OPTICAL PROPERTIES OF INDIUM OXIDE AND INDIUM TIN OXIDE FILMS PREP ARED BY SPUTTERING

  • Thin films of indium oxide and indium tin oxide have been prepared by d.c. magnetron sputtering onto the fused silica substrates kept at 90, 200 and $300^{\circ}C$. In order to elucidate the optical absorption process in low energy region below 3 eV, we have analyzed the absorption coefficients obtained from reflectance and transmittance measurements for these films based on the Lucovsky model. It has been found for the first time that a defect center in the band gap is located at 0.8~1.4 eV below the Fermi level in all films and arises from oxygen vacancies in their films. The optical absorption in low energy region is explained to be dominated by the transition of electrons trapped at the positively charged (+2e) oxygen vacancies with s-like nature to the conduction band formed from the 5s-orbit in indium atoms.

  • 청주 근처 토양에서 분리한 에너지 대사 저해제인 2,4-dinitrophenol (DNP)에 내성을 보이는 Streptomyces CM 001 균주가 생산하는 항진균성 물질(CUA)을 순수하게 분리 정제하고 화학분석을 통한 구조의 규명, 항진균활성 연구 및 유전적 변이원성(mutagenicity)등을 살펴보았다. CM 001 균주를 베네트 배지와 같이 영양성이 좋은 배지에서 배양시 항진균 활성이 동반 생성되는 색소와 비례적으로 증가했다. 배양 상등액 속의 항진균성 물질들을 염산처리, 유기용매처리 후 Amberlite XAD-4 column chromatography, silica gel adsorption-Sephadex LH-20 chromatography 과정을 통해 색소가 제거된 부분 정제 백색분말로 얻었다 이로부터 Prep. HPLC과정을 통하여 8종의 화합물을 각기 분리하였는데 이 화합물들은 동일한 UV spectrum과 216, 260nm에서 최대 흡수파장을 보였다.

  • The present study is concerned with the effect of PM process variables on the microstructure by using atomized superalloy powders. It is suggested that the inhomogeneity of composition is strongly dependent on the process variables. The contents of segregation elements of plasma rotating electrode process (PREP) powders are larger than those of Ar atomization (AA) powders. As HIP treatment temperature in-increases, the secondary phases on the prior particle boundaries (PPB) have continuous,uniform distribution and high density, but the amount of PPB decreases suddenly at 1150$^{\circ}$C. Segregated phases on the PPB are identified to be MC type carbide. Brittle MC type carbides on the PPB provide fracture initiation sites and preferred fracture path, thereby leading to intergranular type brittle fracture.

  • A method for isolation of some phenolic acids from Korean ginseng(Panax ginseng C.A.Meyer)was studied using silicic acid column chromatography. preparative thin layer chromatography and high performance liquid chromatography. Two phenolic compounds were isolated and identified as ferulic acid, mp $156-157^{\circ}C$ and vanillic acid. mp $154-156^{\circ}C$ by spectral data of Mass and NMR spectroscopy.

  • Heteroepitaxial growth of cubic silicon carbide films on Si(001) and Si(111) substrates at temperatures 900-$1000^{\circ}C$ has been achieved by high vacuum chemical vapor deposition using the single precursor 1, 3-disilabutane without carrying out the carbonization process of the substrate surfaces. The deposition temperature range is much lowered compared with conventiontional chemical vapor deposition where separate sources for silicon and carbon are employed. The deposition procedure is quite simple and safe. The qualities of the films were found to be very good judging from the results obtained by various characterization techniques including reflection high energy electron diffraction, X-ray diffraction, X-ray pole figure analysis, Rutherford backscattering spectrometry, Auger depth profiling, and transmission electron diffraction.

  • Ginsenosides are responsible for the pharmacological and biological activities of ginseng. In this study, ginsenoside Rh1 and compound K were isolated and purified from fermented ginseng substrate and their anti-allergic effects were assessed in compound 48/80-induced anaphylactic shock model. The fermented ginseng substrate was extracted by methanol and ginsenoside Rh1 and compound K were efficiently purified by preparative high performance liquid chromatography (prep HPLC). Their quality and quantity were analyzed by liquid chromatography-mass spectrometer (LC-MS) and HPLC. Ginsenoside Rh1 showed better anti-allergic effects than compound K in compound 48/80-induced anaphylactic shock model. This study suggested that fermented ginseng extracts with enriched Rh1 may be utilized as a potential biomaterial of functional food for the alleviation of allergic symptoms.

  • I. Objectives This study investigated the effect of rewetting agent on dentinal microtensile bond strengths(${\mu}TBS$), using one bottle adhesives. II. Materials and Methods Non caries human molars were sectioned to exposed the superficial dentin surfaces, etched 15 seconds using 32% phosphoric acid and 10 seconds rinsed. Samples were randomly divided into two groups according to adhesives (Single Bond, One-Step) used. Each group were subdivided into five groups by different dentin surface treatment:15 second dry(D), blot dry(BD) or 15 second dry and rewetted with distrilled water(DW), Gluma Desensitizer(GD) and Aqua-Prep(AP) during 30 second, respectively.(omitted)

  • Liquid chromatography mass spectrometry is widely employed in proteomics studies. One of such instruments is the Liquid Chromatography (LC)-Matrix-assisted laser desorption ionisation (MALDI)-Time of flight (TOF) or LC-MALDI-TOF/TOF. In this study, this instrument was used to identify the membrane proteins of a protozoan parasite namely Entamoeba histolytica. It causes amoebiasis in human. The E. histolytica trophozoites were cultured prior to the membrane protein extraction using the conventional method, $ProteoPrep^{(R)}$ and $ProteoExtract^{(R)}$ kits. Then, the membrane protein extracts were trypticdigested and analysed by LC-MALDI-TOF/TOF. Approximately, 194 proteins were identified and 27.8% (54) were predicted as membrane proteins having 1 to 15 transmembrane regions and signal peptides by combining all three extraction methods. Also, this study has discovered 3 unique proteins as compared to our previous study which merit further investigation.

  • Four phenolic compounds were isolated from the wood and bark of Magnolia sieboldii. The structures were determined as : costunolide (I), syringin (II), 1, 2-dihydroxyxanthone (III), and vanillic acid (IV), respectively, on the basis of spectroscopic data.

Gene Cloning and Expression of Thermostable DNA Polymerase from Thermus thermophilus HJ6 (Thermus thermophilus HJ6 유래 내열성 DNA Polymerase의 유전자 클로닝 및 발현)

  • The gene encoding Thermus thermophilus HJ6 DNA polymerase (Tod) was cloned and sequenced. The open reading frame (ORF) of the Tod gene was composed of 2,505 nucleotides and encoded a protein (843 amino acids) with a predicted molecular weight of 93,795 Da. The deduced amino acid sequence of Tod showed 98% and 86% identities to the Thermus thermophilus HB8 DNA pol and Thermus aquaticus DNA pol, respectively, The Tod gene was expressed under the control of the bacteriophage $\lambda$ promoters PR and PL on the expression vector pJLA503 in Escherichia coli strain BL21 (DE3) codon plus. The expressed enzyme was purified by heat treatment, $HiTrap^{TM}$ Q column, and $HiPrep^{TM}$ Sephacryl S-200 HR 26/60 column chromatographies. The optimal temperature and pH for DNA polymerase activity were found to be $75{\sim}80^{\circ}C$ and 9.0, respectively. The optimal concentrations of $Mg^{2+}$ and $Mn^{2+}$ were 2.5 mM and 1 mM, respectively. The enzyme activity was activated by divalent cations, and was inhibited by monovalent cations. The result of the PCR experiment with Tod DNA polymerase indicates that this enzyme might be useful in DNA amplification and PCR-based applications.
