The Improvement of Sensing Performance of ISFET Glucose and Sucrose Sensors by Using Platinum Electrode and Photo-crosslinkable Polymers (백금전극과 감광성 고분자를 이용한 ISFET 포도당 및 자당센서의 감지성능 개선)

  • The ISFET glucose and sucrose sensors containing platinum electrode and photopolymeric enzyme membrane were fabricated. The platinum working electrode was used for the electrolysis of hydrogen peroxide, which was the other product of the enzyme reaction, to improve sensing characteristics of the sensors. In order to improve response time, photo-crosslinkable polymer(PVA-SbQ) was used to the matrix for the enzyme immobilized membrane. The characteristics of glucose and sucrose sensors were investigated according to the variation of platinum electrode area. The response time was about $3{\sim}5$ minutes and determinations of glucose and sucrose in the range of about $30{\sim}300mg/dl$ could be possible.

  • A new functional organic thin film optical waveguide ion sensor is designed, which can select a specific ion, i.e., $Ca^{2+}$ -ion. The sensing membrane was composed of PVC-PVAC-PVA copolymer matrix based on anionic cation-selective chromoionophor(ETH5294), neutral ionophore(K23El), anionic site and plasticizer and it was coated on the etched glass substrate as embeded type optical waveguide itself. The sensor sensitivity dependence on waveguide length and thickness, contence of chromoionophore, and each mode was investigated. And this sensor could detect $Ca^{2+}$ ion in concentrations ranging from 1$\times$10­6~1M(with 0.05M tris-HCI buffer solution of pH7.4) by measuring the absorbance change at 514nm of light. Utilizing thin film membrane, the fast response time and high sensitivity are obtained. Also, it is expected that this sensor can be applied to various biochemical important ions.ons.

Studies on Preparation and Ion Exchange Characteristics of Humic Acid Membranes (Humic Acid 분리막의 제조와 이온교환 특성에 관한 연구)

  • Humic acid has been extracted and purified from biologically nondegradable humic substances. Using the ion exchange capability of carboxylic acids which are the main component of the humic acids, a membrane was prepared with poly(viny1 alcohol). Its transport behavior of biologically active ions, $K^+$and $Na^+$, were investigated. The ion transport velocity increased with hydrogen ion concentration, especially, in the range of $10^-1$~$10^0$. The selectivity increased with increasing the concentrations of $K^{+}$ and Na$^{+}$, In particular, the transport velocity of $K^+$ increased twice compared to that of $Na^+$ at the 100 hydrogen ion concentration. In this regards, humic acid may be used as a new material for ion exchange membranes.

  • The effects of interactions between permeant molecules or permeant and membrane material have been investigated on the permeation behavior of permeants in pervaporation of water/alcohol mix¬tures. A poly(vinyl alcohol) (PVA) membrane crosslinked with glutaraldehyde was employed in this study. A homologous series of alcohols in aqueous solution were used as feed. The pervaporation experiments were carried out with feed having 70-97 wt.% of alcohol contents and at various feed temperatures. In a high alcohol content above 92 wt.%, the permeation rate was increased in the order of interaction strength between alcohol and water in feed. However, in a low alcohol content below 90 wt.%, the tendency of the permeation rate was found to be opposite. These observations were discussed in terms of changes in interaction between permeant/permeant or permeant/membrane in varying feed composition and feed temperature.

  • pH-ISFETs, the semiconductor pH sensors, were combined with immobilized enzyme membranes to prepare FET type urea and glucose sensors and its operational characteristics were investigated. Photolithography techniques were applied to immobilize enzymes on the $H^{+}$ sensing membrane of the pH-ISFET with photo-sensitive polymers, PVA-SbQ. Fabricated urea and glucose sensors could determine $0.5{\sim}50{\;}mg/dl$ urea concentrations and $10{\sim}1000{\;}mg/dl$ glucose concentrations, respectively.

  • Prussian blue (PB) is well known for its excellent $Cs^+$ ions adsorption capacity. Due to the high dispersibility of PB in aqueous phase, composite materials imbedding PB in supporting materials have been introduced as a solution. However, building PB particles inside porous supporting materials is still difficult, as PB particles are not fully formed and elute out to water. In this study, we suggest layer-by-layer (LBL) assembly to provide better immobilization of PB on supporting materials of poly vinyl alcohol sponge (PVA) and cellulose filter (CF). Three different PB attachment methods, ex-situ/in-situ/LBL assembly, were evaluated using PB leaching test as well as $Cs^+$ adsorption test. Changes of surface functionality and morphology during PB composite preparation protocols were monitored through Fourier transform infrared spectroscopy and scanning electron microscopy. The results indicate that LBL assembly led to better PB attachment on supporting materials, bringing less eluting PB particles in aqueous phase compared to other synthesis methodologies, such as ex-situ and in-situ synthesis. By enhancing the stability of the adsorbent, adsorption capacity of PVA-PB with LBL improved nine times and that of CF-PB improved over 20 times. Therefore, the results suggest that LBL assembly offers a better orientation for growing PB particles on porous supporting materials.

Preparation of Ion Exchange Membranes for Fuel Cell Based on Crosslinked Poly(vinyl alcohol) with Poly(acrylic acid-co-maleic acid)

  • Crosslinked poly(vinyl alcohol) (PVA) membranes were prepared at various crosslinking temperatures using poly(acrylic acid-co-maleic acid) (PAM) containing different PAM contents. The thermal properties of these PVA/PAM membranes prepared at various reaction temperatures were characterized using differential scanning calorimetry (DSC). The proton conductivity and methanol permeability of PVA/PAM membranes were then investigated as PAM content was varied from 3 to 13 wt%. It was found that the proton and methanol transport were dependent on PAM content in their function both as crosslinking agent and as donor of hydrophilic -COOH groups. Both these properties decreased monotonously with increasing PAM concentration. The proton conductivities of these PVA/PAM membranes were in the range from $10^{-3}\;to\;10^{-2}S/cm$ and the methanol permeabilities from $10^{-7}\;to\;10^{-6}cm^{2}/sec$. In addition, the effect of operating temperature up to $80^{\circ}C$ on ion conductivity was examined for three selected membranes: 7, 9 and 11 wt% PAM membranes. Ion conductivity increased with increasing operating temperature and showed and S/cm at $80^{\circ}C$, respectively. The effects of crosslinking and ionomer group concentration were also examined in terms of water content, ion exchange capacity (IEC), and fixed ion concentration. In addition, the number of water molecules per ionomer site was calculated using both water contents and IEC values. With overall consideration for all the properties measured in this study, $7{\sim}9\;wt%$ PAM membrane prepared at $140^{\circ}C$ exhibited the best performance. These characteristics of PVA/PAM membranes are desirable in applications related to the direct methanol fuel cell (DMFC).

  • In recent years, polymer membranes, which are actively used in various industrial fields, have the advantage of being able to impart unique properties through the control of chemical structures and physical properties in the film-fabrication process, as well as through fabricating blend membranes mixed with various materials. In this study, the solubility parameter, which can be used as an index of miscibility with other materials, was calculated using molecular dynamics using a silkworm (Bombyx mori) silk polymer which has a wide potential to be used as an eco-friendly natural material. When the solubility parameter of polyvinylalcohol (PVA), which is also environmentally friendly and biocompatible, was calculated by molecular dynamics and compared with each other, it was confirmed that the two polymer materials had similar solubility parameter values. In conclusion, it was theoretically proved that the two polymers could blend well with each other, which was confirmed through experiments.

Hydrophilization of hydrophobic membrane surfaces for the enhancement of water flux via adsorption of water-soluble polymers

  • In this study, to improve the water flux of porous hydrophobic membranes, various water-soluble polymers including neutral, cationic and anionic polymers were adsorbed using 'salting-out' method. The adsorbed hydrophobic membrane surfaces were characterized mainly via the measurements of contact angles and scanning electron microscopy (SEM) images. To enhance the durability of the modified membranes, the water-soluble polymers such poly(vinyl alcohol) (PVA) were crosslinked with glutaraldehyde (GA) and found to be resistant for more than 2 months in vigorously stirred water. The water flux was much more increased when the ionic polymers used as the coating materials rather than the neutral polymer and in this case, about 70% of $0.31L/m^2{\cdot}h$ (LMH) to 0.50 LMH was increased when 300 mg/L of polyacrylamide (PAAm) was used as the coating agents. Among the cationic coating polymers such as poly(styrene sulfonic acid-co-maleic acid) (PSSA_MA), poly(acrylic acid-comaleic acid) (PAM) and poly(acrylic acid) (PAA), PSSA_MA was found to be the best in terms of contact angle and water flux. In the case of PSSA_MA, the water flux was enhanced about 80%. The low concentration of the coating solution was better to hydrophilize while the high concentration inclined to block the pores on the membrane surfaces. The best coating condition was found: (1) coating concentration 150 to 300 mg/L, (2) ionic strength 0.15, (3) coating time 20 min.

Influence of the Substrate and Inhibitors Related to Phosphatidylinositol Metabolism in the Maturation Processes of Porcine Oocytes (돼지 난모세포의 성숙과정에서 Phosphatidylinositol 대사의 기질 및 억제인자의 영향)

  • We evaluated the effects of the substrate and inhibitors related to phosphatidylinositol metabolism on in vitro maturation and fertilization of porcine oocytes. Cumulus-oocyte complexes were cultured in mTLP-PVA medium supplemented with or without inositol (250 mM) fur 46h. Subsequently, these oocytes were inseminated with fresh boar semen in mTALP-PVA medium for 6h. At 6h after insemination, oocytes were cultured for further 12 h in TCM-199 supplemented with 10% FBS (fetal bovine serum). The higher percentage of oocytes in inositol-supplemented medium reached metaphase of the second meiotic division compared to those in control (81.4% vs. 67.3%; P<0.()5). following 18 h of insemination, more number of male pronuclei were formed in the oocytes matured in inositol-supplemented medium than in those of control experiment (42.0% vs. 27.3%; P<0.05). When oocytes were cultured in medium with 10mM LiCl (chloride lithium) or 0.5mM dbcAMP (dibutyryl cyclic adenosine monophosphate) to determine the role of inositol on the maturation of oocytes, these two drugs inhibited the meiotic division of oocytes (P<0.05). However, addition of inositol to the culture medium did overcome the inhibitory effect of these drugs on the oocyte maturation. DbcAMP and verapamil supplemented synergistically arrested the meiotic division of oocytes. Addition of verapamil did not inhibit germinal vesicle breakdown, but it severly inhibited the second meiotic division of oocytes. These results suggest that inositol exert its improving effects on maturation, by activating the PI (phosphatidylinositol) cycle and causing beneficial changes in both cytoplasm and membrane of oocytes.
