In order to enhance the clinical efficacy of 5-aminolevulinic acid-induced photodynamic therapy (ALA-PDT), liposomal formulations using bulk hydrogenated phospholipids from soybean were introduced. Three types of lipids, S75-3, S100-3, and SL80-3 were used for formulating ALA. The pH of all the liposomal ALA is 4.5~5.5 and the size is 50~200 nm. All the liposomal formulations gave better ex vivo ALA skin penetration using nude mice skin in Franz cell than free ALA did. Among them, SL80-3 including 22% of lyso-phosphocholine achieved excellent ALA penetration when compared with those of S75-3 and S100-3 which have only 1~2% of lyso-phospholipids. S100-3 showed a little better results than S75-3 did. Addition of humectants (glycerine, propylene glycol, butylene glycol, betaine) in liposomal ALA formulated with SL80-3 produced little enhancing effect in ALA penetration. On the other hand, addition of surfactants (Tween 20, 60, Brij 72, 76, 78) in same liposomal system produced significant increase in ALA penetration. Among them, transferosomal system of lyso-phospholipid, SL80-3 and the surfactant, Brij76 showed the highest ALA penetration. Furthermore, this system also established the highest in vivo PpIX biosynthesis in hairy mice skin of C57BL/6. These results concluded that the transferosome of SL80-3 and Brij76 produced the best results in both ALA penetration and PpIX biosynthesis, and proved good correlation between them.
Polar psychrophiles which thrive under extreme conditions such as cold temperature, high salinity, and high dose ultraviolet light, emerge as novel targets for biotechnology. To prevent genetic drift and the possibility of contamination by subculturing, cryopreservation was employed for two psychrophilic microalgae, Porosira sp. (KOPRI AnM0008) and Pyramimonas sp. (KOPRI AnM0046), which have anti-freeze activities. Five cryoprotectants (dimethyl sulphoxide, ethylene glycol, glycerol, methanol and propylene glycol) showed toxicity at 20-30% (v/v). The optimal cryoprotectant concentration and equilibration time were less than 20% and 10 min, respectively. Cryopreservation was carried out in the presence of cryoprotectants either by direct freezing in liquid nitrogen ($LN_2$) or controlled freezing using a controlled rate freezer followed by storage in the $LN_2$ tank. As a result, Pyramimonas sp. (KOPRI AnM0046), a psychrophilic chlorophyta was revived. Cryopreserved Porosira sp. was not revived from either freezing protocols probably due to the silicic cell wall and its relatively large cell size. In the case of Pyramimonas sp. (KOPRI AnM0046), the controlled freezing method showed higher revival yield than the direct freezing method.

Yew (Taxus cuspidata Sieb.) chose that grow as medicine, food, decorative plant in Korea's Kyong-Gi province surroundings. Extracts of yew extracted leaf of 250 g and stems of 300 g with 1,3-butylene glycol (1,3-BG), propylene glycol (PG) and water. As results, external appearance of leaf extract of yew was slightly brown clear extract. The pH was 5.3$\pm$0.5, and specific gravity was 1.012$\pm$0.05, and refractive index was 1.375$\pm$0.05. Also, appearance of stem's extract was slightly brown clear extract, and the pH was 5.4$\pm$0.5, and specific gravity was 1.016$\pm$0.05, and refractive index was 1.358$\pm$0.05. Oil of yew separated from seeds, and extracted polysaccharide high purity from fruits. As a result, specific gravity of oil was 0.987, and obtained 40% of yield. Total polyphenols amount of yew extract is detected 0.563% in leaves, 0.325% in stems, whereas total tannins amount contained 0.054% and 0.037% each in leaves and stems. As effect in cosmetics, the anti-oxidative effect by DPPH method is 75.0% in leaves, and stems was 64.0%. Collagen synthesis rate was shown high activity by 54.16% in stem's extract, 33.18% in leaves' extract. Also, PPE-inhibitory activities were 13.7% and 23.5% each in leaves and stems. Anti-inflammatory effect of yew seed oil displayed superior effect of 41 % than control. Polysaccharide's molecular weight that is gotten from fruits was 5$\times$10$^4$~ 3$\times$10$^{5}$ dalton, and got 20.0$\pm$5% of yield.
Objectives: This study was designed to evaluate exposure levels of various chemicals used in wafer fabrication product lines in the semiconductor industry where work-related leukemia has occurred. Methods: The research focused on 9 representative wafer fabrication bays among a total of 25 bays in a semiconductor product line. We monitored the chemical substances categorized as human carcinogens with respect to leukemia as well as harmful chemicals used in the bays and substances with hematologic and reproductive toxicities to evaluate the overall health effect for semiconductor industry workers. With respect to monitoring, active and passive sampling techniques were introduced. Eight-hour long-term and 15-minute short-term sampling was conducted for the area as well as on personal samples. Results: The results of the measurements for each substance showed that benzene, toluene, xylene, n-butyl acetate, 2-methoxy-ethanol, 2-heptanone, ethylene glycol, sulfuric acid, and phosphoric acid were non-detectable (ND) in all samples. Arsine was either "ND" or it existed only in trace form in the bay air. The maximum exposure concentration of fluorides was approximately 0.17% of the Korea occupational exposure limits, with hydrofluoric acid at about 0.2%, hydrochloric acid 0.06%, nitric acid 0.05%, isopropyl alcohol 0.4%, and phosphine at about 2%. The maximum exposure concentration of propylene glycol monomethyl ether acetate (PGMEA) was 0.0870 ppm, representing only 0.1% or less than the American Industrial Hygiene Association recommended standard (100 ppm). Conclusion: Benzene, a known human carcinogen for leukemia, and arsine, a hematologic toxin, were not detected in wafer fabrication sites in this study. Among reproductive toxic substances, n-butyl acetate was not detected, but fluorides and PGMEA existed in small amounts in the air. This investigation was focused on the air-borne chemical concentrations only in regular working conditions. Unconditional exposures during spills and/or maintenance tasks and by-product chemicals were not included. Supplementary studies might be required.
Yew (Taxus cuspidata Sibe) selected cultivation as drug, food and decorative plant in Gyong-gi province in Korea. To extract the water soluble active ingredients, as a extracting method, there was extracted with 20g of dried seeds with each 20g of butylene glycol(BG) and propylene glycol(PG), and 40 mL of water mixing 72 hours at $40{\pm}5^{\circ}C$, and then they were filtrated by 400 mesh. Appearance of extract of seeds was pale brown, $pH=4.5{\pm}0.5$, $gravity=1.013{\pm}0.05$, a reflective $index=1.373{\pm}0.05$, and yield=75%. Also, to extract the high purity oil from seeds, it minutely pulverized the dried seeds and added the hexane, mixing 2 hours at $20{\pm}57^{\circ}C$. And then, this filtrated it with 400-mesh. It got the purified oil through evaporating them at $55^{\circ}C$ during under vacuum. As the results, appearance was slightly brown, gravity=0.922 acid value=0.12, saponification value=192, and it should be obtained the $40{\pm}5%$ of yield. As the efficacy evalution of cosmetic field, the antioxidative activities by NBT method were stronger 86.0% from extract of talus seeds than 52.0% from green tea extract and 35.0% from skullcap extract as well as the antioxidative activities by DPPH method were stronger 93.7% from extract of seed than 60.3% from extract of green tea and 27.1% from extract of skullcap. These are more effective than other plant extracts. The collagen synthesis rate on the activating fibroblast for Taxus cuspidata Sibe extract showed 35.43%. As the activity of the skin elasticity, PPE(porcine pancreatic elastase)-inhibitory activities of talus extract was 50.8%. Anti-inflammatory activity was more effective to be taken 41.1% of taxus seed oil than 24.2% of steady glycyrrhetinate (SG) as a control.
Methyl methacrylate-butyl methacrylate copolymer (MMBM)-dibutyl phthalate (DBP) films were investigated as a potential topical drug delivery system for the controlled release of nitrofurazone. The kinetic analysis of release data indicated that drug release followed a diffusion-controlled granular matrix model, where the quantity released per unit area is proportional to the square root of time. DBP of several hydrophobic plasticizers selected was found to give the highest release of nitrofurazone. However, hydrophilic plasticizers such as propylene glycol and polyethylene glycol 400 had no controlled release properties and acceptable film formation. The effects of changes in film composition, drug concentration, film thickness, pH of release medium, and temperature on the in vitro release of nitrofurazone were analyzed both theoretically and experimentally. The release rate constant (k') was found to be proportional to DBP content, pH, and the temperature of release medium, but independent of film thickness, and drug concentration in a range of 0.1-0.4% by weight. The linear relationship was found to exist between the log k' and DBP content. The release of nitrofurazone from MMBM-DBP (8:2) films was found to be an energy-linked process. Two energy terms were calculated ; the activation energy for matrix diffusion was 13.45 kcal/mole, and the heat of drug crystal solvation was 27.26-29.34 kcal/mole. Observation of scanning electron micrographs and microscopic photographs showed that the incorporation of DBP in films increased markedly the particle size of nitrofurazone dispersed in the film matrix, comparing with the fine dispersion of nitrofurazone in pure MMBM film alone.
본 연구에서는 캐비테이션에 의한 부식에 강한 도료를 개발하기 위하여 고탄성의 우레탄 수지에 내마모 성능을 향상시키기 위한 첨가제로서 Polypropylene glycol(PPG), Polycarbonate diol(PCD), Polycaprolactone polyol(PCL-1), Polycaprolactone-tetramethylene glycolether(PCL-2) 등 4가지 종류의 Polyol을 첨가하여 제조한 도료의 물성과 캐비테이션 저항성을 평가하였다. 합성된 도료의 물성을 비교한 결과 Polyol 중에서는 PCD을 첨가한 경우 경도와 내마모성이 높아 캐비테이션 저항성이 높을 수 있는 물성을 가진 것으로 나타났으나 고점도를 가지고 있어서 도료화에 어려움이 있을 것으로 예상되어 점도가 낮은 도료를 위하여 PCL-1을 적절한 첨가제로 선정하였다. PCL-1이 첨가된 도료의 캐비테이션 저항성을 평가한 결과 저항성이 높은 것으로 나타났고 SEM을 이용하여 표면 분석을 통한 기공의 침식현상을 관찰하였다.
The present study was carried out to evaluate the antioxidant and anti-inflammatory activities of three extracts (hot water, 50% ethanol and mixed solvent;water, ethanol, butylene glycol, propylene glycol) of dried chestnut, chestnut shell, chestnut leaves and dried chestnut leaves obtained from Castanea crenata tree. When conducted DPPH assay, radical scavenging activity of ethanol extract of chestnut shell was the highest with $IC_{50}$$10.8{\mu}g/mL$ among four extracts from these parts (p < 0.05). In additional results by the xanthine oxidase assay, antioxidant activity showed that water extract of chestnut leaves showed the highest xanthine oxidase inhibitory activity in the tested extracts (p < 0.05). Futhermore, extracts of chestnut shell and leaves exhibited no cytotoxicity in RAW 264.7 cells (p < 0.05). Also, anti-inflammatory activity by NO assay showed LPS-induced NO was significantly inhibited following treatment with extracts of chestnut shell and leaves of 3mg/mL (p < 0.05). These data suggest that extract of chestnut shell have antioxidant and anti-inflamantory activity including chestnut leaves. Therefore, it is considered that Castanea crenata research range and selection of functional material can broaden chestnut shell to other fractions such as chestnut and chestnut leaves.
The goal of this study was to evaluate the effects of cryoprotectants [dimethyl sulfoxide (DMSO), methanol, polyethylene glycol, and propylene glycol], cryoprotectant concentrations (10% and 20%), equilibration time (3, 10, and 30 min), cooling rate ($3^{\circ}$, $5^{\circ}$, $7^{\circ}$, and $10^{\circ}/min$), and straw size (0.25 and 0.5 mL) for sperm cryopreservation of tetraploid Pacific oysters. There was a significant difference among the four cryoprotectants, with 10% DMSO yielding the highest post-thaw survival and activity index of sperm. A significant relationship was observed between the cryoprotectant and its concentration. The sperm with equilibration times of 30 min yielded higher post-thaw survival and activity indices than those with 3 and 10 min equilibration times. The sperm cooled at a rate of $5^{\circ}/min$ yielded the highest post-thaw survival and activity index, and the results were significantly different from those observed for cooling at $7^{\circ}$ and $10^{\circ}/min$. Post-thaw survival and activity indices of sperm using a 0.25-mL straw were significantly higher than those using a 0.5-mL straw.
