Molecular Identification of the Toxic Alexandrium tamiyavanichii (Dinophyceae) by the Whole-cell FISH Method

  • The dinoflagellate Alexandrium tamiyavanichii Balech, a producer of toxins causing paralytic shellfish poisoning (PSP), has recently been considered as one of main organisms responsible for toxication of shellfish in Japan. In this study, A. tamiyavanichii was subjected to a molecular phylogenetic analysis inferred from 28S rDNA D1-D2 sequences and a species-specific LSU rRNA-targeted oligonucleotide DNA probe was designed to identify A. tamiyavanichii using the whole cell-FISH (fluorescence in situ hybridization). The sequences of the 28S rDNA D1-D2 region of A. tamiyavanichii showed no difference from A. cohorticular AF1746l4 (present name A. tamiyavanichii) and formed a distinct clade from the 'tamarensis species complex'. The probe, TAMID2, reacted specifically with A. tamiyavanichii cultured cells, without any cross-reaction with other species belonging to the same genus, including A. tamarense, A. catenella, A. affine, A. fraterculus, A. insuetum and A. pseudogonyaulax. In a test of cross-reactivity with a field sample, TAMID2 reacted consistently with only A. tamiyavanichii, indicating that the present protocol involving the TAMID2 probe might be useful for detecting toxic A. tamiyavanichii in a simple and rapid manner.

  • In order to introduce the hydrogen economy and increase supply, research in the field of hydrogen production is being actively conducted. Among the hydrogen production methods, the method of steam reforming from natural gas and producing it currently accounts for about 50% of the global hydrogen production. In the method of steam reforming process, hydrogen can be produced by adding a reformer to an existing natural gas supply pipe. Because of these advantages, it is evaluated as a realistic production method at present in Korea, where the city gas supply chain is well established. But there is concern in that it is highly likely to be installed in downtown areas and residential spaces. In this study, the risk of the process of steam reforming to produce hydrogen was reviewed.

광통신 시스템에서 PDL로 인한 PMD 보상 수준 한계에 대한 연구 (Analysis on the PMD compensator's limitation from PDL in the optical communication system)

  • 본 논문에서는 10Gb/s의 광전송 시스템에서 편광모드별 광손실(PDL)이 개입되었을 때, 전송 시스템이 받는 영향과 편광 모드분산에 대한 보상회로의 실효성이 어떻게 감소되는지를 컴퓨터 모의 실험을 통해 분석하였다. 그 결과 편광모드별 광손실의 양이 늘어남에 따라서 시스템의 성능은 비례하여 감소되는 형태를 보였으며 편광모드 분산에 대한 보상회로의 성능 또한 감소되었다. 이는 모드별 광손실의 효과가 포함되었을 때에는 단순한 속도 차이 외에 펄스의 모양이 그대로 유지되는 축인 PSP의 직교성이 상실되기 때문에 펄스의 속도 차이에 왜곡이 더해져서 보상이 어렵기 때문이다. 따라서 PDL 소자가 다수 포함되는 장거리 광대역 광전송 시스템에서의 편광모드 분산에 대한 보상회로 설계 시에 이에 대한 숙고가 필요함을 알수있었다.

그룹게임이 유아의 친사회적 사고 및 사회적 능력에 미치는 영향 (The Effect of the Group Games on Young Children's Prosocial Thinking and Social Ability)

  • The purpose of this study was to investigate the effect of the group games on children's prosocial thinking and social ability. The subjects of this study were 30 four-five years old boys and' girls in S daycare center in Iksan of Jeonbuk province. Children in this study were divided into two groups. In order to measure children's pro social thinking. 'Skill Situation Measure' developed by McGinnis & Goldstein(l990) was used. In order to measure children's social ability, 'Preschool Socio-affective Profile(PSP)' developed by La Freniere. Dumas, Capuano & Dubeau(1992) was used. The tools were applied in two groups in pre-test, and the experimental group was given a total of 20 sessions (twice a week. for 10 weeks). The results of the study were as follows: 1. The experimental group children with group games showed a significant improvement in their prosocial thinking. 2. The experimental group children with group games showed a significant improvement in their social ability.

코안다 효과를 이용한 평판 슬롯의 막냉각 성능 향상 (Improvement of Film Cooling Performance of a Slot on a Flat Plate Using Coanda Effect)

  • In this study, the Coanda effect inducing bump was applied to improve the film cooling effectiveness on the flat plate with $30^{\circ}$ and $45^{\circ}$ angled rectangular slots. The slot length to width ratio was 6. A cylindrical cap shaped structure, called Coanda bump, was installed at the exit of the slot to generate Coanda effect. The width and height of the bump was 10.5 mm and 1 mm, respectively. The film cooling effectiveness was measured at the fixed blowing ratio, M=2.0, using pressure sensitive paint (PSP) technique. The mainstream velocity was 10 m/s and the turbulence intensity was about 0.5%. Results showed that the film cooling effectiveness for case of $30^{\circ}$ angled slot was higher than that of $45^{\circ}$ angled slot. It was found that there was no positive effect of Coanda effect on the overall averaged film cooling effectiveness for the $30^{\circ}$ angled slot. On the other hand, for the $45^{\circ}$ angled slot, the film cooling effectiveness was improved with the installation of the Coanda bump.

Identification of a pr 1-like Gene of Entomopathogenic Fungus, Beauveria bassiana F-101 Isolated from Thecodiplosis japonensis

  • Beauveria bassiana F-101, which has high toxicity toward Acantholyda parki as well as Thecodiplosis japonensis, was an isolate to develop an alternative control system against the major forest pests. Up to now, in B. bassiana, only one pr1 gene has been isolated and characterized. Therefore, we here reported the identification of a pr1-like gene, which would be a factor of toxicity from B. bassiana F-101. The oligonucleotides for the amplification of the pr1-like gene, were chosen based on the conserved regions of the subtilisin family enzymes, pr1 genes of B. bassiana and Metarhizium anisopliae, and proteinase K of Tritirachium album. The cloned PCR fragment had 1111 bp including 52 bp intron. The deduced Pr1-like peptide showed a low identity with Pr1s of entomopathogenic fungi such as B. bassiana Pr1 (BbPr1) and M. anisopliae Pr1 (MaPr1) as well as the proteinase K of T. album (TaPrK). Instead, the deduced peptide had a substantially high amino acid sequence identity $(>65\%)$ with the serine proteases of Magnaporthe grisea (MgSPM1) and Podospora anserina (PaPspA). These results, therefore, appear to suggest that the putative Pr1-like peptide of B. bassiana F-101 belongs to the subtilisin-like serine protease family and may be a novel gene.

자색고구마 습관성캘러스로부터 안토시아닌 고생합성 세포주 선발 (Selection of High Anthocyanin-Producing Cells from Habituated Callus Derived from Purple Sweetpotato)

  • 자색고구마 (Ipomoea batatas cv. Jami) 잎 조직배양으로 유도된 캘러스는 계대배양시 생장량이 감소하였다. 빠르고 간편하게 생장하는 습관성캘러스를 $1{\mu}M\;2,4-D$ 첨가된 MSBM에서 계대배양 없이 6개월간 지속배양하여 선발하였다. 습관성캘러스는 생장조절제가 첨가되지 않는 MSBM의 고체, 액체 배지에서 계대배양 횟수의 제한 없이 빠르게 지속생장이 가능하였다. 암조건의 액체배양 6 개월 후에는 자색생합성 부위가 생겼으며, 이를 2년 여간 분리하여 자색습관성세포괴를 선발하였다. 안토시아닌 함량은 자미의 괴근에서 추출되는 1.5 mg/ml에 근접한 1.0 mg/ml으로 나타났다. 자색습관성 세포괴는 안토시아닌 기내대량에 효과적 세포주로 사용 될 수 있을 것으로 기대한다.

자연 기흉의 치료에서 8 French 도관삽입의 치료 효과에 대한 장기적 관찰 (The Long-term Follow-up Study of Therapeutic Effects of 8 French Catheter for Spontaneous Pneumothorax)

  • 연구배경 : 자연 기흉은 기저 질환이 없는 건강한 사람에서 발생하는 원발성 자연 기흉과, 결핵이나 만성폐쇄성 폐질환에서 발생하는 속발성 자연기흉으로 나뉘어지며, 자연 기흉에 대한 치료는 다양하지만 흔히 흉관 삽입을 우선적으로 고려하고 있다. 흉관삽입법은 특별한 술식과 삽입시 통증, 피하기종, 감염 등의 합병증이 문제되는 바 본 저자들은 흉관에 비해 직경이 훨씬 작은 8 French 도관을 자연 기흉의 치료에 적용했을 때 단기적인 치료 효과가 있음을 이전 연구에서 확인한 바 있다. 본 연구는 의인성 기흉의 치료에 이용되어져 왔던 작은 직경의 도관을 자연 기흉의 치료에 적용하였을 때, 치료 성공후 추적관찰을 통하여 재발율을 조사하여 장기적인 치료적 효과를 살펴보고자 하였다. 방법 : 1990년 1월부터 1996년 1월까지 중앙대학교 부속병원 내과와 흉부외과에서 8 French 도관 또는 흉관 삽입법으로 치료받은 원발성 또는 속발성 자연 기흉 환자 62명을 대상으로 하였으며 이들은 기흉의 크기가 가 25% 이상, 기흉의 크기에 관계없이 호흡곤란이나 흉통이 발생하였을 때, 기흉의 크기가 증가할 때, 다시 재발한 자연 기흉 환자를 대상으로 하였다. 긴장성 기흉, 혈흉, 화학적 흉막유착술이나 흉강경하 기포절제술을 한 경우는 대상에서 제외되었다. 임상특성(성별, 연령별, 과거 기흉의 유무, 기흉의 크기, 기저 질환의 유무)을 조사하고 합병증, 유치기간, 의무기록이나 개인적 접촉을 통하여 관찰 후 재발율을 비교 분석 하였다. 결과 : 8 French 삽입군과 흉관 삽입군의 추적 관찰기간의 중앙값은 각각 28개월, 22개월로 양군간에 유의한 차이를 보이고 있지 않았다. 8 French 도관 삽입군과 흉관 삽입군의 대상환자들의 임상적 특성-성별, 연령별, 기저 질환의 유무, 기흉의 크기의 분포에는 유의한 차이가 없었다. 원발성 및 속발성을 포함한 자연 기흉을 대상으로 비교한 결과, 도관 또는 흉관의 유치기간은 8 French 도관 삽입군이 $6.2{\pm}3.8$일로 흉관삽입군의 $9.1{\pm}7.5$일에 비해 유의하게 짧았다(p=0.047). 원발성 자연 기흉을 대상으로 두군을 비교하여, 8 French 도관 삽입군의 치료와 관련된 합병증은 6.25%로 흉관삽입군의 23.8%에 비해 더 적은 경향을 관찰할 수 있었다(0.041 ; one-tailed, p=0.053, two-tailed). 8 French 도관 삽입군과 흉관 삽입군에서 재발율에 있어서는 유의한 차이를 나타내지 않았다. 결론 : 이상의 결과로 원발성 또는 속발성 자연 기흉에 대한 치료로 지금까지의 흉관삽입법에 대하여 더 작은 직경의 도관을 삽입하여 치료에 의한 합병증의 빈도를 줄이면서, 치료기간을 줄이고 장기적인 재발을 예방할 수 있을 것으로 기대되며, 더 많은 환자를 대상으로하여 전향적인 연구가 앞으로 필요할 것으로 사료된다.

The effects of noise reduction, sharpening, enhancement, and image magnification on diagnostic accuracy of a photostimulable phosphor system in the detection of non-cavitated approximal dental caries

  • Purpose: Contrast, sharpness, enhancement, and density can be changed in digital systems. The important question is to what extent the changes in these variables affect the accuracy of caries detection. Materials and Methods: Forty eight extracted human posterior teeth with healthy or proximal caries surfaces were imaged using a photostimulable phosphor (PSP) sensor. All original images were processed using a six-step method: (1) applying "Sharpening 2" and "Noise Reduction" processing options to the original images; (2) applying the "Magnification 1:3" option to the image obtained in the first step; (3) enhancing the original images by using the "Diagonal/"option; (4) reviewing the changes brought about by the third step of image processing and then, applying "Magnification 1:3"; (5) applying "Sharpening UM" to the original images; and (6) analyzing the changes brought about by the fifth step of image processing, and finally, applying "Magnification 1:3." Three observers evaluated the images. The tooth sections were evaluated histologically as the gold standard. The diagnostic accuracy of the observers was compared using a chi-squared test. Results: The accuracy levels irrespective of the image processing method ranged from weak (18.8%) to intermediate (54.2%), but the highest accuracy was achieved at the sixth image processing step. The overall diagnostic accuracy level showed a statistically significant difference (p=0.0001). Conclusion: This study shows that the application of "Sharpening UM" along with the "Magnification 1:3" processing option improved the diagnostic accuracy and the observer agreement more effectively than the other processing procedures.

진해만 해역에서 지중해담치 (Mytilus galloprovincialis)의 마비성패독 독화 양상 (Paralytic Shellfish Poisoning of Mediterranean mussels from Jinhae Bay in Korea)

  • This study looked at toxicity of Mediterranean mussels, Mytilus galloprovincialis, which had accumulated paralytic shellfish toxins (PST) from early March to late May 2005 at Jinhae Bay, Korea. Alexandrium sp. was observed in low densities (< 1,000 cells/L) at the beginning of the study in March, increased rapidly in April, declined rapidly and disappeared in May. Although low densities of Alexandrium sp. were observed in March, mussel toxicity exceeded regulation level ($80{\mu}g$ STXeq. /100 g). Peak PSP (Paralytic Shellfish Poisoning) toxicity in the mussels occurred during high Alexandrium sp. cell densities in April. Mussels toxicity decreased with decline of Alexandrium sp. cell density. Major toxin components identified were $GTX_1$, $GTX_4$, followed by $C_1$, $C_2$, $GTX_2$, $GTX_3$ and neoSTX. Trace or sporadic toxin components were STX, $GTX_5$, $dcGTX_2$, $dcGTX_3$ and dcSTX. Toxin component analysis from the middle to end of the study showed that $11{\beta}$-epimers ($GTX_{3,4}$, $C_2$) were converted into $11{\alpha}$-epimers ($GTX_{1,2}$, $C_1$) and started to determine STX.
