluates the performance of the packed bed scrubber and proposes the optimization of spray system for improvements of collection efficiency. The packed bed scrubber is used primarily in the semiconductor manufacturing process. The mean diameter of entering solid particles in scrubber is the submicron. The impaction between water droplets and solid particles is an important factor in removing the solid particles. Thus, the coverage area of spray system influences on the collection efficiency. The collection efficiency of a single droplet is calculated through the mathematical model and numerical calculations are performed for coverage area for each nozzle type (Droplet diameters: 500, 319.5, $289.5{\mu}m$) and injected directions (0, 15, $30^{\circ}$). In case of nozzle type 3, the collection efficiency of a single droplet is highest but the collection efficiency of spray system has lowest value because the ratio of flow rate between the gas and water is below 0.1. The results show the coverage area ratio is about 85% in the case of nozzle type 3 and downward sirection $15^{\circ}$. It was shown that a coverage area increase by two times than an existing spray system. In simulation of demister, collection efficiency by demister is predicted about 80% and the pressure drop in demister is below 3.5 Pa.

Production of γ-Aminobutyric Acid Using Immobilized Glutamate Decarboxylase from Lactobacillus plantarum (Lactobacillus plantarum 유래 글루탐산 탈탄산효소의 고정화를 이용한 γ-aminobutyric acid의 생산)

  • The glutamate decarboxylase gene (gadB) from Lactobacillus plantarum WCFS1 was cloned and expressed as an N-terminal hexa-histidine-tagged fusion protein in Escherichia coli BL21 (DE3) as the host strain. Purified glutamate decarboxylase (GAD) was immobilized onto porous silica beads by covalent coupling. The pH dependence of activity and stability of the immobilized GAD was significantly altered, when compared to those of the free enzyme. Immobilized GAD was stable in the range of pH 3.5 to 6.0. The resulting packed-bed reactor produced 41.7 g of γ-aminobutyric acid/l·h at 45℃.

  • This study was carried out to investigate the heat storage/release characteristics of the thermochemical reaction of the calcined dolomite with the packed bed shape experimental apparatus for development of chemical heat pump system. In the present study, it was found that MgO of the calcined dolomite was not hydrated during the hydration process under the experimental conditions. Therefore, the MgO of the calcined dolomite can be regard as an inert material. As a result, it was found that all of CaO packed kept the reaction temperature of about $510^{\circ}C$ through the entire part of the bed. The dehydration reaction was incurred first at the wall side area as the supplied heat was transferred through the wall side into the packed bed. As a result of the temperature and concentration spread, the reaction was completed at the wall side progressed into the center.

Characteristics of Organic Compounds Removal and Microbe Attachment in Packed Bed Column Reactor Using Surface-modified Media (표면개질 담체를 이용한 충전탑 반응기에서 유기물 제거 및 미생물 부착 특성)

  • This study was accomplished using packed bed column reactors that contain nonsurface-modified polypropylene media and surface-modified media from hydrophobic surface property into hydrophilic property by ion beam irradiation. The objectives of this research was investigated the characteristics of organic compounds removal and microbe attachment from sewage of school cafeteria in these reactors. In 736.8 mg/L of the average inflow $COD_{Cr}$ concentration the reactors with and without surface modification showed 81.8% and 70.3% of average $COD_{Cr}$ removal efficiencies, respectively, which proves the $COD_{Cr}$ removal efficiency of surface-modified media reactor is higher than that of nonsurface-modified media reactor. After 90 days, there were maximum differences between modified system and non-modified system. In that time the maximum removal efficiency of $COD_{Cr}$ was 96.5% in modified system and was 85.2% in non-modified system that showed removal efficiency of surface-modified media system is 11.3% higher than that of nonsurface-modified media system. The average removal efficiency of SS was 80.4% for the surface modified system and 61.6% for the non-modified system under same condition. Also, the reactor of surface-modified media has advantage on microbe attachment and biofilm formation.

  • In this experiment, an attempt to use pellets($BaTiO_3$, $TiO_2$, ${\gamma}-Al_2O_3$, sludge) for $NO_x$ removal was conducted The effect of pellets on NO removal from simulated flue gas was experimentally investigated for packed-bed reactor of plate-plate geometry. An experimental investigation has been conducted for NO concentration of 50ppm balanced by air, and gas flow rate of $5{\ell}/min$. Ceramic pellets were used for surface discharge and the sludge pellets was added on $BaTiO_3$ and $TiO_2$ to increase $NO_x$ removal rate. In the result, $NO_x$ removal rate using $TiO_2$ was better than other pellets. $NO_2$ segnificatly generated by using $BaTiO_3$ pellets and sludge pellets used with $BaTiO_3$ decreased $NO_2$ generation.

  • An experimental investigation has been carried out to evaluate the characteristics in wastewater treatment using an anaerobic packed bed bioreactor with ceramics, rubber sponge, soft stone A, and soft stone B as carrier. The results of the work have shown that soft stone A as a carrier was superior to other carriers in methane production, then the content of methane for soft stone A was about 70%. First of all, soft stone A had higher efficiency of the COD removal than the others in response of passing the operation, as well as it had a low volatile acid in reactor. In addition, the slope of methane production with respect to the removal of COD ($m^{3}CH_{4}$/kgCOD) was 0.58 for soft stone A. In biomass hold-up equation for each carriers, the equation of soft stone A was m$_{p}$=714 ($C_{o}/0.41+C_{o}$) and it was the largest in this experimented carriers. Based upon the results obtained, it is suggested that the major effective carrier in wastewater treatments within the packed bed bioreactor used in this experimental work by soft stone A.

Characteristics of Organics and Nitrogen Removal with the Recycle Ratio in Anoxic / Oxic Packed Bed Process (충전탑형 무산소/호기 공정에서 반송비에 따른 유기물 및 질소 제거 특성)

  • This study was focused on the investigation of the characteristics of organics and nitrogen removal with the recycle ratio in anoxic/oxic(A/O) packed bed process that consisted of the anoxic reactor and the aerobic reactor. As increasing the recycle ratio by 0.5, 1.0, 2.0, the COD removal efficiency increased by 94.0%, 98.5%, 98.8% respectively. The aerobic reactor showed the perfect nitrification efficiency by 98.5%, 99.2%, 98.0% respectively. The T-N removal efficiency with the recycle ratio, increased by 56%, 67%, 70% respectively. As increasing the recycle ratio by 0.5, 1.0, 2.0, T-P removal efficiency decreased by 62.1%, 57.4%, 51.3% respectively. The process by releasing the stored phosphorus in the anoxic reactor and uptaking the excess phosphorus in the aerobic reactor, occurred well comparatively when recycle ratio is 0.5. But this process did not occur when the recycle ratio is 1.0 and 2.0. And optimum pH of nitrification was about 6~7 and alkalinity decreased as nitrification rate increased. As increasing the recycle ratio in the anoxic reactor, DO concentration and ORP increased.

Laccase Production Using Pleurotus ostreatus 1804 Immobilized on PUF Cubes in Batch and Packed Bed Reactors: Influence of Culture Conditions

  • The feasibility of laccase production by immobilization of Pleurotus ostreatus 1804 on polyurethane foam (PUF) cubes with respect to media composition was studied in both batch and reactor systems. Enhanced laccase yield was evidenced due to immobilization. A relatively high maximum laccase activity of 312.6 U was observed with immobilized mycelia in shake flasks compared to the maximum laccase activity of free mycelia (272.2 U). It is evident from this study that the culture conditions studied, i.e. biomass level, pH, substrate concentration, yeast extract concentration, $Cu^{2+}$ concentration, and alcohol nature, showed significant influence on the laccase yield. Gel electrophoretic analysis showed the molecular weight of the laccase produced by immobilized P. ostreatus to be 66 kDa. The laccase yield was significantly higher and more rapid in the packed bed reactor than in the shake flask experiments. A maximum laccase yield of 392.9 U was observed within 144 h of the fermentation period with complete glucose depletion.

Continuous Production of Lactosucrose by Immobilized Sterigmatomyces elviae Mutant

  • In this study, in order to develop a continuous production process of lactosucrose in a packed-bed reactor, Sterigmatomyces elviae ATCC 18894 was selected and mutated. The mutant strain of S. elviae showed 54.3% higher lactosucrose production than the wild type. Reaction conditions such as temperature, pH, substrate concentration and flow rate were also optimized. Under optimized reaction conditions ($50^{\circ}C$, pH 6.0, 25% sucrose and 25% lactose as substrate, flow rate 1.2 ml/min), the maximum concentration of lactosucrose (192 g/l) was obtained. In a packed-bed reactor, continuous production of lactosucrose was performed using S. elviae mutant immobilized in calcium alginate, and about 180 g/l of lactosucrose production was achieved for 48 days.
