Effect of Molecular Weights and Mixture Ratios of Polyvinylpyrrolidone on the Bioavailability of Ipriflavone Solid Dispersion (이프리플라본 고체분산체의 생체이용률에 미치는 폴리비닐피롤리돈의 분자량 및 혼합비율의 영향)

  • Ipriflavone (3-phenyl-7-isopropoxy-4H-1-benzopyran-4-one, IP) is a well-known antiosteoporotic drug with poor bioavailability. In the previous study, we reported that the IP formulation prepared by spray-drying method with polyvinylpyrrolidone (PVP) (SIP) was very effective in improving the bioavailability of IP. In this study, we examined the effects of molecular weight and mixture ratios of PVP to IP on the systemic absorption of IP following oral administration of SIP at a dose of 50 mg/kg to rats. In the effect of molecular weight, the Cmax of spray-dried IP with PVP K30 (SIP-K30) was significantly higher than those of spray-dried IP with PVP 360 (SIP-360), spray-dried IP with PVP K90 (SIP-K90), and spray-dried IP with PVP K17 (SIP-K17) (p<0.05). The AUC of SIP-K30 was about 2, 3, and 5.5 times higher than those of SIP-360, SIP-K90, and SIP-K17, respectively. The AUC value of SIP-K30 was significantly greater than those of SIP-K17 and SIP-K90 (p<0.05) except for SIP-360. In the ratio of PVP K30 to drug, the $C_{max}$ and the AUC value of 3 : 7 IP-PVP solid dispersion were similar to those of 5 : 5 IP-PVP and significantly higher than those of the other solid dispersions (p<0.05). It was concluded that the spray-dried IP with PVP K30 at the ratio of 3:7 (w/w) was the best formulation for improving the bioavailability of IP.

  • Hydrogels for wound dressing were manufactured using poly(vinylalcohol)(PVA) and poly(N-vinylpyrrolidone)(PVP). The hydrogels were obtained by exposing to $^{60}$ Co${\gamma}$-rays after freezing and thawing of aqueous solutions of PVA and PVP to improve mechanical strength. Mechanical properties such as gelation, water absorptivity and gel strength were examined after repeating the \"freezing and thawing\" of PVA/PVP hydrogels, and then irradiating them at 40 kGy. The PVA/PVP ratio was in the range of 30:70 ~ 100:0, and the solid concentration of PVA/PVP was 20 wt%. The gelation and strength of hydrogels were much higher when \"freezing and thawing\" and the irradiation process were used than when only the irradiation process was utilized.   • Electrospun of PVP (polyvinylpyrrolidone) ultra fine fibers were fabricated using various solvents including methanol, ethanol, 2-propanol, butanol, acetone, methylene chloride, and DMF, which possess different properties such as boiling point, dielectric constant, and dipole moment. Electrospun PVP fiber was influenced by viscosity, conductivity, and surface tension of spinning solution. Therefore, the electrospun PVP fiber was successfully prepared under critical conditions of viscosity > $0.114kg/m{\cdot}s$, conductivity > 1.02 mS/m, surface tension < 30.0 mN/m. In case of an ethanol solvent system, average diameter of PVP fiber increased from 1701 nm to 5454 nm as increased the applied voltage from 10 kV to 20 kV.

  • In this paper, the characteristics of a humidity sensor implemented by Mach Zehnder Interferometer (MZI) in a Planar Lightwave Circuit (PLC) have been designed and demonstrated. The humidity outside is detected with polyvinylpyrrolidone (PVP) coated on the etched arm of the MZI. The length of the etched arm is 10 mm and the PVP was coated by dip-coating into the etched region. As the refractive index of the PVP changes with the surrounding humidity, the PVP-coated humidity sensor presented changes in the interferogram depending on RH (Relative Humidity) around the PLC. The measured results show that the proposed humidity sensor works successfully in the range of 30% to 80% of RH.

Effect of Various Supplements on Embryo Development and Quality of Bovine Embryos during In Vitro Maturation (한우 난포란의 체외성숙 시 여러 가지 첨가물이 배 발생과 품질에 미치는 영향)

  • This study was examined the effects of concentrations of polyvinylpyrrolidone(PVP) and supplementation of EGF, cysteine and PVP during in vitro maturation on the development of bovine embryos. In experiment 1, 0.1 to 3.0% PVP was supplemented to IVM medium before IVF. The development rates to the blastocyst stage was significantly higher in 0.5% PVP group than 3.0% PVP group (P<0.05). In experiment 2, EGF, rysteine and PVP were supplemented to IVM medium. The hight cleavage rate was obtained from cysteine group, but blastocyst formation rates did not differ among groups. The highest total cell number and inner cell mass (ICM) cell number were observed in cysteine group. In PVP group, ICM cell number was significantly low than those of cysteine and control groups (P<0.05). After embryo transfer, pregnancy rate was significantly low in PVP group compared to other groups (P<0.05). These results indicate that the supplementation of PVP in IVM medium support the embryo development, but has a deteriorate effect on the blastocyst quality.

  • Global warming due to indiscriminate carbon dioxide emissions has a profound impact on human life by causing abnormal climate change and ecosystem destruction. As a way to reduce carbon dioxide emissions, in this study, we presented a polymeric membrane prepared by blending a self-crosslinkable P(GMA-g-PPG)-co-POEM (SP) copolymer and commercial polymer polyvinylpyrrolidone (PVP). As the content of PVP increased, it was observed that the gas permeance decreased and $CO_2/N_2$ selectivity increased. At 30 wt% PVP content, the $CO_2$ permeance of the membrane decreased from 72.9 GPU of pure SP polymer to 12.6 GPU, while $CO_2/N_2$ selectivity improved by 79% from 28.1 to 50.4. It results from the hydrogen bonding between the SP copolymer and PVP, leading to more compact structure of the polymer chains, which was confirmed by FT-IR, TGA, XRD and SEM analysis. Therefore, we suggest that the permeance and selectivity of the membranes can be easily adjusted as desired by controlling the PVP content in the SP/PVP polymer blend.

Studies on Solid Dispersion System of Ketoconazole (케토코나졸의 고체분산계에 관한 연구)

  • Dissolution profiles of ketoconazole in solid dispersion system (SDS) were investigated in the second fluid (pH 6.8) for the dissolution test (KPV). PVP K-30 and methyl cellulose were used as carriers in SDS, and chloroform as a solvent. Computer optimization technique was applied to obtain an optimum formula of SDS, and the following results were obtained; 1) Dissolution rate of ketoconazole in SDS was larger than that of pure ketoconazole in the second fluid of pH 6.8. 2) PVP K-30 and methyl cellulosr were good carriers for SDS of ketoconazole. Moerover PVP K-30 was better than methyl cellulose. 3) The optimum formula of ketoconazole SDS obtained from computer optimization technique was chloroform 175 ml, PVP K-30 5g and methyl cellulose 0.2g per 1g of ketoconazole. 4) The experimental value of $A_{60}$ (Amount released from ketoconazole tablet during 60 minutes) obtained from SDS by optimum formula agreed well with the value calculated by polynomial regression equation.

  • Objectives This study aims to evaluate the size-dependent toxicity of spherical silver nanoparticles (Ag NPs) to an endemic benthic organism, Glyptotendipes tokunagai. Methods Ag nanoparticles of three nominal sizes (50, 100, and 150 nm) capped with polyvinyl pyrrolidone (PVP-Ag NPs) were used. Their physicochemical properties, acute toxicity (48 hours), and bioaccumulation were measured using third instar larvae of G. tokunagai. Results The aggregation and dissolution of PVP-Ag NPs increased with exposure time and concentration, respectively, particularly for 50 nm PVP-Ag NPs. However, the dissolved concentration of Ag ions was not significant compared with the median lethal concentration value for $AgNO_3$ (3.51 mg/L). The acute toxicity of PVP-Ag NPs was highest for the smallest particles (50 nm), whereas bioaccumulation was greatest for the largest particles (150 nm). However, larger PVP-Ag NPs were absorbed and excreted rapidly, resulting in shorter stays in G. tokunagai than the smaller ones. Conclusions The size of PVP-Ag NPs significantly affects their acute toxicity to G. tokunagai. In particular, smaller PVP-Ag NPs have a higher solubility and stay longer in the body of G. tokunagai, resulting in higher toxicity than larger PVP-Ag NPs.

  • The shelf life of 1,2-indandione/zinc (1,2-IND/Zn) solution and polyvinylpyrrolidone (PVP) solution, which are known as reagents for developing latent fingermarks deposited on the surface of thermal paper, was studied. The standard latent fingermarks used for comparisons were artificial latent fingermarks printed on thermally sensitive and non-sensitive surfaces with the same intensity. Upon treatment of standard latent fingermarks with the pre-mixed 1,2-IND/Zn and PVP solutions, the fingermarks could be successfully developed until 3 days after the preparation of the mixture. However, from the third day after mixing the reagents, blackening was observed on the surface of the thermal paper, indicating deterioration of the reagent performance. The 1,2-IND/Zn and PVP solutions separately stored without mixing in advance were mixed immediately before use, and the development efficiency of the latent fingermarks deposited on the surface of thermal paper was observed. The performance of the PVP solution decreased after 20 days from the preparation of the reagent. It was also found that the shelf life of 1,2-IND/Zn and PVP mixture was determined by the PVP solution. The effect of oxygen and moisture on the degradation of PVP was investigated. It was found that the performance of the PVP solution deteriorated because of the influence of moisture, though it was not affected by oxygen.

Survival and In Vitro Development of Immature Bovine Oocytes Cryopreserved by Vitrification

  • The present study was undertaken to investigate the effects of PVP concentration and exposure temperature to vitrification solution on the post-thaw survival, in vitro maturation and development of immature bovine oocytes (germinal vesicle stage). The vitrification solution (VS) consisted of 40% ethylene glycol (EG)+0.5 M sucrose (S)+10% FBS. PVP was added to VS: 0%, 5% or 10%. The cumulus-oocyte complexes (COCs) were diluted in VS as one step, after 2 min the COCs were loaded in straw and vitrified by direct immersion into liquid nitrogen. For thawing, the straws were plunged into $30^{\circ}C$ water bath for 10s. After thawing, the oocytes were diluted in 0.5 M (in DPBS with 10% FBS) sucrose solution for 5 min. The survival rate (FDA-test and trypan blue) of immature bovine oocytes was measured. The survival rate was higher in 5% PVP (91.5%) than in 0% (64.2%) or in 10% PVP (79.7%). The proportion of metaphase II formation was 69.35% in control (no vitrified COCs), 9.3% in 40% EG+0.5 M S+0% PVP and 21.05% in 40% EG+0.5 M S+5% PVP (p<0.05). The effect of room temperature ($25^{\circ}C$ for 10 min) and cold temperature ($4^{\circ}C$ for 10 min) on COCs were determined in this study. After IVF, the cleavage and blastocysts rate of oocytes exposed to room temperature and cold temperature in VS+5% PVP was significantly different (2 cell: 63.20% vs 37.97%, blastocysts: 18.40% vs 2.53%). The cleavage rates of frozen-thawed oocytes were 20.53% with PVP and 22.13% without PVP (p>0.05). Two out of 151 oocytes (1.32%) developed to blastocyst stage after frozen-thawed with 5% PVP (p>0.05). Development of oocytes after frozen-thawing to the 2 cell were not significantly affected with or without PVP following IVF. However, the vitrification of immature bovine oocytes with PVP maintained the ability to develop to the blastocyst stage after IVM-IVF and IVC, while no blastocysts were obtained from oocytes vitrified without PVP. These results suggested that PVP has a protective role for vitrification of immature bovine oocytes as far as survival is concerned, however, the protection was not sufficient enough to support blastocyst formation.
