A Review on Clinical Studies on Gynecological Disease by Using a Pulse Diagnosis Device (맥진기를 활용한 부인과 질환 대상 임상연구 고찰)

  • Objectives: The purpose of this study was to survey the status of clinical use of a pulse tonometric device (PTD).Methods: We searched domestic and international articles using the PTD from online medical databases including OASIS, RISS, DBpia and PubMed. We selected articles on clinical application of PTD but excluded articles on mechanical design or software programming for developing a new PTD. Finally we found 13 articles and classified the articles according to the disease of clinical study.Results: Out of the 13 articles, 5 were focused on the dysmenorrhea. Three articles were focused on the infertility, two article were focused on the post mastectomy. According to the results, the pulse energy has been widely used in research as the primary outcome.Conclusions: We found out that until now the researches on clinical application of PTD mainly had been performed for producing a variety of PTDs. This results of this study will be used as a useful information during perform a clinical study and clinics. We suggest that the standard operating procedure for PTD will be developed, and researches for development and its application of various new contents will be performed.

  • Effective for the operation of fire department management of occupational stress than what organizational members perceive and to identify factors that would cause job stress. Dysfunction caused by job stress or minimize that by eliminating the stress factor that can enhance organizational effectiveness research is needed. The results of the study the impact on job stress of firefighters of the psychological environment of the most influential factor is the variable. Next, organizational culture, physical environment, orderly, and administration and human relations were not significant statistically.

  • Though protein transduction domains (PTDs) are well known for the delivery of exogenous therapeutic proteins into living cells, the overall low efficiency of transduction is a serious obstacle. We investigated the effect of bog blueberry anthocyanins (BBA) on protein transduction efficiency and found that BBA enhanced the transduction efficiencies of Tat-SOD fusion protein into HeLa cells and mice skin. The enzymatic activities in the cells and skin tissue in the presence of BBA were markedly increased compared to controls. Further, BBA did not demonstrate any cell toxicity at various concentrations. Although the mechanism is not fully understood, we suggest that BBA might alter the conformation of the membrane, which would indicate that BBA can be used as a protein transduction enhancer for the efficient delivery of therapeutic proteins for a variety of disorders.

  • Many proteins with poor transduction efficiency were reported to be delivered to cells by fusion with protein transduction domains (PTDs). In this study, we investigated the effect of levosulpiride on the transduction of PEP-1 ribosomal protein S3 (PEP-1-rpS3), and examined its influence on the stimulation of the therapeutic properties of PEP-1-rpS3. PEP-1-rpS3 transduction into HaCaT human keratinocytes and mouse skin was stimulated by levosulpiride in a manner that did not directly affect the cell viability. Following 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced inflammation in mice, levosulpiride alone was ineffective in reducing TPA-induced edema and in inhibiting the elevated productions of inflammatory mediators and cytokines, such as cyclooxygenase-2, inducible nitric oxide synthase, interleukin-6 and -1${\beta}$, and tumor necrosis factor-${\alpha}$. Anti-inflammatory activity by PEP-1-rpS3 + levosulpiride was significantly more potent than by PEP-1-rpS3 alone. These results suggest that levosulpiride may be useful for enhancing the therapeutic effect of PEP-1-rpS3 against various inflammatory diseases.

  • Human fibroblast growth factor (FGF) has the potential to be a commercially important therapeutic or cosmeceutical agent due to its ability to generate tissue and heal wounds. Granting permeability into skin tissues increases the therapeutic effects of FGF. Thus, several researchers have attempted the fusion of FGF conjugates with protein transduction domains (PTDs) to investigate the transduction ability and therapeutic effects of FGF. Less is known, however, about whether the location of PTD fused to the N- or C-terminus of FGF proteins has a significant impact on the folding and stability in Escherichia coli, and eventually, on transduction. Here, we report cloning of human basic fibroblast growth factor (FGF2) as a control and FGF2 with PTD fused to the N- or C-terminal ends of FGF proteins by an overlap extension PCR. We performed expression, verified expression properties of recombinant FGF2 without or with PTD fused to the N-terminus and the C-terminus, and investigated transduction ability into tissue by treating the dorsal skin of mice subjects. As a result, FGF2 and FGF2-PTD (fused to C-terminus) fusion protein were expressed as soluble forms suitable for straight-forward purification, unlike insoluble PTD-FGF2 (fused to N-terminus), but only FGF2-PTD fusion protein could transduce into the dorsal skin tissue of the mice subjects. Our results suggest that FGF2 with PTD fused to the C-terminus is more efficient than other options in terms of expression, purification, and delivery into skin tissue, as it does not require labor-intensive, costly, and time-consuming methods.
